To test preschoolers’ development of cognitive flexibility--an ability to solve a problem in one way and to then switch solution strategies, and the mechanism involved in the development, 3-5-year-olds are asked to perform switching tasks in which the experimenter manipulates the way the stimuli are presented: consecutive or simultaneous; the way the switching happens: between dimensions or within a dimension; the conceptual domains involved: shape, color, number and direction; the specific labels used. The main results of this work are presented below: (1) 3-5-year-olds’ cognitive flexibility develops with age, yet its development is not of the same speed in extra-dimensional switch tasks and inter-dimensional reversal tasks. 3-year-olds manifest some cognitive flexibility, but their performance is significantly worse than that of 4- and 5-year-olds. For the 3-year-olds, in reversal tasks, although 80% of the children passed the post-switch phrase in color task; less then 60% children passed the post-switch phrase in shape, number and direction tasks. In extra-dimensional tasks, 3-year-olds performance is worse than that in the reversal tasks. Less than 50% of the children passed the tasks. Children’s cognitive flexibility develops fast from 3-year-olds to 4-year-olds. Both 4-year-olds and 5-year-olds demonstrate high flexibility without significant difference between them. (2) Children’s flexibility in the conceptual domains of shape, color, number and direction follows different developing patterns. In inter-dimensional reversal tasks, 3-year-olds’ performance is not the same in the 4 conceptual domains, but the difference among the domains is insignificant in 4-and-5-year-olds. In extra-dimensional switching tasks, children’s performance on the 4 domain tasks is significantly different from one another in 3-, 4-, and 5-year-olds. (3) The way the stimuli are presented affects children’s development of cognitive flexibility. In inter-dimensional reversal tasks, 3-year-olds’ performance in consecutive presentation is significantly better than that in simultaneous presentation. 4- and 5-year-olds’ performance in the 2 presentations is not significantly different from each other. In extra-dimensional switch tasks, 3-, 4-, and 5-year-olds’ performance in the consecutive presentation is not significantly better than that in the simultaneous presentation (4) 3-, 4-, and 5-year-olds’ self-issued labeling aids their performance on the switching tasks. Children’ performance in the labeling condition is significantly better than that of no labeling. (5) 3-5-year-olds’ cognitive flexibility is highly correlated with their working memory and inhibition. Children’ development of cognitive flexibility is a process that involves activation of working memory and inhibition, in which the complexity of the task also plays a role.

We present numerical simulations of thermosolutal convection for directional solidification of Al-3.5 wt% Ni and Al-7 wt% Si. Numerical results predict that fragmentation of dendrite arms resulting from dissolution could be favored in Al-7 wt% Si, but not in Al-3.5 wt% Ni. Corresponding experiments are in qualitative agreement with the numerical predictions. Distinguishing the two fragmentation mechanisms, namely dissolution and remelting, is critical during experiments on earth, when fluid flow is dominant. (C) 2007 COSPAR. Published by Elsevier Ltd. In this paper, we demonstrated a dual-wavelength competitive output in Nd:Y3SC1.5Al3.5O12 ceramic disk laser. Different dual-wavelength output behaviors for Nd:YSAG and Nd:YAG ceramic disk laser were investigated and discussed. By applying the energy transfer model, we suggested the reasonable explanation for this new phenomenon as the disordered replacing of Al3+ ions by Sc3+ ions. The main advantage of the dual-wavelength ceramic laser is the possibility to serve as the seed source to generate Terahertz radiation. (C) 2008 Elsevier B.V. All rights reserved.

