Biosensors were developed by immobilization of gilo (Solanum gilo) enzymatic extract on chitosan biopolymers using three different procedures: glutaraldehyde, carbodiimide/glutaraldehyde and epichlorohydrin/glutaraldehyde. The best biosensor performance was obtained after the immobilization of peroxidase on chitosan with epichlorohydrin/glutaraldehyde. Linear analytical curves for hydroquinone concentrations from 2.5x10-4 to 4.5x10-3 mol L-1 with a detection limit of 2.0x10-6 mol L-1 and recovery of hydroquinone ranging from 95.1 to 105% were obtained. The relative standard deviation was < 1.0 % for a solution of 3.0x10-4 mol L-1 hydroquinone and 2.0x10-3 mol L-1 hydrogen peroxide in 0.1 mol L-1 phosphate buffer solution at pH 7.0 (n=8). The lifetime of this biosensor was 6 months (at least 300 determinations).

Electrochemical sensors have attracted considerable attention in recent years because they provide data about the chemical state of our surroundings and the dynamics of the chemical transformations in the form a spatially resolved image. Particular interest has been directed to measurements in restricted-volume samples as new technologies enable the fabrication of miniaturized versions of sensors with reproducible characteristics. Taking these aspects into consideration, this review focuses on the use of electrodes of micrometer dimensions to acquire chemical information in microdomains in which concentrations may not be spatially homogeneous. This is possible because microelectrodes allow fast-response measurements with micrometer resolution to be performed. On the other hand, the use of microelectrodes as amperometric sensors presents an inherent drawback owing to the insufficient specificity toward the substrate of interest. Hence, some comments on strategies to enhance the selectivity of amperometric sensors are also made. Finally, recent applications of structurally microscopic electrodes as in vivo sensors are shown, as well as a prospect of the future trend in this field.

Initially, all major factors that affect the rate of the AldH-catalyzed reaction (enzyme concentration, substrate concentration, temperature and pH) were investigated. Optimal activity was observed between pH values of 7.5 and 9.5 in the temperature range of 25 to 50 ºC. Kinetic parameters, such as Km (2.92 µmol L-1) and Vmax (1.33 10-2 µmol min-1) demonstrate a strong enzyme-substrate affinity. The sensors were based on screen-printed electrodes modified with the Meldola Blue-Reinecke salt (MBRS) combination. Operational conditions (NAD+ and substrate contents, enzyme loading and response time) were optimized. Also, two enzyme immobilization procedures were tested: entrapment in poly(vinyl alcohol) bearing styrylpyridinium groups (PVA-SbQ) and crosslinking with glutaraldehyde. Chronoamperometry was employed to observe the biosensor responses during enzymatic hydrolysis of propionaldehyde and also to construct inhibition curves with maneb and zineb fungicides. Best results were found with the following conditions: [NAD+] = 0.25 mmol L-1; [propionaldehyde] = 80 µmol L-1; enzyme loading = 0.8 U per electrode; response time = 10 min, and inhibition time = 10 min. Current intensities around 103 ± 13 nA with the sensors and good stability was obtained for both immobilization procedures. Detection limits, calculated using 10% inhibition were 31.5 µg L-1 and 35 µg L-1 for maneb and zineb, respectively. Results obtained with other MBRS-modified electrodes consisting of mono and bi-enzymic sensors were compared. The ability to catalyze NADH oxidation by MB was also highlighted.

This work presents a new approach to control the flow rate in hydrodynamic flow experiments. The combination of air pressure generated by an aquarium air pump and the pressure generated by a water column were used for this purpose. This device supports a stable flow rate without pulsation for a long period of time. Furthermore, the flow rate can be easily controlled at various values in one or more streams. The performance of this approach was investigated using Fe(CN)6(4-) solutions in flowing systems using amperometric and voltammetric detection in wall-jet configuration. The results showed that the performance of the proposed device was better than a commercial peristaltic pump. It suggests that this approach can be used successfully in flow analysis systems.

Fast, selective, reproducible and reliable detections have been carried out by using enzymatic biosensors in several areas. The enzymatic biosensors based on the inhibition represent an important role in analytical chemistry. Enzymes like cholinesterases, peroxidases, tyrosinases, etc. have been immobilized on electrochemical and optical transducers and the enzymatic activity decreasing in the presence of the inhibitor is related with its concentrations. This article presents a review on the enzymes used on the construction of these sensors, emphasizing the respective applications.

Since 2000, spore dosimetry and spectral photometry have been performed in parallel at the Southern Space Observatory, São Martinho da Serra (Southern Brazil). A comparative study involving data from Punta Arenas - Chile (53.2º S), São Martinho da Serra (29.5º S), Padang - Indonesia (0.9ºS), Brussels - Belgium (50.9º N) and Kiyotake - Japan (31.9º N) from 2000 to 2006 is presented. The Spore Inactivation Doses presented the higher values in summer (973 ± 73 for Punta Arenas and 4,369 ± 202 for São Martinho da Serra, as well 1,402 ± 170 and 3,400 ± 1,674 for Brussels and Kiyotake, respectively). The simplicity, robustness and high resistance of bacterial spores makes the biosensor an potential biological tool for UV-B monitoring.

