Este trabalho analisa a política de participação na Educação de Jovens e Adultos (EJA) no contexto de desenvolvimento capitalista brasileiro, focalizando os programas e as experiências implementadas no município de Belém, Estado do Pará, durante o período de 1997 a 2004. Na análise privilegiaremos as relações entre o nível macro (Orçamento Participativo e Congresso da Cidade) e o nível micro (PROALFA e MOVA) implementado pela Prefeitura de Belém, que são tidos como modelos de instâncias caracterizadas pela participação da população na discussão e elaboração de políticas e projetos de desenvolvimento e o resgate histórico das experiências e programas efetivados nessa modalidade de ensino. É nossa intenção ainda identificar como a SEMEC e outros órgãos municipais se reestruturaram, para criar e fortalecer setores responsáveis por essa modalidade de ensino. No processo investigativo utilizaremos a pesquisa de caráter qualitativo. Dessa forma, para traçar um panorama acerca da temática foram utilizados os seguintes procedimentos: pesquisa bibliográfica, pesquisa documental. Estaremos utilizando as categorias conceituais de participação-poder, e da participação controlada. Em nível macro, apesar dos avanços observados na participação popular em Belém, não se pode afirmar que a sociedade civil tenha assumido o controle sobre o planejamento municipal. Pode-se inferir que este ocorreu de forma compartilhada entre o poder municipal e a sociedade civil no qual a organização e a dinâmica de funcionamento do Orçamento Participativo e o Congresso da Cidade permitiram a descentralização das decisões sobre as políticas públicas que passaram a acontecer dentro do Conselho da Cidade, órgão máximo de decisão no Congresso da Cidade. Essa foi a maneira encontrada pelo Poder Municipal de legitimar a participação da sociedade civil. O PROALFA e o MOVA foram parte da política educacional do Governo do Povo e sua ação política e cultural se afirmam, tomando corpo o conceito de parceria e diálogo entre poder público e sociedade civil. O primeiro papel do Governo do Povo em relação ao PROALFA e ao MOVA foi o de se colocar como impulsionador da criação e da implementação do Movimento, o que se constituiu como participação controlada, pois teve origem na concessão do governo. Essas instâncias como o Orçamento Participativo e o Congresso da Cidade, o PROALFA e o MOVA tiveram suas limitações, da mesma forma que se reconhece que ainda estão muito longe de uma participação massiva plena - participação poder, dada à complexidade de construção desta. Mas é preciso reconhecer que muito se avançou no processo de democratização e descentralização em Belém, tendo um avanço enorme na construção de políticas públicas para essa modalidade de ensino.

This article portrays pathways and studies, arising from research already completed on relationships that constitute the process of appropriation-objectification of the world of human culture by the child at the School for Children, under the gaze of Historical-Cultural Approach. The studies had been configured in accord with a theoretical look that understands the relations as propeller of the humanization process and the child as capable to learn since very small. Initially, there is deepening of theoretical issues concerning the regularities of human development between three and six years old, based on the propositions of the Historical-Cultural Approach. Subsequently, it presents results of the routes methodological that integrated observation sessions of educational practice in schools in early childhood education, involving the relationship between children, adults and objects, as well as sessions of designs and semi-structured interviews with the children. These results show the resulting analysis of the pedagogical implications of the propositions of the HistoricalCultural Approach about the humanization process, mediation and activity.

The article searches to launch some analyses and investigations on the historical reach of produced participative processes in Spain and Brazil. Evidently, both the processes if had uncurled in historical, social, economic contexts and distinct politicians, but, exactly thus, keep some approaches that we intend to give prominence in the present article. For example, the popular mobilization in the two presented cases after occurred a long period of not attendance of social demands socials. In the same way, the processes of democratic renewal of both are related with historical landmarks “end of the Cold War” and the dismantlement of the Walfare State that introduced new rationalists in the relation between governing and governed dynamic, over all in the coinage and offer of public politics. How much to the dismantlement of the Welfare State, we present, still, that in the case of Spain it provoked consequences in the modus operandi of the formularization process and implementation of public politics. It occurred, in other words, a decentralization of its formulator power to decide arrangement of public politics that started to count on the participation of social actors in the called relational city (BECERRA, 2011). In the case of Brazil, the dictatorial period (1964-1985), the fast urban growth and the generation of precarious peripheries in the main cities, the distension and the Constituent (1986-1988) had generated the enough forces of pressure so that, gradually, if it incorporated the participative arrangement in the power to decide process. We identify that in both the cases, kept the had ratios, the instauration of a critical scene of suppression/absence of formulated public politics from an interface with the social movements were the propeller spring of the transformations in the power to decide mechanisms of formularization and implementation of public politics.

