Neoplasias malignas da pele são os cênceres mais comuns da espécie humana. No entanto há tipos raros como o Carcinoma de células de Merkel (CCM), cuja incidência tem aumentado em todo mundo. O CCM possui curso agressivo, com freqüente envolvimento de nódulos linfáticos regionais e metástases à distância. Adicionalmente, afeta predominantemente idosos e imunocomprometidos, fato que levou-se a suspeita de uma possível etiologia infecciosa para essa neoplasia. Nesse foi-se isolado e descrito o MCPyV, um novo poliomavirus humano, diretamente de células tumorais do CCM. O objetivo do presente trabalho é dar continuidade à pesquisa desse novo vírus apresentando dados iniciais da pesquisa do MCPyV em número significativo de casos de CCM de pacientes brasileiros. Para tanto, foram analisadas 24 biópsias de CCM fixadas e incluídas em parafina, das quais foram extraído o material genômico e o produto submetido à PCR convencional com três pares de iniciadores descritos pela literatura (LT1, LT3 e VP1), com a finalidade de se detectar segmentos do vírus. No presente estudo o genoma viral foi detectado em 11/24 (45,8%) das amostras avaliadas, sendo que a positividade para cada par de iniciadores foi de 4/24 (16,7%) para LT1, 11/24 (45,8%) para LT3 e 4/24 (16.7%) para VP1. Essas freqüências são menores do que a relatada pela literatura e essa diferença pode ser devida a diferença nas amostras analisadas e nas técnicas empregadas. Outros estudos são necessários para comprovar a relação de causalidade, assim como desvendar o ciclo do MCPyV

Introduction: Tuberculosis (TB) is a granulomatous disease caused by Mycobacterium tuberculosis. The genus Mycobacteriumhas two different complexes: M. tuberculosis Complex and M. avium Complex. This is a global health epidemic and remains a major global health problem, besides, the clinical severity of TB is significantly higher in transplanted patients. The detection of these mycobacteria complexes in transplanted patients, by molecular methods, is fundamental for quick treatment of patients and can contribute for rapid and accuracy of diagnosis. Objective: To detect mycobacteria DNA of M. tuberculosis and M. avium Complexes in formalin fixed paraffin-embedded samples (FFPE) of two patients groups: non transplanted and transplanted. Materials and Methods: The study includes 40 FFPE biopsies separated in four groups: NTP – presence of epithelioid granuloma and positive ZN, non-transplanted patients – 9 samples; NTN - presence of epithelioid granuloma and negative ZN, non-transplanted patients – 10 samples; TP – positive ZN, transplanted patients – 9 samples; TN – negative ZN, transplanted patients – 7 samples. Sections were cut for DNA extraction. Samples were submitted to PCR for amplification of: a) β-actin, b) IS6110 insertion and c) IS1245 insertion. DNA evaluation was made by spectrophotometry and efficiency and PCR analysis was made by agarose gels under UV light. Results: In all samples processed, 97.1% were positive for human β-actin gene. In22.2% of NTP group were found the IS6110 insertion sequencebut the IS1245 wasn´t. In the NTN group was not found any sequence. In theTP group, 11.1% of the samples were positive for IS6110 and also 11,1% werepositive for IS1245. In the TN group, 14.3% of the samples were positive forIS6110 and for IS1245, 14.3% was also positive. A fluoxetina, pertencente à classe dos inibidores da recaptação seletiva de serotonina, é um dos antidepressivos mais amplamente prescritos para o tratamento da depressão e de transtornos da ansiedade, como o transtorno obsessivo compulsivo. Considerando que, dentre os efeitos colaterais causados pela fluoxetina, têm sido relatados distúrbios na função sexual masculina, foi proposto avaliar a ação deste fármaco sobre a integridade morfológica dos túbulos seminíferos. Foram utilizados 16 ratos adultos, distribuídos em 2 grupos: grupo fluoxetina (GF) e grupo controle (GC). Os animais do GF receberam injeções intraperitoneais de fluoxetina (20mg/Kg) e os animais do GC receberam água destilada. O tratamento se estendeu por 11 dias consecutivos e, ao final do tratamento, foram obtidos os pesos corpóreo e testicular dos animais. Os testículos foram fixados e processados para inclusão em historesina e parafina. Nos cortes de historesina, as frequências de túbulos de acordo com o estágio do ciclo do epitélio seminífero e de túbulos seminíferos contendo células descamadas preenchendo a luz tubular foram obtidas, bem como os seguintes parâmetros morfométricos: área tubular total, área da luz tubular e área do epitélio seminífero. Os cortes em parafina foram submetidos ao método do TUNEL para detecção de morte celular. Os níveis séricos de testosterona também foram avaliados. Os resultados foram submetidos à análise estatística para avaliação das diferenças entre os grupos. O tratamento com fluoxetina causou redução significante do peso corpóreo dos animais e redução, embora não significante, do peso testicular absoluto. Além disso, uma redução significante nos níveis séricos de testosterona foi observada nos animais do GF. Os biossurfactantes, moléculas tenso-ativas sintetizadas por microrganismos, apresentam propriedades biológicas e superficiais que podem ser aplicadas a processos industriais diversos. Apesar das vantagens apresentadas, não são amplamente utilizados pela indústria, devido ao alto custo de produção, baixa produtividade e uso de substratos caros. Bactérias do gênero Pseudomonas são conhecidas por sintetizarem ramnolipídios, moléculas tenso-ativas promissoras em termos de produção industrial, por apresentarem excelentes propriedades tenso-ativas, além de características físico-químicas e biológicas que permitem a obtenção em concentrações superiores a outros biossurfactantes. O uso de substratos alternativos seria uma estratégia que facilitaria o desenvolvimento industrial de sua produção. A proposta deste trabalho é dar continuidade ao estudo da produção de ramnolipídios a partir de substratos não-convencionais (n-parafina, óleos vegetais utilizados em frituras, glicerol) por isolados de P. aeruginosa.

