Este trabalho apresenta uma modificação dos procedimentos descritos nas Farmacopéias Francesa e Européia para a análise de flavonoides de Passiflora incarnata L., Passifloraceae, por espectrometria UV-Visível e propõe a sua aplicação na determinação dos flavonoides totais das folhas da espinheira-santa (Maytenus aquifolium Mart. e Maytenus ilicifolia (Schrad.) Planch., Celastraceae) e do maracujá (Passiflora edulis Sims. e Passiflora alata Curtis, Passifloraceae). Os resultados obtidos por espectrometria no UV-Visível foram comparados aos obtidos por cromatografia líquida de alta eficiência (CLAE-UV), encontrando-se resultados estatisticamente similares entre os métodos espectrométrico modificado da Farmacopéia Francesa e CLAE-UV.

Este trabalho descreve a investigação química e biológica do extrato bruto e das partições hexano e acetato de etila, das folhas de Pyrostegia venusta (Ker Gawl.) Miers, popularmente conhecida como “cipó de São João”. P. venusta é classificada botanicamente como uma liana de porte mediano, tendo como característica uma exuberante floração vermelha, e por isso, sendo utilizada como planta ornamental. Essa planta possui uma larga utilização na medicina popular, sendo utilizada no tratamento de vitiligo, diarreia, bronquite, resfriado, icterícia e infecções. Os objetivos deste trabalho foram identificar as classes de metabólitos secundários presentes, avaliar o potencial antioxidante das amostras de P. vesnuta (extrato bruto, frações acetato de etila e hexano), quantificar o teor de flavonoides no extrato bruto, verificar a segurança do uso dessa planta, em termos de viabilidade celular (VC) frente à macrófagos murinos (RAW 264.7) (ensaio de imunotoxicidade). Adicionalmente os resultados de viabilidade celular foram comparados com quatro compostos anti-inflamatórios comerciais (ácido acetilsalicílico, indometacina, betametasona e piroxicam), e testar o extrato bruto quanto à inibição de catepsinas K e V. Os testes de identificação fitoquímica confirmaram a presença de flavonoides, cumarinas e esteroides nas amostras. A metodologia cromatográfica associada à análises por espectrometria de massas, levou a identificação dos compostos: fitol (1), sitosterol (2), estigmasterol (3) e campesterol (4). O extrato bruto demonstrou ter atividade inibitória frente as duas catepsinas testadas (K e V). A fração acetato de etila foi a que apresentou maior atividade antioxidante nas metodologias de inibição do radical DPPH (IC50 38,62 μg/mL) e radical ABTS (IC50 27,58 μg/mL). O teor de flavonoides total para o extrato bruto foi de 148,5±7,65 μg/mg (14,85 % (m/m)), o que justifica a observada atividade antioxidante, já que estes possuem atividade antioxidante. As amostras de P. venusta obtiveram valores de VC maiores do que os anti-inflamatórios comerciais, estes apresentaram VC abaixo do controle negativo, assim como o extrato bruto e a fração acetato de etila, a fração hexano obteve valores acima do controle negativo, sendo estes os maiores resultados de VC entre as amostras de P. venusta.

Previous studies have reported that a diet containing 10% cocoa, a rich source of flavonoids, has immunomodulatory effects on rats and, among others effects, is able to attenuate the immunoglobulin (Ig) synthesis in both systemic and intestinal compartments. The purpose of the present study was focused on investigating whether these effects were attributed exclusively to the flavonoid content or to other compounds present in cocoa. To this end, eight-week-old Lewis rats were fed, for two weeks, either a standard diet or three isoenergetic diets containing increasing proportions of cocoa flavonoids from different sources: one with 0.2% polyphenols from conventional defatted cocoa, and two others with 0.4% and 0.8% polyphenols, respectively, from non-fermented cocoa. Diet intake and body weight were monitored and fecal samples were obtained throughout the study to determine fecal pH, IgA, bacteria proportions, and IgA-coated bacteria. Moreover, IgG and IgM concentrations in serum samples collected during the study were quantified. At the end of the dietary intervention no clear changes of serum IgG or IgM concentrations were quantified, showing few effects of cocoa polyphenol diets at the systemic level. However, in the intestine, all cocoa polyphenol-enriched diets attenuated the age-related increase of both fecal IgA and IgA-coated bacteria, as well as the proportion of bacteria in feces. As these effects were not dependent on the dose of polyphenol present in the diets, other compounds and/or the precise polyphenol composition present in cocoa raw material used for the diets could be key factors in this effect.

