The pressure has grown to develop cost-effective emission reduction strategies in the Baltic Sea. The forthcoming stringent regulations of the International Maritime Organization for reducing harmful emissions of shipping in the Baltic Sea are causing increasing expenses for the operators. A market-based attitude towards pricing of economic incentives could be seen as a new approach for a successful application for the additional emission reduction of nitrogen oxides (NOx). In this study the aim is to understand the phenomenon of environmentally differentiated port fees and its effects on shipping companies’ emission reduction investments. The goal is to examine empirically the real-life effects of the possible environmental differentiated port fee system and the effect of environmentally differentiated port fees on NOx reduction investments in the Baltic Sea. The research approach of this study is nomothetical. In this study research questions are answered by analyzing the broad database of the Baltic Sea fleet. Also the framework of theory is confirmed and plays an important role in analyzing the research problem. Existing investment costs of NOx emission reduction technology to ship owners are estimated and compared to investment costs with granted discounts added to the cash flows. The statistical analysis in this study is descriptive. The major statistic examination of this study is the calculation of the net present values of investments with different port fee scenarios. This is done to investigate if the NOx technology investments could be economically reasonable. Based on calculations it is clear that the effect of environmentally differentiated port fees is not adequate to compensate the total investment costs for NOx reduction. If the investment decision is made only with profitability considerations, sources will prefer to emission abatement as long as incomes from the given subsidy exceeds their abatement costs. Despite of the results, evidence was found that shipping companies are nevertheless willing to invest on voluntary emission abatement technology. In that case, investment decision could be made with criteria of, for example, sustainable strategy or brand image. Combined fairway and port fee system or governmental regulations and recommendation could also function as additional incentives to compensate the investment costs. Also, the results imply that the use of NPV is not necessarily the best method to evaluate environmental investments. If the calculations would be done with more environmental methods the results would probably be different.

Tutkimuksen tavoitteena on tutkia kulu- ja verotehokkuutta rahastojen rahastoissa. Tutkimus ontoteutettu yhteistyössä Sampo Rahastoyhtiö Oy:n kanssa. Tutkimuksessa verrataan, miten rahastojen rahastot ovat menestyneet suhteessa yksittäisen sijoittajan itse rakentamaan portfolioon. Tutkimuksessa on käytetty mallirahastona Sampo Kompassi 50 Kasvu -rahastoa. Alkuperäisen rahaston ympärille on rakennettu kuusi Kompassi 50 -rahastoa jäljittelevää rahastoportfoliota. Rahastoportfoliot on rakennettu joko merkitsemällä suoraan yksittäisiä rahastoja tai hyödyntämällä sijoitusvakuutusta. Tutkimus perustuu Sampo Rahastoyhtiö Oy:n tilastoihin kahden vuoden ajanjaksolta. Tutkimustulosten perusteella vertaillaan eri sijoitusvaihtoehtojentuottoja, palkkioita, veroja, riskejä sekä tuotto-riskisuhdetta. Tutkielma jakautuu teoreettiseen ja empiiriseen osaan. Teoriaosuuden tarkoituksena on johdattaa lukija sijoitusrahastojen maailmaan. Samassa osiossa syvennytään lisäksi tutkimuksen kannalta oleellisiin tekijöihin: tuottoon ja riskiin,palkkioihin sekä verotukseen. Tutkielman empiirisessä osassa selvitetään vastausta tutkimusongelmaan matemaattisia laskelmia hyväksikäyttäen. Kyseisestä aiheesta ei ole aiempia empiirisiä tutkimuksia.

