A sensitive and simple system was proposed for the in situ measurement of total aldehyde in outdoor or indoor ambient. The method is based on the use of a reagent drop as an useful interface to preconcentrate the sample prior to determination of total aldehyde as formaldehyde. The drop is formed at the tip of a cylindrical tube that contains two optical fibers placed on opposite sides and in contact with the reagent solution. One optical fiber carries the red light to the drop form a light emitting diode (LED). The transmitted light is measured by a second optical fiber/photodiode system. The analytical signal is read and converted into absorbance. The reagent solution of 3-methyl-2-benzothiazoline hydrazone (MBTH) forms a blue cation during reaction with formaldehyde that can be measured at 660 nm. Some aspects of kinetics reaction formation of dye were reevaluated. The formaldehyde reacts with MBTH and forms the azine in about 12 min. The oxidation of MBTH by Fe (III) and the formation of dye requires 3 min. The absorbance of the reagent drop is proportional to the sampling time and to the analyte concentration. The absorbance signal increases with increased sample gas flow until a maximum is reached then decreases until it forms a plateau. The proposed method was evaluated using both outdoor and indoor samples, and it was shown to viable provide an accurate measure of total aldehyde.

A simple device for glass plate drilling, which is an important step in microfabrication procedures, is described. A reservoir of concentrated hydrofluoric acid with a hole in its bottom is affixed against the glass plate. Leakage is prevented by using a neoprene O-ring. A plastic pipet tip inserted in the reservoir, close to the corrosion region, provides forced convection by pressure variations inside it. A device to make 5 simultaneous holes in a plate is also presented. For a 140 µm thickness alkaline glass lamina and 1/8" O-ring, 5 holes are drilled in 20 min.

Microcystins are non-ribosomal peptides that must be detected for its health concern. Here, microcystin LR and its specific antibody were respectively tethered to the substrate and to the tip of an atomic force microscope, after surface functionalization using 3-aminopropyltriethoxysilane and glutaraldehyde. Functionalization was confirmed comparing topographic images taken on bare and modified tips. Force versus distance curves were successfully used to measure the specific antibody-antigen interactions comparing with a control in which microcystin was initially blocked by incubation with free antibodies. The results showed unequivocally the specific recognition of MLR, suggesting that this method could be useful for biosensor development.

Clonal cleaning, followed by pre-immunization with protective complexes of Citrus tristeza virus(CTV), allowed the commercial cultivation of Pêra sweet orange, a variety that has great importance for Brazilian citriculture but is sensitive to the virus. The use of mild protective isolates in other citrus varieties, even those more tolerant to CTV, can also be of interest to prevent the spread of severe isolates. The aim of this study was to characterize, by means of SSCP (Single Strand Conformational Polymorphism) analysis of the coat protein gene, CTV isolates present in plants of the sweet orange cultivars Pêra, Hamlin and Valencia propagated from four budwood sources: 1) old lines, 2) nucellar lines, 3) shoot-tip-grafted lines, and 4) shoot-tip-grafted lines pre-immunized with the mild CTV protective isolate 'PIAC'. We also evaluated the correlation of the obtained SSCP patterns to stem pitting intensity, tree vigor and fruit yield. SSCP results showed low genetic diversity among the isolates present in different trees of the same variety and same budwood source and, in some cases, in different budwood sources and varieties. Considering tristeza symptoms, lower intensity was noted for plants of new, shoot-tip-grafted and pre-immunized shoot-tip-grafted lines, compared to old lines of the three varieties. The observed SSCP patterns and symptomatology suggested that more severe CTV complexes infect the plants of old lines of all three varieties. The protective complex stability was observed in the SSCP patterns of CTV isolates of some shoot-tip-grafted and pre-immunized clones. It was concluded that the changes detected in other electrophoretic profiles of this treatment did not cause loss of the protective capacity of CTV isolate 'PIAC' inoculated in the pre-immunization.

