Lectin obtained from the marine sponge Tedania ignis was purified and characterized by extraction of soluble proteins (crude extract) in 50mM Borax, pH 7.5. The purification procedure was carried out by crude extract precipitation with ammonium sulfate 30% (FI). The precipitated was resuspended in the same buffer and fractionated with acetone 1.0 volume (F1.0). A lectin was purified from this specific fraction by using an affinity chromatography Sepharose 6B. This lectin preferentially agglutinated human erythrocytes from B type previously treated with papain enzyme. The hemagglutinating activity lectin was dependent of divalent Mn2+ cation and was inhibited by the carbohydrates galactose, xylose and fructose. SDS-PAGE analysis indicated a molecular mass of the lectin around 45 kDa. This protein showed stability until 40°C for 1 h. Further, it showed activity between pH 2.5 and 11.5, with an enhanced activity at pH 7.5. Leishmania chagasi promastigotes stained with Coomassie brilliant blue R-250 were agglutinated by F1,0 and in the presence of galactose this interaction was abolished. These results show that this lectin could be implicated in defense procedures and it will can be used as biological tools in studies with this protozoon

This study presents the Environmental Sensibility Mapping to oil spillings on the Potengi estuary - RN and neighboring coastline through remote sensing data, collecting, treatment and integration of the geomorphologic, oceanographic (temperature, salinity, density, direction and intensity), meteorological (wind speed and direction) and high resolution seismic (bathymetry and sonography) data. The Potengi river estuary is located on the eastern coastline of the Rio Grande do Norte State, being inserted in the geological context for the coastal Pernambuco-Paraíba basin and spreading over 18 km; it shelters the Natal harbor zone and an oil terminal, centralizing, therefore, important oil transport operations that can cause accidental spillings. Under the oceanographic point of view, the Potengi estuary is characterized by the absence of any expressive thermic stratification, being classified as partially mixed, B type according to Pritchard (1955), and 2 type in conformity to the stratification-circulation diagram by Hansen & Rattray (1966). Two main wind systems are responsable for the formation of wave sets that occur in the area. The dynamic tide presents, in the Natal Harbor, mean amplitude in spring and quadrature tides, with around 2.8 and 2m, respectively. The mechanism of saline tide mixing was defined through the salinity which is the main parameter for the identification of this mechanism. Important variations of the salinity mean values (36.32 psu), temperature (28.11ºC) and density (22.96 kg/m3) in the estuarine waters presented features belonging to low latitude regions. The water temperature follows the air temperature variations, in the region, with expressive daily amplitudes. In this study, the identification of the estuarine bed morphology through bathymetric and sonographic analysis, had the purpose to evaluate the influence of the superficial and bottom currents for the bottom shaping. In this way, the use of the side scan sonar showed, to be very useful in the identification of the bottom morphology and its relationship with the predominant action of the tidal currents in the Potengi estuary. Besides, it showed how the sonograms can be a support to the comparison of the several patterns derived from the local hydrodynamic variations. The holocene sediments, which fill the estuarine channel, are predominantly sandy, varying from selected, sometimes silty. The sedimentation is controlled by the environmental hydrodynamic conditions, being recognized two important textural facies: Muddy Facies and Sandy Facies. The distribution of these textural facies apparently oscillates owing to the tidal cycle and flow intensity. Each one of the above mentioned data was integrated in a Geographic Information System (GIS), from which was produced the Environmental Sensibility Map to oil spillings with Coastal Sensibility Index (CSI) to the Potengi estuary. The integrated analysis of these data is essential to oil spilling contingency plans, in order to reduce the spilling environmental consequences and to make efficient the endeavours of contention and cleaning up/removal on the Natal Harbor. This study has the aim to collaborate for the increase of informations about the estuarine environment and contribute to a better management of the question: environment/polluting loads

