Learning and memory are exquisitely sensitive to behavioral stress, but the underlying mechanisms are still poorly understood. Because activity-dependent persistent changes in synaptic strength are believed to mediate memory processes in brain areas such as the hippocampus we have examined the means by which stress affects synaptic plasticity in the CA1 region of the hippocampus of anesthetized rats, Inescapable behavioral stress (placement on an elevated platform for 30 min) switched the direction of plasticity, favoring low frequency stimulation-induced decreases in synaptic transmission (long-term depression, LTD), and opposing the induction of long-term potentiation by high frequency stimulation, We have discovered that glucocorticoid receptor activation mediates these effects of stress on LTD and longterm potentiation in a protein synthesis-dependent manner because they were prevented by the glucocorticoid receptor antagonist RU 38486 and the protein synthesis inhibitor emetine. Consistent with this, the ability of exogenously applied corticosterone in non-stressed rats to mimic the effects of stress on synaptic plasticity was also blocked by these agents, The enablement of low frequency stimulation-induced LTD by both stress and exogenous corticosterone was also blocked by the transcription inhibitor actinomycin D, Thus, naturally occurring synaptic plasticity is liable to be reversed in stressful situations via glucocorticoid receptor activation and mechanisms dependent on the synthesis of new protein and RNA, This indicates that the modulation of hippocampus-mediated learning by acute inescapable stress requires glucocorticoid receptor-dependent initiation of transcription and translation.

The anesthetic, propofol, effectively suppresses excitatory synaptic transmission and facilitates long-term depression (LTD) in the CA1 region of the hippocampus. Here, we have examined whether these effects are different in the developing hippocampus. We found that propofol in suppressing whole-cell excitatory postsynaptic currents (EPSC) was more effective in 21 day old rats than either in 7 day old rats or under the condition of high intracellular chloride concentration in 21 day old rats. Furthermore, the propofol concentration to facilitate the NMDA receptor-dependent LTD was lower at postnatal day 21 than at postnatal day 7. Interestingly, the decay time of EPSC was decreased during the development from postnatal day 7 to 21, but it was increased by the recording condition of high intracellular chloride concentration or by propofol administration. All these effects of propofol were dependent on the chloride channel opening. These observations suggest that propofol may induce differential anesthetic effects in the developing hippocampus, at least partially, depending on the intracellular chloride concentration. 下载PDF阅读器目的 研究三七总皂苷(Panax notoginseng saponins,PNS)对大鼠海马脑片CA1区锥体神经元兴奋性和抑制性突触传递的作用.方法 断头法分离3~4周雄性Wistar大鼠海马半脑,用切片机切出400μm厚度的海马脑片,对CA1区锥体细胞采用"盲法"全细胞膜片钳技术记录,分别检测和分析PNS(0.05~0.4 g/L)对刺激CA1传人纤维引出的兴奋性突触后电流(EPSCs)和抑制性突触后电流(IPSCs)的影响,继而以脉冲间隔为50 ms的配对刺激代替单刺激,通过EPSC2/EPSC1(P2/P1)值的变化观察PNS对双脉冲易化(paired-pulse facilitation,PPF)的影响.结果 0.1~0.4 g/L PNS显著抑制EPSCs(P<0.05),且PNS在抑制P1、P2的同时明显升高P2/P1值(P<0.05),加强了双脉冲易化,但PNS对IPSCs无显著影响(P>0.05).结论 PNS 显著减小大鼠海马CA1区锥体神经元的EPSCs而不影响IPSCs,说明PNS不是通过强化抑制性中间神经元的功能间接地抑制兴奋性神经元,而是对兴奋性突触传递直接产生抑制;PNS明显升高P2/p1值,说明 PNS是通过突触前机制抑制CA1区兴奋性突触传递.

目的研究异丙酚对大鼠海马CA1区神经元兴奋性突触后电流(EPSC)和自发性兴奋性突触后电流(sEPSC)的影响。方法 Wistar大鼠断头后分离海马脑组织,制成400μm厚度的海马脑片,脑片随机分为5组(n=10)。脂肪乳剂Ⅰ组、异丙酚Ⅰ组、SR95531+异丙酚组:记录EPSC 10 min (基础值)后分别加入10%脂肪乳剂90μl、1%异丙酚90μl(相当于100μmol/L)、10μmol/L SR95531+100 μmol/L异丙酚,继续记录EPSC 40 min,分析EPSC幅值的变化。脂肪乳剂Ⅱ组、异丙酚Ⅱ组:细胞破膜后稳定10-15 min,分别加入10%脂肪乳剂90μl和1%异丙酚90μl,记录sEPSC 40 min,分析sEPSC频率、幅值和半衰期的变化。膜钳制电压均为-70 mV。结果与基础值比较,给药后脂肪乳剂Ⅰ组和 SR95531+异丙酚组EPSC幅值差异无统计学意义,异丙酚Ⅰ组EPSC幅值降低;给药后异丙酚Ⅰ组 EPSC幅值比脂肪乳剂Ⅰ组降低(P<0.05)。与脂肪乳剂Ⅱ组比较,异丙酚Ⅱ组sEPSC的频率、幅值降低、半衰期缩短(P<0.05)。结论异丙酚主要通过增强大鼠海马CA...

