This paper describe a Ru(bpy)(3)(2+) based electrochemiluminescence (ECL) method to detect procyclidine in human urine following separation by capillary electrophoresis (CE). An ECL detection cell was designed for post-column addition of Ru(bpy)(3)(2+). Parameters affecting separation and detection were optimized, leading to a detection limit of 1 x 10(-9) mol/l in an on-capillary stacking mode. For application in urine, a cartridge packed with slightly acidic cation-exchange resin was used to eliminate the matrix effects of urine and improve the detection sensitivity. Extraction recovery was nearly 90%.

In the present study, curcumin from Chinese herbal medicine turmeric was determined by capillary electrophoresis with amperometric detection (CE-AD) pretreated by a self-designed, simple, inexpensive solid-phase extraction (SPE) cartridge based on the material of tributyl phosphate resin. An average concentration factor of 9 with the recovery of >80% was achieved when applied to the analysis of curcumin in extracts of turmeric. Under the optimized CE-AD conditions: a running buffer composed of 15 mM phosphate buffer at a pH 9.7, separation voltage at 16 W, injection for 6 s at 9 W and detection at 1.20 V, CE-AD with SPE exhibited low detection limit as 3 - 10(-8) mol/l (SIN = 3), high efficiency of 1.0(.)10(5) N, linear range of 7(.)10(-4) -3(.)10(-6) mol/l (r = 0.9986) for curcumin extracted from light petroleum. The method developed resulted in enhancement of the detection sensitivity and reduction of interference from sample matrix in complicated samples and exhibited the potential application for routine analysis, especially in food, because a relatively complete process of sample treatment and analysis was described.

A new detection scheme for the determination of adsorbable coreactants of Ru(bpy)(3)(2+) electrochemiluminescent reaction is presented. It is based on selective preconcentration of coreactant onto an electrode, followed by Ru(bpy)(3)(2+) electrochemiluminescent detection. The coreactant employed is chlorpromazine. It was sensitively detected after 5-min preconcentration onto a lauric acid-modified carbon paste electrode. The linear concentration range was found to occur from 1 x 10(-8) to 3 x 10(-6) mol L-1 with a detection limit of 3.1 x 10(-9) mol L-1. The total analysis time is less than 10 min. As a result of selective preconcentration and medium exchange, such remarkable selectivity is achieved that reproducible quantitation of chlorpromazine in urine is possible.

The biosensor based on surface plasmon resonance(SPR) technology is a very useful tool to study the interaction between biomolecles. The main advantages of this technique is to "visualize" macromolecular interactions directly in real time, and in a label-free mode rather than indirect methods like enzyme-linked immunosorbent assays (ELISAs). We immobilize human serum albumin (HSA) to the carboxymethyldextran-modified sensor chip surface covalently to detect the activity of anti-HSA in serum, and regenerate the surface with .1 mol/L phosphoric acid. The results show that SPR biosensor can detect the activity of anti-HSA in real-time quickly and the sensor chip can be used over 100 cycles.

A new type of tyrosinase biosensor was developed for the detection of phenolic compounds, based on the immobilization of tyrosinase in a sol-gel-derived composite matrix that is composed of titanium oxide sol and a grafting copolymer of poly(vinyl alcohol) with 4-vinylpyridine. Tyrosinase entrapped in the composite matrix can retain its activity to a large extent owing to the good biocompatibility of the matrix. The parameters of the fabrication process and the variables of the experimental conditions for the enzyme electrode were optimized. The resulting sensor exhibited a fast response (20 s), high sensitivity (145.5 muA mmol(-1) 1) and good storage stability. A detection limit of 0.5 muM catechol was obtained at a signal-to-noise ratio of 3.

A novel glucose biosensor based on capacitive detection has been developed using molecularly imprinted polymers. The sensitive layer was prepared by electropolymerization of o-phenylenediamine on a gold electrode in the presence of the template (glucose). Cyclic voltammetry and capacitive measurements monitored the process of electropolymerization. Surface uncovered areas were plugged with 1-dodecanethiol to make the layer dense, and the insulating properties of the layer were studied in the presence of redox couples. The template molecules and the nonbound thiol were removed from the modified electrode surface by washing with distilled water. A capacitance decrease could be obtained after injection of glucose. The electrode constructed similarly but with ascorbic acid or fructose only showed a small response compared with glucose. The stability and reproducibility of the biosensor were also investigated. Determination of aesculin (AL) and aesculetin (AT) by capillary electrophoresis with end-column amperometric detection using a 33 mu m microdisk carbon fiber electrode is described. The HDVs, the effect of pH, buffer concentration, injection voltage, injection time and separation voltage on the peak current response (i(p)) of the analytes and the number of theoretical plates (N) were studied. The method has high sensitivity and good reproducibility. Under the optimum condition - 10 mM, pH 9.00 phosphate buffer, 4 s at 9 kV injection, separation at 15 kV and +1.0 V as the detection potential - low detection limits (S/N = 3) of 0.06 and 0.3 mu M were obtained for AL and AT, respectively. The calibration curve was linear over three orders of magnitude. The relative standard deviations (n = 15) of peak current and migration time were 3.9% and 4.6%, and 0.96% and 0.75% for 15 consecutive injections of 5 mu M AL and AT, respectively. The use of this method for the separation and detection of the two compounds present in the traditional Chinese medicine and human urine samples is also reported. (C) 1999 Elsevier Science B.V. End-column amperometric detection of 6-mercaptopurine by capillary zone electrophoresis was described with high resolution and speed. The detection conditions were optimized and the electrochemical behavior was observed. Under the optimal conditions: detection potential of 1.2 V ( vs. Ag/AgCl), operating voltage of 15 kV, sample injection of 3 s at 15 kV and 10 mmol/l Na2HPO4 buffer, the detection limit for 6-MP was as low as 1 x 10(-7) mol/L and the linear range was from 5 x 10(-4) to 5 x 10(-6) mol/L with the relative coefficient of 0.995. The RSD of reproducibility for peak current and migration time was 2.5% and 1.2%, respectively. This method was utilized in assay real sample of human mine and bovine serum albumin containing 6-mercaptopurine.

