Marine bacterium Vibrio sp. F-6, utilizing agarose as a carbon source to produce agarases, was isolated from seawater samples taken from Qingdao, China. Two agarases (AG-a and AG-b) were purified to a homogeneity from the cultural supernatant of Vibrio sp. F-6 through ammonium sulfate precipitation, Q-Sepharose FF chromatography, and Sephacryl S-100 gel filtration. Molecular weights of agarases were estimated to be 54.0 kDa (AG-a) and 34.5 kDa (AG-b) by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). The optimum pH values for AG-a and AG-b were about 7.0 and 9.0, respectively. AG-a was stable in the pH range of 4.0-9.0 and AG-b was stable in the pH range of 4.0-10.0. The optimum temperatures of AG-a and AG-b were 40 and 55 degrees C, respectively. AG-a was stable at temperature below 50 degrees C. AG-b was stable at temperature below 60 degrees C. Zn2+, Mg2+ or Ca2+ increased AG-a activity, while Mn2+, Cu2+ or Ca2+ increased AG-b activity. However, Ag+, Hg2+, Fe3+, EDTA and SDS inhibited AG-a and AG-b activities. The main hydrolysates of agarose by AG-a were neoagarotetraose and neoagarohexaose. The main hydrolysates of agarose by AG-b were neoagarooctaose and neoagarohexaose. When the mixture of AG-a and AG-b were used, agarose was mainly degraded into neoagarobiose.

Food Sources of three filter-feeding bivalves from two habitats (intertidal oyster Crassostrea gigas, mussel Mytilus galloprovincialis. and subtidal cultured scallop Chlamys farreri) of Jiaozhou Bay (Qingdao,China) were determined by fatty acid and stable isotope in analysis. Cultured scallop was characterized by significant diatom markets such as 16:1/16:0 close to 1 and high ratio of 20:5(n - 3)/22:6(n - 3), hence we assume that the scallop mainly feeds on diatoms. Fatty acid biomarkers specific to bacteria and terrestrial materials were also found in considerable amounts in scallop tissue, which suggested that there were Substantial bacterial and terrestrial input into the food of the species. Intertidal oyster and mussel, however, exhibited significant flagellate marker. 22:6(n - 3). and lower level of diatom markers. which indicated that flagellates are also part of intertidal bivalves' Planktonic food Sources: meanwhile, high level of Chlorophyta fatty acid marker, Sigma 18:2(n - 6) + 18:3(n - 3), suggested that Ulva pertusa (Chlorophyta) seaweed bed supplied important food sources to intertidal bivalves. Additionally, result of stable isotope analysis showed that phytoplankton contributed 86.2 to 89.0% to intertidal bivalves' carbon budget; macroalga U. pertusa origin source had a contribution of MIX, to 11.0%, which indicated its role Lis in important supplemental food source to intertidal bivalves. From this study. it is concluded that the dietary difference of three bivalves probably relates to the different potential food sources in the scallop farm and intertidal zone in Jiaozhou Bay.

With the development of both seismic theory and computer technology, numerical modeling technology of seismic wave has achieved great advancement during the past half century. The current methods under development include finite differentiation method (FDM), finite element method (FEM), pseudospectral method (PSM), integral equation method (IEM) and spectral element method (SEM). They exert their very important roles in every corner of seismology and seismic prospecting. Large quantity of researches towards spectral element method in the end of last century bring this method to a new era, which results in perfect solution of many difficult problems. However, parts of posterior works such as seismic migration and inversion which base on spectral element method have never been studied widely at least up to the present whereas are of importance to seismic imaging and seismic wave propagation. Based on previous work, this paper uses spectral element method to investigate the characteristics and laws of the seismic wave propagation in isotropic and anisotropic media. By thoroughly studying this high-accuracy method, we implement a kind of reverse-time pre- and post-stack migration based on SEM. In order to verify the validity of the SEM method, we have simulated the propagation of seismic wave in several different models. The simulation results show that: (1) spectral element method can be used to model any complex models and the computational results are comparable with the expected results and the analytic results; (2) the optimum accuracy can be achieved when the rank is between 4 and 9. When it is below 4, the dispersion may occur; and when it is above 9, the time step-length will be changed accordingly with the reducing space step-length in order to keep the computation stability. This will exponentially increase the computation time and at the same time the memory even if simulating the same media. This paper also applies explosive reflection surface imaging technology, time constancy principle of wave-filed extrapolation and least travetime raytracing technology of surface source to SEM pre- and post-stack migration of isotropic and anisotropic media. All imaging results derived by the above methods agree well with the real geological models and the position of interface and inflexions can also return to their right location well. This indicates that the method proposed in this paper is a kind of technology with high accuracy and robust stability. It can serve as an alternative method in real seismic data processing. All these work can boost the development of high-accuracy seismic imaging, and therefore have significant inference value.

