Stromal vascular fraction cell sheets angiogenic potential for tissue engineering and regenerative medicine applications

One of the biggest concerns in the Tissue Engineering field is the correct vascularization of engineered constructs. Strategies involving the use of endothelial cells are promising but adequate cell sourcing and neo-vessels stability are enduring challenges. In this work, we propose the hypoxic pre-conditioning of the stromal vascular fraction (SVF) of human adipose tissue to obtain highly angiogenic cell sheets (CS). For that, SVF was isolated after enzymatic dissociation of adipose tissue and cultured until CS formation in normoxic (pO2=21%) and hypoxic (pO2=5%) conditions for 5 and 8 days, in basal medium. Immunocytochemistry against CD31 and CD146 revealed the presence of highly branched capillary-like structures, which were far more complex for hypoxia. ELISA quantification showed increased VEGF and TIMP-1 secretion in hypoxia for 8 days of culture. In a Matrigel assay, the formation of capillary-like structures by endothelial cells was more prominent when cultured in conditioned medium recovered from the cultures in hypoxia. The same conditioned medium increased the migration of adipose stromal cells in a scratch assay, when compared with the medium from normoxia. Histological analysis after implantation of 8 days normoxic- and hypoxic-conditioned SVF CS in a hindlimb ischemia murine model showed improved formation of neo-blood vessels. Furthermore, Laser Doppler results demonstrated that the blood perfusion of the injured limb after 30 days was enhanced for the hypoxic CS group. Overall, these results suggest that SVF CS created under hypoxia can be used as functional vascularization units for tissue engineering and regenerative medicine.

O ecrã da (hiper) televisão: novos olhares a partir das emissões dedicadas ao Euro 2012 na TV portuguesa

Dando continuidade a uma série de estudos já realizados no âmbito do projecto de investigação em que se insere, o presente trabalho questiona a evolução do ecrã televisivo como dispositivo supostamente convergente. Contrariando uma tendência discursiva dominante, quer na esfera dos media quer na própria produção académica sobre o medium televisivo, clarificam-se aqui alguns dos desenvolvimentos teóricos mais significativos, resultantes dos anos de trabalho que este projecto de investigação já tem. Propõe-se, pois, uma distinção conceptual entre o ecrã prometido pela cultura da convergência, que designamos como ecrã convergente, e o ecrã verdadeiramente resiliente da instituição-televisão, o ecrã centrípeto, que ainda obedece aos princípios de sequência e fluxo enunciados por Raymond Williams nos anos 1970.

Caught between changing tides: Gender and kinship in Cape Verde

Gender ideologies in Cape Verde are shifting. Individuals find themselves caught between changing tides, pushed and pulled in opposite directions by divergent gendered expectations. The article examines the different ways in which young women and men take recourse to tactics in response to the tensions that arise as they deal with changing gender ascriptions in the midst of their relations with community and kin. Women, in particular, are unevenly affected by traditional demands and expectations whilst they cross the boundaries of traditional gender roles in their pursuit of enhanced education and more sexual freedom. Yet, their actions are not characterized by an outright rejection of traditional gender ideologies, but rather by piecemeal tactical manoeuvres to plot a route through the centrifugal forces at play. keywords

Using OpenFOAM to aid the design of complex profile extrusion dies

[Extrat] Thermoplastic profiles are very attractive due to their inherent design freedom. However, the usual methodologies employed to design extrusion forming tools, based on experimental based trial–and–error procedures, are highly dependent on the designer’s experience and lead to high resources consumption. Despite of the relatively low cost of the raw materials employed on the production of this type of profiles, the resources involved in the die design process significantly increase their cost. These difficulties are even more evident when a complex geometry profile has to be produced and there is no previous experience with similar geometries. Therefore, novel design approaches are required, in order to reduce the required resources and guarantee a good performance for the produced profile. (...)