This work provides a numerical and experimental investigation of fatigue crack growth behavior in steel weldments including crack closure effects and their coupled interaction with weld strength mismatch. A central objective of this study is to extend previously developed frameworks for evaluation of crack clo- sure effects on FCGR to steel weldments while, at the same time, gaining additional understanding of commonly adopted criteria for crack closure loads and their influence on fatigue life of structural welds. Very detailed non-linear finite element analyses using 3-D models of compact tension C ( T ) fracture spec- imens with center cracked, square groove welds provide the evolution of crack growth with cyclic stress intensity factor which is required for the estimation of the closure loads. Fatigue crack growth tests con- ducted on plane-sided, shallow-cracked C ( T ) specimens provide the necessary data against which crack closure effects on fatigue crack growth behavior can be assessed. Overall, the present investigation pro- vides additional support for estimation procedures of plasticity-induced crack closure loads in fatigue analyses of structural steels and their weldments

The aim of this Doctoral Thesis is to develop a genetic algorithm based optimization methods to find the best conceptual design architecture of an aero-piston-engine, for given design specifications. Nowadays, the conceptual design of turbine airplanes starts with the aircraft specifications, then the most suited turbofan or turbo propeller for the specific application is chosen. In the aeronautical piston engines field, which has been dormant for several decades, as interest shifted towards turboaircraft, new materials with increased performance and properties have opened new possibilities for development. Moreover, the engine’s modularity given by the cylinder unit, makes it possible to design a specific engine for a given application. In many real engineering problems the amount of design variables may be very high, characterized by several non-linearities needed to describe the behaviour of the phenomena. In this case the objective function has many local extremes, but the designer is usually interested in the global one. The stochastic and the evolutionary optimization techniques, such as the genetic algorithms method, may offer reliable solutions to the design problems, within acceptable computational time. The optimization algorithm developed here can be employed in the first phase of the preliminary project of an aeronautical piston engine design. It’s a mono-objective genetic algorithm, which, starting from the given design specifications, finds the engine propulsive system configuration which possesses minimum mass while satisfying the geometrical, structural and performance constraints. The algorithm reads the project specifications as input data, namely the maximum values of crankshaft and propeller shaft speed and the maximal pressure value in the combustion chamber. The design variables bounds, that describe the solution domain from the geometrical point of view, are introduced too. In the Matlab® Optimization environment the objective function to be minimized is defined as the sum of the masses of the engine propulsive components. Each individual that is generated by the genetic algorithm is the assembly of the flywheel, the vibration damper and so many pistons, connecting rods, cranks, as the number of the cylinders. The fitness is evaluated for each individual of the population, then the rules of the genetic operators are applied, such as reproduction, mutation, selection, crossover. In the reproduction step the elitist method is applied, in order to save the fittest individuals from a contingent mutation and recombination disruption, making it undamaged survive until the next generation. Finally, as the best individual is found, the optimal dimensions values of the components are saved to an Excel® file, in order to build a CAD-automatic-3D-model for each component of the propulsive system, having a direct pre-visualization of the final product, still in the engine’s preliminary project design phase. With the purpose of showing the performance of the algorithm and validating this optimization method, an actual engine is taken, as a case study: it’s the 1900 JTD Fiat Avio, 4 cylinders, 4T, Diesel. Many verifications are made on the mechanical components of the engine, in order to test their feasibility and to decide their survival through generations. A system of inequalities is used to describe the non-linear relations between the design variables, and is used for components checking for static and dynamic loads configurations. The design variables geometrical boundaries are taken from actual engines data and similar design cases. Among the many simulations run for algorithm testing, twelve of them have been chosen as representative of the distribution of the individuals. Then, as an example, for each simulation, the corresponding 3D models of the crankshaft and the connecting rod, have been automatically built. In spite of morphological differences among the component the mass is almost the same. The results show a significant mass reduction (almost 20% for the crankshaft) in comparison to the original configuration, and an acceptable robustness of the method have been shown. The algorithm here developed is shown to be a valid method for an aeronautical-piston-engine preliminary project design optimization. In particular the procedure is able to analyze quite a wide range of design solutions, rejecting the ones that cannot fulfill the feasibility design specifications. This optimization algorithm could increase the aeronautical-piston-engine development, speeding up the production rate and joining modern computation performances and technological awareness to the long lasting traditional design experiences.