Finnish food producers' trade with Russia has experienced profound changes since the collapse of the Soviet Union. Simultaneously, the distribution systems of foodstuffs have changed remarkably. This study sheds some light into these changes and analyses the current situation in distribution systems of foodstuffs in Russia. In addition, the study discusses the possibilities of Finnish food producers to get more of their products to the shelves of Russian food retail stores. Before the 1998 financial crisis, the import of foreign foodstuffs was booming in Russia due to the overvalued rouble. As a result of the financial crisis, food import collapsed. The export of Finnish foodstuffs to Russia has been slowly recovering during the past few years, but in the most important product categories the pre-crisis levels have so far not been reached and maybe will not be reached. In certain product categories the growth has been only marginal. It seems that starting localproduction will become increasingly important in the future. This is further encouraged by the fact that Russian consumers favour domestic food products. Russian consumers are very price conscious and demand quality in food products. The perceived price-quality ratio is an important criterion in the purchase decision.The majority of foodstuff retail is still conducted via unorganised forms of trade (e.g. kiosks and marketplaces) but modern retail chains are developing at a fast pace in Russia. They are also expected to dominate the retail trade in foodstuffs over the unorganised forms of trade in the future. This will change the distribution systems as well. The retail chains are trying to shorten the distribution chain, similarly to what has been seen in the Western countries. This together with the strengthening of retail chains is likely to shrink the role of wholesalers, as the chains increasingly want to work directly with the producers. Many large retail chains are acquiring or have already acquired a distribution centre or centres in order to boost efficiency and control the flow of products. The strengthening of the retail chains also gives them power in negotiations, which the producers and distributors have to adjust to. For example store entry fees and retail chains' own private label products pose challenges to the food producers. In the food production sector the competition is fierce, as large Russianand foreign producers want to ensure their piece of the market. The largest producers utilise their size: they invest in big marketing campaigns and are willing to pay high entry fees to retail chains in order to secure a place on the store shelves and to build a strong brand in Russia. This complicates the situation from the viewpoint of small producers. Currently, the most popular type of distribution system among the interviewed Finnish food producers is based on a network of local distributors. There is, however, a strong consensus on the importanceof starting local production in order to be a serious actor in Russia in the future. Factors that hinder the starting of local production include the lack of local infrastructure and qualified staff, and the low risk tolerance of Finnish firms. Major barriers for entry in Russia are the actions of authorities, fierce competition, fragmented market and Finnish producers' heavy production costs. The suggested strategies for increasing the market share include focusing geographically or segment-wise, introducing new products, starting local production, andcooperation between Finnish producers. Smallness was one reason why Finnish producers had to cut down their operations in Russia due to the 1998 crisis. Smaller producers had fewer resources to tolerate losses during the period of crisis. Smallness is reflected also on trade negotiations with retail chains and distributors. It makes it harder to cope with the store entry fees and to differentiatefrom the mass of products propped up by expensive advertising. Finally, it makes it harder for Finnish producers to start or expand local production, as it is more difficult for a small producer to get financing and to tolerate the increased risks. Compensating for the smallness might become the crucial factor determining the future success of Finnish food producers in the Russian market.

Tässä diplomityössä tutkitaan tehdasjärjestelmien uusintaan vaikuttavia tekijöitä, kun tarkoituksena on siirtyä yhtenäisiin tuotepohjaisiin järjestelmäratkaisuihin usean paperitehtaan ympäristössä. Paperitehtaan tehdasjärjestelmän avulla hallitaan tehtaan toimintaa alkaen tilauksen saapumisesta tehtaalle aina valmiin tuotteen varastointiin sekä asiakkaalle toimitukseen saakka. Työn pääasiallinen tarkoitus on tutkia erilaisten teknisten arkkitehtuurien, kuten keskitetyn tai hajautetun palvelinympäristön vaikutusta järjestelmän käyttövarmuuteen, käytettävyyteen sekä järjestelmän elinkaaren aikaisiin kustannuksiin. Tietojärjestelmien keskittämiseen ja konsolidointiin liittyen tutkitaan mm. vaikutuksia liityntöjen ja raportoinnin hallintaan sekä tietoverkoille asetettavia vaatimuksia. Työssä tutkitaan myös tuotepohjaisten järjestelmien räätälöintitarpeita ja siihen liittyvää vaatimusmäärittelyä. Työssä esitettyjä näkemyksiä voidaan käyttää päätösten tukena, kun pohditaan toteutettavan tietojärjestelmän teknisen infrastruktuurin optimaalisinta rakennetta.

