The porcine stress syndrome or malignant hyperthermia is an inherited autosomic recessive disease, which results in neuromuscular disorders leading to death in homozygous individuals and is associated with deterioration of meat quality. The defect in susceptible animals results from modifications in the calcium release channel or Ryanodine Receptor (RYR1), with a mutation leading to a C to T transition in nucleotide 1843 of the gene. The objective of this work was to develop a method based on analysis of SNPs to detect the mutation described in the RYR1 locus in pigs, and study polymorphisms of the gene in four exotic (Large White, Landrace, Duroc and Pietrain) and three native (Bísaro, Alentejano and Malhado de Alcobaça) breeds of pigs in Portugal. The method was successful in identifying the mutation by analysis of SNPs, and results indicate a high incidence of the mutant allele in Pietrain (0.75) and, to a lesser degree, in Malhado de Alcobaça (0.34) and Landrace (0.28); frequencies in Alentejano, Bísaro and Large White ranged between 0.04 and 0.09. These results suggest the need to establish breeding programs aimed at eliminating the susceptibility allele from those populations.

Background: Native pig breeds in the Iberian Peninsula are broadly classified as belonging to either the Celtic or the Mediterranean breed groups, but there are other local populations that do not fit into any of these groups. Most of the native pig breeds in Iberia are in danger of extinction, and the assessment of their genetic diversity and population structure, relationships and possible admixture between breeds, and the appraisal of conservation alternatives are crucial to adopt appropriate management strategies. Methods: A panel of 24 microsatellite markers was used to genotype 844 animals representing the 17 most important native swine breeds and wild populations existing in Portugal and Spain and various statistical tools were applied to analyze the results. Results: Genetic diversity was high in the breeds studied, with an overall mean of 13.6 alleles per locus and an average expected heterozygosity of 0.80. Signs of genetic bottlenecks were observed in breeds with a small census size, and population substructure was present in some of the breeds with larger census sizes. Variability among breeds accounted for about 20% of the total genetic diversity, and was explained mostly by differences among the Celtic, Mediterranean and Basque breed groups, rather than by differences between domestic and wild pigs. Breeds clustered closely according to group, and proximity was detected between wild pigs and the Mediterranean cluster of breeds. Most breeds had their own structure and identity, with very little evidence of admixture, except for the Retinto and Entrepelado varieties of the Mediterranean group, which are very similar. Genetic influence of the identified breed clusters extends beyond the specific geographical areas across borders throughout the Iberian Peninsula, with a very sharp transition from one breed group to another. Analysis of conservation priorities confirms that the ranking of a breed for conservation depends on the emphasis placed on its contribution to the betweenand within-breed components of genetic diversity. Conclusions: Native pig breeds in Iberia reveal high levels of genetic diversity, a solid breed structure and a clear organization in well-defined clusters.

- The aim of this study was the process of construction and validation of an observation instrument. In this context, we focused the analysis on a fundamental requirement - the validity and reliability of the instrument. This instrument will serve as basis for the registration of behaviors observed during the execution of twenty gestural cycles of the breaststroke technique. The instrument consisted of a Field Formats system (Oliveira, Campaniço & Anguera, 2001), based on references of the observational methodology and Swimming biomechanical models (Persyn & Colman, 1993; Silva & Alves, 2000; Louro, Garrido, Ferraz, Marinho, Conceição, Tolentino, Barbosa & Silva, 2009), giving special emphasis to five taxonomic criteria that aggregate, in the form of alpha-numeric codes, crucial information to describe behaviors that define the breaststroke technique. To ensure the instrument’s accuracy and objectivity, an analysis of the instrument’s quality and validity was made, using the Kappa Index (Anguera, 1993; Blanco 1993, 1997). This analysis was based on records of five trained observers (inter-observer, 96.9%) and, for accuracy purposes, another expert (intraobserver, 97.8%). In conclusion, it is possible to determine that, given the high reliability values, the validity of this qualitative instrument is evident, making it a suitable instrument to observe the technical behavior in breaststroke swimmers.
