应用RT-PCR技术检测草鱼出血病病毒。建立了一种快速、简易而可靠的dsRNA模板的制备方法。应用该方法制备模板进行RT-PCR扩增,可以有效地检测出病鱼组织及病毒感染的培养细胞裂解液中的草鱼出血病病毒,且全过程只需3~4h,大大缩短了检测时间

从氧化塘系统中分离出能降解对硫磷的细菌Pseudomonas sp.代号CTP-01,能将对硫磷分解成对硝基酚和二乙基硫代磷酸酯,并进一步分解对硝基酚。在有Cu~(++)存在的情况下,酶比活可以达到1×10~4毫微克分子/毫克蛋白/分钟,Cu~(++)对酶有激活作用,并对温度和pH影响有保护作用。对硫磷水解酶反应最适温度为40—50℃,超过50℃活性急剧降低,80℃完全失活。 CTP-01的对硫磷水解酶大部分是同膜片结合状态存在,超声破碎的无细胞酶制剂中,只有37.2%的活力存在于可溶性蛋白部分。

Argonaute 2 gene plays a pivotal role in RNAi in many species. Herein is the first report of the cloning and characterization of Argonaute 2 gene in fish. The full-length cDNA of Gobiocypris rarus Argonaute 2 (GrAgo2) consisted of 3073 nucleotides encoding 869 amino acid residues with a calculated molecular weight of 98.499 kDa and an estimated isoelectric point of 9.18. Analysis of the deduced amino acid sequence showed the presence of two signature domains, PAZ and Piwi. RT-PCR analysis indicated that GrAgo2 mRNA expression could be detected in widespread tissues. After infection with grass carp reovirus, GrAgo2 expression was up-regulated from 12 h post-injection (p < 0.05) and returned to control levels at 48 h post-injection (p > 0.05). These data imply that GrAgo2 is involved in antiviral defense in rare minnow. (C) 2008 Published by Elsevier Ltd.

A new freshwater phototrophic species of the dinoflagellate genus Peridiniopsis, P. niei sp. nov., is described based on morphology. The new species appeared during spring with densities up to 1.48 x 10(7) cells L-1 in some tributaries and gullies of Three Gorge Reservoir and Lake Donghu, China, forming red tides. Peridiniopsis niei is a cyst-producing freshwater dinoflagellate that belongs to the group Penardii. The plate tabulation is po+x+4 '+0a+6 ''+5c+5s+5 '''+2 '''' and the plate pattern is symmetric. The cells of P. niei are pentagonal in ventral view, the epitheca is larger than the hypotheca, making up about 2/3 the length of the cell. Plate 3 ' is hexangular. The closest species to P. niei is P. penardii (Lemmermann) Bourrelly, but cells of the former are pentagonal, very compressed dorsoventrally, and the hypotheca is truncated with one transparent, robust spine on each antapical plate.

Cyprinidae is the largest fish family in the world and contains about 210 genera and 2010 species. Appropriate DNA markers must be selected for the phylogenetic analyses of Cyprinidae. In present study, the 1st intron of the S7 ribosomal protein (r-protein) gene is first used to examine the relationships among cyprinid fishes. The length of the 1st intron obtained by PCR amplification ranges from 655 to 859 by in the 16 cyprinid species investigated, and is 602 by in Myxocyprinus asiaticus. Out of the alignment of 925 nucleotide sites obtained, the parsimony informative sites are 499 and occupy 54% of the total sites. The results indicate that the 1st intron sequences of the S7 r-protein gene in cyprinids are rich in informative sites and vary remarkably in sequence divergence from 2.3% between close species to 66.6% between distant species. The bootstrap values of the interior nodes in the NJ (neighbor-joining) and MP (most-parsimony) trees based on the present S7 r-protein gene data are higher than those based on cytochrome b and the d-loop region respectively. Therefore, the 1st intron sequences of the S7 r-protein gene in cyprinids are sensitive enough for phylogenetic analyses, and the 1st intron is an appropriate genetic marker for the phylogenetic reconstruction of the taxa in different cyprinid subfamilies. However, attempts to discuss whether the present S7 r-protein gene data can be applied to the phylogeny of the taxa at the level of the family or the higher categories in Cypriniformes need further studies.

Thickness effect of immiscible alloy InAlAs as matrix layer on the morphology of InAs nanostructure grown on InAlAs/InP (0 0 1) by solid-source molecular-beam epitaxy has been studied. Experiments demonstrate that InAs nanostructure grown on thin InAlAs matrix layer forms randomly distributed quantum dot, whereas, grown on thick InAlAs matrix layer forms one-dimension ordered mixture of quantum wire and quantum dot. This drastic modification in the nanostructure morphology is attributed to the generation of composition modulation in the immiscible InAlAs alloy with the increase of the layer thickness. 为了揭示黄土高原地区人工油松林自然化发育过程中的植物叶片与土壤养分库和碳库的变化规律,采用典型样地法,以子午岭林区天然油松群落为参照,选择了不同立地条件的人工油松林,分别对其土壤的氮库、碳库及植物叶片养分库和碳库进行了分析。结果表明,该区油松针叶C、N、P的含量平均值分别为(499.5±63.75)mg/g、(8.53±0.50)mg/g和(0.94±0.64)mg/g,叶片C含量大小依次为阳坡天然林>阴坡天然林>阴坡人工林>阳坡人工林,阳坡人工林叶片N、P含量显著高于阳坡天然林。不同立地条件下油松林叶片C/N、C/P差异显著,叶片N、P和N/P均达到极显著水平,但是叶片C含量差异不显著。油松叶片C含量与N、P含量均呈极显著负相关,N和P之间的呈显著正相关。不同立地的油松林除40~60cm土层土壤C、N含量无显著差异外,0~20cm和20~40cm两个土层的人工林土壤C、N含量显著高于天然林,同时阳坡人工林土壤C、N含量显著高于阴坡人工林。0~20cm、20~40cm两个土层土壤C含量与N含量均呈极显著正相关,与植物N、P均呈显著正相关。子午岭林区阳坡的人工油松林不仅叶片养分含量较高,而且林地土壤是该区土壤的...

对宁夏云雾山草原自然保护区植物种群分布格局的分形计盒维数进行了研究。结果表明:本氏针茅、铁杆蒿和百里香是该区的优势种。3种优势种的计盒维数在1.7左右,表明它们具有较大的空间占据能力。但是它们的计盒维数最大值出现的时间不同,本氏针茅、百里香计盒维数最大值出现在封育20a的样地中,铁杆蒿出现在封育25a的样地中。