The problem of oil and gas migration and accumulation have been investigated for many years in petroleum geology field. However, it is still the most weak link. It is a challenge task to research the question about the dynamics of hydrocarbon migration and accumulation. The research area of this article,Chengbei step-fault zone is the important exploration area of Dagang oil field. The oil distribution is complicated in this area because of abundant faults and rock-reservoir-cap assemblage. In recent years, oil shows is often discovered, but no large-scale pool is found. The most important problem influencing exolore decision is lake of kowning about accumulation process of oil and resources potential. According to the geology characteristic and exolore difficult, the analysis principles of dynamics is used in this paper. The course from source to reservoir is considered as main research line, and relation of valid source rcok, migration dynamic and heterogeneous distribution of carrier is discussed especially in key time. By use of numerial model the couling of migration and passage is realized and dynamic process of oil migration is analysed quantitatively. On the basis of other research about structure and sendiment, basin model is built and parameters are choiced. The author has reconstructed characteristic and distribution of fluid dynamical in main pool-forming time by numerical model. The systems of oil migration and acuumulaiton are divided according to distribution of fluid potential. Furthermore, the scope of valid sourece rock and scale of discharging hydrocarbon is studied in geology history by the method of producting hydrocarbon poential. In carrier research, it is outstanding to analyse the function that fault controls the oil-gas migration and accumulation. According to the mechanism of fault sealing, the paper author puts forward a new quantitative method evaluating fault opening and sealing properties-fault connective probability by using the oil and gas shows in footwall and hangwall reservoir as the index of identifying fault sealing or non-sealing. In this method, many influencing factors are considered synthetically. Then the faut sealing propery of different position in third deimention of faults controlling hydrocarbon acummulation are quantitative evaluated, and it laies a foundation for building compex carrier systems. Ten models of carrier and dynamical are establishe by analysis of matching relation of all kinds of carriers in main pool-forming period. The forming process and distribution of main pathway has been studied quantitatively by Buoyancy-Percolation mode, which can conbine valid source rock, migration dynamical and carrier. On the basis of oil-gas migration and accumulation model, the author computes the loss of hydrocarbon in secondary migration, ahead of cap formation, and the quantity of valueless accumulation according to the stage of migration and accumulation and the losing mechamism. At the same time, resource potential is evaluated in every migration and accumulation system. It shows that the quanlity of middle systems arrive to 5.67×108t, which has a huge explore potential prospect. Finally, according to the result of quantitve analysis above mentioned, the favorable explore aims are forcasted by the way of overlapping migration pathway and valid trap and considering factors of pool-forming. The drilling of actual wells proved that the study result is credible. It would offer strong support to optimize explore project in Chengbei step-fault zone.

