Os sucroésteres são empregados como surfactantes, tensoativos, substitutos de gordura e antibióticos. Este trabalho tem como objetivo sintetizar um novo sucroéster derivado do ácido picolínico (ácido 2-piridinocarboxílico), o picolinato de sacarose, e estudar sua atividade antibacteriana in vitrosobre a bactéria Gram-negativa patogênica Escherichia coli. A síntese do picolinato de sacarose foi processada a partir da transesterificação da sacarose com picolinato de metila em condição anidra à 80oC, utilizando dimetil sulfóxido (DMSO) como solvente e K2CO3 como catalisador. A separação dos isômeros formados foi realizada por HPLC no modo semipreparativo e cinco frações cromatográficas foram coletadas e aplicadas em testes de atividade antibacteriana, por disco-difusão em meio sólido, nas concentrações de 150; 300; 450; 600; 750 e 900 μg/mL. As frações 2, 4 e 6 foram ativas contra E.coli. e a fração 4 (900 μg/mL) foi a mais eficiente, sendo selecionada para testes em sinergia com EDTA nas concentrações de 250, 500 e 750 μg/mL, com melhor resultado quando empregado EDTA em 750 μg/mL. Neste caso, os discos apresentaram halos de inibição de crescimento igual ao da Tetraciclina (30 μg/mL) e superior aos produzidos pelos discos com Gentamicina (10 μg/mL). A fração 4 foi caracterizada por FTIR e espectrometria de massas (ESI-MS) e os resultados indicam que se trata sucroéster monossubstituído. Palavras-chave:Sucroquímica. Sucroéster. Sacarose. ABSTRACT Study of antimicrobial activity of sucrose picolinate against Escherichia coli Sucrose esters are generally used as surfactants, fat substitutes and antibiotics. The aim of the present study was to synthesize new sucrose esters derived from picolinic acid (2-pyridine carboxylic acid) and study in vitro antimicrobial activity on the Gramnegative pathogenic bacterium Escherichia coli. The synthesis of sucrose picolinate was performed through the transesterification of sucrose with methyl picolinate under anhydrous conditions at 80 oC using dimethyl sulfoxide (DMSO) as the solvent and K2CO3as the catalyst. The separation of the formed isomers was performed by HPLC in a semi-preparative chromatograph system. Five fractions were collected and applied to a disc-diffusion antibiogram in solidmedium tests at concentrations of 150, 300, 450, 600, 750 and 900 μg/mL. Fractions 2, 4 and 6 were active against E. coli. Fraction 4 (900 μg/mL) was the most efficient and was selected for the determination of antimicrobial activity in synergistic tests with EDTA at concentrations of 250, 500 and 750 μg/mL. The best result was obtained with 750 μg/mL of EDTA. Fraction 4 was characterized as a monosubstituted sucrose ester by FTIR and mass spectrometry (ESI-MS). Keywords: Sucrochemistry. Sucrose. Sucrose esters are biodegradable, non pollutant and safe for health; moreover, they have shown great potential in the pest control. We tested a crude mix of sucrose esters on the survivorship and oviposition of Calacarus heveae Feres (Acari: Eriophyidae) females, an important rubber tree pest mite. The females were collected from rubber leaflets of the GT 1 and RRIM 600 clones. The mites were kept on stock arenas placed in rearing chamber at 28 + 0,1°C, 80 + 10% of relative humidity and 12h of light phase about two weeks before the assays. After this period, the mites were transferred to assay arenas made with leaflets from the same clone of stock arenas. The survivorship of females sprayed with sucrose esters in the concentrations of 1, 2, 3 and 4 g/L and their oviposition effect with 0.5 and 1 g/L were analyzed. The mites sprayed with 4 g/L had about 80% of mortality, while those with 1 g/L around 60%. All mites that died in the treatments with sucrose ester became dark with wrinkled tegument and decreased their body volume, suggesting dehydration. We also observed the efficacy of 1 g/L concentration to decrease the female oviposition in about 50%, in the third day after spraying. No differences were observed in the mortality and oviposition between females kept on both clones.

