The distribution of parvalbumin (PV), calretinin (CR), and calbindin (CB) immunoreactive neurons was studied with the help of an image analysis system (Vidas/Zeiss) in the primary visual area 17 and associative area 18 (Brodmann) of Alzheimer and control brains. In neither of these areas was there a significant difference between Alzheimer and control groups in the mean number of PV, CR, or CB immunoreactive neuronal profiles, counted in a cortical column going from pia to white matter. Significant differences in the mean densities (numbers per square millimeter of cortex) of PV, CR, and CB immunoreactive neuronal profiles were not observed either between groups or areas, but only between superficial, middle, and deep layers within areas 17 and 18. The optical density of the immunoreactive neuropil was also similar in Alzheimer and controls, correlating with the numerical density of immunoreactive profiles in superficial, middle, and deep layers. The frequency distribution of neuronal areas indicated significant differences between PV, CR, and CB immunoreactive neuronal profiles in both areas 17 and 18, with more large PV than CR and CB positive profiles. There were also significantly more small and less large PV and CR immunoreactive neuronal profiles in Alzheimer than in controls. Our data show that, although the brain pathology is moderate to severe, there is no prominent decrease of PV, CR and CB positive neurons in the visual cortex of Alzheimer brains, but only selective changes in neuronal perikarya.

Abstract OBJECTIVE To understand the family dynamics when there is a member in the residence with Alzheimer's disease. METHOD A study of qualitative approach, using the creative sensitive method (CSM), and with participation of two families who had a member with Alzheimer's disease at home. RESULTS Three categories emerged: Effects of Alzheimer's disease and the family dynamics; Development process of Alzheimer's disease and Coping strategies in face of the disease. CONCLUSION It was possible to know the manifestations and consequences of Alzheimer's disease in the family, such as mutual help, the mobilization of resources to activate memories of the past, spirituality and faith. There was also understanding of the structure of family dynamics.

Caspase cleaved amyloid precursor protein (APPcc) and SET are increased and mislocalized in the neuronal cytoplasm in Alzheimer Disease (AD) brains. Translocated SET to the cytoplasm can induce tau hyperphosphorylation. To elucidate the putative relationships between mislocalized APPcc and SET, we studied their level and distribution in the hippocampus of 5 controls, 3 Down syndrome and 10 Alzheimer patients. In Down syndrome and Alzheimer patients, APPcc and SET levels were increased in CA1 and the frequency of both localizations in the neuronal cytoplasm was high in CA1, and low in CA4. As the increase of APPcc is already present at early stages of AD, we overexpressed APPcc in CA1 and the dentate gyrus neurons of adult mice with a lentiviral construct. APPcc overexpression in CA1 and not in the dentate gyrus induced endogenous SET translocation and tau hyperphosphorylation. These data suggest that increase in APPcc in CA1 neurons could be an early event leading to the translocation of SET and the progression of AD through tau hyperphosphorylation.

Lipoprotein(a) [Lp(a)] is an enigmatic lipoprotein particle present in the plasma from humans, great apes and hedgehogs. Plasma levels of Lp(a) vary widely between individuals and are largely determined by specific sequences within the gene encoding apo(a), the unique highly polymorphic glycoprotein attached to apoB of low density lipoprotein (LDL) to form Lp(a). Elevated plasma concentrations of LP(a) are associated with the premature development of atherosclerosis. A major goal of our laboratory is to better understand the metabolism of Lp(a) and its function in humans. We have identified unexpected and large variations in plasma Lp(a) levels during renal disease, HIV-infection and in sepsis. Moreover, we have observed an association between Lp(a) and Alzheimer disease. Taken together, our observations suggest that Lp(a) may constitute a novel target in our fight against cardiovascular and neurodegenerative disorders.