 目的 观察500μmol/ L 丙泊酚对大鼠海马CA1 区电刺激诱发的兴奋性突触后电流 ( EPSC) 的影响,分析丙泊酚的可能作用机制。方法 断头法分离Wistar 大鼠(13~19 d) 海马半脑, 用切片机切出400μm 厚度的海马脑片,全细胞膜片钳技术记录CA1 区锥体神经元EPSC。实验分 两组:脂肪乳剂组( n = 6) 和丙泊酚组( n = 10) 。先以50μmol/ L 印防己毒素预孵脑片30 min 后,记录 基础EPSC 10 min ,然后加入450μl 脂肪乳剂或丙泊酚(相当于500 μmol/ L ) , 继续记录EPSC 40 min ;继而以配对刺激代替单刺激,观察EPSC2/ EPSC1 比率的变化;改变膜钳制电压( - 80~ + 60 mV) ,观察电流2电压( I2V) 曲线的变化。结果 脂肪乳剂对EPSC 无影响,500μmol/ L 丙泊酚降低 大鼠海马CA1 区EPSC 值,25~30 min 左右达最大抑制效果,EPSC 幅值下降至基础值的6715 % ,明 显低于脂肪乳剂组( P < 0105) ;而且500μmol/ L 丙泊酚明显降低EPSC2/ EPSC1 比率,也使I2V 曲线 左移,降低反转电位至- 35 mV 左右。结论 500μmol/ L 丙泊酚对大鼠海马CA1 区兴奋性突触传 递产生抑制作用,这可能与其增强突触前膜、突触后膜GABAA 受体活性有关。

Experience-dependent long-lasting increases in excitatory synaptic transmission in the hippocampus are believed to underlie certain types of memory(1-3). Whereas stimulation of hippocampal pathways in freely moving rats can readily elicit a long-term potentiation (LTP) of transmission that may last for weeks, previous studies have failed to detect persistent increases in synaptic efficacy after hippocampus-mediated learning(4-6). As changes in synaptic efficacy are contingent on the history of plasticity at the synapses(7), we have examined the effect of experience-dependent hippocampal activation on transmission after the induction of LTP, We show that exploration of a new, non-stressful environment rapidly induces a complete and persistent reversal of the expression of high-frequency stimulation-induced early-phase LTP in the CA1 area of the hippocampus, without affecting baseline transmission in a control pathway. LTP expression is not affected by exploration of familiar environments. We found that spatial exploration affected LTP within a defined time window because neither the induction of LTP nor the maintenance of long-established LTP was blocked. The discovery of a novelty-induced reversal of LTP expression provides strong evidence that extensive long-lasting decreases in synaptic efficacy may act in tandem with enhancements at selected synapses to allow the detection and storage of new information by the hippocampus.

The subiculum, which is the primary target of CA1 pyramidal neurons and sending efferent fibres to many brain regions, serves as a hippocampal interface in the neural information processes between hippocampal formation and neocortex. Long-term depression (LTD) is extensively studied in the hippocampus, but not at the CA1-subicular synaptic transmission. Using whole-cell EPSC recordings in the brain slices of young rats, we demonstrated that the pairing protocols of low frequency stimulation (LFS) at 3 Hz and postsynaptic depolarization of -50 mVelicited a reliable LTD in the subiculum. The LTD did not cause the changes of the paired-pulse ratio of EPSC. Furthermore, it did not depend on either NMDA receptors or voltage-gated calcium channels (VGCCs). Bath application of the G-protein coupled muscarinic acetylcholine receptors (mAChRs) antagonists, atropine or scopolamine, blocked the LTD, suggesting that mAChRs are involved in the LTD. It was also completely blocked by either the Ca2+ chelator BAPTA or the G-protein inhibitor GDP-beta-S in the intracellular solution. This type of LTD in the subiculum may play a particular role in the neural information processing between the hippocampus and neocortex. Microstructure characterization is important for controlling the quality of laser welding. In the present work, a detailed microstructure characterization by transmission electron microscopy was carried out on the laser welding cast Ni-based superalloy K418 turbo disk and alloy steel 42CrMo shaft and an unambiguous identification of phases in the weldment was accomplished. It was found that there are gamma-FeCrNiC austenite solid solution dendrites as the matrix, (Nb, Ti) C type MC carbides, fine and dispersed Ni-3 Al gamma' phase as well as Laves particles in the interdendritic region of the seam zone. An analytical model for size-dependent interface phonon transmission and thermal conductivity of nanolaminates is derived based on the improved acoustic mismatch theory and the Lindemann melting theory by considering the size effect of phonon velocity and the interface lattice mismatch effect. The model suggests that the interface phonon transmission is dominant for the cross-plane thermal conductivity of nanolaminates and superlattices, and the intrinsic variety of size effect of thermal conductivity for different systems is proposed based on the competition mechanism of size effect of phonon transport between two materials constituting the interfaces. The model's prediction for thermal conductivity of nanolaminates agrees with the experimental results. (C) 2008 American Institute of Physics.

Severe acute respiratory syndrome (SARS) is a serious disease with many puzzling features. We present a simple, dynamic model to assess the epidemic potential of SARS and the effectiveness of control measures. With this model, we analysed the SARS epidemic data in Beijing. The data fitting gives the basic case reproduction number of 2.16 leading to the outbreak, and the variation of the effective reproduction number reflecting the control effect. Noticeably, our study shows that the response time and the strength of control measures have significant effects on the scale of the outbreak and the lasting time of the epidemic.

We investigate the optical transmission properties of a combined system which consists of two quantum-dot-nanocavity subsystems indirectly coupled to a waveguide in a planar photonic crystal. A Mollow-like triplet and the growth of sidebands are found, reflecting intrinsic optical responses in the complex microstructure.
