Lettuce mottle virus (LeMoV) and dandelion yellow mosaic virus (DaYMV) infect lettuce in South America and Europe, respectively. LeMoV and DaYMV possess isometric particles, occur at low concentrations in plants and have narrow host ranges. Partial genome sequences of both viruses were obtained using purified viral preparations and universal primers for members of the family Sequiviridae. DaYMV and LeMoV sequences were analyzed and showed identity with other members of the family. Universal primers that detect both viruses and specific primers for LeMoV and DaYMV were designed and used in RT-PCR-based diagnostic assays. These results provide the first molecular data on the LeMoV and DaYMV genomes and suggest that LeMoV is a member of the genus Sequivirus, probably distinct from DaYMV.

Knowledge of structural and physiological differences among the prostatic lobes (PL) is the basis for development of experimental studies in traditional laboratory rodents. Although Mongolian gerbil reproductive organs have been increasingly investigated, its prostate structure is far from being properly known, and investigations of this organ focused on the ventral lobe (VL). Thus, the present study provides a thorough morphological description of prostatic complex in the male adult gerbil on the basis of topographic, histological, and ultrastructural analysis and ductal branching. Like other rodents, four pairs of PL were observed. However, in contrast to the rat and mouse, the VL is the least voluminous component and the dorsolateral lobe (DLL) is the most prominent and spatially isolated from remaining PL. The occurrence of a dorsal lobe (DL), hidden between bladder and insertion of seminal vesicles, has not been mentioned in previous reports with Mongolian gerbil. Collagenase digestion followed by microdissection revealed that, except for DL, which has a tubularacinar organization, all PL exhibit tubular organization and variable ductal branching. Distinct histological and ultrastructural features such as secretory epithelium, aspect of luminal secretion and stromal organization are reported for each PL and are confirmed by morphometric and stereological methods. Histological sections showed at least three intralobar segments in VL and DL. Ultrastructural analysis evidenced that, although luminal epithelial cells of PL share typical features of exocrine secretory cells, there are striking lobe phenotypical variations. Both merocrine and apocrine pathways are observed in variable rates in all PL, with the predominance of the former in the DLL and the latter in the CG. The morphological observations presented herein point to distinct structural identities for each PL, which probably reflects,specific functional compromise of seminal fluid secretion. These data also point to the gerbil as a good model for investigations concerning the regulation of prostate development and homeostasis, mainly with regard to the dorsal and dorsolateral PL.

The aim of this paper was to apply a multiresidue method using Supercritical Fluid Extraction (SFE) and capillary gas chromatography with electron capture and mass spectrometry detections in the analysis of the levels of pesticide residues in fruits and vegetables. Single laboratory validation of both solid-liquid and supercritical fluid extraction methods was carried out for 32 compounds selected from four pesticide classes (organochlorine, organonitrogen, organophosphorus and pyretroid) in blank and fortified samples of fresh lettuce, potato, apple and tomato. Recoveries for the majority of pesticides from fortified samples at fortification level of 0.04-0.10 mg kg -1 ranged 74-96% for both methods and confirmation of pesticide identity was performed by gas-chromatography-mass spectrometry in a selected-ion monitoring mode. Both methods showed good limits of detection (less 0.01 mg kg-1, depending on the pesticide and matrix) and the SFE method minimized environmental concerns, time, and laboratory work. ©2005 Sociedade Brasileira de Química.

DBMODELING is a relational database of annotated comparative protein structure models and their metabolic, pathway characterization. It is focused on enzymes identified in the genomes of Mycobacterium tuberculosis and Xylella fastidiosa. The main goal of the present database is to provide structural models to be used in docking simulations and drug design. However, since the accuracy of structural models is highly dependent on sequence identity between template and target, it is necessary to make clear to the user that only models which show high structural quality should be used in such efforts. Molecular modeling of these genomes generated a database, in which all structural models were built using alignments presenting more than 30% of sequence identity, generating models with medium and high accuracy. All models in the database are publicly accessible at http://www.biocristalografia.df.ibilce.unesp.br/tools. DBMODELING user interface provides users friendly menus, so that all information can be printed in one stop from any web browser. Furthermore, DBMODELING also provides a docking interface, which allows the user to carry out geometric docking simulation, against the molecular models available in the database. There are three other important homology model databases: MODBASE, SWISSMODEL, and GTOP. The main applications of these databases are described in the present article. © 2007 Bentham Science Publishers Ltd.