In this work, solid-state NMR methods suitable for the investigation of supramolecular systems were developed and improved. In this context, special interest was focussed on non-covalent interactions responsible for the formation of supramolecular structures, such as pi-pi interacions and hydrogen-bonds. In the first part of this work, solid-state NMR methods were presented that provide information on molecular structure and motion via the investigation of anisotropic interactions, namely quadrupole and dipole-dipole couplings, under magic-angle spinning conditions. A two-dimensional 2H double quantum experiment was developed, which is performed under off magic-angle conditions and correlates 2H isotropic chemical shifts with quasistatic DQ-filtered line shapes. From the latter, the quadrupole coupling parameters of samples deuterated at multiple sites can be extracted in a site-selective fashion. Furthermore, 7Li quadrupole parameters of lithium intercalated into TiO2 were determined by NMR experiments performed under static and MAS conditions, and could provide information on the crystal geometry. For the determination of 7Li-7Li dipole-dipole couplings, multiple-quantum NMR experiments were performed. The 1H-13C REREDOR experiment was found to be capable of determining strong proton-carbon dipole-dipole couplings with an accuracy of 500~Hz, corresponding to a determination of proton-carbon chemical-bond lengths with picometer accuracy In the second part of this work, solid-state NMR experiments were combined with quantum-chemical calculations in order to aid and optimise the interpretation of experimental results. The investigations on Calix[4]hydroquinone nanotubes have shown that this combined approach can provide information on the presence of disordered and/or mobile species in supramolecular structures. As a second example, C3-symmetric discs arranging in helical columnar stacks were investigated. In these systems, 1H chemical shifts experience large pi-shifts due to packing effects, which were found to be long-ranged. Moreover, quantum-chemical calculations revealed that helicity in these systems is induced by the propeller-like conformation of the core of the molecules.

Identifizierung, Sequenzierung und Charakterisierung des Dmxl1-Gen in Mus musculus sowie die funktionelle Analyse durch Knock-OutrnrnBei Dmxl1 handelt es sich um ein neuartiges Gen aus Mus musculus. Das ebenfalls in der vorliegenden Arbeit bioinformatisch untersuchte Gen DMXL1 ist das zu Dmxl1 homologe Gen des Menschen. Beide Gene bestehen aus 43 Exons, das murine Dmxl1 codiert für eine mRNA von 10992 bp bzw. 12210 bp, das humane DMXL1 kodiert für eine cDNA von 11082 bp, der offene Leserahmen umfasst bei der Maus 9042 bp. In der Maus konnte ein mögliches alternatives Polyadenylierungssignal identifiziert werden. Zwischen beiden Spezies sind die Exonpositionen und ihre Längen hoch konserviert. Dmxl1 liegt auf dem Crick-Strang von Chromosom 18 Bande C, der translatierte Bereich erstreckt sich auf genomischer Ebene über 129558 bp und die Orientierung verläuft in Richtung Centromer. Dmxl1 und DMXL1 gehören damit zu den größten bekannten Genen in Maus und Mensch. Bei beiden Spezies liegen die DmX-Homologen genomisch innerhalb eines Bereichs der Isochoren-Klasse L1 in einer Gen-armen Region. Die Anzahl der repetitiven Elemente innerhalb der Genregion von Dmxl1 liegt 6% unter dem erwarteten Wert eines L1 Isochors, die Anzahl beim Menschen liegt 4% über dem erwarteten Wert. Um die mögliche Promotorstruktur von Dmxl1 darzustellen, wurden umfangreiche in silico-Analysen der Region um den putativen Transkriptionsstart vorgenommen. Mit Hilfe der gewonnenen Daten konnte ein Transkriptionstartpunkt identifiziert werden. Zudem wurde eine Promotorstruktur erarbeitet, bei der angenommen werden kann, dass sie eine gute Näherung an die tatsächlich vorhandenen Bindungsstellen von Transkriptionsfaktoren darstellt. Die mit bioinformatischen Werkzeugen erzeugte virtuelle Promotor- und Enhancerstruktur zeigt das Potenzial, Dmxl1 basal und ubiquitär zu exprimieren. Gleichzeitig zeigen diese Daten, dass Dmxl1 vermutlich in einigen Geweben der Keimbahn, im Fettgewebe, dem blutbildenen System und während der Embryogenese hochkomplex reguliert werden kann. Eine regulierte Expression zur Steuerung des Energiestoffwechsels ist ebenfalls wahrscheinlich. Diese Ergebnisse passen sehr gut zu den experimentell ermittelten Daten und den beobachteten Phänotypen Dmxl1-chimärer Mäuse.rnDie abgeleitete Aminosäuresequenz umfasst in der Maus 3013 AS, im Menschen 3027 AS, der Vergleich der abgeleiteten Aminosäuresequenzen zeigt eine Identität von 89,3 % und eine Similarität von 94,7 % zwischen beiden Spezies. Im Dmxl1/DMXL1-Protein von Maus und Mensch konnten mindestens 24 und maximal 36 WD-Wiederholungseinheiten identifiziert werden, zudem wurden eine Reihe weiterer konservierter Proteinmotive gefunden. Die in silico-Strukturanalysen beider abgeleiteter Aminosäuresequenzen lässt vermuten, dass sich C- und N-terminal WD-Propellerstrukturen befinden. In dieser Arbeit gelang eine C-terminale Rekonstruktion einer 10-blättrigen Propellerstruktur, denkbar ist jedoch auch eine Struktur mit mindestens drei WD-Propellern, wenn eine prädominante Struktur mit Propellern aus jeweils sieben Propellerblättern angenommen wird.rnDas primäre Ziel dieser Arbeit, die Etablierung einer stabilen Mauslinie mit diruptiertem Dmxl1-Gen konnte aufgrund einer beobachteten Haploinsuffizienz nicht erreicht werden. Trotz zahlreicher Transformationen von Maus-Stammzelllinien konnte letztlich nur eine stabil transformierte Linie mit einem Dmxl1-Null-Allel identifiziert werden, was auch zu den theoretischen Daten und den angenommenen Aufgaben von Dmxl1 als komplex und diffizil reguliertes Multifunktions-Protein passt. Aus der transformierten Mauszelllinie konnten chimäre Mäuse entwickelt werden, die in Abhängigkeit von dem Ausmaß des Chimärismus phänotypisch massive Schädigungen aufwiesen. Neben einer Teilsterilität wurden massive Fettleibigkeit und ein ausgeprägter Hypogonadismus beobachtet. Keines der Tiere war in der Lage das Dmxl1-Null-Allel zu transduzieren. Die Tiere waren nur sehr eingeschränkt fertil, die wenigen Nachkommen entsprachen genotypisch und phänotypisch ausschließlich den verwendeten Blastocysten.rn

