Traditional Chinese Medicine (TCM) based on natural products is one important part of the Chinese civilization.Owing to the complexity of the composition,the study on medical effective components and curative effects are very difficult;Fuzi (Radix Aconiti Lateralis Praeparata) has been widely used for cardiotonic and analgesics in China,however,the component-aconitine in which is very toxic and may cause some side-effect.This paper reported the application of electrospray iohization mass spectrometric(ESIMS) technique on study of the compound prescriptions containing fuzi,Renshensini concoction and Baweidihuang concoction,to explore the mechanism of synergy between fuzi and other herbs.From chemical point of view,the detoxification mechanism can be attributed to the hydrolysis of the solubility of toxic diester-alkaloids from concoctions of fuzi.Radix Glycyrrhizae Praeparata,Rhizoma Zingiberis,Radix Ginsengp promote the hydrolysis to produce the less toxic monoester-alkaloids and Fructus Corni reduce the solubility of hypaconitine from fuzi.

Simple, convenient, sensitive and accurate analytical methods are needed for the structural characterization and identification of alkaloid components in Rhizoma Coptidis in traditional Chinese herbal medicine, which has important bioactivity. In this work, the identification of alkaloid compounds in Rhizoma Coptidis was investigated by obtaining molecular mass information using electrospray ionization mass spectrometry (ESI-MS). Multi-stage tandem mass spectrometric (ESI-MSn) data for the alkaloid compounds were used for detailed structural characterization, then structure information was obtained by comparison of the fragmentation mechanisms of both alkaloids in Rhizoma Coptidis and standard samples of berberine, palmatine, coptisine and jatrorrhizine by MS. Based on the results obtained, the structure of a novel compound was elucidated. The results of the experiments demonstrate that ESI-MSn is a sensitive, selective and effective tool for the rapid determination of alkaloids in Rhizoma Coptidis.

CE/tris(2,2-bipyridyl) ruthenium(ll) (Ru(bpy)(3)(2+)) electrochemiluminescence (ECL), CEECL, with an ionic liquid (IL) detection system was established for the determination of bioactive constituents in Chinese traditional medicine opium poppy which contain large amounts of coexistent substances. A minimal sample pretreatment which involves a one-step extraction approach avoids both sample loss and environmental pollution. As the nearby hydroxyl groups in some alkaloid such as morphine may react with borate to form complexes and IL, as a high-conductivity additive in running buffer, could cause an enhanced field-amplified effect of electrokinetic injection. Running buffer containing 25 mM borax-8 mM 1-ethyl-3-methylimidazolium tetrafluoroborate (EMImBF(4)) IL (pH 9.18) was used which resulted in significant changes in separation selectivity and obvious enhancement in ECL intensities for those alkaloids with similar structures. Sensitive detection could be achieved when the distance between the Pt working electrode and the outlet of separation capillary was set at 150 mu m and the stainless steel cannula was fixed approximately 1 cm away from the outlet of the capillary. Quantitative analysis of four alkaloids was achieved at a detection voltage of 1.2 V and a separation voltage of 15 kV in less than 7 min.

The fragmentations of four strychnos alkaloids have been investigated by electrospray ionization Fourier transform ion cyclotron resonance mass spectrometry (ESI-FTICR-MS) in the positive ion mode. Experiments using multi-stage tandem mass spectrometry (ESI-FT-ICR-MSn) allowed us to obtain precise elemental compositions of product ions at high mass resolution. The experimental data demonstrated that the nitrogen bridge and the coordinated oxygen atom on the nitrogen bridge in the alkaloid compounds were the active sites in the MS2 fragmentations. The loss of CH3 or the OCH3 group in those alkaloids, which have an OCH3 substituent, was the dominant fragmentation mode in the MS3 fragmentations. Logical fragmentation schemes for strychnos alkaloids have been proposed and these should be useful for the identification of these compounds.

The chemical investigation of the crude extract of the marine-derived Streptomyces sp. M491 yielded three new sesquiterpenes, namely, 10 alpha,11-dihydroxyamorph-4-ene (4), 10 alpha,15-dihydroxyamorph-4-en-3-one (6), and 5 alpha,10 alpha,11-trihydroxyamorphan-3-one (7). In addition, the known compounds 10 alpha-hydroxyamorph-4-en-3-one (2), o-hydroxyacetanilide, genistein, prunetin, and indole-3-carbaldehyde and the macrolide antibiotic chalcomycin A were identified. The structures were determined on the basis of spectroscopic analysis, especially 1D and 2D NMR data. This is the first report of these sesquiterpenes from bacteria.

Two strains of Penicillium, DQ25 and SC10, isolated from marine sponge Haliclona angulata (Bowerbank) and Hymeniacidon sp. respectively, were subjected to stationary cultivation under GYP medium for 30 days. The fermentation extracts were undergone bioactivities assays against human pathogens, phytopathogenic fungi and brine shrimp (Artemia salina). Bioassays-guided compounds isolation was performed by Silica gel columns and Sephadex LH-20 chromatography. Spectroscopic methods were used to structures elucidation of the compounds. Results showed the activities of secondary metabolites of strain DQ25 were generally stronger than that of strain SC10. Major bioactive molecules isolated from strain DQ25 were a 1,4-naphthoquinone derivative and an unidentified alkaloid. The two components were not isolated from the extract of strain SC10. ITS sequences revealed that these two species have the greatest similarity with Penicillium vinaceum and Penicillium granulatum respectively.