Flavonoidit ovat kasvien polyfenolisia sekundäärimetaboliitteja, jotka koostuvat 15-hiilisestä C6-C3-C6-perusrungosta. Niillä on havaittu useita terveyden kannalta edullisia ominaisuuksia kuten antioksidatiivisia, antikarsinogeenisia, antiallergeenisia, anti-inflammatorisia, estrogeenisia sekä antimikrobiaalisia vaikutuksia. Kasvimateriaalia käyttävän elintarviketeollisuuden sivuvirrat ovat flavonoidi-pitoisten jakeiden arvokkaita raaka-aineita. Tässä työssä tarkoituksena oli tutkia marjojen puristusjäännöksen sisältämien flavonoidien uuttamista ja uuttautumiseen vaikuttavia tekijöitä kuten liuottimen koostumusta ja määrää, uuttolämpötilaa ja -aikaa sekä liuottimen lisäaineita. Uutteet analysointiin HPLC:lla käyttäen ODS-2-kolonnia ja gradienttieluointia. Komponentit detektoitiin niille tyypillisillä UV-VIS-aallonpituuksilla ja identifioitiin vertaamalla flavonoidiprofiililtaan tunnettujen marjojen kromatogrammeihin. Antosyaanien kvantitatiiviseen analysointiin käytettiin spektrofotometrista pH-eromenetelmää ja tulokset ilmoitettiin syanidiini-3-glukosidina. Uutteiden kestävyyttä tutkittiin säilyttämällä näytteitä eri olosuhteissa sekä lisäämällä niihin uuton saantoon ja komponenttien säilyvyyteen vaikuttavia aineita. Useimmat tutkituista hapoista paransivat flavonoidien saantoa ja uutteiden säilyvyyttä, mutta esim. askorbiinihapon lisääminen nopeutti tiettyjen uutteen sisältämien flavonoidien tuhoutumista.

Logistiikan ulkoistaminen on strateginen valinta, joka mahdollistaa yrityksen keskittymisen ydinosaamiseensa. Ulkoistavien yritysten tarpeiden kasvaessa muuttuu myös ulkoistettavien prosessien laajuus. Palvelut pyritään hankkimaan yhdeltä toimittajalta. Tämä lisää palveluntarjoajien haasteita ja johtaa enenevässä määrin neljännen osapuolen logistiikkaratkaisujen käyttöön. Tässä tutkimuksessa etsittiin vastausta kysymykseen: Mikä on logistiikan kokonaispalveluratkaisun arvo asiakkaalle? Teoriaosassa paneuduttiin logistiikan, ulkoistamisen ja suorituskyvyn mittaamisen käsitteisiin ja keskeisiin ilmiöihin. Empiirisessä osassa tutkittiin logistiikan ulkoistamiselle asetettuja tavoitteita ja niiden toteutumisen seurantaa. Aineisto kerättiin postikyselyn ja haastattelujen avulla. Tutkimuksessa ilmeni, että onnistuneen ulkoistamisratkaisun toteuttaminen edellyttää palveluntarjoajalta asiakastarpeiden syvällistä ymmärtämistä, prosessien integrointi- ja koordinointikykyä sekä taitoa johtaa logistista verkostoa. Olennaista on myös IT-järjestelmien jatkuva kehittäminen ja suorituskyvyn mittausjärjestelmän käyttöönotto. Ratkaisun toimivuuteen vaikuttaa myös ulkoistavan yrityksen ja palveluntarjoajan välinen suhde. Sen tulee olla kumppanuussuhde, joka perustuu luottamukseen, tiedon avoimeen jakamiseen ja toiminnan aktiiviseen kehittämiseen yhteistyössä.

This work describes the mechanism of action of some reactive oxygen species (ROS) and reactive nitrogen species (RNS) in the oxidative stress of the human body, and their consequences on damage to DNA, RNA, proteins and lipids. It also illustrates the defense system of our organism against these ROS and RNS species. The action of nonenzymatic protection systems is reported, with emphasis on micromolecules like Q10 coenzyme, vitamin C, alpha-tocopherol, carotenoids and flavonoids. The importance of flavonoids is also emphasized, and their body protection mechanism is detailed.