River is a major component of the global surface water and CO2 cycles. The chemistry of river waters reveals the nature of weathering on a basin-wide scale and helps us understand the exogenic cycles of elements in the continent-river-ocean system. In particular, geochemical investigation of large river gives important information on the biogeochemical cycles of the elements, chemical weathering rates, physical erosion rates and CO2 consumption during the weathering of the rocks within the drainage basin. Its importance has led to a number of detailed geochemical studies on some of the world's large and medium-size river systems. Flowing in the south of China, the Xijiang River is the second largest river in the China with respect to its discharge, after the Yangtze River. Its headwaters drain the YunGui Plateau, where altitude is approximately 2000 meters. Geologically, the carbonate rocks are widely spread in the river drainage basin, which covers an area of about 0.17xl06 km2, i.e., 39% of the whole drainage basin. This study focuses on the chemistry of the Xijiang river system and constitutes the first geochemical investigation into major and trace elements concentrations for both suspended and dissolved loads of this river and its main tributaries, and Sr isotopic composition of the dissolved load is also investigated, in order to determine both chemical weathering and mechanical erosion rates. As compared with the other large rivers of the world, the Xijiang River is characterized by higher major element concentration. The dissolved major cations average 1.17, 0.33, 0.15, and 0.04 mmol I"1 for Ca, Mg, Na, and K, respectively. The total cation concentrations (TZ+) in these rivers vary between 2.2 and 4.4 meq I'1. The high concentration of Ca and Mg, high (Ca+Mg)/(Na+K) ratio (7.9), enormous alkalinity and low dissolved SiO2/HCO3 ratio (0.05) in river waters reveal the importance of carbonate weathering and relatively weak silicate weathering over the river drainage basin. The major elements in river water, such as the alkalis and alkaline-earths, are of different origins: from rain water, silicate weathering, carbonate and evaporite weathering. A mixing model based on mass budget equation is used in this study, which allows the proportions of each element derived from the different source to be calculated. The carbonate weathering is the main source of these elements in the Xijiang drainage basin. The contribution of rainwater, especially for Na, reaches to approximately 50% in some tributaries. Dissolved elemental concentration of the river waters are corrected for rain inputs (mainly oceanic salts), the elemental concentrations derived from the different rock weathering are calculated. As a consequence, silicate, carbonate and total rock weathering rates, together with the consumption rates of atmospheric CO2 by weathering of each of these lithologies have been estimated. They provide specific chemical erosion rates varying between 5.1~17.8 t/km2/yr for silicate, 95.5~157.2 t/km2/yr for carbonate, and 100.6-169.1 t/km2/yr for total rock, respectively. CO2 consumptions by silicate and carbonate weathering approach 13><109and 270.5x10 mol/yr. Mechanical denudation rates deduced from the multi-year average of suspended load concentrations range from 92-874 t/km2/yr. The high denudation rates are mainly attributable to high relief and heavy rainfall, and acid rain is very frequent in the drainage basin, may exceed 50% and the pH value of rainwater may be <4.0, result from SO2 pollution in the atmosphere, results in the dissolution of carbonates and aluminosilicates and hence accelerates the chemical erosion rate. The compositions of minerals and elements of suspended particulate matter are also investigated. The most soluble elements (e.g. Ca, Na, Sr, Mg) are strongly depleted in the suspended phase with respect to upper continent crust, which reflects the high intensity of rock weathering in the drainage basin. Some elements (e.g. Pb, Cu, Co, Cr) show positive anomalies, Pb/Th ratios in suspended matter approach 7 times (Liu Jiang) to 10 times (Nanpan Jiang) the crustal value. The enrichment of these elements in suspended matter reflects the intensity both of anthropogenic pollution and adsorption processes onto particles. The contents of the soluble fraction of rare earth elements (REE) in the river are low, and REE mainly reside in particulate phase. In dissolved phase, the PAAS-normalized distribution patterns show significant HREE enrichment with (La/Yb) SN=0.26~0.94 and Ce depletion with (Ce/Ce*) SN=0.31-0.98, and the most pronounced negative Ce anomalies occur in rivers of high pH. In the suspended phase, the rivers have LREE-enriched patterns relative to PAAS, with (La/Yb) SN=1 -00-1 .40. The results suggest that pH is a major factor controlling both the absolute abundances of REE in solution and the fractionation of REE of dissolved phase. Ce depletion in river waters with high pH values results probably from both preferential removal of Ce onto Fe-Mn oxide coating of particles and CeC^ sedimentation. This process is known to occur in the marine environment and may also occur in high pH rivers. Positive correlations are also observed between La/Yb ratio and DOC, HCO3", PO4", suggesting that colloids and (or) adsorption processes play an important role in the control of these elements.

