Objectives. Standard pharmaceutical capsules are designed to dissolve in the acidic environment of the stomach releasing the encapsulated contents for absorption. When release is required further along the gastrointestinal tract capsules can be coated with acid insoluble polymers to enable passage through the stomach and dissolution in the intestine. This paper describes formulations that have the potential to be used to produce two-piece hard capsules for post-gastric delivery without the requirement of an exterior coat. Methods. The formulation uses three polysaccharides: sodium alginate, hypromellose and gellan gum to provide acid insolubility and the ability to form capsules using standard industrial equipment. Key findings. The rheological profile, on cooling, of the base material, water content and thickness of the films were shown to be comparable with those of commercial capsules. The capsules remained intact for 2 h in 100 mm HCl at pH 1.2, and within 5 min of being removed from the acid and submerged in phosphate-buffered saline at pH 6.8 were ruptured. Conclusions. Selected formulations from this study have potential for use as delayed release capsules.

In this study the effect of structure and amount of polyglactin fibre incorporation into a brushite forming calcium phosphate cement system and the effect of mechanical compaction on the fibre modified system were investigated. In comparison the effect of resorbable polycaprolactone surface coating of cement specimens was investigated. The results showed that, apart from the mechanical properties of the reinforcing material, the structure of the incorporated fibres, regular or random, is crucial for the resulting flexural strength and modulus of elasticity. Fibre reinforcement could also be combined with mechanical compaction of the cement/fibre composite paste leading to a possible 7-fold increase in flexural strength or an almost 5-fold increase in modulus of elasticity. Reinforcement of the tensile surface of cement grafts may ultimately improve strength where required, especially in conjunction with bone fixation devices. © 2005 Acta Materialia Inc. Lyophilised orally disintegrating tablets (ODTs) have achieved a great success in overcoming dysphagia associated with conventional solid dosage forms. However, the extensive use of saccharides within the formulation limits their use in treatment of chronic illnesses. The current study demonstrates the feasibility of using combination of proline and serine to formulate zero sacharide ODTs and investigates the effect of freezing protocol on sublimation rate and tablets characteristics. The results showed that inclusion of proline and serine improved ODT properties when compared to individual counterparts. Additionally, annealing the ODTs facilitated the sublimation process and shortened the disintegration time. © 2010 by the authors; licensee Molecular Diversity Preservation International, Basel, Switzerland.

The exponential growth of studies on the biological response to ocean acidification over the last few decades has generated a large amount of data. To facilitate data comparison, a data compilation hosted at the data publisher PANGAEA was initiated in 2008 and is updated on a regular basis (doi:10.1594/PANGAEA.149999). By January 2015, a total of 581 data sets (over 4 000 000 data points) from 539 papers had been archived. Here we present the developments of this data compilation five years since its first description by Nisumaa et al. (2010). Most of study sites from which data archived are still in the Northern Hemisphere and the number of archived data from studies from the Southern Hemisphere and polar oceans are still relatively low. Data from 60 studies that investigated the response of a mix of organisms or natural communities were all added after 2010, indicating a welcomed shift from the study of individual organisms to communities and ecosystems. The initial imbalance of considerably more data archived on calcification and primary production than on other processes has improved. There is also a clear tendency towards more data archived from multifactorial studies after 2010. For easier and more effective access to ocean acidification data, the ocean acidification community is strongly encouraged to contribute to the data archiving effort, and help develop standard vocabularies describing the variables and define best practices for archiving ocean acidification data.

Nesta dissertação apresentamos um trabalho de desenvolvimento e utilização de pulsos de radiofreqüência modulados simultaneamente em freqüência, amplitude e fase (pulsos fortemente modulados, SMP, do inglês Strongly Modulated Pulses) para criar estados iniciais e executar operações unitárias que servem como blocos básicos para processamento da informação quântica utilizando Ressonância Magnética Nuclear (RMN). As implementações experimentais foram realizas em um sistema de 3 q-bits constituído por spins nucleares de Césio 133 (spin nuclear 7/2) em uma amostra de cristal líquido em fase nemática. Os pulsos SMP´s foram construídos teoricamente utilizando um programa especialmente desenvolvido para esse fim, sendo o mesmo baseado no processo de otimização numérica Simplex Nelder-Mead. Através deste programa, os pulsos SMP foram otimizados de modo a executarem as operações lógicas desejadas com durações consideravelmente menores que aquelas realizadas usando o procedimento usual de RMN, ou seja, seqüências de pulsos e evoluções livres. Isso tem a vantagem de reduzir os efeitos de descoerência decorrentes da relaxação do sistema. Os conceitos teóricos envolvidos na criação dos SMPs são apresentados e as principais dificuldades (experimentais e teóricas) que podem surgir devido ao uso desses procedimentos são discutidas. Como exemplos de aplicação, foram produzidos os estados pseudo-puros usados como estados iniciais de operações lógicas em RMN, bem como operações lógicas que foram posteriormente aplicadas aos mesmos. Utilizando os SMP\'s também foi possível realizar experimentalmente os algoritmos quânticos de Grover e Deutsch-Jozsa para 3 q-bits. A fidelidade das implementações experimentais foi determinadas utilizando as matrizes densidade experimentais obtidas utilizando um método de tomografia da matriz densidade previamente desenvolvido.