Este trabalho trata da importância do setor florestal para o desenvolvimento socioeconômico brasileiro por meio dos modelos econômicos de equilíbrio geral multissetoriais, através das análises das matrizes de insumo-produto (MIP) e de contabilidade social (MCS). As principais fontes dos dados foram as Tabelas de Insumo-Produto (TIP) do Instituto Brasileiro de Geografia e Estatística (IBGE), além de informações obtidas no Banco Nacional de Desenvolvimento Econômico e Social (BNDES), no Banco Central (BACEN) e na Secretaria da Receita Federal (SRF), referentes ao ano de 1995. Observou-se que tão importante quanto contribuir para a geração e formação do PIB, de emprego, impostos, salários e balança comercial é o poder que um setor produtivo tem a mais que o outro de multiplicar estes indicadores quando uma unidade monetária é gasta a mais pelos consumidores finais na demanda por produtos deste determinado setor. Os resultados indicaram que o setor florestal é um dos setores da economia brasileira que apresentou os maiores efeitos multiplicadores para os indicadores socioeconômicos, sendo superiores aos de outros como o da indústria automobilística, de equipamentos elétricos e eletrônicos, de máquinas e equipamentos e de produtos químicos e petróleo.

ABSTRACTThe study was conducted with shoot tip explants of neem (Azadirachta indica A. Juss) to identify a viable regenerative process. Shoot tips were obtained from neem embryos cultured alternatingly in DKW medium supplemented with BAP and medium without hormones. Initial shoot development was influenced by cotyledon presence. Basal callus, excised from in vitro stem base, also presented organogenic potential. In some cases, plant lines, obtained from each seed, presented different characteristics. The most common characteristic observed in vitro was callus formation at the stem base. However, the rarest characteristics were stem callus formation and leaf senescence. The regenerated shoot tips were further subculture and rooted on a medium supplemented with IBA so that complete plants could be obtained. The rooted plants were transplanted to a greenhouse and successfully acclimatized. No significant differences in in vivo development were observed between neem plants from callus and from shoot tip propagation.

The furrow openers for no-till system should be easy to penetrate the soil, generate low levels of soil mobilization and require a little traction force. Thus, the aim of this research was to compare six hoe-type furrow openers, four of industrial manufacturing and two handcrafted models, which were used in planters in the region of Pelotas, Brazil. The parameters for comparison among these furrow openers were the horizontal (Fh) and vertical (Fv) forces acting on the tip of the hoe and the cross-sectional area mobilized in the furrow. The experimental design was randomized, with six treatments (furrow openers) from T1 to T6, with four repetitions, constituted by their passage through plots of 20 x 3 m. The force data were collected by load cells and a signal conditioner. The determination of the mobilized area in the furrow was done by a micro soil profilometer. It was concluded that the furrow openers T1, T4 and T6 presented the lowest values of horizontal force (statistically equal and ranging between 1,034 and 1,230 N) and that T1 and T2 produced the highest vertical forces downwards (statistically identical and varying between 749 and 845 N). The furrow openers T1, T2, T4, T5 and T6 generated soil mobilization statistically equal and between 0.006993 and 0.008933 m².

Tissues from cattle, mice, rats and guinea pigs experimentally intoxicated by Riedeliella graciliflora were studied histologically. Cattle lymph nodes, spleen, Peyer patches and peribronchial lymphoid tissues had diffuse necrosis of lymphocytes, mainly in the germinal centers of the follicles. This lesion was less severe in laboratory animals. All species had severe enteritis with infiltration of the lamina propria by mononuclear cells. Some cells in this infiltrate were necrotic. Degeneration and necrosis of the epithelial cells, mainly in the tip of the villi, and detachment of the epithelial lining from the lamina propria were also observed. In the liver the trabecular structure was disrupted and the hepatocytes had some degree of individual necrosis and degeneration. A tubular nephrosis was observed in the kidneys. Liver, lung, kidney, intestine and lymph nodes had different degrees of congestion. Those lesions are similar than those caused by Polygala klotzschii, a plant that contains 5-metoxi-podophyllotoxin.