Lopputyössä vertailtiin vesikiertoisen ja kaapeleilla toteutetun lattialämmityksen erilai-sia ominaisuuksia. Vertailuparametreinä olivat lattialämmitysten mitoitus, säätö, asen-nus, investointikustannukset, käyttökustannukset, vikaantuminen ja korjaus, asumis-viihtyvyys, käyttöön liittyvät tekijät, markkinat ja toimijat sekä lattian päällysteet. Näistä lat-tialämmityksien asumisviihtyvyys, säätö ja kustannukset otettiin tarkempaan vertailuun. Lämmitysjärjestelmien kokonaiskustannuksia vertailtiin 25 vuoden jaksolla ja 4 % ko-rolla. Investointikustannuksiin sisältyi lämmitysjärjestelmien lisäksi lkv-varaajan ja tek-nisen tilan kustannukset sekä liittymismaksut. Käyttökustannusten vertailussa otettiin huomioon lämmityskulujen lisäksi myös lämmityksen hyötysuhde, yö- ja päiväsähköosuudet ja -energian hinnat, taloussähkön kustannukset, perusmaksut sekä huoltokulut. Tutkimuksen mukaan halvin pientalojen lämmitysmuoto on osittain varaava huonekoh-tainen sähkölämmitys, jonka vuosittaisiksi kustannuksiksi tuli 11452 mk. Sähkölämmi-tyksen tekee vertailtavista halvimmaksi lämmitysjärjestelmäksi pientaloille (n. 150 m2) sen pienet pääomakustannukset, lämmityksen hyvä hyötysuhde sekä varsin kilpailuky-kyiset käyttökustannukset. Vertailun kallein oli öljyä energialähteenä käyttävä vesikiertoi-nen lattialämmitys, jonka vuosittaisiksi kustannuksiksi tuli 13080 mk. Öljy on energia-hinnaltaan halpa, mutta öljylämmityksen huono hyötysuhde sekä varsin korkeat pää-omakustannukset tekevät siitä vertailtavista kalleimman lämmitysmuodon. Energian hin-tojen herkkyysanalyysissä huomattiin sähkön säilyttävän kilpailukykynsä, vaikka öljyn ja maakaasun hinnat laskisivatkin huomattavasti. Asumismukavuuden ja säädön kohdalla vertailtiin lämpötilojen tasaisuutta huonetilois-sa. Mittauskohteiden huonelämpötiloja on seurattu eri projekteissa jatkuvatoimisesti jo-pa yli kahden vuoden ajan. Mittaustulosten mukaan vesikiertoisella lattialämmityksellä huonelämpötilat ovat tasaisemmat kuin osittain varaavalla huonekohtaisella sähköläm-mityksellä. Tämä johtuu sähkölämmityksen varaavuudesta. Hyvillä yhdistelmätermostaateilla päästään tosin myös huonekohtaisen sähkölämmityksen yhteydessä tasaisiin huonelämpötiloihin. Molempien lämmitysmuotojen säädön heikkoutena on hidas reagointi sisäisten lämmönlähteiden aiheuttamaan nopeaan sisälämpötilan nousuun.

The main purpose of this study was to examine and compare the possibilities of profit repatriation from the point of view of tax planning of an international corporation, in such a case that a Finnish parent company has a subsidiary in Poland. The main research problem was divided into two sub research problems: 1) to examine concepts and principles of international taxation and tax planning from the point of view of international corporations and 2) to discuss the main features of Polish Companies-, Accounting- and Tax Act from the point of view a Finnish parent company. The research method of this study is mainly decision making, comparative analysis. In this study have been discussed the possibilities of international profit repatriation for supporting the decision making of the management of a Finnish parent company. In addition different repatriation possibilities have been compared. In this study has been noticed that a Finnish parent company can repatriate profit of its Polish subsidiary either directly as dividends or by using indirect methods such as interests, royalties, management fees and transfer pricing of goods. The total tax burden of dividends is heavier than the tax burden of indirect methods. It was also concluded that during the last years the Polish legislation has been renewed in order to prevent hidden dividend distribution. This has been done by implementing new rules of transfer pricing and thin capitalization.

Tutkimuksessa on tarkoitus selvittää velallisen asemaa perinnässä ja perintäkulujen merkitystä. Tutkimus jakautuu teoreettiseen ja empiiriseen osaan, jossa selvitetään teoreettisesti velkaan ja perintään liittyviä käsitteitä ja lainsäädäntöä. Laki saatavien perinnästä sääntelee yleislakina perintätoimintaa riippumatta siitä suorittaako perintää itse velkoja vai perimistoimisto. Lakiin otettiin vuonna 2005 kuluttajasaatavia koskevia perimiskuluja rajoittavat enimmäismäärät, jotka on porrastettu saatavan suuruuden mukaan. Empiirisessä osassa tutkitaan oikeudenkäyntiaineistosta kerätyistä tiedoista perintään kuuluvia osapuolia, perittäviä saatavia ja perimiskuluja. Velallisen asema näyttää olevan riittävän hyvin turvattu oikeusjärjestyksessämme, joskin perimiskulujen ja oikeudenkulujen määrä nousee usein itse perittävää pääomaa suuremmaksi. Rahasaatavien perintää koskevien tapausten määrä käräjäoikeuksissa on kasvanut huomattavasti vuonna 2007. Tällaisia velkomuksia käsiteltiin käräjäoikeuksissa yli 200.000 kappaletta vuonna 2007. Tämän kasvun osaltaan selittää uudeksi velkaongelmien aiheuttajaksi nousemassa olevat pikavipit, joiden määrä kaikista perittävistä saatavista oli noin 10%. Näiden pääomat olivat keskiarvoltaan noin 145 euroa, ja näiden perintäkulut noin kaksinkertaisia verrattuna pääomaan Koko aineistossa perityt pääomat olivat keskiarvoltaan noin 2900 euroa ja perintäkulut noin 270 euroa.