The effects of ocean acidification and elevated seawater temperature on coral calcification and photosynthesis have been extensively investigated over the last two decades, whereas they are still unknown on nutrient uptake, despite their importance for coral energetics. We therefore studied the separate and combined impacts of increases in temperature and pCO2 on phosphate, ammonium, and nitrate uptake rates by the scleractinian coral S. pistillata. Three experiments were performed, during 10 days i) at three pHT conditions (8.1, 7.8, and 7.5) and normal temperature (26°C), ii) at three temperature conditions (26°, 29°C, and 33°C) and normal pHT(8.1), and iii) at three pHT conditions (8.1, 7.8, and 7.5) and elevated temperature (33°C). After 10 days of incubation, corals had not bleached, as protein, chlorophyll, and zooxanthellae contents were the same in all treatments. However, photosynthetic rates significantly decreased at 33°C, and were further reduced for the pHT 7.5. The photosynthetic efficiency of PSII was only decreased by elevated temperature. Nutrient uptake rates were not affected by a change in pH alone. Conversely, elevated temperature (33°C) alone induced an increase in phosphate uptake but a severe decrease in nitrate and ammonium uptake rates, even leading to a release of nitrogen into seawater. Combination of high temperature (33°C) and low pHT(7.5) resulted in a significant decrease in phosphate and nitrate uptake rates compared to control corals (26°C, pHT = 8.1). These results indicate that both inorganic nitrogen and phosphorus metabolism may be negatively affected by the cumulative effects of ocean warming and acidification.

Currently, there is increasing use of nanomaterials in the food industry thanks to the many advantages offered and make the products that contain them more competitive in the market. Their physicochemical properties often differ from those of bulk materials, which require specialized risk assessment. This should cover the risks to the health of workers and consumers as well as possible environmental risks. The risk assessment methods must go updating due to more widespread use of nanomaterials, especially now that are making their way down to consumer products. Today there is no specific legislation for nanomaterials, but there are several european dispositions and regulations that include them. This review gives an overview of the risk assessment and the existing current legislation regarding the use of nanotechnology in the food industry.

We investigate the application of time-reversed electromagnetic wave propagation to transmit energy in a wireless power transmission system. “Time reversal” is a signal focusing method that exploits the time reversal invariance of the lossless wave equation to direct signals onto a single point inside a complex scattering environment. In this work, we explore the properties of time reversed microwave pulses in a low-loss ray-chaotic chamber. We measure the spatial profile of the collapsing wavefront around the target antenna, and demonstrate that time reversal can be used to transfer energy to a receiver in motion. We demonstrate how nonlinear elements can be controlled to selectively focus on one target out of a group. Finally, we discuss the design of a rectenna for use in a time reversal system. We explore the implication of these results, and how they may be applied in future technologies.

Cysteine is susceptible to a variety of modifications by reactive oxygen and nitrogen oxide species, including glutathionylation; and when two cysteines are involved, disulfide formation. Glutathione-cysteine adducts may be removed from proteins by glutaredoxin, whereas disulfides may be reduced by thioredoxin. Glutaredoxin is homologous to the disulfide-reducing thioredoxin and shares similar binding modes of the protein substrate. The evolution of these systems is not well characterized. When a single Cys is present in a protein, conjugation of the redox buffer glutathione may induce conformational changes, resulting in a simple redox switch that effects a signaling cascade. If a second cysteine is introduced into the sequence, the potential for disulfide formation exists. In favorable protein contexts, a bistable redox switch may be formed. Because of glutaredoxin's similarities to thioredoxin, the mutated protein may be immediately exapted into the thioredoxin-dependent redox cycle upon addition of the second cysteine. Here we searched for examples of protein substrates where the number of redox-active cysteine residues has changed throughout evolution. We focused on cross-strand disulfides (CSDs), the most common type of forbidden disulfide. We searched for proteins where the CSD is present, absent and also found as a single cysteine in protein orthologs. Three different proteins were selected for detailed study-CD4, ERO1, and AKT. We created phylogenetic trees, examining when the CSD residues were mutated during protein evolution. We posit that the primordial cysteine is likely to be the cysteine of the CSD which undergoes nucleophilic attack by thioredoxin. Thus, a redox-active disulfide may be introduced into a protein structure by stepwise mutation of two residues in the native sequence to Cys. By extension, evolutionary acquisition of structural disulfides in proteins can potentially occur via transition through a redox-active disulfide state.