The growth of maize (Zea mays L.) kernels depends on the availability of carbon (C) and nitrogen (N) assimilates supplied by the mother plant and the capacity of the kernel to use them. Our objectives were to study the effects of N and sucrose supply levels on growth and metabolism of maize kernels. Kernel explants of Pioneer 34RO6 were cultured in vitro with varying combinations of N (5 to 30 mM) and sucrose (117 to 467 mM). Maximum kernel growth was obtained with 10 mM N and 292 mM sucrose in the medium, and a deficiency of one assimilate could not be overcome by a sufficiency of the other. Increasing the N supply led to increases in the kernel sink capacity (number of cells and starch granules in the endosperm), activity of certain enzymes (soluble and bound invertases, sucrose synthase, and aspartate aminotransaminase), starch, and the levels of N compounds (total-N, soluble protein, and free amino acids), and decreased the levels of C metabolites (sucrose and reducing sugars). Conversely, increasing the sucrose supply increased the level of endosperm C metabolites, free amino acids, and ADPG-PPase and alanine transaminase activities, but decreased the activity of soluble invertase and concentrations of soluble protein and total-N. Thus, while C and N are interdependent and essential for accumulation of maximum kernel weight, they appear to regulate growth by different means. Nitrogen supply aids the establishment of kernel sink capacity, and promotes activity of enzymes relating to sucrose and nitrogen uptake, while sucrose regulates the activities df invertase and ADPG-PPase. Different culture conditions for Protaminobacter rubrum and enzymatic reaction parameters were evaluated with the goal of improving isomaltulose production. P. rubrum was grown in a medium with 1% (w/v) cane molasses and 0.5% yeast extract and achieved a maximum cell yield Y(x/s) of 0.295 g of cells/g sucrose and a specific growth rate (mu) of 0.192 h(-1). The immobilization of P. rubrum cells was carried out with calcium alginate, glutaraldehyde and polyethyleneimine. Stabile immobilized cell pellets were obtained and used 24 times in batch processes. Enzymatic conversion was carried out at different sucrose concentrations and in pH 6 medium with 70% (w/v) sucrose at 30 degrees C an isomaltulose yield of 89-94% (w/v) was obtained. The specific activity of the P. rubrum immobilized pellets in calcium alginate at 30 degrees C ranged from 1.6 to 4.0 g isomaltulose g(-1) pellet h(-1), respectively with 70% and 65% sucrose solution, while in lower sucrose concentration had higher specific activities presumably due to substrate inhibition of the isomaltulose synthase in higher sucrose concentrations. Two botryosphaerans, exopolysaccharides (EPS) secreted by the ascomyceteous fungus Botryosphaeria rhodina, when grown on sucrose and fructose as sole carbon sources, were structurally compared after their isolation from the culture medium. Both EPS were submitted to trypsin digestion, and eluted as a single peak on gel filtration. Total acid hydrolysis yielded only glucose, and data from methylation analysis and Smith degradation indicated that both EPS constituted a main chain of glucopyranosyl beta(1 -> 3) linkages substituted at O-6. The products obtained after partial acid hydrolysis demonstrated side chains consisting of glucosyl- and gentiobiosyl- linked beta(1 -> 6) residues. C-13-NMR spectroscopy studies showed that all glucosidic linkages were of the beta-configuration. The carbon source affected the side chain structures of botryosphaeran but not the main chain makeup. Sucrose produced less branching (21%) than fructose (31%). (c) 2005 Published by Elsevier Ltd.

Potentially bioactive N-(aminoalkyl)lactamic amino acids and esters were synthesized in satisfactory to good yields by SNAr reactions of aromatic acids with N-(3-aminopropyl)lactams followed by esterification with tertiary amino alcohols. The addition-elimination SNAr mechanism was confirmed by NMR and MS measurements.