Aspectos relacionados à transferência de imunidade passiva (TIP) foram estudados em 290 cordeiros recém-nascidos, cruzas Texel, manejados extensivamente em contato permanente com as suas mães, e criados em clima tropical. Amostras de sangue foram colhidas dos cordeiros uma única vez entre 24 e 36 horas de vida para obtenção do soro sanguíneo. Foram determinadas a atividade da gama glutamiltransferase e as concentrações de proteínas totais por refratometria e colorimetria, de albumina, alfa, beta e gamaglobulinas por eletroforese e de IgG estimada pela turvação pelo sulfato de zinco. Os cordeiros foram agrupados de acordo com o sexo, o número de partos das ovelhas, o escore de condição corporal (ECC) da ovelha, o número de cordeiros nascidos, o peso e a vitalidade ao nascimento. Os fatores de risco para a ocorrência de falha na transferência de imunidade passiva (FTIP) foram identificados e a sua associação com a mortalidade de cordeiros foi testada. Foram estabelecidas correlações entre as variáveis estudadas. A TIP não foi influenciada pelo sexo, pelo número de partos ou pelo ECC das ovelhas. Houve diferenças entre os cordeiros únicos e gêmeos e entre os que nasceram com peso >3 kg e < 3 kg. A FTIP foi pouco frequente (12,4%), mais provável em gêmeos (p=0,026) e em cordeiros leves (p<0,001) e esteve fortemente associada com a mortalidade dos cordeiros até os 60 dias de vida (p<0,001). A taxa de mortalidade geral foi de 11,3%; e 30,5% dos cordeiros com FTIP morreram, quase todos no primeiro mês de vida. A concentração de proteína total determinada por refratometria se correlacionou (p<0,001) com as concentrações de gamaglobulinas (r=0,816) e de IgG (r=0,810) e o valor crítico < 5,0g/dL para a proteína total pode ser admitido como indicador de FTIP. Sob condições de clima tropical a FTIP pode ser considerada pouco frequente em cordeiros mestiços de corte, entretanto, a vigilância e os cuidados devem ser intensificados no caso do nascimento de cordeiros gêmeos com peso reduzido.

Most descriptions of the ostrich oropharynx and oesophagus are superficial and supply little meaningful morphological data. The aim of this investigation is describe the ostrich oropharingeal cavity, in order to supply the deficiency of macroscopic data about this important animal. Five heads of 12 to 14-month-old ostriches of either sex were anatomically dissected to expose the oropharynx. The ostrich oropharynx was "bell-shaped" composed by the maxillary and mandibular ramphoteca. The roof and floor presented two distinct regions different in colour of the mucosa. The rostral region was pale pink contrasting to creamy-pink coloured caudal region. The median longitudinal ridge extended rostrally from the apex of the choana to the tip of the beak in the roof and it is clearly more prominent and rigid than the homolog in the floor that appeared thin and stretched rostrally, continuing caudally surrounding the tongue and the laryngeal mound eventually merging with the oesophageal mucosa. The floor was formed by the interramal region, tongue and laryngeal mound containing shield-shaped glottis. It can be concluded that the present study, in addition to confirming the basic features of the oropharynx previously described for the ostrich, clarified the contradictory information presented in the literature and also provided new, unreported morphological data, some of which may be important when studying nutrition and health in these birds.

The rhea (Rhea americana americana) is an american bird belonging to Ratite's family. Studies related to its morphology are still scarce. This study aims to describe the macroscopic structures of the oropharyngeal cavity. Five heads (2 to 6 months old) formalin preserved were anatomically dissected to expose the oropharynx. The oropharynx of the rhea was "bell-shaped" composed by the maxillary and mandibular rhamphotheca. The roof and floor presented two distinct regions different in colour of the mucosa. The rostral region was pale pink contrasting to grey coloured caudal region. The median longitudinal ridge extended rostrally from the apex of the choana to the tip of the beak in the roof and it is clearly more prominent and rigid than the homolog in the floor that appeared thin and stretched merely along the rostral portion of the regio interramalis. The floor was formed by the interramal region, (regio interramalis) tongue and laryngeal mound containing glove-shaped glottis. This study confirmed the basic morphology of the oropharinx of the rhea. However, important morphological information not previously described is highlighted and contradictory information present in the literature is clarified.