Several tests that evaluate the quality of seeds are destructive and require time, which is considered long and expensive in the processes that involves the production and marketing of seed. Thus, techniques that allow reducing the time related to assess the quality of seed lots is very favorable, considering the technical, economic and scientific point of view. The techniques images of seed analyzed both by X-ray such as digital images, represent alternative for this sector, and are considered reproducible and fast, giving greater flexibility and autonomy to the activities of production systems. Summarily, the objective was to analyze the internal morphology of seeds of this species through x-rayed images and the efficiency of weed seed area increased during soaking through image analysis and compare them with the results of germination tests and force the evaluation of physiological seed quality. For X-ray tests, the seeds were exposed for 0.14 seconds at radiation 40kV and 2.0 mAs. Were analyzed images using the ImageJ program and subsequently put to germinate in B.O.D chamber at 27 ° C, in which there was the comparison of results for germination. To determine the test area increase (% IA), seeds were used with and without seed coat, maintained the B.O.D chamber at 15 ° to 20 ° C, the seeds were photographed before and after the soaking period, the results were compared to the germination rates. For the X-ray test, it was observed that seeds with empty area greater than 20%, showed a higher percentage of abnormal seedlings. Camu-camu (Myrciaria dubia H.B.K. (McVaugh)) is a native Amazon fruit, recognized worldwide as one of the main natural sources of ascorbic acid. Due to its great acidity, this fruit is generally consumed after processing into juice or as ingredient in food preparations. As a co-product of the camu-camu processing, a significant amount of agroindustrial residue is generated. Despite the studies showing the bioactive value and biological potential of the fruit, few studies have approached the possible processing techniques, transformation and preservation of camu-camu fruits and its agroindustrial pomace. Therefore, the present work has the objective of evaluating two different drying processes applied to camu-camu pomace (peel and seeds with residual pulp), freeze drying and hot air drying, in order to obtain a functional fruit product. This thesis was divided into three stages: the first one shows the studies related to the freeze drying and hot air drying, where we demonstrated the impact of the selected drying techniques on the bioactive components of camu-camu, taking the fresh pomace as the control group. Among the investigated conditions, the groups obtained at 50ºC and 4 m/s (SC50) and 80ºC and 6 m/s (SC80) were selected as for further studies, based on their ascorbic acid final content and Folin-Ciocalteau reducing capacity. In addition to SC50 and SC80, the fresh pomace (RF) and freeze dried (RL) samples were also evaluated in these further stages of the research. Overall, the results show higher bioactive concentration in the RF samples, followed by RL, SC50 and SC80. On the second step of the research, the antioxidant, antimicrobial and antienzymatic activities were evaluated and the same tendency was observed. It was also reported, for the first time in the literature, the presence of syringic acid in dried camu-camu pomace. In the third and final stage of the research, it was investigated the effect of dried camu-camu on aging and neuroprotective disorders, using the in vivo model C.elegans. It was observed that camu-camu extracts were able to modulate important signaling genes relevant to thermal and oxidative stresses (p < 0.05). The polar acid, polar basic and polar neutral fractions obtained from the low molecular extracts of SC50 were able to extend the lifespan of wild type N2 C. elegans in 20% and 13% (p < 0.001). Results also showed that the paralysis induced by the β1-42 amyloid was significantly (p < 0.0001) retarded in CL4176 worms. Similarly, the camu-camu extracts attenuated the dopaminergic induction associated to Parkinson’s disease. Finally, a global analysis of the data presented here reveal that the camu-camu pomace, a co-product obtained from the industrial processing of a native Brazilian fruit, is a relevant natural source of health relevant compounds. This thesis, shows for the first time, the multifunctionality of camu-camu pomace, a natural resource still underexploited for scientific, commercial and technological purposes.

This work proposes a computational methodology to solve problems of optimization in structural design. The application develops, implements and integrates methods for structural analysis, geometric modeling, design sensitivity analysis and optimization. So, the optimum design problem is particularized for plane stress case, with the objective to minimize the structural mass subject to a stress criterion. Notice that, these constraints must be evaluated at a series of discrete points, whose distribution should be dense enough in order to minimize the chance of any significant constraint violation between specified points. Therefore, the local stress constraints are transformed into a global stress measure reducing the computational cost in deriving the optimal shape design. The problem is approximated by Finite Element Method using Lagrangian triangular elements with six nodes, and use a automatic mesh generation with a mesh quality criterion of geometric element. The geometric modeling, i.e., the contour is defined by parametric curves of type B-splines, these curves hold suitable characteristics to implement the Shape Optimization Method, that uses the key points like design variables to determine the solution of minimum problem. A reliable tool for design sensitivity analysis is a prerequisite for performing interactive structural design, synthesis and optimization. General expressions for design sensitivity analysis are derived with respect to key points of B-splines. The method of design sensitivity analysis used is the adjoin approach and the analytical method. The formulation of the optimization problem applies the Augmented Lagrangian Method, which convert an optimization problem constrained problem in an unconstrained. The solution of the Augmented Lagrangian function is achieved by determining the analysis of sensitivity. Therefore, the optimization problem reduces to the solution of a sequence of problems with lateral limits constraints, which is solved by the Memoryless Quasi-Newton Method It is demonstrated by several examples that this new approach of analytical design sensitivity analysis of integrated shape design optimization with a global stress criterion purpose is computationally efficient

