Amyotrophic lateral sclerosis (ALS) is a progressive degenerative disorder affecting motoneurons and the SOD1(G93A) transgenic mice are widely employed to study disease physiopathology and therapeutic strategies. Despite the cellular and biochemical evidences of an early motor system dysfunction, the conventional behavioral tests do not detect early motor impairments in SOD1 mouse model. We evaluated early changes in motor behavior of ALS mice by doing the analyses of tail elevation, footprint, automatic recording of motor activities by means of an infrared motion sensor activity system and electrophysiological measurements in male and female wild-type (WT) and SOD1(G93A) mice from postnatal day (P) 20 up to endpoint. The classical evaluations of mortality, weight loss, tremor, rotometer, hanging wire and inclined plane were also employed. There was a late onset (after P90) of the impairments of classical parameters and the outcome varied between genders of ALS mice, being tremor, cumulative survival, weight loss and neurological score about 10 days earlier in male than female ALS mice and also about 20 days earlier in ALS males regarding rotarod and hanging wire performances. While diminution of hindpaw base was 10 days earlier in ALS males (P110) compared to females, the steep length decreased 40 days earlier in ALS females (P60) than ALS males. The automatic analysis of motor impairments showed substantial late changes (after P90) of motility and locomotion in the ALS females, but not in the ALS males. It was surprising that the scores of tail elevation were already decreased in ALS males and females by P40, reaching the minimal values at the endpoint. The electrophysiological analyses showed early changes of measures in the ALS mouse sciatic nerve, i.e., decreased values of amplitude (P40) and nerve conduction velocity (P20), and also an increased latency (P20) reaching maximal level of impairments at the late disease phase. The early changes were not accompanied by reductions of neuronal protein markers of neurofilament 200 and ChAT in the ventral part of the lumbar spinal cord of P20 and P60 ALS mice by means of Western blot technique, despite remarkable decreases of those protein levels in P120 ALS mice. In conclusion, early changes of motor behavior and electrophysiological parameters in ALS mouse model must be taken into attention in the analyses of disease mechanisms and therapeutic effects. There is a high prevalence of leprosy in the Amazon region of Brazil. We have developed a distance education course in leprosy for training staff of the Family Health Teams (FHTs). The course was made available through a web portal. Tele-educational resources were mediated by professors and coordinators, and included the use of theoretical content available through the web, discussion lists, Internet chat, activity diaries, 3-D video animations (Virtual Human on Leprosy), classes in video streaming and case simulation. Sixty-five FHT staff members were enrolled. All of them completed the course and 47 participants received a certificate at the end of the course. At the end of the course, 48 course-evaluation questionnaires were answered. A total of 47 participants (98%) considered the course as excellent. The results demonstrate the feasibility of an interactive, tele-education model as an educational resource for staff in isolated regions. Improvements in diagnostic skills should increase diagnostic suspicion of leprosy and may contribute to early detection.

This is a study in the rat of the distribution of specific neurotransmitters in neurones projecting from the substantia nigra reticulata (SNR) to the ventrolateral (VL) and ventromedial (VM) thalamic nuclei. Individual axons projecting from the SNR to these thalamic nuclei have also been reconstructed following small injection of the anterograde tracer dextran biotin into the the SNR. Analysis of reconstructions revealed two populations of SNR neurones projecting onto the VL and VM thalamic nuclei. One group projects directly onto the VM and VL, and the other projects to the VM/VL and to the parafascicular nucleus. In another set of experiments Fluoro-Gold was injected into the VL/VM to label SNR projection neurones retrogradely, and immunohistochemistry was performed to determine the distribution of choline acetyltransferase (ChAT), vesicular acetylcholine transporter (VAChT), gamma -aminobutyric acid (GABA), and glutamate in Fluoro-Gold-labelled SNR projection neurones. Most SNR-VL/VM thalamic projection neurones were immunoreactive to acetylcholine or glutamate, whereas only 25% of the projection neurones were found to be immunoreactive to GABA. We wished to identify the different types of retinal neurons on the basis of their content of neuroactive substances in both larval tiger salamander and mudpuppy retinas, favored species for electrophysiological investigation. Sections and wholemounts of retinas were labeled by immunocytochemical methods to demonstrate three calcium binding protein species and the common neurotransmitters, glycine, GABA and acetylcholine. Double immunostained sections and single labeled wholemount retinas were examined by confocal microscopy. Immunostaining patterns appeared to be the same in salamander and mudpuppy. Double and single cones, horizontal cells, some amacrine cells and ganglion cells were strongly calbindin-immunoreactive (IR). Calbindin-IR horizontal cells colocalized GABA. Many bipolar cells, horizontal cells, some amacrine cells and ganglion cells were strongly calretinin-IR. One type of horizontal cell and an infrequently occurring amacrine cell were parvalbumin-IR. Acetylcholine as visualized by ChAT-immunoreactivity was seen in a mirror-symmetric pair of amacrine cells that colocalized GABA and glycine. Glycine and GABA colocalized with calretinin, calbindin and occasionally with parvalbumin in amacrine cells. (C) 2001 Elsevier Science Ltd. All rights reserved.