Biosensors based on laccase immobilized on microparticles of chitosan crosslinked with tripolyphosphate (biosensor I) and glyoxal (biosensor II) obtained by spray drying for the determinations of rutin in pharmaceutical formulations were developed. Under optimized operational conditions (pH 4.0, frequency of 30 Hz, pulse amplitude of 40 mV and scan increment of 2.0 mV) two analytical curves were obtained for both biosensors showing a detection limit of 6.2x10-8 mol L-1 for biosensor (I) and 2.0x10-8 mol L-1 for biosensor (II). The recovery of rutin from pharmaceutical sample ranged from 90.7 to 105.0% and the lifetime of these biosensors were 4 months (at least 400 determinations).

Studies about the inorganic nanoparticles applying for non-viral release of biological and therapeutic species have been intensified nowadays. This work reviews the preparation strategies and application of layered double hydroxides (LDH) as carriers for storing, carrying and control delivery of intercalated species as drugs and DNA for gene therapy. LDH show low toxicity, biocompatibility, high anion exchange capacity, surface sites for functionalization, and a suitable equilibrium between chemical stability and biodegradability. LDH can increase the intercalated species stability and promote its sub-cellular uptake for biomedical purposes. Concerning the healthy field, LDH have been evaluated for clinical diagnosis as a biosensor component.

A fluid conducting composite material prepared from graphite powder, commercial epoxy resin Araldite®, and cyclohexanone has been developed. The composition was optimized considering the mechanical properties as conductivity and adhesiveness using response surface methodology. This work employed cyclic voltammetry and amperometry to investigate the characteristics of such composite electrodes without and with the insertion of Prussian blue in the electrode body (bulk modified electrode). The composite electrodes were also successfully used for the amperometric detection of hydrogen peroxide at 0.0V vs Ag/AgCl.

Microcystins are non-ribosomal peptides that must be detected for its health concern. Here, microcystin LR and its specific antibody were respectively tethered to the substrate and to the tip of an atomic force microscope, after surface functionalization using 3-aminopropyltriethoxysilane and glutaraldehyde. Functionalization was confirmed comparing topographic images taken on bare and modified tips. Force versus distance curves were successfully used to measure the specific antibody-antigen interactions comparing with a control in which microcystin was initially blocked by incubation with free antibodies. The results showed unequivocally the specific recognition of MLR, suggesting that this method could be useful for biosensor development.

Giant extracellular hemoglobins are considered the summit of complexity in systems that carry oxygen, constituting an extraordinary model system to the study of hemoproteins. This class includes the hemoglobin of the annelid Glossoscolex paulistus that presents high cooperativity, great oligomeric and redox stabilities and ability of oligomeric reassociation. These properties have motivated evaluations about its utilization as prototype of artificial blood and biosensor. Kinetic studies involving autoxidation and detailed spectroscopic characterizations of its ferrous and ferric species have propitiated information about the structure-activity relationship of this macromolecule. The present review analyzes several biochemical issues, evaluating the state-of-art of this subject.

A simple flow-injection amperometric procedure using a three-electrode-integrated sensor for the determination of H2O2 in antiseptic mouthwash is reported. This method involves the use of a working composite electrode modified with Prussian Blue (PB) particles that was easily adapted as detector in FIA. The best amperometric response was observed for a composite containing 30% of graphite modified with PB particles (GAP) and 70% of pure graphite (GR). The proposed method presents a linear response in the range of 10 to 200 μmol L-1. The detection and quantification limits were 0.8 and 2.6 μmol L-1, respectively.

In this work, we describe the immobilization of the dinuclear compound [Cu2(apyhist)2Cl2](ClO4)2 (1) and its derived cations complexes, obtained in water solution or by deprotonation of the imidazolate moiety in the ligand leading to a cyclic tetranuclear species, in the Nafion® membrane on glass carbon electrode surface. After that, we studied the influence of the equilibrium in the electrocatalytic activity towards the reduction of H2O2 in the development of an amperometric sensor for the analytical determination of hydrogen peroxide. This strategy proved successful, and the electrochemical behaviour of the all complexes formed within the Nafion® coatings was characterized. We also provide evidence that its related cyclic tetranuclear imidazolate-bridged complex acts as a catalysts for the intramolecular, two-electron reduction of H2O2.

The potentialities and applications of the Multiple Pulse Amperometric detection (MPA) coupled with Flow Injection Analysis (FIA) are evaluated. Important aspects as cleaning and activation of electrode surface, indirect and simultaneous analysis of electroactive compounds and the use of the internal standard method for quantifications utilizing FIA-MPA are presented. The main parameters concerning the detection of electroactive analytes by multiple pulse amperometric detection in flowing solutions were also discussed. In addition, aspects such as flow rate, sample volume, application time of the potential pulses and instrumentation necessary for implementing of the method were also addressed.

Biscationic amidines bind in the DNA minor groove and present biological activity against a range of infectious diseases. Two new biscationic compounds (bis-α,ω-S-thioureido, amino and sulfide analogues) were synthesized in good yields and fully characterized, and their interaction with DNA was also investigated. Isothermal titration calorimetry (ITC) was used to measure the thermodynamic properties of binding interactions between DNA and these ligands. A double stranded calf thymus DNA immobilized on an electrode surface was used to study the possible DNA-interacting abilities of these compounds towards dsDNA in situ. A remarkable interaction of these compounds with DNA was demonstrated and their potential application as anticancer agents was furthered.