Das Ziel der vorliegenden Arbeit waren die Synthese und Untersuchung von Modellverbindungen zur Sauerstoffaktivierung auf der Basis neuer Ligandensysteme des 1,3,4-Thiadiazols unter Ausarbeitung einer Synthesestrategie zur Derivatisierung der heteroaromatischen 1,3,4-Thiadiazol-Liganden, deren Koordinationsverhalten in Abhängigkeit ihres 2,5-Substitutionsmusters untersucht wurde, sowie die fortführende Bearbeitung bereits bekannter Ligandensysteme zur Erzeugung von homo- und heterovalenten Übergangsmetallkomplexverbindungen.rnDie unter der Verwendung der modifizierten Liganden TPDE, H1TPDP und H1BPMP resultierenden dinuklearen Komplexverbindungen zeigen unterschiedlich starke antiferromagnetische Wechselwirkungen in Abhängigkeit der vorhandenen Brückenliganden. In der Verbindung [Fe6O2(OH)(L´)2(OOCMe3)9(OEt)2] trat eine Fragmentierung des Liganden H1TPDP auf. Das cisoide Ligandensubstitutionsmuster der entstandenen sechskernigen Verbindung ist verantwortlich für die interessanten magnetischen Eigenschaften des Komplexes. rnNeue Perspektiven zur Erzeugung von Modellverbindungen zur Sauerstoffaktivierung wurden mit dem Mono-Chelatliganden H1ETHP und den Bis-Chelatliganden HL2H, H2L2H und H2BATP aufgezeigt. Die Umsetzung von H1ETHP mit verschiedenen Übergangsmetallsalzen resultierte für die Metalle Cr(III), Fe(III), Co(III) und Ni(II) in mononuklearen Verbindungen des Typs [M(ETHP)2]X (X = ClO4, FeCl4, OMe, Cl, Br) sowie in zwei tetranuklearen Verbindungen mit Mn(II) und Cu(II). [Mn4(ETHP)6] besitzt ein propellerförmiges, planares [Mn4O6]2+-System mit einen Spingrundzustand von S = 5. In allen Verbindungen von H1ETHP konnte eine mono-κN-Koordination des 1,3,4-Thiadiazol-Rückgrates über eines seiner beiden endozyklischen Stickstoffdonoratome beobachtet werden. rnAus Umsetzungen der Bis-Chelatliganden wurden fast ausschließlich polynukleare Übergangsmetallkomplexe erhalten. Insbesondere der Ligand H2L2H zeigt eine ausgeprägte Tendenz zur Ausbildung trinuklearer, linearer Komplexe, welche auf Grund ihrer ungeraden Anzahl von Übergangsmetallionen einen Spingrundzustand S ≠ 0 aufweisen.rn Die mit dem Liganden HL2H erhaltenen Verbindungen unterstreichen die hohe Flexibilität dieser Systeme hinsichtlich der Erzeugung polynuklearer und heterovalenter Komplexverbindungen. So konnten in Abhängigkeit vom verwendeten Übergangsmetallsalz trinukleare, pentanukleare, aber auch hepta- und oktanukleare Verbindungen synthetisiert werden. Insbesondere die Komplexe des Mangans und des Cobalts zeigen ein heterovalentes [MnIIMnIII4]- bzw. [CoII2CoIII3]-Motiv, was sich in Spingrundzuständen von S ≠ 0 äußert. Der diamagnetische, achtkernige Fe8-Cluster besitzt eine pseudo C3-symmetrische Anordnung der Metall-Zentren, während für die heptanukleare Cu7-Kette durch ihre stark unterschiedlichen Kupfer-Koordinationsgeometrien interessante magnetische Austauschwechselwirkungen beobachtet werden konnten. Der dreikernige µ3-oxo-verbrückte Komplex des Liganden H2BATP zeigt als interessante strukturelle Eigenschaft ein ein µ3-Verbrückungsmuster des eingesetzten Sulfat-Anions. rnIn allen Komplexen der Bis-Chelatliganden HL2H, H2L2H und H2BATP konnte ein µ2-κN,κN-Koordiantionsmodus des 1,3,4-Thiadiazols und somit eine Abhängigkeit der Verbrückung vom Ligandensubstitutionsmuster beobachtet werden.rn