Quantum feedback can stabilize a two-level atom against decoherence (spontaneous emission), putting it into an arbitrary (specified) pure state. This requires perfect homodyne detection of the atomic emission, and instantaneous feedback. Inefficient detection was considered previously by two of us. Here we allow for a non-zero delay time tau in the feedback circuit. Because a two-level atom is a non-linear optical system, an analytical solution is not possible. However, quantum trajectories allow a simple numerical simulation of the resulting non-Markovian process. We find the effect of the time delay to be qualitatively similar to chat of inefficient detection. The solution of the non-Markovian quantum trajectory will not remain fixed, so that the time-averaged state will be mixed, not pure. In the case where one tries to stabilize the atom in the excited state, an approximate analytical solution to the quantum trajectory is possible. The result, that the purity (P = 2Tr[rho (2)] - 1) of the average state is given by P = 1 - 4y tau (where gamma is the spontaneous emission rate) is found to agree very well with the numerical results. University students spelled low-frequency words to dictation and subsequently made lexical decisions to them. In Experiment I, lexical decisions were slower on words students had spelled incorrectly relative to words they had spelled correctly, and there A as a larger repetition benefit 101 incorrectly spelled words. In experiment 2, the latency advantage for items spelled correctly was replicated when words were presented for only 200 ms and also in a spelling recognition task, In Experiment 3. masked identity and form priming effects were similar for words that had been spelled correctly and incorrectly, Item spelling accuracy tracked word frequency effects in the way chat it combined with repetition and priming effects. we inter that an individuals learning with a word's orthography underlies word frequency and item spelling accuracy effects and that a single orthographic lexicon serves visual word recognition and spelling. Trata-se de uma pesquisa-intervenção com o objetivo principal de colocar em análise a atividade na vigilância em saúde do trabalhador ancorada nos pressupostos da formação pelo trabalho. Tem como aporte conceitual o referencial teórico-metodológico da clínica da atividade proposta por Yves Clot e colaboradores. A pesquisa foi realizada com a equipe multiprofissional que atua na vigilância em saúde do trabalhador do Centro de Referência Estadual em Saúde do Trabalhador do Espírito Santo (Cerest/ES). As rodas de conversa, a partir da clínica da atividade, foram a estratégia metodológica privilegiada. O gênero profissional vigilante em saúde do trabalhador, ao se confrontar com o estudo das diretrizes da clínica da atividade, foi produzindo deslocamentos que o levou a (re)pensar, analisar a sua atividade e o processo de trabalho no qual estava inserido. A cada encontro no diálogo com a clínica da atividade, eram experimentados os modos de como esses profissionais atuavam na vigilância, como também todo o atravessamento desse gênero na assistência aos trabalhadores, nas atividades educativas e gestão do trabalho. No desenvolvimento das rodas, ficou evidenciado o quanto esses encontros se tornaram equipamentos para esses profissionais. As atividades desenvolvidas produziram análise coletiva do trabalho e um processo de formação pelo trabalho. Buscaram incorporar os métodos utilizados pela clínica da atividade no cotidiano do Cerest dialogando com os efeitos desse processo. Nas rodas também emergiram propostas de mudanças na maneira como vinham ocorrendo as atividades realizadas, a gestão do trabalho nessa equipe e as estratégias de atuação na vigilância, ou seja, intervenção no processo de trabalho do grupo, produzindo potência para o poder de agir do gênero. O referencial teórico-metodológico da clínica da atividade foi um dispositivo de formação importante. Permitiu a experimentação e o desenvolvimento do gênero vigilantes em saúde do trabalhador na produção de análise da atividade, na constituição de uma clínica dialógica em transformação. Também possibilitou contemplar na análise as dimensões integrantes da atividade de trabalho, fazendo a interlocução para o encontro entre os saberes da experiência instituídos no cotidiano do serviço de saúde e o saber acadêmico na produção de conhecimento.

Multiple-Choice items are used in many different kinds of tests in several areas of knowledge. They can be considered an interesting tool to the self-assessing or as an alternative or complementary instrument to the traditional methods for assessing knowledge. The objectivity and accuracy of the multiple-choice tests is an important reason to think about. They are especially useful when the number of students to evaluate is too large. Moodle (Modular Object-Oriented Dynamic Learning Environment) is an Open Source course management system centered around learners' needs and designed to support collaborative approaches to teaching and learning. Moodle offers to the users a rich interface, context-specific help buttons, and a wide variety of tools such as discussion forums, wikis, chat, surveys, quizzes, glossaries, journals, grade books and more, that allow them to learn and collaborate in a truly interactive space. Come together the interactivity of the Moodle platform and the objectivity of this kind of tests one can easily build manifold random tests. The proposal of this paper is to relate our journey in the construction of these tests and share our experience in the use of the Moodle platform to create, take advantage and improve the multiple-choices tests in the Mathematic area.

O desafio que nos colocamos prende-se antes com a (re)descoberta da didacticidade.