This review describes aspects of the Bromeliaceae family dealing the traditional applications, biological activities and distribution of secondary metabolites in distinct subfamilies. Some species are used with medicinal purposed in the treatment of respiratory, diabetes or inflammation diseases, and gastrointestinal disorders. Special emphasis on cycloartane triterpenoids and flavonoids, typical metabolites of this family, are presented. Bromeliaceae is unique amongst the monocotyledons in the frequency and variety of flavonoids with hydroxylation or methoxylation at the 6-position. Other compound classes as steroids, hidroxycinnamic acids, phenylpropane diglycerides, lignans, are presented.

Phytochemical investigations on three Brazilian Lauraceae species from the Cerrado region of São Paulo State, Ocotea corymbosa (Meins) Mez., O. elegans Mez. and Persea pyrifolia Nees & Mart. ex Nees resulted in the isolation of flavonoids, an ester of the 4-O-E-caffeoylquinic acid, an aromatic sesquiterpene besides furofuran lignans. This is the first chemical study on the leaves of Ocotea elegans and O. corymbosa as well as the first report of non-volatile compounds from Persea pyrifolia.

This paper reports on a modification of the procedures originally described in the French Pharmacopoeia for the UV-visible spectrometric analysis of flavonoids, and proposes a validation of the method and its application in the determination of total flavonoids from sugarcane (Saccharum officinarum) leaves and vinasse. An analysis of precision and accuracy revealed a low relative standard deviation (< 5.0%) and a good recovery percentages (99.79 and 98.34%). A comparison of the spectrometric results against those obtained by high performance liquid chromatography (HPLC-UV) demonstrated complete compatibility between the modified French Pharmacopoeia (spectrometric) and HPLC-UV methods

Interest in analytical methods for quality control of herbal drugs has grown sharply due to the scarcity of monographs in official manuals. Thus, the aim of the present study was to evaluate analytical procedures for quantitative determination of flavonoids from leaves of Bauhinia forficata Link (pata-de-vaca). Two procedures for quantification of total flavonoids (with and without acid hydrolysis) by spectrophotometry were tested. The proposed methods proved to be specific, sensitive, precise, accurate and robust, being suitable for routine laboratory use.

The current study aims to verify the best method for a rapid and efficient extraction of flavonoids from Alpinia zerumbet. Dried leaves were extracted using distillated water and ethanol 70% by extraction methods of shaking maceration, ultrasonic, microwave and stirring. By the application of TLC and reversed-phase HPLC techniques the rutin and kaempferol-3-O-glucuronide were detected. Ethanol 70% was more efficient for flavonoids extraction than water. No significant yielding variation was verified for ultrasonic, microwave and stirring methods using ethanol 70% (11 to 14%). The relative concentration of rutin and kaempferol-3-O-glucuronide, respectively, was higher by ultrasonic (1.5 and 5.62 mg g-1 dried leaves, respectively) and by microwave (1.0 and 6.64 mg g-1 dried leaves) methods using ethanol. Rapid and simplified extraction proceeding optimize phytochemical work and acquisition of secondary metabolites.

Tämän diplomityön tarkoituksena on kehittää menetelmä, jolla voidaan seurata inhalaatiovalmisteen sisältämiä epäpuhtauksia. Menetelmä kehitetään erittäin korkean suorituskyvyn kromatografialaitteistolle (Ultra High Performance Liquid Chromatography, UHPLC) jo olemassa olevan HPLC-epäpuhtausmenetelmän pohjalta. Uusi menetelmä kehitetään analyysiajan lyhentämiseksi ja erotuksen resoluution parantamiseksi. Työn kirjallisuusosa esittelee lyhyesti inhalaatiovalmisteet sekä nestekromatografian perusteet. Korkean erotuskyvyn nestekromatografia ja analysoinnin apuna käytettävät parametrit selvitetään laskukaavoineen. Kirjallisuusosa keskittyy epäpuhtausmenetelmän kehittämisen kulkuun ja menetelmän validoinnissa suoritettaviin kokeisiin. Soveltavassa osassa Orion Oyj:n kehitteillä olevalle kahden vaikuttavan aineen inhalaatiovalmisteelle kehitetään UHPLC-epäpuhtausmenetelmä kirjallisuusosiossa esitellyn menetelmäkehitysrungon pohjalta. Menetelmäkehityksen kulku ja analyysin olosuhteiden valinta esitellään pääpiirteittäin, jonka jälkeen kehitetty menetelmä validoidaan sille tehdyn validointisuunnitelman mukaisesti. Kehitetystä UHPLC-epäpuhtausmenetelmästä saatiin 16 minuuttia lyhyempi kuin vastaava HPLC-epäpuhtausmenetelmä. Myös analyysin resoluutio parantui merkittävästi. Toistuvien injektioiden jälkeen kromatogrammissa esiintyi kuitenkin häntimistä, joka johti merkittävään resoluution heikkenemiseen. Häntimisen syyksi epäiltiin toisen vaikuttavan aineen kiinnittymistä kolonnimateriaaliin, mutta sen estämiseksi ehdotetut toimenpiteet eivät sopineet kehitettyyn menetelmään.