Cutaneous melanoma is a malignancy caused by the uncontrolled proliferation of melanocytes. Has been increasing in incidence and mortality over the years. This type of tumor may arise in the skin or any other place where there was migration of neural crest cells. Melanocytes are located in the basal layer of the epidermis forming the epidermal melanin unit, involving a melanocyte to 36 keratinocytes. The proliferation of melanocytes depends on this relationship with the keratinocytes, gap junctions, E-cadherin and desmoglein. The basal membrane is responsible for promoting the integrity of the skin, their adhesion structures shown to be altered in several skin diseases. Some molecules that play the role of adherence in this membrane are the laminins, type IV collagen and type XVII and perlecan. Analysis of protein expression of cell adhesion molecules in melanoma by immunohistochemistry on tissue microarray (TMA). We selected paraffin blocks of patients who were diagnosed with cutaneous melanoma in the period 1995 to 2010, reaching a sample of 124 cases. All these have gone through review of preparation for subsequent histological analysis of tissue microarray (TMA). The protein expression will be analyzed by immunohistochemistry, using markers anti-laminin, anti-collagen type IV and type XVII and anti-perlecan. There was a predominance of female patients, Caucasians and southeastern Brazil. There were frequent in type of extensive superficial ulcers with Breslow depth greater than 4 mm, Clark level IV and mitotic index greater than 6 mitoses per cga. The patients, most of them were female with mean age of 57.13 years and Caucasian. The most frequent histological type was extensive superficial followed by acral lentiginous. Predominated cases very deep, with Breslow thickness greater than 4 mm, Clark IV, ulcerated and on average by 9 cga mitosis, leading to worse prognosis in this sample. Cyclosporin A (CsA) is a potent immunosuppressor used in organ transplantation and in the management of various autoimmune diseases. Recent studies have shown that CsA stimulates deposition of cementum on root surfaces. The aim of this study was to evaluate the periapical cementum thickness and the apical foramen width in CsA-treated rats. Rats weighing 50 g were treated with a daily injection of 10 mg/kg body weight of CsA in the chow for 60 days. The cementum of the mandibular 1st molars was histologically and morphometricaly examined by analysis of 5-microm-thick serial buccolingual paraffin sections stained with hematoxylin and eosin. Histometric and stereologic analyses revealed the presence of large amounts of cementum in all root surfaces, particularly abundant in the periapical region and obliterating the foramen. The volume density of cementoblasts did not increase. Five to 90 days after the termination of CsA therapy, there was no reduction of cementum thickness. These results suggest that cementum deposition is not reversible after cessation of CsA treatment.

The biocompatibility of two dentin adhesives (Scotchbond MultiPurpose- 3M and Optibond Multi-Use - Kerr) was evaluated in rats subcutaneous connective tissue implants. Polyethylene tubes filled with the adhesives were surgically implanted into the dorsal subcutaneous tissues of rats. The tubes were left implanted for periods of 14, 30, 60 and 84 days, following wich the animais were killed and the implants, together with the surrounding tissue, were excised. The specimens were then fixed in 10 per cent formalin, sectioned serially and stained with hematoxylin and eosin and then, examined by light microscopy. After 14 and 30 days the tubes were surrounded by a mild to moderate inflammatory infiltration. After 60 and 84 days microscopic examination revealed the formation of a connective tissue capsule around each tube. The microscopic examination of the biopsy specimens allowed to conclude that: a) both dentin adhesives are considered biologically acceptable when placed in contact with rats subcutaneous connective tissue; b) in the observation period of 30 days there were moderate tissue reaction to the Optibond Multi-Use -light cure- (Kerr); c) after 60 and 84 days the use of both dentine adhesives did not interfere in the healing of the surrounding connective tissue

Allogeneic, fresh-frozen bone has been used in order to replace bone autografts. However, its osteoinduction and osteoconduction properties are not well-defined in the scientific literature. This work aimed to evaluate samples of homogenous bone grafts in humans by qualitative histological and immunohistochemical analysis. For this, ten pre-selected patients underwent surgical augmentation of bone defects. The homogenous fresh frozen block bone graft was stabilized and fixed by bicortical screws. After six months, the reopening procedure was performed for installation of osseointegrated implants. At this time surgical bone graft samples were removed by means of drill trephine. The samples were fixed in 10% formalin, processed with decalcified paraffin, and stained with hematoxylin and eosin. Immunohistochemistry was performed for the expression of Caspase 3 enzyme. The slides were brought to light microscopy for qualitative histology and immunohistochemistry. The results showed non-vital bone tissue, with few areas of deposition of new bone formation on the amorphous matrix, presence of chronic inflammatory infiltrate with areas of osteomyelitis, and expressive immunolabeling of Caspase 3. Given the methods employed and the results it was concluded that the allograft fresh-frozen block is not incorporated into the recipient bed after a healing period of six months.