Auditor independence is a cornerstone of the auditing profession and the basic principle that underpins the reputation of the auditing profession in the public eye. Indeed, it is the attribute most demanded from auditors by the public. Therefore, the sustainability of the auditing profession depends on how auditors can protect this principle. This dissertation consists of four interrelated essays concerned with auditor independence. Specifically, it examines situations that can threaten and impair auditor independence. In addition, this dissertation also examines several variables that may enhance and protect auditor independence. The first essay aims to examine the impact of social pressures occurring within audit firms on auditors’ judgment in the setting of a society with “high power distance” and “low individualism” cultural dimensions. The social pressures consisted of obedience pressure exerted by an auditor’s superior and conformity pressure exerted by an auditor’s colleague. Moreover, two moderating variables—a multi-dimensional professional commitment and locus of control—were included as moderator variables in the relationship between the social pressures faced by auditors and their judgment. The findings show that obedience and conformity pressures influence auditor judgment. Auditors who face the social pressures will make a judgment that may be even diametrically opposite to the independence principle. The findings also indicate that a multi-dimensional professional commitment and locus of control may potentially influence auditor judgment in a situation with social pressures. The second essay aims to investigate the association of advocacy and familiarity threats caused by auditor fee dependence and auditor tenure on auditor independence based on Finnish data, law, and auditing environment. This essay was motivated by the Green Paper on Audit Policy, published by the European Commission in 2010 that questions whether the maximum fee collected from a client should be regulated and whether consecutive assignments should be limited, among others. Contrary to popular belief, this essay does not find evidence that audit fees and long auditor tenure will jeopardise auditor independence. Therefore, the findings do not support policies to regulate auditor fees or limit auditor tenure in Finland. The third essay aims to examine the effect of client intimidation on auditor independence in an audit-client conflict situation. Intimidation threat is one of five independence threats that are explicitly referenced in the IFAC’s independence framework. Client intimidation was manifested in the client threatening to replace the auditor if the auditor did not adopt the client’s position. In addition, this essay examines the role of auditor’s perceived pressure and multi-dimensions of professional commitment as moderator variables. The findings suggest that auditors who experience client intimidation in an audit conflict situation are more likely to have their independence impaired than those who are in a similar situation but without client intimidation. Moreover, auditors who experience client intimidation perceive higher pressure than those who do not experience intimidation. Finally, auditors’ affective and continuance professional commitment dimensions moderate the relationship between auditors’ perceived pressures and auditor independence. The aim of the fourth essay is twofold. First, it aims to develop a scale for measuring auditors’ reputation awareness. Second, it aims to examine the correlation between the levels of auditor reputation awareness and auditor independence. A seven-item scale was developed as the reputation awareness scale. The findings indicate that the scale consists of one dimension. It also has a level of satisfactory reliability and a high level of validity. The findings show that there is a positive correlation between the level of auditors’ reputation awareness and auditor independence.

In this thesis different parameters influencing critical flux in protein ultrafiltration and membrane foul-ing were studied. Short reviews of proteins, cross-flow ultrafiltration, flux decline and criticalflux and the basic theory of Partial Least Square analysis (PLS) are given at the beginning. The experiments were mainly performed using dilute solutions of globular proteins, commercial polymeric membranes and laboratory scale apparatuses. Fouling was studied by flux, streaming potential and FTIR-ATR measurements. Critical flux was evaluated by different kinds of stepwise procedures and by both con-stant pressure and constant flux methods. The critical flux was affected by transmembrane pressure, flow velocity, protein concentration, mem-brane hydrophobicity and protein and membrane charges. Generally, the lowest critical fluxes were obtained at the isoelectric points of the protein and the highest in the presence of electrostatic repulsion between the membrane surface and the protein molecules. In the laminar flow regime the critical flux increased with flow velocity, but not any more above this region. An increase in concentration de-creased the critical flux. Hydrophobic membranes showed fouling in all charge conditionsand, furthermore, especially at the beginning of the experiment even at very low transmembrane pressures. Fouling of these membranes was thought to be due to protein adsorption by hydrophobic interactions. The hydrophilic membranes used suffered more from reversible fouling and concentration polarisation than from irreversible foul-ing. They became fouled at higher transmembrane pressures becauseof pore blocking. In this thesis some new aspects on critical flux are presented that are important for ultrafiltration and fractionation of proteins.