Among marine bacteria isolated from the cytotoxic sponge Hymeniacidon perleve, one strain NJ6-3-1 classified as Pseudomonas sp. showed both cytotoxic and antimicrobial activities. Fatty acid analysis indicated that the bacterial strain consists mainly of C16:1, C16:0, C18:1, C18:0, C15:0, C14:0. One unusual 9,10-cyclopropane-C17:0 fatty acid and C26:0 also constitute major components, as well as the existence of squalene, the precursor of triterpenoids. The major metabolites in the culture broth were identified as alkaloids, including diketopiperazines and indole compounds, namely 3,6-diisopropylpiperazine-2,5-dione, 3-benzyl-3-isopropylpiperazine-2,5-dione, 3,6-bis-(2-methylpropyl)-piperazine-2,5-dione, indole-3-carboxaldehyde, indole-3-carboxylic acid methyl ester, indole-3-ethanol, and quinazoline-2,4-dione.

Two new brominated diterpenes, namely, laurendecumtriol (1) and 11-O-deacetylpinnaterpene C (2), one new polybromoindole, 2,3,4,6-tetrabromo-1-methyl-1H-indole (7), and six known natural products were isolated and identified from the marine red alga Laurencia decumbens. Their structures were elucidated on the basis of detailed spectroscopic and mass-spectrometric analysis as well as by comparison with literature data. Based on 2D-NMR experiments, the previously reported NMR data for pinnaterpene C (3) were reassigned.

海洋是一个巨大的天然产物宝库,约占地球表面积70%的海洋蕴藏着80%的生物资源。由于海洋生态环境的特殊性,导致海洋生物能够产生大量结构独特多变和活性特殊多样的代谢产物。我国海域辽阔,海洋资源丰富,为寻找结构新颖、生理活性独特的先导化合物,加强对海洋资源的开发利用,本论文对中国沿海的三种海洋红藻和两株放线菌次生代谢产物以及生物活性进行研究,为新药研究与开发提供模式结构和药物前体。 对红藻似瘤凹顶藻Laurencia similis乙酸乙酯萃取物进行分离纯化,从中得到单体化合物35个,通过波谱学方法(IR、MS、NMR等)鉴定了他们的结构。分别为:2, 2, 5, 5, 6, 6-sixibromo-3, 3-bi-1H-indole (1),3,5-dibromo- 1-methyl-indole (2),3',5',6,6'-tetrabromo-2,4-dimmethyldiphenyl ether (3),1,2,5- tribromo-3-bromoamino-7-bromomethylnaphthalene (4),2,5,8-tribromo-3-bromo- amino-7-bromomethylnaphthalene (5),2,5,6-tribromo-3-bromoamino-7-bromo- methylnaphthalene (6), 2,5,6,5',6'-pentabromo-3,4,3',4'-tetramethoxybenzophenone (7), (4E)-1-bromo-5-[(1'S*,3'R*)-3'-bromo-2',2'-dimethyl-6'-methylenecyclohexyl] -3-methylpent-4-ene-2,3-diol (8),4-hydroxy-Palisadin C (9),Isopalisol (10),Luzonensol (11),Palisadin B (12),Aplysistatin (13),Palisadin A (14),5-Acetoxypalisadin B (15),Aristolan-1(10)- en-9-ol (16),Aristol-8-en-1-one (17),Aristolan-9-en-1-one (18),Aristolan-1(10)-en- 9-one (19),Aristofone (20),Aristolan-1(10)-8-diene (21),Aristolan-1,9-diene (22),10-Hydroxyaristolan-9-one (23),7,11,15-trimethyl-3-methylene-hexadecan-1,2-diol (24),3β-Hydroxyergosta- 5,24(28)-dien-7-one (25),Isofucosterol (26),β-sitosterol (27),豆甾-4-烯-3α,6β-二醇 (28),Cholesta-5-en-3β-ol (29),Stigmasterol (30),2,3,5,6-四溴-吲哚 (31),2,3,6-tribromo-1H-indole (32),3,5,6-tribromo-1-methylindole (33),3,5,6-tribromo -1H-indole (34),2,3,5-tribromo-1-methylindole (35),其中化合物1-9为新化合物,化合物10-15、20和化合物24-30均为首次从该种海藻中得到。对新化合物1-9进行PTP1B酶抑制剂活性筛选,新化合物1、3、7显示强的PTP1B酶抑制活性。 对红藻齐藤凹顶藻Laurencia saitoi乙酸乙酯萃取物进行分离纯化,从中得到单体化合物11个,通过波谱学方法(IR、MS、NMR等)鉴定了他们的结构,分别为:2-hydroxyl-Luzofuranone (1),2-hydroxyl-Luzofuranone B (2),4-hydroxyl-Palisudin C (3),2-bromo-γ-ionone (4),Aplysistatin (5),5-Acetoxypalisadin B (6),Palisadin B (7),Palisadin A (8),Pacifigorgiol (9),豆甾-4-烯-3α,6β-二醇 (10),2, 3, 5, 6-四溴-吲哚 (11),其中化合物1-4为新化合物,所有化合物均为首次从该种海藻中得到。通过MTT法对分离得到的新化合物1-4进行肿瘤细胞毒活性筛选,结果显示4个新化合物对所测肿瘤细胞株均无明显的活性。 对红藻瘤状软骨凹顶藻Chondrophycus papillous乙酸乙酯萃取物进行分离纯化,从中得到单体化合物5个,通过波谱学方法(MS、NMR等)鉴定了他们的结构,分别为邻苯二甲酸二丁酯 (1),邻苯二甲酸二异辛酯 (2),胆甾醇 (3),3,7,11,15-tetramethyl-hexadec-2-en-1-ol (4),4-羟基苯甲醛 (5),所有化合物均为首次从该种海藻中得到。 对海洋放线菌M159乙酸乙酯萃取物进行分离纯化,从中得到单体化合物13个,通过波谱学方法(MS、NMR等)鉴定了他们的结构,分别为:5-(4',6'-dihydroxy-6-methyloctyl)furan-2(5H)-one (A),phenethyl alcohol (1),4-羟基苯甲醛(2),anthranilic acid (3),4-Hydroxy-3-methoxy- phenyl-propionic acid (4),5-(6,7-dihydroxy-6-methyloctyl)furan-2(5H)-one (5),p-Hydroxyphenylethyl alcohol (6),3-Indoleacrylic acid (7),Indol-3-carboxylic acid (8),Adenine cordyceposide (9),腺嘌呤核苷(10),尿嘧啶核苷(11),Thymidine (12),其中化合物A为新化合物。所有化合物均为首次从该株放线菌中得到。 对海洋放线菌L211乙酸乙酯萃取物进行分离纯化,从中得到单体化合物15个,通过波谱学方法(MS、NMR等)鉴定了7个结构,分别为:spatozoate (1),anthranilic acid (2),3-Indolylethanol (3),1-Acetyl-β-carbolin (4),p-Hydroxyphen- ylethyl alcohol (5),Indole-3-acetic acid (6),Indol-3-carboxylic acid (7),所有化合物均为首次从该株放线菌中得到。

