A novel amperometric biosensor for the detection of hydrogen peroxide was described. The biosensor was constructed by electrodepositing HRP/PPy membrane on the surface of ferrocenecarboxylic acid mediated sol-gel derived composite carbon electrode. The biosensor gave response to hydrogen peroxide in a few seconds with detection limit of 5.0 x 10(-5) M (based on signal:noise = 3). Linear range was upto 0.2 mM. A water-soluble endohedral metallofullerol, Pr@C82Om(OH)(n)(m approximate to 10 and n approximate to 10), was successfully synthesized through the reaction of a pure endohedral metallofullerene, Pr@C-82, with a concentrated nitric acid and a subsequent hydrolysis process. The formation of endohedral metallofullerols Pr@C82Om(OH)(n) is thought to involve a NO2 radical formation step, in much the same way as the reaction of empty fullerenes. FT-IR, XPS, and LD-TOF MS techniques were employed to characterize the structure of the endohedral metallofullerol from the above reaction.

The laser-desorption-ionization time-of-flight mass spectrometry and desorption-electron-ionization mass spectrometry were employed for the characterization of metallofullerenes extract. it was found that the relative intensities of metallofullerenes in this positive-ion, negative-ion LD-TOF MS and DEI MS were much different. This phenomenon should have relationship with the peculiar ionization energies and electron affinities of metallofullerenes.

Endohedral metallofullerenes Tb@C-2n were synthesized and extracted with high-yield by K-H carbon-are evaporation and an effective pyridine extraction technique at high-temperature high-pressure. Laser-desorption-ionization time-of-flight (LD-TOF) mass spectrometry, X-ray photoelectron spectroscopy (XPS), solid-state fluorescent emission spectroscopy and gas phase derivation reaction with the self-chemical ionization mass spectrometric ion system of vinyl acetate were employed for studying the electronic structures, fluorescent properties and gas phase reactivities of metallofullerenes Tb@C-2n. The experimental results suggest that endohedral metallofullerenes Tb@C-2n would have the approximate structures of Tb3+@C-2n(3-) similar to other metallofullerenes, good fluorescent emission properties and active reactivities in gas phase ion-molecular reactions.

Endohedral metallofullerenes Tm@C-2n were synthesized with the carbon-arc discharge method of activating the Tm2O3-containing graphite anode in situ and back-burning technique, Toluene soxhlet extraction, toluene and pyridine high-temperature and high-pressure extraction technique were employed for the effective extraction of Tm@C-2n. The formation of Tm@C-2n was confirmed by LD-TOF and DEIMS characterization. The mass spectrometric results indicate that Tm@C-2n have a better solubility in toluene and worse solubility in pyridine than other metallofullerenes.

The novel polyetheretherketone (PEK-C) prepared from phenolphthalein in our institute is an amorphous, rigid, tough material with good mechanical properties over a wide temperature range. To improve its water vapor permeability for the application of gas drying, PEK-C was sulfonated with concentrated sulfuric acid and transferred in sodium, cupric, and ferric salt forms. Sulfonation degree can be regulated by controlling the temperature and reaction time. Characterization of sulfonated PEK-C in sodium form was made by infrared spectroscopy. Some properties of the sulfonated PEK-C, such as solubility, glass transition temperature, thermal stability, mechanical properties, and transport properties to nitrogen and water vapor, are also discussed. It is found that there is a linear relationship between log P-w, and the parameter term V-f/0.5 E(coh) [1+(delta(w) - delta(p))(2)/delta(p)(2), from the water permeability (P-w) data of 21 polymers covering 4 orders of magnitude. This correlation may be useful in choosing membrane materials for dehumidification of gases.

A method for the prediction of gas permeabilities (P) through polymers from their chemical structure has been developed on the basis of the ratio of molar free volume to molar cohesive energy, V(f)/E(coh). The permeation of small gas molecules through polymer membranes is dependent on the chain packing density measured by V(f) and segmental motion of polymer chains measured by E(coh). But no simple relationship between P and V(f) or E(coh) alone was found. The permeability data of more than 60 polymers covering 7 orders of magnitude for six gases have been treated with linear regression analysis. All plots of log P vs. V(f)/E(coh) gave good straight lines. It is also found that a linear relationship holds when plotting both the intercepts and slopes of log P vs. V(f)/E(coh) lines against square of the diameters of gas molecules. Therefore, the permeabilities of all the non-swelling gases through a great variety of polymers can be estimated using two correlations above. Moreover, this method is more accurate than others in the literature and may found useful for the selection of gas separation or barrier membrane materials.