Tourism is one of the biggest industry branches with billions of tourists traveling every year around the world. Therefore, solutions providing tourist information have to be up to date with both changes in the industry and the world’s technological progress. The aim of this thesis is to present a design and a prototype of a tourist mobile service which is individual-oriented, cost-free for the end user, and secure. On the information providers’ side, the solution is implemented as a Webbased database. The end users access the information through a Bluetooth application on their mobile devices. The Bluetooth-based solution allows to avoid any costs for the end users, that is tourists. The study shows that, even with small data transfers, the tourists could save significantly when compared to possible roaming charges for data transfer. Also, the proposed mobile service is not intrusive, as it is provided through an application installed by tourists voluntarily on their mobile devices. Through design and implementation this work shows that it is possible to build a system which can be used to provide information services to tourists through mobile phones. The work achieved a successful ongoing synchronization between the client and the server databases. Implementation and usage were limited to smart phones only, as they provide better technological support for the solution having features like maps, GPS, Wi-Fi, Bluetooth and Databases. Moreover, the design of this system shows how Bluetooth technology can be used effectively as a means of communication while minimizing its shortcomings and risks, such as security, by bypassing Bluetooth server service discovery protocol (SDP) and connecting directly to the device. Apart from showing the design and implementation of the end-user costfree mobile information service, the results of this work also highlight the possible business opportunities to the provider of the service.

Purpose of the study is to evaluate performance of active portfolio management and the effect of stock market trend on the performance. Theory of efficient markets states that market prices reflect all available information and that all investors share a common view of future price developments. This view gives little room for the success of active management, but the theory has been disputed – at least the level of efficiency. Behavioral finance has developed theories that identify irrational behavior patterns of investors. For example, investment decisions are not made independent of past market developments. These findings give reason to believe that also the performance of active portfolio management may depend on market developments. Performance of 16 Finnish equity funds is evaluated during the period of 2005 to 2011. In addition two sub periods are constructed, a bull market period and a bear market period. The sub periods are created by joining together the two bull market phases and the two bear market phases of the whole period. This allows for the comparison of the two different market states. Performance of the funds is measured with risk-adjusted performance by Modigliani and Modigliani (1997), abnormal return over the CAPM by Jensen (1968), and market timing by Henriksson and Merton (1981). The results suggested that in average the funds are not able to outperform the market portfolio. However, the underperformance was found to be lower than the management fees in average which suggests that portfolio managers are able to do successful investment decisions to some extent. The study revealed substantial dependence on the market trend for all of the measures. The risk-adjusted performance measure suggested that in bear markets active portfolio managers in average are able to beat the market portfolio but not in bull markets. Jensen´s alpha and the market timing model also showed striking differences between the two market states. The results of these two measures were, however, somewhat problematic and reliable conclusions about the performance could not be drawn.

Consumers increasingly demand convenience when dealing with companies and therefore it is important to provide professional, diverse and speedy service via customer’s preferred communication channel. These interactions between the customer service and customer have a critical role in customer’s future purchasing decisions. Those customers who don't receive satisfactory customer service are willing to do business with another company that charges more but offers better customer service. This study identifies the critical success factors for the customer service in order to improve the customer service according to the company’s mission and meet customer expectations. Case study is used as a research method and data is collected via observation, archival records and interviews during a time span of fourteen months. The analysis suggests three critical success factors: voice support, scalable and flexible customer service and customer service champions. The study further analyzes the improvement measures according to the critical success factors concluding the Business Process Outsourcing to be the most proper to proceed with. As a conclusion of the study, critical success factors enable achieving the goals of the customer service and align operations according to the company’s mission. Business Process Outsourcing plays important role in improving the customer service by allowing fast expansion of new service offering and obtaining specialized workforce.