La maladie d'Alzheimer (MA), forme de démence la plus fréquente, est caractérisée précocement par des troubles de la mémoire, puis par une détérioration cognitive progressive, corrélée avec la progression des lésions cérébrales que sont les dépôts de protéine ß-amyloïde, notamment dans les plaques séniles, et la dégénérescence neurofibrillaire qui touche en priorité les grands neurones pyramidaux de l'hippocampe et du cortex cérébral. La perte ou les dommages synaptiques sont aussi prépondérants et conduisent à la mort neuronale et à la perte de réseaux fonctionnels notamment au niveau des protéines présynaptiques comme la synaptophysine et postsynaptiques comme PSD-95 (Leuba et al. 2008), ainsi que des récepteurs NMDA (NMDAR) liés à PSD-95 et jouant un rôle prédominant dans le fonctionnement synaptique. Notre étude s'est portée une des régions du cortex frontal correspondant à l'aire de Brodmann 10, encore peu étudiée dans la littérature scientifique, qui peut être touchée de manière plus ou moins importante dans la MA, entraînant des troubles de l'humeur et du comportement ainsi que des répercussions sur les fonctions exécutives. L'étude a pour but d'identifier et de quantifier les dépôts de protéine ß-amyloïde et les lésions neurofibrillaires ainsi que les changements de protéines synaptiques et de récepteurs NMDA dans cette région entre une population contrôle et AD, et de la comparer avec d'autres régions déjà partiellement étudiées dans le laboratoire, notamment l'aire 9 qui lui est adjacente et les aires cingulaires 24 et 25. L'analyse est faite de manière qualitative et semi-quantitative au microscope optique sur des coupes colorées avec des méthodes immunohistochimiques. De possibles corrélations anatomo-cliniques sont recherchées dans les cas AD. La région FC10 est touchée par la MA avec une présence de plaques séniles et de dégénérescences neurofibrillaires plus marquées chez les cas atteints de la MA ce qui n'est pas le cas des protéines synaptiques. Le comportement des deux marqueurs pathologiques dans FC10 est comparable aux autres régions cérébrales étudiées notamment à la région adjacente, FC9, contrairement aux marqueurs synaptiques qui selon la région ont un comportement plus variable. L'effet de l'âge dans l'évolution de la physiopathologie de la MA pour les marqueurs pathologiques a été mis en évidence dans la région FC10. Les régions EC et FC9 n'ont pas montré de microhémorragie synonyme d'une possible contribution vasculaire. L'étude de cette région a permis de mettre en évidence l'implication de FC10 dans la MA. Elle montre des points communs avec les autres régions cérébrales notamment vis-à-vis des plaques séniles et des DNF.

We investigated how synaptic plasticity is related to the neurodegeneration process in the human dorsolateral prefrontal cortex. Pre- and postsynaptic proteins of Brodmann's area 9 from patients with Alzheimer's disease (AD) and age-matched controls were quantified by immunohistochemical methods and Western blots. The main finding was a significant increase in the expression of postsynaptic density protein PSD-95 in AD brains, revealed on both sections and immunoblots, while the expression of spinophilin, associated to spines, remained quantitatively unchanged despite qualitative changes with age and disease. Presynaptic protein alpha-synuclein indicated an increased immunohistochemical level, while synaptophysin remained unchanged. MAP2, a somatodendritic microtubule protein, as well as AD markers such as amyloid-beta protein and phosphorylated protein tau showed an increased expression on immunosections in AD. Altogether these changes suggest neuritic and synaptic reorganization in the process of AD. In particular, the significant increase in PSD-95 expression suggests a change in NMDA receptors trafficking and may represent a novel marker of functional significance for the disease.

Alzheimer's disease (AD) is a neurodegenerative disease characterized by cognite impairment and personality changes. The development of drugs for the treatment of the cognitive deficits of AD has focused on agents which counteract loss in cholinergic activities. These symptons of AD have been successfully treated with acetylcholinesterase (AchE) inhibitors (eg. galanthamine). There still is great interest in finding better AchE inhibitors. We use Ellmann's microplate assay and silica gel thin-layer chromatography (TLC) to screen natural products from plants as new sources of AchE inhibitors.

Hypertension (HT) is the most prevalent cardiovascular risk factor associated with dementia and Alzheimer disease (AD). The evidence about the association within hight blood pressure (BP) level in the middle age and dementia and Alzheimer’s disease incidence in the advanced age has increased. Longitudinal studies show that in the previous years of onset AD, BP is similar or lower in the patients who develop AD with regard to those who not develop. When patients has developed AD the case is the same that the previous one. Most studies show that BP reduction is beneficious to prevent cognitive impairment, dementia and AD. Is spite of some discordant studies, health authorities recommend to treat isolated systolic HT with an ‘A’ evidence degree, level 1, to prevent AD. Animal experimentation prove different ways to act of antihypertensive drugs in the AD prevention and has established the pathophysiological bases in this relation, until now only showed through various clinical studies