A rapid and sensitive detection method for the determination of 5-fluorouracil(5-FU) in real samples such as human urine and bovine serum albumin (BSA) was described. A carbon fiber microdisk electrode was used to perform end-column amperometric detection in capillary zone electrophoresis. The detection limit was as low as 2.5x10(-7) M and the wider linear range for the concentration was between 5x10(-6) and 1x10(-4) M with a correlation coefficient of 0.995.

Capillary electrophoresis (CE)/electrochemical detection (EC) for the simultaneous detection of hydrazine, methylhydrazine, and isoniazid has been developed with a 4-pyridyl hydroquinone self-assembled microdisk platinum electrode. Such an electrode has very high catalytic ability for hydrazines and they could be detected even at 0.0 V. The responses for hydrazine, methylhydrazine, and isoniazid are linear over 3 orders of detected concentration and of magnitude of 0.2-400 mu M, 0.2-400 mu M, 0.5 mu M-2 mM, with correlation coefficients of 0.9998, 0.9991, and 0.9982, respectively. And they could be detected to levels of 0.1, 0.1 and 0.2 mu M, respectively. This modified electrode was found to be very stable and reproducible when continuously used as detector for capillary electrophoresis for period of at least 4 weeks with no apparent loss of response. Matrix-assisted laser desorption ionization (MALDI) mass spectrometry is difficult for the characterization of noncovalent complexes hitherto because of the limitations in acidic matrix, sample preparation, laser-induced polymerization and adduct formation with matrix. Under our experimental conditions, sinapinic acid is used as a matrix, the specific noncovalent interactions of protein with fullerenols were observed by MALDI mass spectrometry. Some mass spectrometric features, such as mass shifts, broad adduct peaks and stoichiometries, showed that the specific non-covalent complexes between protein and fullerenols have been formed at a ratio of 1 : 4 for hemoglobin-fullerenols or 1 : 1 for myoglobin-fullerenols. The results implied that fullereneols could be used to protect partly hemoglobin from decomposition in acidic media, and therefore, it is possible to realize the molecular weight determination of a quaternary protein by MALDI mass spectrometry via the addition of specific organic compound in the matrix.

A poly(4-vinyl)pyridine (PVP)/Pd film electrode was constructed for the electrocatalytic detection of hydrazine. The preparation of the PVP/GC electrode was performed by electropolymerization of the monomer 4-vinylpyridine onto the surface of a glassy carbon electrode. Subsequently, palladium is electrodeposited onto the polymer modified electrode surface. The ion-exchange function of PVP polymer is helpful to this process in view of the tetrachlorapalladate anion. Compared with the Pd/GC electrode, the modified electrode displays a better mechanical stability in a flowing stream. The PVP/Pd film electrode exhibits higher sensitivity when detecting hydrazine with a detection limit of 0.026 ng (S/N=3).

Chromatography-amperometric detection of nitrite with a polypyrrole modified glassy carbon electrode doped with tungstodiphosphate anion (Dawson-type P2W18O626-/PPy/GC electrode) based on its electrocatalytic reduction of nitrite is described. The cyclic and hydrodynamic voltammetry of nitrite at the P2W18O626-/PPy/GC electrode was studied. The factors affecting the detection of nitrite and the analytical performance of the modified electrode in flowing stream were investigated. The results show that the modified electrode has a good sensitivity (the limit of detection is 1 mu mol dm(-3)) and a satisfactory reproducibility (RSD = 3.78%, N = 21). The modified electrode was used in the chromatographic detection of nitrite spiked in the liquid from a tin of mushrooms and the mineralized spring water. It was found that the modified electrode exhibited good selectivity for nitrite.

A reversed-phase high-performance liquid chromatography with series dual glassy carbon electrodes for the amperometric detection of water-soluble menadione is described. The complex post-column derivatization reaction and the high background currents were avoided. The menadione sodium bisulfite was reduced at -0.3 V vs. SCE at the upstream (generator) electrode and oxidized at +0.2V vs. SCE at the downstream (collector) electrode. The mobile phase was 0.2moll(-1) HAc-NaAc aqueous buffer (pH 5.50) and 40% (v/v) methanol. The linear response was in the range of 35 ng to 15 mu g, with a detection Limit of 15 ng (S/N=3). The correlation coefficient was 0.9997 (n=6). The electrochemical detection with series dual electrodes has a higher selectivity for menadione (vitamin K-3) compound than with UV detection.

A palladium particle-modified carbon fiber microdisk array electrode was designed and employed in capillary electrophoresis for the simultaneous detection of hydrazine, methylhydrazine, and isoniazid. The Pd-modified microdisk electrode had high catalytic ability for hydrazines and exhibited good reproducibility and stability. The response for hydrazine was linear over 3 orders of magnitude with a correlation coefficient of 0.993. The detection limits far hydrazine, methylhydrazine, and isoniazid were 1.2, 2.1, and 6.2 pg, respectively.