ABSTRACTInadequate herbicide application can result in failures in weed control and/or poisoning of the crops, resulting in yield losses. In this research were assessed the effects of the sprayer nozzle boom height in the distribution of the spray solution for weed control, influencing intoxication of beans and crop yield. Experiments were conducted in laboratory and field conditions. In laboratory, the performance of flat spray tip TT 11002 was assessed at heights 0.20, 0.30, 0.40 and 0.50 meters with respect to the target surface. In the field the same heights were assessed in applications of herbicides fomesafen, fluazifop-P-butyl and fomesafen + fluazifop-P-butyl. There was an inverse relationship between the height of the spray boom and the coefficients of variation of the patterns. The mixture better efficiency in a tank of fluazifop-P-butyl + fomesafen was obtained with the height of 0.50 m from the target. This treatment resulted in better weed control, lower poisoning of the bean plants and better crop yield rates.

Karyotypes of 14 populations including eight species of the genus Lobelia were studied using root tip mitotic metaphases. All populations were tetraploid with 2n = 28 chromosomes. The chromosome base number x = 7 was confirmed for the genus. Karyotype analysis showed that chromosome size varied from 1.05 µm to 2.02 µm with predominance of M and SM chromosome types. The karyotypes were similar among themselves with small intra- and interspecific variations on the size of haploid sets, symmetry indexes and centromere position of some chromosome pairs. These results showed that karyotypes of Brazilian lobelias of the subgenus Tupa were probably due to polyploidy associated with chromosomal rearrangements probably in small chromatin segments.

Karyotypes of six species of the genus Stevia from Southern Brazil were studied, utilizing root tip metaphases. All species were diploid with 2n = 22 chromosomes. It was possible to identify each species by chromosome morphology. The basic chromosome number for Brazilian species of Stevia is X = 11. This number is also found in almost all South American species. We suggest that in Stevia there is an evolutionary trend toward chromosomal rearrangement, caused mainly by pericentric inversions. It was found that, in addition to aneuploidy and polyploidy, chromosomal rearrangements are common in the tribe Eupatorieae.

The effects of dorsomedial hypothalamic (DMH) nucleus lesion on body weight, plasma glucose levels, and the gastric emptying of a liquid meal were investigated in male Wistar rats (170-250 g). DMH lesions were produced stereotaxically by delivering a 2.0-mA current for 20 s through nichrome electrodes (0.3-mm tip exposure). In a second set of experiments, the DMH and the ventromedial hypothalamic (VMH) nucleus were lesioned with a 1.0-mA current for 10 s (0.1-mm tip exposure). The medial hypothalamus (MH) was also lesioned separately using a nichrome electrode (0.3-mm tip exposure) with a 2.0-mA current for 20 s. Gastric emptying was measured following the orogastric infusion of a liquid test meal consisting of physiological saline (0.9% NaCl, w/v) plus phenol red dye (6 mg/dl) as a marker. Plasma glucose levels were determined after an 18-h fast before the lesion and on the 7th and 15th postoperative day. Body weight was determined before lesioning and before sacrificing the rats. The DMH-lesioned rats showed a significantly faster (P<0.05) gastric emptying (24.7% gastric retention, N = 11) than control (33.0% gastric retention, N = 8) and sham-lesioned (33.5% gastric retention, N = 12) rats, with a transient hypoglycemia on the 7th postoperative day which returned to normal by the 15th postoperative day. In all cases, weight gain was slower among lesioned rats. Additional experiments using a smaller current to induce lesions confirmed that DMH-lesioned rats had a faster gastric emptying (25.1% gastric retention, N = 7) than control (33.4% gastric retention, N = 17) and VMH-lesioned (34.6% gastric retention, N = 7) rats. MH lesions resulted in an even slower gastric emptying (43.7% gastric retention, N = 7) than in the latter two groups. We conclude that although DMH lesions reduce weight gain, they do not produce consistent changes in plasma glucose levels. These lesions also promote faster gastric emptying of an inert liquid meal, thus suggesting a role for the DMH in the regulation of gastric motility
