Xylanases with hydrolytic activity on xylan, one of the hemicellulosic materials present in plant cell walls, have been identified long back and the applicability of this enzyme is constantly growing. All these applications especially the pulp and paper industries require novel enzymes. There has been lot of documentation on microbial xylanases, however, none meeting all the required characteristics. The characters being sought are: higher production, higher pH and temperature optima, good stabilities under these conditions and finally the low associated cellulase and protease production. The present study analyses various facets of xylanase biotechnology giving emphasis on bacterial xylanases. Fungal xylanases are having problems like low pH values for both enzyme activity and growth. Moreover, the associated production of cellulases at significant levels make fungal xylanases less suitable for application in paper and pulp industries.Bacillus SSP-34 selected from 200 isolates was clearly having xylan catabolizing nature distinct from earlier reports. The stabilities at higher temperatures and pH values along with the optimum conditions for pH and temperature is rendering Bacillus SSP-34 xylanase more suitable than many of the previous reports for application in pulp and paper industries.Bacillus SSP-34 is an alkalophilic thertmotolerant bacteria which under optimal cultural conditions as mentioned earlier, can produce 2.5 times more xylanase than the basal medium. The 0.5% xylan concentration in the medium was found to the best carbon source resulting in 366 IU/ml of xylanase activity. This induction was subjected to catabolite repression by glucose. Xylose was a good inducer for xylanase production. The combination of yeast extract and peptone selected from several nitrogen sources resulted in the highest enzyme production (379+-0.2 IU/ml) at the optimum final concentration of 0.5%. All the cultural and nutritional parameters were compiled and comparative study showed that the modified medium resulted in xylanase activity of 506 IU/ml, 5 folds higher than the basal medium. The novel combination of purification techniques like ultrafiltraton, ammonium sulphate fractionation, DEAE Sepharose anion exchange chromatography, CM Sephadex cation exchange chromatography and Gel permeation chromatography resulted in the purified xylanase having a specific activity of 1723 U/mg protein with 33.3% yield. The enzyme was having a molecular weight of 20-22 kDa. The Km of the purified xylanase was 6.5 mg of oat spelts xylan per ml and Vmax 1233 µ mol/min/mg protein.Bacillus SSP-34 xylanase resulted in the ISO brightness increase from 41.1% to 48.5%. The hydrolytic nature of the xylanase was in the endo-form.Thus the organism Bacillus SSP-34 was having interesting biotechnological and physiological aspects. The SSP-34 xylanase having desired characters seems to be suited for application in paper and pulp industries.

The Sultanate of Oman is located on the south-eastern coast of the Arabian Peninsula, which lies on the south-western tip of the Asian continent. The strategic geographical locations of the Sultanate with its many maritime ports distributed on the Indian Ocean have historically made it one of the Arabian Peninsula leaders in the international maritime trade sector. Intensive trading relationships over long time periods have contributed to the high plant diversity seen in Oman where agricultural production depends entirely on irrigation from groundwater sources. As a consequence of the expansion of the irrigated area, groundwater depletion has increased, leading to the intrusion of seawater into freshwater aquifers. This phenomenon has caused water and soil salinity problems in large parts of the Al-Batinah governorate of Oman and threatens cultivated crops, including banana (Musa spp.). According to the Ministry of Agriculture and Fisheries, the majority of South Al-Batinah farms are affected by salinity (ECe > 4 dS m-1). As no alternative farmland is available, the reclamation of salt-affected soils using simple cultural practices is of paramount importance, but in Oman little scientific research has been conducted to develop such methods of reclamation. This doctoral study was initiated to help filling this research gap, particularly for bananas. A literature review of the banana cultivation history revealed that the banana germplasm on the Arabian Peninsula is probably introduced from Indonesia and India via maritime routes across the Indian Ocean and the Red Sea. In a second part of this dissertation, two experiments are described. A laboratory trial conducted at the University of Kassel, in Witzenhausen, Germany from June to July 2010. This incubation experiment was done to explore how C and N mineralization of composted dairy manure and date palm straw differed in alkaline non-saline and saline soils. Each soil was amended with four organic fertilizers: 1) composted dairy manure, 2) manure + 10% date palm straw, 3) manure + 30% date palm straw or 4) date palm straw alone, in addition to un-amended soils as control. The results showed that the saline soil had a lower soil organic C content and microbial biomass C than the non-saline soil. This led to lower mineralization rates of manure and date palm straw in the saline soil. In the non-saline soil, the application of manure and straw resulted in significant increases of CO2 emissions, equivalent to 2.5 and 30% of the added C, respectively. In the non-amended control treatment of the saline soil, the sum of CO2-C reached only 55% of the soil organic C in comparison with the non-saline soil. In which 66% of the added manure and 75% of the added straw were emitted, assuming that no interactions occurred between soil organic C, manure C and straw C during microbial decomposition. The application of straw always led to a net N immobilization compared to the control. Salinity had no specific effect on N mineralization as indicated by the CO2-C to Nmin ratio of soil organic matter and manure. However, N immobilization was markedly stronger in the saline soil. Date palm straw strongly promoted saprotrophic fungi in contrast to manure and the combined application of manure and date palm straw had synergistic positive effects on soil microorganisms. In the last week of incubation, net-N mineralization was observed in nearly all treatments. The strongest increase in microbial biomass C was observed in the manure + straw treatment. In both soils, manure had no effect on the fungi-specific membrane component ergosterol. In contrast, the application of straw resulted in strong increases of the ergosterol content. A field experiment was conducted on two adjacent fields at the Agricultural Research Station, Rumais (23°41’15” N, 57°59’1” E) in the South of Al-Batinah Plain in Oman from October 2007 to July 2009. In this experiment, the effects of 24 soil and fertilizer treatments on the growth and productivity of Musa AAA cv. 'Malindi' were evaluated. The treatments consisted of two soil types (saline and amended non-saline), two fertilizer application methods (mixed and ring applied), six fertilizer amendments (1: fresh dairy manure, 2: composted dairy manure, 3: composted dairy manure and 10% date palm straw, 4: composted dairy manure and 30% date palm straw, 5: only NPK, and 6: NPK and micronutrients). Sandy loam soil was imported from another part of Oman to amended the soil in the planting holes and create non-saline conditions in the root-zone. The results indicate that replacing the saline soil in the root zone by non-saline soil improved plant growth and yield more than fertilizer amendments or application methods. Particularly those plants on amended soil where NPK was applied using the ring method and which received micronutrients grew significantly faster to harvest (339 days), had a higher average bunch weight (9.5 kg/bunch) and were consequently more productive (10.6 tonnes/hectare/cycle) compared to the other treatments.

