O Citocromo P450 2D6 (CYP 2D6) é uma importante enzima metabolizadora de fármacos. Apesar de representar apenas 2% do total das isoenzimas CYP, o CYP 2D6 tem um papel importante pois é responsável pela metabolização de cerca de 20 a 25% dos fármacos mais frequentemente utilizados. (Ramamoorthy, 2010) É sabido que nem todos os indivíduos respondem da mesma maneira a um fármaco, podendo alguns sofrer reação adversa devido à toma deste e outros nem apresentar qualquer efeito terapêutico. Tais diferenças na resposta terapêutica devem-se a variações genómicas interindividuais, nos genes que codificam as enzimas responsáveis pelo metabolismo do fármaco. Os alelos polimórficos podem levar a uma redução ou aumento na capacidade metabólica, ao passo que um aumento do número de cópias do gene CYP 2D6 pode conduzir a um aumento da atividade metabólica. Os indivíduos comportam-se de acordo com o seu fenótipo, como metabolizadores lentos, rápidos ou ultra-rápidos (Abraham, 2001) Visto existirem grandes alterações nesta enzima metabolizadora de fármacos, é importante perceber quais as situações onde a segurança e a eficácia estão alteradas, pois qualquer terapêutica tem sempre como base o binómio risco/beneficio. Para além do seu papel bem estabelecido na segurança e eficácia terapêutica, estudos mais recentes relatam que o CYP 2D6 desempenha um papel importante no aparecimento de algumas doenças, condicionando a predisposição individual para patologias, de que é exemplo a esclerose sistémica. (Sanjay Harhang & al, 2001) Como desenvolvimento desta monografia pretende-se estudar os efeitos a nível de segurança e eficácia em determinadas situações terapêuticas e ainda avaliar a associação dos polimorfismos no CYP 2D6 com a predisposição individual para patologias.

The present dissertation examines how grammatical aspect and mood are handled by machine translation (MT) systems within the scope of imperative sentences (orders, recommendations) when dealing with the language pair French-Greek (unidirectional, towards Greek). As the grammatical category of aspect is not expressed in the same way in both languages, choosing the correct aspect value when translating a verb from French to Greek can pose problems. We are interested in describing the types of errors that occur and their frequency in a corpus taken from texts pertaining to the security domain and from technical manuals, where imperative sentences are very common. In order to further delimit our research, our sample consists of sentences that comply with the general principles of simplicity and readability provided by several controlled language guidelines and aimed at higher translatability when having MT in mind. In a second phase, this study aims at discovering how modifying some of the control rules would help (or not) the MT systems better decide upon the translation of aspect and mood.

The aquaculture industry aims at replacing significant amounts of marine fish oil by vegetable oils in fish diet. Dietary lipids have been shown to alter the fatty acid composition of bone compartments, which would impact the local production of factors controlling bone formation. Knowledge on the mechanisms underlying the nutritional regulation of bone metabolism is however scarce in fish. Two in vitro bone-derived cell systems developed from seabream (an important species for aquaculture in the Mediterranean region) vertebra, capable of in vitro mineralization and exhibiting prechondrocyte (VSa13) and pre-osteoblast (VSa16) phenotype, were used to assess the effect of certain polyunsaturated fatty acids (PUFAs; arachidonic (AA), eicosapentaenoic (EPA) and docosahexaenoic (DHA) acids) on cell proliferation, extracellular matrix (ECM) mineralization and gene expression. While all PUFAs promoted morphological changes in both cell lines, VSa16 cell proliferation appeared to be stimulated by PUFAs in a dose dependent manner until 100M, whereas proliferation of VSa13 cells was impaired at concentrations above 10M. AA, EPA and DHA inhibited VSa13 ECM mineralization, alone and in combination, while VSa16 ECM mineralization was only inhibited by AA and EPA. DHA had the opposite effect, increasing mineralization almost by 2 fold. When EFAs were combined, DHA apparently compensated for the inhibitory effect of AA and EPA. Expression of marker genes for bone and lipid metabolisms has been investigated by qPCR and shown to be regulated in pre-osteoblasts exposed to individual PUFAs. Our results show that PUFAs are effectors of fish bone cell lines, altering cell morphology, proliferation and mineralization when added to culture medium. This work also demonstrates the suitability of our in vitro cell systems to get insights into mineralization-related effects of PUFAs in vivo and to evaluate the replacement of fish oils by vegetable oil sources in fish feeds.