Introduction: Les aidants des personnes âgées (PA) atteintes de la démence de type Alzheimer (DTA) sont confrontés à de nombreux défis lors du soin de leurs proches, y compris ceux liés à l’alimentation. Cependant, ces défis restent méconnus et les stratégies créées pour les gérer sont encore peu efficaces. Objectifs: Identifier les difficultés rencontrées par les aidants pendant la gestion de l’alimentation des PA atteintes de la DTA ayant participé à une intervention nutritionnelle (l’étude NIS) et dégager leurs opinions concernant cette intervention. Sujets: Trente-trois aidants des PA avec DTA du groupe intervention de l’étude NIS ont été ciblés. Méthodes: L’approche qualitative a été employée lors des entrevues individuelles auprès de ces aidants. Les entrevues ont été transcrites et le verbatim fut soumis à une analyse thématique. Résultats: Vingt-quatre aidants ont été interviewés. Quelque 58,4 % avaient 70 ans et plus et 58,3 % étaient des conjoint (es) des patients affectés. Quatre catégories de thèmes furent dégagées menant à l’identification des défis alimentaires suivants: les changements des habitudes alimentaires (altération des préférences); les perturbations du comportement alimentaire (ex. l’oubli de repas); la dépendance à la préparation des repas. L’utilité des conseils, la gentillesse et la compétence du personnel NIS, la documentation écrite offerte et la durée du suivi ont été appréciées par les aidants. Conclusion: Une meilleure compréhension de l'expérience de soin vécu par l’aidant est essentielle au développement des interventions nutritionnelles adaptées aux besoins des aidants et des PA atteintes de la DTA.

Esta revisión de la literatura tuvo como objetivo describir las actitudes hacia el VIH/SIDA, el cáncer y la Enfermedad de Alzheimer desde el modelo tripartito. Se revisaron 109 artículos publicados entre 2005 y 2015 en algunas bases de datos especializadas y herramientas de análisis de impacto. También se incluyeron fuentes secundarias ampliándose la búsqueda a los últimos 20 años (1995-2015). Los resultados mostraron que la mayoría de los estudios realizados sobre las actitudes hacia estas tres enfermedades son de tipo cuantitativo y la información se analizó con base en los componentes del modelo tripartito. Algunos aspectos sociodemográficos como el sexo y la edad están asociados con las actitudes hacia las tres enfermedades y predominan las creencias erróneas sobre ellas respecto a sus causas, curso y tratamiento. También predominan actitudes negativas hacia las tres enfermedades y las conductas e intenciones conductuales son diversas hacia cada una de ellas. No se hallaron antecedentes empíricos del estudio de la estructura de las actitudes propuesta por el modelo tripartito hacia las tres enfermedades. La Salud Pública ha liderado la investigación con base en el modelo de conocimientos, actitudes y prácticas propuesto por la OMS.

Comparar a nivel cognitivo, funcional y conductual el efecto de la modalidad de intervención cognitiva, individual frente a la grupal, en dos grupos de pacientes con Enfermedad de Alzheimer (EA) que recibieron un programa de estimulación cognitiva y un grupo de pacientes con EA que estaba en lista de espera para recibir el mismo programa de intervención cognitiva. Se ha trabajado con una muestra conformada por 60 participantes que asistieron a la Asociación de Familiares de Enfermos de Alzheimer (AFA) Salamanca durante el año 2007. De la muestra, 15 participantes eran sujetos sanos a nivel cognitivo, los 45 restantes tenían diagnóstico de demencia según los criterios del Manual Diagnóstico y Estadístico de las Enfermedades Mentales. El rango de edad de los participantes oscilaba entre los 64 y los 86 años. La media de edad para el total de la muestra era de 75.45 años y una desviación típica de 4.50 años. Participaron 27 hombres y 33 mujeres. El grupo de pacientes con EA estaba conformado por 20 hombres y 25 mujeres. Los 45 pacientes con EA fueron distribuidos en 3 grupos, cada uno con 15 integrantes; de los cuales 15 participantes recibieron el programa de estimulación cognitiva en el formato individual (EAI), 15 pacientes recibieron el mismo programa en el formato grupa (EAG), y los demás estaban en lista de espera para recibir el mismo programa de intervención (EANT). El programa de estimulación cognitiva se llevo a cabo en un periodo de tres meses por grupo, con una frecuencia de tres veces por semana y una duración de una hora por sesión, para un total de 36 sesiones. Al inicio del programa de estimulación cognitiva, los participantes de los tres grupos estaban equiparados en cuento a rendimiento cognitivo evidenciado en las puntuaciones pretest en el Alzheimer Disease Assessment Scale, asimismo en cuanto al rendimiento funcional en la Escala de Evaluación Rápida de Discapacidad y en el rendimiento conductual Neuropsychiatric Inventory Questionary y en la Escala de Depresión de Cornell. Recibir un programa de estimulación cognitiva resulta especialmente significativo para los pacientes con EA respecto a la progresión del declive que a nivel cognitivo, funcional y conductual van experimentando. Al realizar las comparaciones respecto al formato de intervención: individual frente a grupal puede notarse que el formato de intervención individual ofrece mayores beneficios a los pacientes con EA reduciendo especialmente la sintomatología depresiva. Los resultados indican que tanto los pacientes que recibieron el programa de estimulación cognitiva en el formato individual como grupal demostraron mayor rendimiento a nivel cognitivo, funcional y conductual que los pacientes que se encontraban en lista de espera para recibir el mismo programa de intervención cognitiva. Asimismo al realizar las comparaciones tanto entre los pacientes EAI frente a los EAG, se pudo observar que el formato de intervención cognitiva individual ofrece mayores beneficios a los pacientes con EA, especialmente a nivel cognitivo y conductual reduciendo la manifestación de la sintomatología depresiva.