Neuroinflammation plays an integral role in the progression of neurodegeneration. In this study we investigated the anti-inflammatory effects of different classes of flavonoids (flavanones, flavanols and anthocyanidins) in primary mixed glial cells. We found that the flavanones naringenin and hesperetin and the flavols (+)-catechin and (-)-epicatechin, but not the anthocyanidins cyanidin and pelargonidin, attenuated LPS/IFN-gamma-induced TNF-alpha production in glial cells. Naringenin also inhibited LPS/IFN-gamma-induced iNOS expression and nitric oxide production in glial cells, thus showing the strongest antiinflammatory activity among all flavonoids tested. Moreover, naringenin protected against inflammatory-induced neuronal death in a primary neuronal-glial co-culture system. Naringenin also inhibited LPS/IFN-gamma-induced p38 mitogen-activated protein kinase (MAPK) phosphorylation and downstream signal transducer and activator of transcription-1 (STAT-1) in LPS/IFN-gamma stimulated primary mixed glial cells. Taken together, our results suggest that naringenin may produce an anti-inflammatory effect in LPS/IFN-gamma stimulated glial cells that may be due to its interaction with p38 signalling cascades and the STAT-I trascription factor. Emerging evidence suggests that the cellular actions of flavonoids relate not simply to their antioxidant potential but also to the modulation of protein kinase signalling pathways. We investigated in primary cortical neurons, the ability of the flavan-3-ol, (-)epicatechin, and its human metabolites at physiologically relevant concentrations, to stimulate phosphorylation of the transcription factor cAMP-response element binding protein (CREB), a regulator of neuronal viability and synaptic plasticity. (-)Epicatechin at 100-300 nmol/L stimulated a rapid, extracellular signal-regulated kinase (ERK)- and PI3K-dependent, increase in CREB phosphorylation. At micromolar concentrations, stimulation was no longer apparent and at the highest concentration tested (30 mu mol/L) (-)epicatechin was inhibitory. (-)Epicatechin also stimulated ERK and Akt phosphorylation with similar bell-shaped concentration-response characteristics. The human metabolite 3 '-O-methyl-(-)epicatechin was as effective as (-)epicatechin at stimulating ERK phosphorylation, but (-)epicatechin glucuronide was inactive. (-)Epicatechin and 3 '-O-methyl-(-)epicatechin treatments (100 nmol/L) increased CRE-luciferase activity in cortical neurons in a partially ERK-dependent manner, suggesting the potential to increase CREB-mediated gene expression. mRNA levels of the glutamate receptor subunit GluR2 increased by 60%, measured 18 h after a 15 min exposure to (-)epicatechin and this translated into an increase in GluR2 protein. Thus, (-)epicatechin has the potential to increase CREB-regulated gene expression and increase GluR2 levels and thus modulate neurotransmission, plasticity and synaptogenesis.

Phytochemical-rich foods have been shown to be effective at reversing age-related deficits in memory in both animals and humans. We show that a supplementation with a blueberry diet (2% w/w) for 12 weeks improves the performance of aged animals in spatial working memory tasks. This improvement emerged within 3 weeks and persisted for the remainder of the testing period. Memory performance correlated well with the activation of cAMP-response element-binding protein (CREB) and increases in both pro- and mature levels of brain-derived neurotrophic factor (BDNF) in the hippocampus. Changes in CREB and BDNF in aged and blueberry-supplemented animals were accompanied by increases in the phosphorylation state of extracellular signal-related kinase (ERK1/2), rather than that of calcium calmodulin kinase (CaMKII and CaMKIV) or protein kinase A. Furthermore, age and blueberry supplementation were linked to changes in the activation state of Akt, mTOR, and the levels of Arc/Arg3.1 in the hippocampus, suggesting that pathways involved in de novo protein synthesis may be involved. Although causal relationships cannot be made among supplementation, behavior, and biochemical parameters, the measurement of anthocyanins and flavanols in the brain following blueberry supplementation may indicate that changes in spatial working memory in aged animals are linked to the effects of flavonoids on the ERK-CREB-BDNF pathway.