The Palma Project is an experiment in the use of cultural identity as a social trigger to address ecological degradation. The research methodology draws from environmental, social and urban analyses to unveil the best strategy to address the ecological, river restoration and water treatment challenges in Berkeley, California’s “Sister City” in southeast Cuba, Palma Soriano. The objective is to provide a better quality of life and to create new opportunities for the local community to reconnect with natural cycles of water and the cultivation of their own land. The project aim is to promote the strength and capacity of local communities to protect their own environment based upon a master plan, which includes natural wastewater treatment, reforestation, urban agriculture and the facilitation and utilization of a public space bordering the major river which flows by Palma Soriano, the Cauto. This project will contribute and produce healthy water recycling for Palma, provide a potable water source for the city, encourage ecological restoration of the riparian zone of the Cauto, and provide new opportunities for food production. It is designed to preserve the cultural identity of the local community, and to restore the essential balance between the community’s need to sustain both itself and the natural environment.

This exploratory study evaluated biophysical, cultural and socio-economic factors affecting crop production and land utilisation in the Nkonkobe Municipality, South Africa. The study sought to establish what farmers in the area perceive as serious threats to crop production, drivers for land abandonment, and how best current agricultural production could be intensified. The farmers’ perspectives were assessed through interviews using semi-structured and open-ended questionnaires. The results of the study revealed declining crop productivity and increase in land abandonment in the Municipality. The biophysical drivers of land abandonment were low and erratic rainfall and land degradation while the socio-economic drivers were labour shortages due to old age and youth movement to cities, lack of farming equipment and security concerns. The most abandoned crops were maize, sorghum and wheat. This trend was attributed to the labour intensiveness of cereal production and a shift in dietary preference to purchased rice. These findings should be factored in any programmes that seek to increase land utilisation and crop productivity in the Municipality.

Using the case of an economically declined neighbourhood in the post-industrial German Ruhr Area (sometimes characterized as Germany’s “Rust Belt”), we analyse, describe and conclude how urban agriculture can be used as a catalyst to stimulate and support urban renewal and regeneration, especially from a socio-cultural perspective. Using the methodological framework of participatory action research, and linking bottom-up and top-down planning approaches, a project path was developed to include the population affected and foster individual responsibility for their district, as well as to strengthen inhabitants and stakeholder groups in a permanent collective stewardship for the individual forms of urban agriculture developed and implemented. On a more abstract level, the research carried out can be characterized as a form of action research with an intended transgression of the boundaries between research, planning, design, and implementation. We conclude that by synchronously combining those four domains with intense feedback loops, synergies for the academic knowledge on the potential performance of urban agriculture in terms of sustainable development, as well as the benefits for the case-study area and the interests of individual urban gardeners can be achieved.

dTwo genetic constructs used to confer improved agronomic characteristics, namely herbicide tolerance (HT) in maize and soyabean and insect resistance (Bt) in maize, are considered in respect of feeding to farm livestock, animal performance and the nutritional value and safety of animal products. A review of nucleic acid (DNA) and protein digestion in farm livestock concludes that the frequency of intact transgenic DNA and proteins of GM and non-GM crops being absorbed is minimal/non existent, although there is some evidence of the presence of short fragments of rubisco DNA of non-GM soya in animal tissues. It has been established that feed processing (especially heat) prior to feeding causes significant disruption of plant DNA. Studies with ruminant and non-ruminant farm livestock offered GM feeds demonstrated that animal performance and product composition are unaffected and that there is no evidence of transgenic DNA or proteins of current GM in the products of animals consuming such feeds. On this evidence, current HT and Bt constructs represent no threat to the health of animals, or humans consuming the products of such animals. However as new GM constructs become available it will be necessary to subject these to rigorous evaluation.

This article presents an ongoing study of educational policy enactment in Singapore lower primary English classrooms. It explores how teachers react to and interpret educational reforms in their classroom practices against a backdrop ofvtraditional cultural values. Using a prescribed coding scheme, the article presents the instructional organisational patterns and participation structures of the lessons.Through a systematic analysis of the enacted curricula, the paper examines classroom practices as well as teaching and learning activities in Primary 1 (7–8 years) and Primary 2 (8–9 years) English lessons in Singapore. The results suggest that there are cultural clashes between major educational reforms which emphasise independent/critical thinking and ‘Asian values’ which promote respect for authority and conformity.