The development of novel therapies is essential to lower the burden of complex diseases. The purpose of this study is to identify novel therapeutics for complex diseases using bioinformatic methods. Bioinformatic tools such as candidate gene prediction tools allow identification of disease genes by identifying the potential candidate genes linked to genetic markers of the disease. Candidate gene prediction tools can only identify candidates for further research, and do not identify disease genes directly. Integration of drug-target datasets with candidate gene data-sets can identify novel potential therapeutics suitable for repositioning in clinical trials. Drug repositioning can save valuable time and money spent in therapeutic development of complex diseases.

Introducción Las pacientes con miomas uterinos pueden llegar a sufrir de síntomas urinarios y de disfunción sexual. Es para nosotros importante conocer la frecuencia de estas patologías en pacientes con miomas con indicación de cirugía atendidos en el Hospital Universitario Mayor Méderi y la relación entre estas tres entidades. Metodología Estudio cuasi experimental de antes-después. El estudio se encuentra dividido en dos fases, en esta primera fase a las pacientes se les aplicó los cuestionarios FSFI, IIQ-7 y UDI-6 antes de realizar el procedimiento quirúrgico. En una segunda fase se realizará un nuevo abordaje a los 6 y 12 meses donde se aplicarán los mismos instrumentos. Se utilizó coeficiente de Spearman y Kruskall-Wallis para evaluar la relación. Resultados En esta primera fase se incluyeron 81 participantes, con una mediana de años de 46 (RIQ=42-49) mínimo 33 y máximo 71 años. La mediana de miomas fue de 1 (RIQ1-2) máximo 5 miomas. El resultado total de la FSFI fue de 21(RIQ=18,5-25,5). La mediana de la escala UDI -6 fue de 50,4 (RIQ=0-31,2) y la mediana de IIQ-7 fue de 4,75 (RIQ=0-23,7). Se presentó una correlación negativa débil entre los puntajes de FSFI y los cuestionarios UDI-6 (-0.3604) e IIQ 7 (-0.3530), con una prevalencia de riesgo de disfunción sexual de 61%. Conclusiones En esta primera fase de la investigación se pudo observar una existencia de correlación entre la función sexual y la sintomatología urinaria. La prevalencia de disfunción sexual es mayor que en población de mujeres sin patología de miomas uterino.

Las enfermedades raras o huérfanas corresponden a aquellas con baja prevalencia en la población, y en varios países tienen una definición distinta de acuerdo con el número de pacientes que afectan en la población. La Organización Mundial de la Salud (OMS), las define como un trastorno que afecta de 650 a 1.000 personas por millón de habitantes, de las que se han identificado alrededor de 7.000. En Colombia su prevalencia es menor de 1 por cada 5.000 personas y comprenden: las enfermedades raras, las ultra-huérfanas y las olvidadas. Los pacientes con este tipo de enfermedades imponen retos a los sistemas sanitarios, pues si bien afectan a un bajo porcentaje de la población, su atención implica una alta carga económica por los costos que involucra su atención, la complejidad en su diagnóstico, tratamiento, seguimiento y rehabilitación. El abordaje de las enfermedades raras requiere un manejo interdisciplinar e intersectorial, lo que implica la organización de cada actor del sistema sanitario para su manejo a través de un modelo que abraque las dinámicas posibles entre ellos y las competencias de cada uno. Por lo anterior, y teniendo en cuenta la necesidad de formular políticas sanitarias específicas para la gestión de estas enfermedades, el presente trabajo presenta una aproximación a la formulación de un modelo de gestión para la atención integral de pacientes con enfermedades raras en Colombia. Esta investigación describe los distintos elementos y características de los modelos de gestión clínica y de las enfermedades raras a través de una revisión de literatura, en la que se incluye la descripción de los distintos actores del Sistema de Salud Colombiano, relacionados con la atención integral de estos pacientes para la documentación de un modelo de gestión integral.