The greatest threat that the biodegradable waste causes on the environment is the methane produced in landfills by the decomposition of this waste. The Landfill Directive (1999/31/EC) aims to reduce the landfilling of biodegradable waste. In Finland, 31% of biodegradable municipal waste ended up into landfills in 2012. The pressure of reducing disposing into landfills is greatly increased by the forthcoming landfill ban on biodegradable waste in Finland. There is a need to discuss the need for increasing the utilization of biodegradable waste in regional renewable energy production to utilize the waste in a way that allows the best possibilities to reduce GHG emissions. The objectives of the thesis are: (1) to find important factors affecting renewable energy recovery possibilities from biodegradable waste, (2) to determine the main factors affecting the GHG balance of biogas production system and how to improve it and (3) to find ways to define energy performance of biogas production systems and what affects it. According to the thesis, the most important factors affecting the regional renewable energy possibilities from biodegradable waste are: the amount of available feedstock, properties of feedstock, selected utilization technologies, demand of energy and material products and the economic situation of utilizing the feedstocks. The biogas production by anaerobic digestion was seen as the main technology for utilizing biodegradable waste in agriculturally dense areas. The main reason for this is that manure was seen as the main feedstock, and it can be best utilized with anaerobic digestion, which can produce renewable energy while maintaining the spreading of nutrients on arable land. Biogas plants should be located close to the heat demand that would be enough to receive the produced heat also in the summer months and located close to the agricultural area where the digestate could be utilized. Another option for biogas use is to upgrade it to biomethane, which would require a location close to the natural gas grid. The most attractive masses for biogas production are municipal and industrial biodegradable waste because of gate fees the plant receives from them can provide over 80% of the income. On the other hand, directing gate fee masses for small-scale biogas plants could make dispersed biogas production more economical. In addition, the combustion of dry agricultural waste such as straw would provide a greater energy amount than utilizing them by anaerobic digestion. The complete energy performance assessment of biogas production system requires the use of more than one system boundary. These can then be used in calculating output–input ratios of biogas production, biogas plant, biogas utilization and biogas production system, which can be used to analyze different parts of the biogas production chain. At the moment, it is difficult to compare different biogas plants since there is a wide variation of definitions for energy performance of biogas production. A more consistent way of analyzing energy performance would allow comparing biogas plants with each other and other recovery systems and finding possible locations for further improvement. Both from the GHG emission balance and energy performance point of view, the energy consumption at the biogas plant was the most significant factor. Renewable energy use to fulfil the parasitic energy demand at the plant would be the most efficient way to reduce the GHG emissions at the plant. The GHG emission reductions could be increased by upgrading biogas to biomethane and displacing natural gas or petrol use in cars when compared to biogas CHP production. The emission reductions from displacing mineral fertilizers with digestate were seen less significant, and the greater N2O emissions from spreading digestate might surpass the emission reductions from displacing mineral fertilizers.

Antiviral nucleosides are compounds that are used against viruses, such as human immunodeficiency virus (HIV) and hepatitis C virus (HCV). To act as therapeutic agent, the antiviral nucleoside needs to be phosphorylated to nucleotide in the body in three consecutive phosphorylation steps by cellular or viral enzymes. The first phosphorylation to the nucleoside monophosphate is often inefficient and leads to poor antiviral activity. The antiviral efficacy can be improved by applying a prodrug strategy and delivering the antiviral nucleoside directly as its monophosphate. In prodrug strategies of antiviral nucleotides, the negative charges on the phosphate moiety are temporarily masked with protecting groups. Once inside the cell, the protecting groups are removed by enzymatic or chemical processes. Many prodrug strategies apply biodegradable protecting groups, the removal of which is triggered by esterase enzymes. Several studies have, however, demonstrated that the removal rate of the second and subsequent esterase labile protecting groups significantly slows down after the first protecting group is removed due to the negative charge on the phosphodiester intermediate, which disturbs the catalytic site of the enzyme. In this thesis, esterase labile protecting group strategies where the issue of retardation could be avoided were studied. Prodrug candidates of antiviral nucleotides were synthesized and kinetic studies on the chemical and enzymatic stability were carried out. In the synthesized compounds, the second protecting group is cleaved from the monophosphate some other mechanism than esterase triggered activation or the structure of prodrug requires only one protecting group. In addition, esterase labile protecting group which is additionally thermally removable was studied. This protecting group was cleaved from oligomeric phosphodiesters both enzymatically and thermally and seems most attractive of the studied phosphate protecting groups. However, the rate of the thermal removal still is too slow to allow efficient protection of longer oligonucleotides and needs optimization.