A field population of Ulva pseudocurvata Koeman et C. Hoek (hereafter termed Ulva) at Sylt Island (North Sea, Germany) exhibited biweekly peaks of gametophytic reproduction during the colder seasons and approximately weekly peaks during summer. The reproductive events lasted 1-5 d and were separated from each other by purely vegetative phases. Under constant conditions in the laboratory, a free-running rhythm was observed with reproductive peaks occurring approximately every 7 d. When artificial moonlight was provided every 4 weeks, fewer reproductive events occurred, and the reproductive rhythm became synchronized to the environmental artificial moonlight rhythm. In the laboratory, apical disks were entirely converted into reproductive tissue after 8 d cultivation, while almost all basal disks stayed vegetative, which prevented the entire loss of the vegetative thallus during reproductive events. Seasonal size reduction of the thallus occurred from late autumn onward and was determined to be controlled by a genuine photoperiodic response, since size reduction could be induced from May onward by experimental short-day (SD) treatment but was prevented in a long-day (LD) or night-break regime (NB). A daily fine-tuning occurred with gamete release early in the morning at the first sign of daylight, following an obligatory dark ("night") period of at least 1 h duration. No release took place if the overnight dark phase was replaced by continuous light. Blue, green, or red light all triggered gamete release after a dark phase at an irradiance of 0.1 mu mol photons . m(-2) .s(-1), while 0.001 mu mol photons . m(-2) . s(-1) was equivalent to a dark control.

Edwardsiella tarda is an important Gram-negative enteric pathogen affecting both animals and humans. It possesses a type III secretion system (T3SS) essential for pathogenesis. EseB, EseC and EseD have been shown to form a translocon complex after secretion, while EscC functions as a T3SS chaperone for EseB and EseD. In this paper we identify EscA, a protein required for accumulation and proper secretion of another translocon component, EseC. The escA gene is located upstream of eseC and the EscA protein has the characteristics of T3SS chaperones. Cell fractionation experiments indicated that EscA is located in the cytoplasm and on the cytoplasmic membrane. Mutation with in-frame deletion of escA greatly decreased the secretion of EseC, while complementation of escA restored the wild-type secretion phenotype. The stabilization and accumulation of EseC in the cytoplasm were also affected in the absence of EscA. Mutation of escA did not affect the transcription of eseC but reduced the accumulation level of EseC as measured by using an EseC-LacZ fusion protein in Ed. tarda. Co-purification and co-immunoprecipitation studies demonstrated a specific interaction between EscA and EseC. Further analysis showed that residues 31-137 of EseC are required for EseC-EscA interaction, Mutation of EseC residues 31-137 reduced the secretion and accumulation of EseC in Ed. tarda. Finally, infection experiments showed that mutations of EscA and residues 31-137 of EseC increased the LD50 by approximately 10-fold in blue gourami fish. These results indicated that EscA functions as a specific chaperone for EseC and contributes to the virulence of Ed. tarda.

Two photoperiodic responses, the development of sporophylls and hairs, have been quantified in sporophytes of the brown alga Undaria pinnatifida. In a final experiment, the algae were cultivated in outdoor, 2000-L seawater tanks in a greenhouse for up to 12 weeks, and daylength was regulated by automatic blinds mounted on top of the tanks. Vegetative young sporophytes were treated under short-day (SD; 8 h light per day) or long-day conditions (LD; 16 h light per day), at 12 h light per day or in a night-break regime (NB; 8 h light per day, 7.5 h dark, 1 h light, 7.5 h dark). The earliest sporophyll development was observed 6, 7 or 9 weeks under LD, NB or SD conditions, respectively. After 12 weeks the sporophylls were significantly longer and wider under LD or NB conditions than in the SD regime, and only half of the experimental algae had formed sporophylls under SD conditions, but all algae under LD or NB conditions. In a foregoing 7-week culture experiment performed in 300-L indoor tanks, enhanced sporophyll formation had also been observed under LD and not under SD conditions (NB omitted). In both experiments, blade elongation rates remained high until the end of the experiments in SD, but declined during sporophyll initiation in LD, NB or at 12 h light per day. Another difference caused by photoperiod was observed in regard to the development of surface hair spots which occurred in both experiments on the blades in LD, NB or at 12 h light per day with identical densities, but were completely lacking under SD conditions. It is concluded that U. pinnatifida is a facultatative long-day plant in regard to reproduction forming vigorously sporophylls in long days, and an obligate long-day plant in regard to hair formation.