It is widely recognized that protein restriction in utero may cause metabolic and endocrine adaptations, which may be of benefit to the neonate on a short-term basis but may cause adverse long-term conditions such as obesity, Type 2 diabetes, metabolic syndrome, hypertension and cardiovascular diseases. Adequate foetal and early post natal nutrient and energy supply is therefore essential for adult animal health, performance and life span. In this project it was investigated the progressive adaptations of the hepatic proteome in male mink offspring exposed to either a low protein (FL) or an adequate protein (FA) diet in utero fed either on a low protein (LP) or on an adequate (AP) diet from weaning until sexual maturity. Specifically, the aim was to determine the metabolic adaptations at selected phases of the animal’s first annual cycle and establish the metabolic priorities occurring during those phases. The three different morphological stages studied during the first year of development included, end of bone growth at 4 months of age, maximal fat accretion at 6 months of age and sexual maturity at 12 months of age. A reference proteome of mink liver coming from these different animal groups were generated using 2D electrophoresis coupled to MALDI-TOF analysis and the way in which dietary treatment affect their proteome was established. Approximately 330 proteins were detected in the mink liver proteome. A total of 27 comparisons were carried out between all different animal groups which resulted in 20 differentially expressed proteins. An extensive survey was conducted towards the characterization of these proteins including their subcellular localization, the biological processes in which they are involved and their molecular functions. This characterization allowed the identification of proteins in various processes including the glycolysis and fatty acid metabolism. The detailed analysis of the different dietary treatment animal groups was indicative of differences in metabolism and also to changes associated with development in mink.

Cytochromes P450 constitute a super-family of enzymes involved in the metabolism of Xenobiotics, where human cytochrome P450 3A4 is the most abundant of all P450s, accounting for about 50% of all human liver cytochromes. This membrane anchored protein is responsible for the metabolization of a wide array of environmental drugs and intoxicants, mainly due to its haem domain properties, and active site cavity volume. These properties make this protein an excellent subject for biosensor application, although CYO3A4 enzyme is also famous for its instability. Enzyme inactivation at room temperature is a normal conversion process that this enzyme undergoes, that may hamper any biosensing approach.

A avaliação e gestão do desempenho organizacional são consideradas, atualmente, ferramentas essenciais para a gestão de qualquer empresa, pública ou privada. O setor da saúde não tem estado alheio a esta corrente e muitas são as publicações que apontam as potenciais vantagens associadas à avaliação e gestão de desempenho neste setor como forma de garantir a sustentabilidade das organizações. No entanto, não se conhece qual o estado de implementação dos sistemas de avaliação de desempenho nas organizações prestadoras de cuidados de saúde em Portugal, nem se conhece a perceção dos profissionais sobre a utilidade destes sistemas de avaliação. Esta dissertação procura assim dar um pequeno contributo para esta temática, fazendo um primeiro levantamento sobre o estado da implementação dos sistemas de avaliação de desempenho nos hospitais portugueses. Em específico, neste trabalho, estudamos, entre outros aspetos, qual a percentagem de hospitais e centros hospitalares públicos, privados e em parceria que estão a adotar modelos de avaliação de desempenho, que tipo de modelos estão a ser implementados, que tipo de indicadores estão a ser usados, o grau de eficácia percecionado da avaliação de desempenho, tal como razões subjacentes à não utilização da avaliação de desempenho ou à sua ineficácia. A metodologia usada para dar resposta aos objetivos desta investigação centrou-se no envio de questionários em formato papel para o Conselho de Administração de todos os hospitais e centros hospitalares a operar em Portugal no ano de 2012. Obtivemos uma taxa de resposta global de 58%, englobando os vários tipos de modelos de gestão dos hospitais portugueses. Os resultados obtidos revelam que a maioria dos hospitais da nossa amostra (87,5%) procede à avaliação de desempenho e que a utiliza como uma ferramenta de gestão. A minoria dos hospitais que não procede à avaliação regular de desempenho (na sua totalidade hospitais privados) aponta como principais justificações, o facto de usar outras ferramentas de controlo, o elevado investimento inicial requerido e a morosidade no processo de implementação. Na maioria dos hospitais que usa a avaliação de desempenho como uma ferramenta de gestão, esta é considerada como fundamental ou importante, permitindo não só a avaliação da organização como um todo, mas também a avaliação dos funcionários e a identificação de erros e medidas corretivas. A minoria dos hospitais que refere alguma ineficácia no sistema de avaliação de desempenho aponta como principais justificações para essa ineficácia, o fraco envolvimento das chefias, a elevada burocracia associada ao processo, a falta de concordância com os indicadores utilizados e a não execução da avaliação de acordo com o que tinha sido planeado inicialmente. Estes resultados são consistentes com os resultados de outros estudos internacionais relativos à implementação de sistemas de avaliação de desempenho em organizações públicas e privadas, e permitem enfatizar, por um lado, o potencial associado à avaliação de desempenho nas organizações prestadoras de cuidados de saúde, e por outro, a importância de não descurar certos aspetos de modo a garantir a sua eficácia.