Miocene catfishes from Lisbon are dealt with. Two distinct sets of pectoral and dorsal pterygiophores are described. That from the Langhian V-b is referred to Arius sp. probably close to A. heudeloti. Another ser from the uppermost Burdigalian V-a may be ascribed to a bagrid, cf. Chrysichthys sp., identified for the first time in this region. The catfish and Lates association is sctrikingly similar to African, nilotic or sudanian ones as far as freshwaters are concerned. In marine, coastal environments, stenotherm warm-water forms (Polynemids, large barracudas and several sharks) indicate, as a model, faunas like chose from Cape Verde to northern Angola. There is some gradation for brackish waters (fig. 1). Catfishes and Lates probably migrated into the Iberian Peninsule in the lower Miocene. They are unknown after Langhian V-b except for a reappearance of Arius in the middle Tortonian VII-b. Decreasing temperatures and aridity account for local extinction at least in freshwaters. Expansion of these fishes have been made easier owing to the displacement of land masses chat narrowed or closed the marine waterway between Europe and Africa. Salinity tolerance is not necessarily the sole explanation for migration. Catfishes plus Lates associations colonized inland waters from both sides of the Paleomediterranean. Local extinction may have weighed more in the development of modern distribution patterns than migration.

A União Europeia tem vindo a promover a intercompreensão entre países, povos e culturas que fazem parte da sua esfera comunicativa. Esta promoção passa por projectos educativos como o ERASMUS, COMENIUS e SOCRATES, permitindo a livre mobilidade de cidadãos em prol da troca de saberes e experiência de realidades educativas. Contudo, os elevados custos que tais programas acarretam, levaram a União Europeia a procurar alternativas e, como tal, nada melhor que utilizar os recursos das novas tecnologias da informação e comunicação, que constituem um inovador instrumento do ambiente de ensino-aprendizagem, já que contribuem para um profundo e vasto desenvolvimento cognitivo. Esta “mobilidade on-line” (Cruz & Melo, 2004) beneficia de uma grande independência temporal, uma vez que os chatantes actuam em momentos distintos e também de uma independência local, visto que, os intervenientes podem estar em locais diversos. Contudo, é necessário ao cidadão comum saber como se deve mover nos meandros da Internet, ou seja, é necessário que domine não só a tecnologia adjacente à navegação na Internet e salas de chat, bem como também alguns dos códigos linguísticos que fazem partem da esfera comunicativa da União Europeia. Refiro-me aqui a uma literacia ou multiliteracia electrónica que engloba os códigos e usos próprios do mundo virtual e a mobilização de uma competência plurilingue e intercultural aquando da comunicação síncrona mediatizada por computador.

O ensino precoce de línguas estrangeiras no resto da Europa encontrase numa fase mais avançada do que em Portugal. Pretendemos, neste estudo, fazer o ponto de situação em Portugal quanto ao ensino precoce de línguas, definir o perfil do professor de línguas estrangeiras no Ensino Básico e perceber qual a idade ideal para a aquisição de línguas estrangeiras. De forma a enriquecer este estudo, centrámos a nossa atenção nas razões para a aquisição de línguas neste nível de ensino e realizámos um estudo de caso que compreende a realização de conversações por chat entre alunos portugueses e suíços do Ensino Básico. In the rest of Europe the foreign languages early teaching is more advanced than in Portugal. With this study we aim to define both the situation Portugal is living regarding this issue, understand which the profile of the teacher of foreign languages is at an early stage and perceive which the ideal age to start studying foreign languages is. In order to enrich this study, we focused our attention in the reasons for the acquisition of languages at a primary school level and made a study case which is related to the analysis of chat conversations between Portuguese and Swiss pupils.

Inibidores contra as amostras de poliovírus do tipo 1, Mahoney e CHAT, foram estudados no sistema de gel de hidróxido de alumínio. O inibidor comportou-se como uma macroglobulina do tipo 19 S (IgM), através filtração em gel (Sephadex G-200), partição em DEAE-Celulose e sedimentação por ultra-centrifugação em gradiente de sacarose. O inibidor foi ainda destruído através o tratamento com 2-Mercapto-Etanol. O inibidor une-se à superfície do vírus na ausência de células. O complexo formado pelo vírus e o inibidor pode ser precipitado por um sôro preparado contra a globulina bovina; o mesmo complexo dissocia-se em idêntico valor de pH ao do complexo vírus-anticorpo 19 S. A chamada infecciosidade residual (cêrca de 10%) que permanece após a neutralização de poliovírus com sôro bovino representa uma dissociação parcial do inibidor da partícula de vírus, quando em presença das células em cultura. Foram obtidas amostras de vírus resistentes à ação específica de soros bovinos. Estas amostras permanecem sensíveis à ação de outros soros bovinos ativos, bem como à ação de anticorpos 7 S e 19 S. Através experiências de adsorção de atividade inibidora, foi possível mostrar a existência de inibidores monoespecíficos, com diferentes especificidades contra as amostras CHAT e Mahoney, respectivamente.