Massively Multiplayer Online Role-Playing Games (MMORPGs) are role-playing games that, through the Internet, can integrate thousands of players interacting at the same time in at least one virtual world. This way, these games can provide, further than fun, a greater familiarity with the additional language and opportunity to improve the linguistic proficiency in a real context. Hence, what is proposed in this study is extended knowledge about the learning of an additional language mediated by MMORPGs for teachers to know how, if relevant, to present, use or encourage this practice to their students. Based on this major purpose, we seek to answer the following research questions: (a) what distinguishes the learning profile of the gamers and non-gamers; (b) if MMORPGs can, through a hybrid and systematic approach, assist the development of proficiency of the additional language and (c) what the think-aloud protocols show about the learning mediated by the MMORPG Allods Online. Following an experimental method (NUNAN, 1997), 16 students of the curricular component Reading and Writing Practices in English Language have comprised the control group and 17 students of the same class formed the experimental group and were submitted to a pre and post-test adapted from the Key English Test (KET) by the Cambridge University (2008). The tests were conducted before and after a period of 5 weeks of 3 hours of practice with Allods Online a week (experimental group), and classes of the curricular component (both groups). A quantitative analysis of the questionnaires about the exposure to English profiles of the participants, a quantitative analysis of the tests scores and a qualitative analysis of the thinkaloud protocols collected during the experiment were conducted based on the theories of (a) motivation (GARDNER, 1985, WILLIAMS & BURDEN, 1997, BROWN, 2007, HERCULANO-HOUZEL, 2005); (b) active learning (GASS, 1997, GEE, 2008, MATTAR, 2010); (c) interaction and collaborative learning (KRASHEN, 1991, GASS, 1997, VYGOTSKY, 1978); (d) situated learning (DAMASIO, 1994; 1999; 2003, BROWN, 2007, GEE, 2003) and (e), tangential learning (PORTNOW, 2008; MATTAR, 2010). The results indicate that the participants of the experimental group (gamers) seem to be more engaged in tangential English learning activities, such as playing games, listening to music in English, communicating with foreigners and reading in English. We also deduced that the period of experiment possibly generated positive results on the gamers proficiency scores, mainly in the parts related to orthographic development, reading and comprehension, writing with focus on content and orthographic accuracy. Lastly, the think-aloud protocols presented evidences that the gamers have engaged in active English language learning, they have interacted in English with other players, and learned linguistic aspects through the experience with the MMORPG Allods Online

The Oral Epithelial Dysplasia (OED) is the lesion that precedes or co-exists with the Oral Squamous Cell Carcinoma (OSCC), presenting molecular and/or histological similar alterations. The divergences about the malignization potential of OEDs and the role of inflammation in this process make hard the early diagnosis and evaluation of OSCCs aggressiveness. Thus, it became the goal of this study to evaluate the role of inflammation in oral carcinogenesis and tumoral aggressiveness. For this purpose a morphological study was performed in 20 OED cases and 40 OSCC cases to detect the malignization potential of OEDs and the histologic malignancy grading (HMG) of OSCCs, analyzing superficial masses for dismorphism evaluation and the invasive front for evaluation of tumoral growing; and immunohistochemical, using anti-CD8, anti-FOXP3, anti-TGFβ, anti-TNFα and anti-NF-кB antibodies, comparing their with the types lesion, histological degree and intensity of the inflammatory infiltrate. The results were statistically significant for the parameters: cell maturity (p=0,0001), masses presence (p=0,038) and dismorphism (p=0,037), when associated to HMG. To compare the expression of the markers with the types lesion, a significantly higher expression of CD8 (p=0,001) and NF-кB (p=0,002) in the OED, and also a smaller expression of the epithelial TGFβ in the severe OEDs (p=0,011), without significant expression between OSCC degrees. By relating the expression of the studied markers with the inflammatory infiltrate intensity, a positive relation was observed with: inflammatory TNFα(p=0,003), epithelial TNFα and NF-кB (p=0,051 and p=0,004), in OEDs; and with CD8 (p=0,021) and TNFα (p=0,015) in conjunctive OSCCs; and a negative relation with epithelial TNFα (p=0,034) in OSCCs. No significant relation was found between FOXP3 with any of the studied variables. These findings lead to the conclusion that, the study of the invasive front is as important as the study of superficial masses for the evaluation of tumoral aggressiveness; the intensity of the inflammatory infiltrate has no use as a parameter for prognostic evaluation of OSCC in routine exams, but, the molecular events detected in this study may be necessary to give basis for determining the malignant potential in OEDs and aggressiveness in OSCCs