The human genome has millions of genetics variants that can affect gene expression. These variants are known as cis-regulatory variants and are responsible for intra-species phenotypic differences and individual susceptibility to disease. One of the diseases affected by cis-regulatory variants is breast cancer. Breast cancer is one of the most common cancers, with approximately 4500 new cases each year in Portugal. Breast cancer has many genes mutated and TP53 has been shown to be relevant for this disease. TP53 is one of the most commonly mutated genes in human cancer and it is involved in cell cycle regulation and apoptosis. Previous work by Maia et al has shown that TP53 has differential allelic expression (DAE), which suggests that this gene may be under the influence of cis-regulatory variants. Also, its DAE pattern is totally altered in breast tumours with normal copy number. We hypothesized that cis-regulatory variants affecting TP53 may have a role in breast cancer development and treatment. The present work aims to identify the cis-regulatory variants playing a role in TP53 expression, using in silico, in vitro and in vivo approaches. By bioinformatic tools we have identified candidate cis-regulatory variants and predicted the possible transcription factor binding sites that they affect. By EMSA we studied DNA-protein interactions in this region of TP53. The in silico analysis allowed us to identified three candidate cis-regulatory SNPs which may affect the binding of seven transcription factors. However, the EMSA experiments have not been conclusive and we have not yet confirmed whether any of the identified SNPs are associated with gene expression control of TP53. We will carry out further experiments to validate our findings.

This work describes the synthesis of nanosized metal sulfides and respective SiO2 and/or TiO2 composites in high yield via a straightforward process, under ambient conditions (temperature and pressure), by adding to aqueous metals a nutrient solution containing biologically generated sulfide from sulfate-reducing bacteria (SRB). The nanoparticles‘ (NPs) morphological properties were shown not to be markedly altered by the SRB growth media composition neither by the presence of bacterial cells. We further extended the work carried out, using the effluent of a bioremediation system previously established. The process results in the synthesis of added value products obtained from metal rich effluents, such as Acid Mine Drainage (AMD), when associated with the bioremediation process. Precipitation of metals using sulfide allows for the possibility of selective recovery, as different metal sulfides possess different solubilities. We have evaluated the selective precipitation of CuS, ZnS and FeS as nanosized metal sulfides. Again, we have also tested the precipitation of these metal sulfides in the presence of support structures, such as SiO2. Studies were carried out using both artificial and real solutions in a continuous bioremediation system. We found that this method allowed for a highly selective precipitation of copper and a lower selectivity in the precipitation of zinc and iron, though all metals were efficiently removed (>93% removal). This research has also demonstrated the potential of ZnS-TiO2 nanocomposites as catalysts in the photodegradation of organic pollutants using the cationic dye, Safranin-T, as a model contaminant. The influence of the catalyst amount, initial pH and dye concentration were also evaluated. Finally, the efficiency of the precipitates as catalysts in sunlight mediated photodegradation was investigated, using different volumes of dye-contaminated water (150 mL and 10 L). This work demonstrates that all tested composites have the potential to be used as photocatalysts for the degradation of Safranin-T.