The protective effect of gallic acid and its esters, methyl, propyl, and lauryl gallate, against 2,2'-azobis(2-amidinopropane)hydrochloride (AAPH)-induced hemolysis and depletion of intracellular glutathione (GSH) in erythrocytes was studied. The inhibition of hemolysis was dose-dependent, and the esters were significantly more effective than gallic acid. Gallic acid and its esters were compared with regard to their reactivity to free radicals, using the DPPH and AAPH/pyranine free-cell assays, and no significant difference was obtained. Gallic acid and its esters not only failed to inhibit the depletion of intracellular GSH in erythrocytes induced by AAPH but exacerbated it. Similarly, the oxidation of GSH by AAPH or horseradish peroxidase/H(2)O(2) in cell-free systems was exacerbated by gallic acid or gallates. This property could be involved in the recent findings on proapoptotic and pro-oxidant activities of gallates in tumor cells. We provide evidence that lipophilicity and not only radical scavenger potency is an important factor regarding the efficiency of antihemolytic substances.

Special diets are used to mitigate many human diseases. When these diets require changes in carbohydrate content, then sweetness becomes an important characteristic. The range of low-calorie sweeteners available to the food industry is expanding. It is essential to have an exact knowledge of the relative sweetness of various sweeteners in relation to different sucrose concentrations. The objective of this study was to determine the variation on the relative sweetness of aspartame (APM), stevia [Stevia rebaudiana (Bert.) Bertoni] leaf extract (SrB) and the mixture cyclamate/saccharin - two parts of cyclamate and one part of saccharin - (C/S) with the increase in their concentrations, and in neutral and acid pH in equi-sweet concentration to 10% sucrose, using magnitude estimation. Sweetness equivalence of SrB in relation to sucrose concentrations of 20% or higher and of APM and C/S to sucrose concentrations of 40% or higher could not be determined, because a bitter taste predominated. The potency of all sweeteners decreased as the level of sweetner increased. In equi-sweet concentration of sucrose at 10%, with pH 7.0 and pH 3.0, the potency was practically the same for all sweeteners evaluated.

This study examined whether sucrose-rich diet (SRD)-induced hyperglycaemia, dyslipidemia and oxidative stress may be inhibited by N-acetylcysteine (C5H9-NO3S), an organosulfur from Allium plants. Male Wistar 40 rats were divided into four groups (n = 10): (C) given standard chow and water; (N) receiving standard chow and 2 mg/l N-acetylcysteine in its drinking water; (SRD) given standard chow and 30% sucrose in its drinking water; and (SRD-N) receiving standard chow, 30% sucrose and N-acetylcysteine in its drinking water. After 30 days of treatment, SRD rats had obesity with increased abdominal circumference, hyperglycaemia, by dyslipidemia and hepatic triacylglycerol accumulation. These adverse effects were associated with oxidative stress and depressed lipid degradation in hepatic tissue. The SRD adverse effects were not observed in SDR-N rats. N-Acetylcysteine reduced the oxidative stress, enhancing glutathione-peroxidase activity, and normalizing lipid hydroperoxyde, reduced glutathione and superoxide dismutase in hepatic tissue of SRD-N rats. The beta-hydroxyacyl coenzyme-A dehydrogenase and citrate-synthase activities were increased in SRD-N rats, indicating enhanced lipid degradation in hepatic tissue as compared to SRD. SRD-N rats had reduced serum oxidative stress and diminished glucose, triacylglycerol, very-low-density lipoprotein (VLDL), oxidized low-density lipoprotein (alpha-LDL) and cholesterol/highdensity lipoprotein (HDL) ratio in relation to SRD. In conclusion, NAC offers promising therapeutic values in prevention of dyslipidemic profile and alleviation of hyperglycaemia in high-sucrose intake condition by improving antioxidant defences. N-Acetylcysteine had also effects preventing metabolic shifting in hepatic tissue, thus enhancing fat degradation and reducing body weight gain in conditions of excess sucrose intake. The application of this agent in food system via exogenous addition may be feasible and beneficial for antioxidant protection. (c) 2006 Elsevier B.V All rights reserved.