Episodic memory refers to the recollection of what, where and when a specific event occurred. Hippocampus is a key structure in this type of memory. Computational models suggest that the dentate gyrus (DG) and the CA3 hippocampal subregions are involved in pattern separation and the rapid acquisition of episodic memories, while CA1 is involved in memory consolidation. However there are few studies with animal models that access simultaneously the aspects ‗what-where-when . Recently, an object recognition episodic-like memory task in rodents was proposed. This task consists of two sample trials and a test phase. In sample trial one, the rat is exposed to four copies of an object. In sample trial two, one hour later, the rat is exposed to four copies of a different object. In the test phase, 1 h later, two copies of each of the objects previously used are presented. One copy of the object used in sample trial one is located in a different place, and therefore it is expected to be the most explored object. However, the short retention delay of the task narrows its applications. This study verifies if this task can be evoked after 24h and whether the pharmacological inactivation of the DG/CA3 and CA1 subregions could differentially impair the acquisition of the task described. Validation of the task with a longer interval (24h) was accomplished (animals showed spatiotemporal object discrimination and scopolamine (1 mg/kg, ip) injected pos-training impaired performance). Afterwards, the GABA agonist muscimol, (0,250 μg/μl; volume = 0,5 μl) or saline were injected in the hippocampal subregions fifteen minutes before training. Pre-training inactivation of the DG/CA3 subregions impaired the spatial discrimination of the objects (‗where ), while the temporal discrimination (‗when ) was preserved. Rats treated with muscimol in the CA1 subregion explored all the objects equally well, irrespective of place or presentation time. Our results corroborate the computational models that postulate a role for DG/CA3 in spatial pattern separation, and a role for CA1 in the consolidation process of different mnemonic episodes

PLCs (acronym for Programmable Logic Controllers) perform control operations, receiving information from the environment, processing it and modifying this same environment according to the results produced. They are commonly used in industry in several applications, from mass transport to petroleum industry. As the complexity of these applications increase, and as various are safety critical, a necessity for ensuring that they are reliable arouses. Testing and simulation are the de-facto methods used in the industry to do so, but they can leave flaws undiscovered. Formal methods can provide more confidence in an application s safety, once they permit their mathematical verification. We make use of the B Method, which has been successfully applied in the formal verification of industrial systems, is supported by several tools and can handle decomposition, refinement, and verification of correctness according to the specification. The method we developed and present in this work automatically generates B models from PLC programs and verify them in terms of safety constraints, manually derived from the system requirements. The scope of our method is the PLC programming languages presented in the IEC 61131-3 standard, although we are also able to verify programs not fully compliant with the standard. Our approach aims to ease the integration of formal methods in the industry through the abbreviation of the effort to perform formal verification in PLCs

In this work it is assessed the performance of Portland cement-based mortar to the grouting of type II ceramic plates with the addition of unusable tire rubber powder. It is presented a bibliographical review about the subject in which is done the theoretical and methodological foundation of the whole investigative process. The analyzed universe comprises a sample of mortar to the grouting of conventional ceramic plates type II (reference sample) and five more samples to the grouting of ceramic plates type II, which were made up of the addition of unusable tire rubber powder in the respective proportion (in mass) of 4%, 8%, 12%, 16% and 20%. These mortar samples were subject to the trials of determination of the consistency index (Brazilian Standard NBR 13276:2005), water retention ( Brazilian Standard NBR 14992 Attachment B:2003), permeability in 240 minutes (Brazilian Standard NBR 14992 Attachment G:2003), absorption of water by immersion (Brazilian Standard NBR 9781:2013), resistance to compression (Brazilian Standard NBR14992 Attachment D: 2003), resistance to traction in the flexion (Brazilian Standard NBR 13279:2005), resistance of traction adherence (Brazilian Standard NBR 14081 part 4:2012) and hardened mass density (Brazilian Standard NBR 13280: 2005). It has been found out from the analyzes of the results in the trial the following situation: the reference mortar used met the established requirements in the norms of specifications corresponding to only six from the eight parameters assessed in the research; the mortar with addition of 4,0% of tire rubber powder met the established requirements corresponding to only the resistance to compression and the resistance of adherence to traction. Thus, the other kinds of mortar with addition of 8,0 %, 12,0 %, 16,0 % and 20,0 % of tire rubber powder met the requirements of specifications corresponding to only the resistance to compression and the resistance of adherence to traction. This result concludes that the adding of tire rubber powder does not grant improvement to the mortar of type II grouting to the laying of ceramic plates.