All systems found in nature exhibit, with different degrees, a nonlinear behavior. To emulate this behavior, classical systems identification techniques use, typically, linear models, for mathematical simplicity. Models inspired by biological principles (artificial neural networks) and linguistically motivated (fuzzy systems), due to their universal approximation property, are becoming alternatives to classical mathematical models. In systems identification, the design of this type of models is an iterative process, requiring, among other steps, the need to identify the model structure, as well as the estimation of the model parameters. This thesis addresses the applicability of gradient-basis algorithms for the parameter estimation phase, and the use of evolutionary algorithms for model structure selection, for the design of neuro-fuzzy systems, i.e., models that offer the transparency property found in fuzzy systems, but use, for their design, algorithms introduced in the context of neural networks. A new methodology, based on the minimization of the integral of the error, and exploiting the parameter separability property typically found in neuro-fuzzy systems, is proposed for parameter estimation. A recent evolutionary technique (bacterial algorithms), based on the natural phenomenon of microbial evolution, is combined with genetic programming, and the resulting algorithm, bacterial programming, advocated for structure determination. Different versions of this evolutionary technique are combined with gradient-based algorithms, solving problems found in fuzzy and neuro-fuzzy design, namely incorporation of a-priori knowledge, gradient algorithms initialization and model complexity reduction.

Cardiogenesis is a delicate and complex process that requires the coordination of an intricate network of pathways and the different cell types. Therefore, understanding heart development at the morphogenetic level is an essential requirement to uncover the causes of congenital heart disease and to provide insight for disease therapies. Mouse Cerberus like 2 (Cerl2) has been defined as a Nodal antagonist in the node with an important role in the Left-Right (L/R) axis establishment, at the early embryonic development. As expected, Cerl2 knockout mice (Cerl2-/-) showed multiple laterality defects with associated cardiac failure. In order to identify the endogenous role of Cerl2 during heart formation independent of its described functions in the node, we accurately analyzed animals where laterality defects were not present. We thereby unravel the consequences of Cerl2 lossof- function in the heart, namely increased left ventricular thickness due to hyperplasia of cardiomyocytes and de-regulated expression of cardiac genes. Furthermore, the Cerl2 mutant neonates present impaired cardiac function. Once that the cardiac expression of Cerl2 is mostly observed in the left ventricle until around midgestration, this result suggest a specific regulatory role of Cerl2 during the formation of the left ventricular myoarchitecture. Here, we present two possible molecular mechanisms underlying the cardiac Cerl2 function, the regulation of Cerl2 antagonist in activation of the TGFßs/Nodal/Activin/Smad2 signaling identified by increased Smad2 phosphorilation in Cerl2-/- hearts and the negative feedback between Cerl2 and Wnt/ß-catenin signaling in heart formation. In this work and since embryonic stem cells derived from 129 mice strain is extensively used to produce targeted mutants, we also present echocardiographic reference values to progressive use of juveniles and young adult 129/Sv strain in cardiac studies. In addition, we investigate the cardiac physiology of the surviving Cerl2 mutants in 129/Sv background over time through a follow-up study using echocardiographic analysis. Our results revealed that Cerl2-/- mice are able to improve and maintain the diastolic and most of systolic cardiac physiologic parameters as analyzed until young adult age. Since Cerl2 is no longer expressed in the postnatal heart, we suggest that an intrinsic and compensatory mechanism of adaptation may be active for recovering the decreased cardiac function found in Cerl2 mutant neonates. Altogether, these data highlight the role of Cerl2 during embryonic heart development in mice. Furthermore, we also suggest that Cerl2-/- may be an interesting model to uncover the molecular, cellular and physiological mechanisms behind the improvement of the cardiac function, contributing to the development of therapeutic approaches to treat heart failures.