The modeling of petroleum flow path (petroleum charging) and the detail of corresponding software development are presented in this paper, containing principle of petroleum charging, quantitative method, and practical modeling in two oil fields. The Modeling of Petroleum Flow Path is based on the result of basin modeling, according to the principle of petroleum migrating along the shortest path from the source to trap, Petroleum System Dynamics (Prof. Wu Chonglong, 1998), the concept of Petroleum Migration and Dynamic Accumulation (Zhou Donyan, Li Honhui, 2002), etc. The simulation is done combing with all parameters of basin, and considering the flow potential, non-uniformity of source and porous layer. It's the extending of basin modeling, but not belong to it. It is a powerful simulating tool of petroleum system, and can express quantitatively every kind of geology elements of a petroleum basin, and can recuperate dynamically the geology processes with 3D graphics. At result, we can give a result that the petroleum flow shows itself the phenomena of main path, and without using the special theory such as deflection flow in fractures(Tian Kaiming, 1989, 1994, Zhang Fawang, Hou Xingwei, 1998), and flow potential(England, 1987). The contour map of petroleum flow quantitative show clearly where the coteau - dividing slot is, and which convergence region are the main flow path of petroleum, and where is the favorable play of petroleum. The farsighted trap can be determined if there are enough information about structural diagram and can be evaluated, such as the entrapment extent, spill point, area, oil column thickness, etc. Making full use of the result of basin modeling with this new tool, the critical moment and scheme of the petroleum generation and expulsion can be showed clearly. It's powerful analysis tool for geologist.

Although studies on carbon burial in lake sediments have shown that lakes are disproportionately important carbon sinks, many studies on gaseous carbon exchange across the water-air interface have demonstrated that lakes are supersaturated with CO2 and CH4 causing a net release of CO2 and CH4 to the atmosphere. In order to more accurately estimate the net carbon source/sink function of lake ecosystems, a more comprehensive carbon budget is needed, especially for gaseous carbon exchange across the water-air interface. Using two methods, overall mass balance and gas exchange and carbon burial balance, we assessed the carbon source/sink function of Lake Donghu, a subtropical, eutrophic take, from April 2003 to March 2004. With the overall mass balance calculations, total carbon input was 14 905 t, total carbon output was 4950 1, and net carbon budget was +9955 t, suggesting that Lake Donghu was a great carbon sink. For the gas exchange and carbon burial balance, gaseous carbon (CO2 and CH4) emission across the water-air interface totaled 752 t while carbon burial in the lake sediment was 9477 t. The ratio of carbon emission into the atmosphere to carbon burial into the sediment was only 0.08. This low ratio indicates that Lake Donghu is a great carbon sink. Results showed good agreement between the two methods with both showing Lake Donghu to be a great carbon sink. This results from the high primary production of Lake Donghu, substantive allochthonous carbon inputs and intensive anthropogenic activity. Gaseous carbon emission accounted for about 15% of the total carbon output, indicating that the total output would be underestimated without including gaseous carbon exchange. (C) 2007 Elsevier Ltd. High (42.5%) indium content GaInNAs/GaAs quantum wells with room temperature emission wavelength from 1.3 mu m to 1.5 mu m range were successfully grown by Radio Frequency Plasma Nitrogen source assisted Molecular Beam Epitaxy. The growth parameters of plasma power and N-2 How rate were optimized systematically to improve the material quality. Photoluminescence and transmission electron microscopy measurements showed that the optical and crystal quality of the 1.54 mu m GaInNAs/GaAs QWs was kept as comparable as that in 1.31 mu m.