This work is an investigation related to issues of those who take home care of people who suffer from Alzheimer disease (AD). Thus, it is justified by the need to acknowledge how these relatives perform this task and in which ways they do this. The study has is analytical and qualitative methodology with the use of a thematic oral history approach. The subjects of the research were nine relatives of those who suffer from AD that participate in the home care group in the Candelária neighborhood in the city of Natal in Rio Grande do Norte-Brazil. The data was collected using a semi-structured questionnaire and interview that was booked in advance and had full support from the care takers. After information collection, three thematic axles were defined. After this procedure, three analisys subcategories were also defined. The first thematic axle emphasizes the so called movement of rite of passage, when the relative becomes a care taker of a person with AD. The second category deals with the care takers strategies, either related to their own behalf or on their relative. It is possible to infer that amongst other forms of help, the care taker needs to rely on a support network, such as health services, groups composed by multiprofessionals that enable better articulation between family and collaborators. The dimension related to faith and spirituality was also observed and pointed out as an important aspect in the emotional support process for these relatives. In the third axle the perspectives of struggle, conquests of the right to health and life quality of those who suffer from AD as well as their relatives was observed. OBJETIVO: Realizar uma revisão sistemática de artigos científicos que analisaram os efeitos da atividade física sistematizada nos sintomas de depressão de pacientes com demência de Alzheimer. MÉTODOS: Foi realizada uma busca nas seguintes bases de dados: Web of Science, PubMed, PsycINFO, MedLine e Biological Abstracts, utilizando-se as seguintes palavras-chave: Alzheimer dementia , Alzheimer disease , Alzheimer, physical activity, physical exercise, motor intervention , physical therapy , exercise , aerobic , strength , fitness , depression , dysphoria , depressive symptoms e depressive episodes. Além da busca nas bases de dados, foi realizada também uma busca manual nas listas de referências dos artigos selecionados. RESULTADOS: Foram encontrados quatro estudos que preencheram todos os critérios de inclusão adotados para o presente trabalho. Dois estudos apresentaram reduções dos sintomas depressivos, e outros dois não encontraram redução desses sintomas. CONCLUSÃO: Com a realização desta revisão sistemática, observou-se que não há consenso em relação aos benefícios da atividade física aos sintomas depressivos em pacientes com demência de Alzheimer.

O P300 desempenha um papel fundamental como método de avaliação e monitoramento das demências, entre elas a doença de Alzheimer. OBJETIVO: Realizar uma busca por artigos que analisaram os valores de latência e amplitude de P300 na doença de Alzheimer. MÉTODOS: Foi realizada uma busca nas seguintes bases de dados: Web of Science, PubMed/Medline, Psyc Info, Biological Abstracts e Scielo. Utilizaram-se as seguintes palavras-chave: speed of information processing, speed of processing, information processing, aged, older, elderly, older people, alzheimer dementia, alzheimer disease, Alzheimer, além de referências cruzadas dos artigos selecionados. RESULTADOS: Foram encontrados oito estudos que preencheram os critérios de inclusão adotados para o presente trabalho. Estes estudos mostraram que existe um consenso em relação ao aumento da latência de P300 de idosos com doença de Alzheimer quando comparados com idosos sem a doença. Porém, verifica-se que, com relação à amplitude de P300, ainda não há um consenso, mas, isso pode estar relacionado às diferentes variáveis metodológicas adotadas nos estudos revisados. CONCLUSÃO: Há necessidade de se padronizar as variáveis envolvidas no método de avaliação do P300 para idosos com doença de Alzheimer, para que seja possível comparar os valores de latência e amplitude de P300 dessa população.