Peritrophin, one of the components of the peritrophic matrix, was first isolated from the intestine of insects. It is thought to protect insects from invasion of microorganisms and to stimulate digestion of food. Peritrophin-like proteins have also been found in crustaceans, as a component of the egg layer. In this study, one fragment of the peritrophin-like gene was obtained from fleshy prawn (Chinese shrimp) (Fenneropenaeus chinensis) by panning the T7 phage display library constructed with the shrimp hemocyte cDNA. The total sequence of the peritrophin cDNA was cloned by modified SMART cDNA and LD-PCR methods. The full cDNA is 1048 bp and the deduced protein is composed of 274 amino acids, including 21 amino acid signal peptide, and four peritrophin A domains and the latter three forming three chitin-binding domains. Similarity analysis results showed that the peritrophin-like protein from F chinensis has significant similarities with peritrophin-like and cortical rod proteins from other shrimp. It was inducing expression in hemocytes, heart, stomach, gut, and gills of the infected shrimp, and constitutive expression in the ovaries. No expression signal was detected in the hepatopancreas of either infected or noninfected shrimp. The recombinant peritrophin-like protein has the activity of binding Gram-negative bacteria and strong binding activity to chitin. Therefore, the bacteria and chitin binding activities of the peritrophin-like protein suggest that it may plays a role in immune defense and other physiological resposes. (c) 2005 Elsevier Ltd. 海带具有很高的营养价值和经济社会价值。自20世纪90年代以来,本实验室在借鉴高等植物基因工程原理和方法的基础上,根据海带自身特点,建立了海带遗传转化体系(海带孢子体表达系统),它的基本原理是利用基因枪法转化海带配子体,经孤雌或受精途径再生幼孢子体后,用氯霉素筛选幼孢子体获得转基因海带,然后进行海上安全栽培和转基因产品的检测与提取。目前该表达系统已成功实现报告基因(β-半乳糖苷酶基因,lacZ)和功能基因(乙肝表面抗原基因,HBsAg)的稳定表达。 由于海带孢子体表达系统需经孢子体再生和海上栽培等阶段,周期较长,而且转基因安全性问题也在一定程度上制约其研究与应用。因此,我们在海带孢子体表达系统的基础上又建立和优化了海带配子体表达系统,并成功实现了报告基因(绿色荧光蛋白基因,GFP)的瞬间表达和功能基因(瑞替普酶基因,rt-PA)的稳定表达。虽然海带配子体表达系统能避免转基因安全性问题,周期较短,但在表达量和生物量积累方面,与孢子体表达系统相比还有较大差距。 本文首先在海带配子体表达系统中成功实现了人酸性成纤维细胞生长因子基因(hafgf)和鲎素基因(tac)的稳定表达,制备了转基因海带配子体,然后将光生物反应器培养技术应用于转基因海带配子体的高效增殖,以期解决阻碍海带配子体表达系统发展的量的问题,并为转基因海带配子体的大规模培养提供试验依据和技术支持。 本文的研究结果为: 1、人酸性成纤维细胞生长因子基因和鲎素基因可以稳定整合到海带配子体基因组中,实现转基因产物的表达。 2、根据转基因海带配子体的生长特点,研制开发了一套培养体积为300 ml的鼓泡式光生物反应器,它具有操作简便、成本低廉、适合海带配子体生长等特点。随后将培养体系扩大到2.5 L,并研究了光对转基因海带配子体生长的影响,试验结果显示,转基因海带配子体在光强为30 μE m-2 s-1时即可达到光饱和生长,最优光周期为14:10 LD,而且蓝光可促进转基因海带配子体的生长。 3、在前期研究工作的基础上,为改善反应器内的传质条件,我们又设计研制了2.5 L气升式光生物反应器用于转基因海带配子体的高效增殖。研究发现,气升式光生物反应器较鼓泡式光生物反应器能明显地改善反应器内的传质状态,实现转基因海带配子体更高密度的培养(生物量可达到1,990 mg L-1),是一套高密度悬浮培养转基因海带配子体的有效装置和设备。