Twenty-nine marine bacterial strains were isolated from the sponge Hymeniacidon perleve at Nanji island, and antimicrobial screening showed that eight strains inhibited the growth of terrestrial microorganisms. The strain NJ6-3-1 with wide antimicrobial spectrum was identified as Pseudoalteromonas piscicida based on its 16S rRNA sequence analysis. The major antimicrobial metabolite, isolated through bioassay-guide fractionation of TLC bioautography overlay assay, was identified as norharman (a beta-carboline alkaloid) by EI-MS and NMR.

A new halogenated biindole and a new apo-carotenone have been isolated from the ethanolic extract of the green alga Chaetomorpha basiretorsa Sethcell. On the basis of chemical and spectroscopic methods including 2D NMR technique, their structures have been elucidated as 4,4'-dichloro-5,5'-dibromo-7,7'-dimethoxy-2,2'-bi-1H-indole and 1'S*,4R*-8-(4'-hydroxy-2',6',6'-trimethylcyclohex-2-enyl)-6-methyloct-3E,5E,7E-trien-2-one, respectively.

To assess the medicinal value of cultural Anisodus tanguticus,the contents of four bioactive tropane alkaloids,anisodine,anisodamine,scopolamine and atropine,in cultural and wild materials were determined by the HPLC method.The results showed that content of each alkaloid in the aboveground parts of cultural and wild samples was lower than that in roots,and this explained why it was not the whole plant but the root that was used as medicinal materials.The content of each alkaloid in the roots of one-year cultural material was lower than that in the two-year plants.The discrepancy of the total of four alkaloids between one-year and wild plants is not significant.Moreover,the total of four alkaloids,and the contents of anisodine,scopolamine,and atropine in two-year plants were higher than those in wild plant.Thus there is medicinal value in the cultivated A.tanguticus as well as wild A.tanguticus,especially in the two-year cultural A.tanguticus.

The protein binding constant, binding sites of the Strychnos alkaloid-strychnine and bovine serum albumin (BSA) was determined by capillary electrophoretic frontal analysis (CE-FA) for the first time. The experiment was carried out in a polyacrylamide-coated fused silica capillary (48.4 cmx50 mu m i.d., 38.1 cm effective length) with 20 mmol/L citrate/MES buffer (pH 6.0, ionic strength 0.17). The applied voltage was 12 kV and detection wavelength was set at 257 nm. The plateau height of the peak was employed to determine the unbound concentration of drug in BSA equilibrated sample solution based on the external drug standard in the absence of protein. The present method provides a convenient, accurate technique for the early stage of drug screening.