The morbilliviruses measles virus (MeV) and canine distemper virus (CDV) both rely on two surface glycoproteins, the attachment (H) and fusion proteins, to promote fusion activity for viral cell entry. Growing evidence suggests that morbilliviruses infect multiple cell types by binding to distinct host cell surface receptors. Currently, the only known in vivo receptor used by morbilliviruses is CD150/SLAM, a molecule expressed in certain immune cells. Here we investigated the usage of multiple receptors by the highly virulent and demyelinating CDV strain A75/17. We based our study on the assumption that CDV-H may interact with receptors similar to those for MeV, and we conducted systematic alanine-scanning mutagenesis on CDV-H throughout one side of the beta-propeller documented in MeV-H to contain multiple receptor-binding sites. Functional and biochemical assays performed with SLAM-expressing cells and primary canine epithelial keratinocytes identified 11 residues mutation of which selectively abrogated fusion in keratinocytes. Among these, four were identical to amino acids identified in MeV-H as residues contacting a putative receptor expressed in polarized epithelial cells. Strikingly, when mapped on a CDV-H structural model, all residues clustered in or around a recessed groove located on one side of CDV-H. In contrast, reported CDV-H mutants with SLAM-dependent fusion deficiencies were characterized by additional impairments to the promotion of fusion in keratinocytes. Furthermore, upon transfer of residues that selectively impaired fusion induction in keratinocytes into the CDV-H of the vaccine strain, fusion remained largely unaltered. Taken together, our results suggest that a restricted region on one side of CDV-H contains distinct and overlapping sites that control functional interaction with multiple receptors.

Morbillivirus cell entry is controlled by hemagglutinin (H), an envelope-anchored viral glycoprotein determining interaction with multiple host cell surface receptors. Subsequent to virus-receptor attachment, H is thought to transduce a signal triggering the viral fusion glycoprotein, which in turn drives virus-cell fusion activity. Cell entry through the universal morbillivirus receptor CD150/SLAM was reported to depend on two nearby microdomains located within the hemagglutinin. Here, we provide evidence that three key residues in the virulent canine distemper virus A75/17 H protein (Y525, D526, and R529), clustering at the rim of a large recessed groove created by beta-propeller blades 4 and 5, control SLAM-binding activity without drastically modulating protein surface expression or SLAM-independent F triggering.