The objective of this study was to use 15N to label microbial cells to allow development of equations for estimating the microbial contamination in ruminal in situ incubation residues of forage produced under tropical conditions. A total of 24 tropical forages were ruminal incubated in 3 steers at 3 separate times. To determine microbial contamination of the incubated residues, ruminal bacteria were labeled with 15N by continuous intraruminal infusion 60 h before the first incubation and continued until the last day of incubation. Ruminal digesta was collected for the isolation of bacteria before the first infusion of 15N on adaptation period and after the infusion of 15N on collection period. To determine the microbial contamination of CP fractions, restricted models were compared with the full model using the model identity test. A value of the corrected fraction A was estimated from the corresponding noncorrected fraction by this equation: Corrected A fraction (ACPC) = 1.99286 + 0.98256 × A fraction without correction (ACPWC). The corrected fraction B was estimated from the corresponding noncorrected fraction and from CP, NDF, neutral detergent insoluble protein (NDIP), and indigestible NDF (iNDF) using the equation corrected B fraction (BCPC) = -17.2181 - 0.0344 × fraction B without correction (BCPWC) + 0.65433 × CP + 1.03787 × NDF + 2.66010 × NDIP - 0.85979 × iNDF. The corrected degradation rate of B fraction (kd)was estimated using the equation corrected degradation rate of B fraction (kdCPC) = 0.04667 + 0.35139 × degradation rate of B fraction without correction (kdCPWC) + 0.0020 × CP - 0.00055839 × NDF - 0.00336 × NDIP + 0.00075089 × iNDF. This equation was obtained to estimate the contamination using CP of the feeds: %C = 79.21 × (1 - e-0.0555t) × e-0.0874CP. It was concluded that A and B fractions and kd of CP could be highly biased by microbial CP contamination, and therefore these corrected values could be obtained mathematically, replacing the use of microbial markers. The percentage of contamination and the corrected apparent degradability of CP could be obtained from values of CP and time of incubation for each feed, which could reduce cost and labor involved when using 15N. © 2013 American Society of Animal Science. Within philosophy and cognitive science, the focus in relation to the problem of personal identity has been almost exclusively on the brain. We submit that the resulting neglect of the body and of bodily movements in the world has been detrimental in understanding how organisms develop a sense of identity. We examine the importance of sensing one’s own movements for the development of a basic, nonconceptual sense of self. More specifically, we argue that the origin of the sense of self stems from the sensitivity to spontaneous movements. Based on this, the organism develops a sense of “I move” and, finally, a sense of “I can move”. Proprioception and kinesthesis are essential in this development. At the same time, we argue against the traditional dichotomy between so-called external and internal senses, agreeing with Gibson that perception of the self and of the environment invariably go together. We discuss a traditional distinction between two aspects of bodily self: the body sense and the body image. We suggest that they capture different aspects of the sense of self. We argue that especially the body sense is of great importance to our nonconceptual sense of self. Finally, we attempt to draw some consequences for research in cognitive science, specifically in the area of robotics, by examining a case of missing proprioception. We make a plea for robots to be equipped not just with external perceptual and motor abilities but also with a sense of proprioception. This, we submit, would constitute one further step towards understanding creatures acting in the world with a sense of themselves.

Harryette Mullen is a contemporary African-American poet whose work has been increasingly analyzed and commented upon in American literary circles. Along her poetic career, one can identify the development of a complex relationship with the construction of the (black) female identity. Early in her career such construction involved the affirmation of a safer, if not “truthful” locus that could encompass the meaning of the female existence, which has ultimately come to develop a deconstruction, in her current poetry, of any centrality or essentiality in the search for a an authentic female identity. Translations of her poems will be presented in order to investigate their implication for understanding the fragmented body of the contemporary woman.

In the mid-nineteenth century, in his fruitful years of the perhaps best-known shortest literary career, Álvares de Azevedo wrote this verse “As ondas são anjos que dormem no mar” (“The waves are angels who sleep in the sea”). In the presentation of João Gilberto Noll’s young adult novel Anjo das ondas (Angel of the waves) in 2010, Ivan Marques states that the character in the novel, Gustavo, “with his adolescent passion, and torn spirit with overflowing lyrics, could even be compared to our tropical Byron”. João Gilberto Noll establishes a connection between him and Álvares de Azevedo that invites the reader to go beyond a first reading level of his work, seeking evidence of dialogue ratification between two apparently distant writers. The subtle intertextuality exceeds the initial clue and can be detected on Noll’s theme development from the history of Gustavo, a troubled teenager in search of his identity. Noll’s affinity to Álvares de Azevedo can still be captured in his narrative texture, in voices coming from his text; however, they also reveal to be unique and original features of his literary production. An apparent exercise in style shows that each author fulfills his mission by expressing, in his own way, the feeling of his time. In addition, it shows that through aesthetic emotion, literature allows us to transcend time and space.