植物通过异戊二烯代谢途径合成多种具有生物活性和功能的三萜及甾醇类化合物,它们在调节植物生长发育、维持膜的完整和功能、抵抗病原微生物侵染中发挥着重要的作用。2,3-氧化鲨烯为三萜和甾醇合成途径的分枝点,参与这一关键步骤的酶被通称为2,3-氧化鲨烯环化酶(OSCs)。本研究系统分了水稻基因组中全部11个OSC基因序列,发现其中四个可能为假基因。亚种间非同义替换率Ka和同义替换率Ks的比值(Ka/Ks)以及进化树的分析表明OsOSC8是单子叶植物特有的功能保守基因,而OsOSC9在水稻两个亚种间发生了功能快速进化,这种快速进化的基因往往参与植物和病原菌相互作用的代谢途径。 根据基因结构、表达谱以及与其它植物已知功能的OSC酶氨基酸序列的比对推测OsOSC3可能具有环阿屯醇合成酶的功能,参与植物甾醇的合成,而OsOSC7、OsOSC10和OsOSC11可能具有β-香树素合成酶的功能,其余OSCs可能参与合成其它三萜化合物。为了进一步分析和验证OSCs酶的功能,将水稻7个OSC基因的开放阅读框(ORF)构建到酵母表达载体并在pichia酵母中表达,发现仅有OsOSC9和OsOSC12能够将酵母内源的2,3-氧化鲨烯分别环化为四环三萜化合物Parkeol和植物中稀有的五环三萜化合物Isoarborinol,目前还未在其它植物中发现参与这两种三萜化合物的基因。另外,水稻所有的OSC基因均不能互补酵母羊毛甾醇缺陷型菌株,表明水稻OSCs不具有合成羊毛甾醇的功能。 RNAi沉默以及启动子融合GUS的表达实验发现OsOSC8可能参与花粉的发育,该基因的下调影响水稻的育性,暗示水稻中存在一个可能与雄性不育有关的三萜代谢途径。水稻其它OSC基因RNAi植株可能在逆境环境和病原菌侵染下才会显现出表型。

通过测定原代培养鲫鱼(Carassius auratus)肝细胞中雌激素受体所介导的卵黄蛋白原(Vtg)生成以及芳香烃受体所介导的CYP1A1基因转录水平的变化, 建立了一种类雌激素体外实验模型. 实验结果表明, Vtg和Vtg mRNA的表达与己烯雌酚(DES)之间均有很好的剂量-效应关系, Vtg和Vtg mRNA均可作为指示类雌激素毒性的生物标志物. TCDD, B[a]P可显著抑制鱼肝细胞中DES诱导的Vtg和Vtg mRNA的表达, 呈明显的抗雌激素效应, 并同时激活了CYP1A1 基因的表达;

The sexual ratio of Gobiocypris rarus exposed to 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and 17 beta -estradiol from embryo to sexually mature revealed feminization and overdevelopment of connective tissue in male fish gonad in 2-30 pg/L TCDD concentration range. Daphnia magna was not sensitive to the high dose of TCDD (0.1-1000 ng/ml), but the reproduction of D. magna treated with TCDD decreased after the 8th day. 7-Ethoxyresorufin-O-deethylase (EROD) activities in newly fertilized eggs of G. rarus exposed to TCDD dosage groups (1000-100,000 pg/L) were significantly induced and increased with TCDD concentrations at the early life stage, while no difference was found between low TCDD dosage groups (<100 pg/L), but a good relationship between the EROD activity and the TCDD concentration was observed during a long-term developmental stage. There was a pericardial edema formed in a 2-week yolk-sac at the concentration of 1000 pg/L TCDD. But in the exposure group (2 pg/L TCDD for 120 days), the cell nuclei of hepatocytes was far from the center and packed toward the cell membrane; the cristae of most mitochondria in the cell dropped and collapsed; the rough endoplasmic reticulum broke into fragments; and numerous lipid droplets formed in the cell. In this study, the possibility of establishing a dual-species biofilm from a bacterium with a high biofilm-forming capability and a 3,5-dinitrobenzoic acid (3,5-DNBA)-degrading bacterium, Comamonas testosteroni A3, was investigated. Our results showed that the combinations of strain A3 with each of five strains with a high biofilm-forming capability (Pseudomonas sp. M8, Pseudomonas putida M9, Bacillus cereus M19, Pseudomonas plecoglossicida M21 and Aeromonas hydrophila M22) presented different levels of enhancement regarding biofilm-forming capability. Among these culture combinations, the 24-h dual-species biofilms established by C. testosteroni A3 with P. putida M9 and A. hydrophila M22 showed the strongest resistance to 3,5-DNBA shock loading, as demonstrated by six successive replacements with DMM2 synthetic wastewater. The degradation rates of 3,5-DNBA by these two culture combinations reached 63.3-91.6% and 70.7-89.4%, respectively, within 6 h of every replacement. Using the gfp-tagged strain M22 and confocal laser scanning microscopy, the immobilization of A3 cells in the dual-species biofilm was confirmed. We thus demonstrated that, during wastewater treatment processes, it is possible to immobilize degrader bacteria with bacteria with a high biofilm-forming capability and to enable them to develop into the mixed microbial flora. This may be a simple and economical method that represents a novel strategy for effective bioaugmentation.
