OBJETIVO: Avaliar em um modelo experimental de isquemia-reperfusão hepática os efeitos da injeção intraluminal de glutamina na capacidade anti-oxidante total em equivalência ao trolox (TEAC) do plasma, verificando a aplicabilidade de modificações ao método original de dosagem. MÉTODOS: Trinta ratos Wistar foram submetidos a laparotomia e confecção de uma alça fechada de 20 cm de comprimento envolvendo o intestinal delgado distal seguido do clampeamento do hilo hepático por 30 minutos e reperfusão por 5 minutos. Na alça fechada foi injetada glutamina (grupo glutamina; n=10) ou água destilada (grupo controle; n=10). Em dez animais (grupo sham) não foi realizado clampeamento hilar. Coletou-se sangue para dosagem da capacidade antioxidante total em equivalência ao trolox em condições modificadas de temperatura, proporções relativas dos reagentes e tempo de leitura sob espectrofotometria. RESULTADOS: A capacidade antioxidante total foi significantemente maior (p<0.05) no grupo glutamina que no grupo controle (1,60[1,55-1,77] vs 1,44[1,27-1,53]) e grupo sham (1,60[1,55-1,77] vs 1,48[1,45-1,59]). Não houve diferenças estatísticas entre o grupo controle e o grupo sham. CONCLUSÃO: A glutamina melhorou a capacidade anti-oxidante total plasmática. O método de dosagem refletiu consistentemente alterações na defesa anti-oxidante nesse modelo experimental.

Introduction: Training for and competing in ultraendurance exercise events is associated with an improvement in endogenous antioxidant defenses as well as increased oxidative stress. However, consequences on health are currently unclear. Purpose: We aimed to examine the impact of training- and acute exercise-induced changes in the antioxidant capacity on the oxidant/antioxidant balance after an ironman triathlon and whether there are indications for sustained oxidative damage. Methods: Blood samples were taken from 42 well-trained male triathletes 2 d before an ironman triathlon, then immediately postrace, 1, 5, and 19 d later. Blood was analyzed for conjugated dienes (CD), malondialdehyde (MDA), oxidized low-density lipoprotein (oxLDL), oxLDL:LDL ratio, advanced oxidation protein products (AOPP), AOPP:total protein (TP) ratio, Trolox equivalent antioxidant capacity (TEAC), uric acid (UA) in plasma, and activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), and catalase (CAT) in erythrocytes. Results: Immediately postrace, there were significant increases in CD, AOPP, TEAC, UA (for all P < 0.001), and AOPP:TP (P < 0.01). MDA rose significantly (P < 0.01) 1 d postrace, whereas CD (P < 0.01), AOPP (P = 0.01), AOPP:TP (P < 0.05), and TEAC (P < 0.001) remained elevated. OxLDL:LDL trended to increase, whereas oxLDL significantly (P < 0.01) decreased 1 d postrace. Except for GSH-Px (P = 0.08), activities of SOD (P < 0.001) and CAT (P < 0.05) significantly decreased postrace. All oxidative stress markers had returned to prerace values 5 d postrace. Furthermore, several relationships between training status and oxidative stress markers, TEAC, and antioxidant enzyme activities were noted. Conclusions: This study indicates that despite a temporary increase in most (but not all) oxidative stress markers, there is no persistent oxidative stress in response to an ironman triathlon, probably due to training- and exercise-induced protective alterations in the antioxidant defense system.

海洋生物具有产生丰富多样的次生代谢产物的能力,其中红藻门松节藻科海藻卤代次生代谢产物以其结构新颖、生物活性独特引起了天然产物化学家的重视。 本论文对海洋红藻多管藻和松节藻进行了化学成分研究,综合利用各种色谱学方法 (硅胶柱层析、反相硅胶柱层析、凝胶Sephadex LH-20柱层析、半制备高效液相色谱以及重结晶等) 和现代波谱学技术 (IR、UV、EI-MS、FAB-MS、HR-ESI-MS、CD、1H-NMR、13C-NMR、DEPT、1H-1H COSY、HSQC、HMBC等),共分离鉴定了100个化合物,发现25个新化合物。 从多管藻中分离鉴定38个化合物 (24个溴酚化合物),其中7个新化合物 (均为溴酚化合物),包括1个菲并呋喃结构溴酚 (P1), 2个二氢菲结构溴酚 (P2, P3),1个含 5,7-dihydrodibenzo[c,e]oxepine 结构溴酚 (P4)和3个简单溴酚 (P5, P6, P7)。P1 (urceolatin) 属首例报道的具有菲并呋喃结构的天然产物,从该种中分离的化合物P12 和 P13 可能是其生源合成的前体。P2和P3为第二例报道的具有二氢菲结构的溴酚化合物。 从松节藻中分离并鉴定了62 个化合物,其中18 个为溴酚类新化合物,44 个为已知化合物。化合物具有多变的取代基团,包括2 个脲基吡咯烷酮溴酚化合物 (R1, R2), 4 个γ-脲基丁酸溴酚化合物 (R3-R6),5 个酰胺溴酚化合物 (R7, R8, R9, R13, R14),1 个溴酚砜化合物 (R12), 1 个Xanthene 溴酚化合物 (R10)和5 个简单溴酚化合物 (R11, R15, R16, R17, R18)。R1、R2 是首例报道的含有脲基吡咯烷酮片段的天然产物,R10 为首次报道的溴代Xanthene 类天然产物。 对分离到的化合物进行了清除DPPH 和ABTS两种自由基活性的筛选。结果发现溴酚类天然产物具有显著的DPPH自由基清除活性,其中R3 的IC50 仅为3.3 μM, 其活性强度约为阳性对照BHT (IC50 为82.1 μM) 的24倍。另外,溴酚类天然产物对ABTS自由基有较强的清除活性,R2 的TEAC(Trolox efficency activity capacity)值为5.2 mM,约为阳性对照 (ascorbic acid, 1.02 mM) 的 5 倍。初步的构效关系研究发现,稠环分子、多羟基和邻位甲氧基等结构特点能有效增强DPPH 自由基清除活性;特殊取代基如脲基、吡咯烷酮等含有氮原子的基团,能有效增强ABTS 自由基清除活性,多羟基、溴代等结构特点也使其活性有所增强。 本研究结果丰富了海藻卤代化合物的结构类型,为多管藻和松节藻的合理利用提供了一定的科学依据。

The effect of coating Arbutus unedo fresh fruit with alginate-based edible coatings enriched with the essential oils compounds (EOC) eugenol (Eug) and citral (Cit) was studied. The minimum inhibitory concentrations (MIC) against the main postharvest pathogens were determined for Eug and Cit giving values of 0.10 and 0.15 (w/v), respectively. Twelve formulations of edible coatings were used: sodium alginate (AL) was tested at 1 and 2% (w/v) with incorporation of Eug and Cit at MIC and double MIC or their combination at MIC. Arbutus berries were dipped in those solutions for 2 min, and then stored at 0.5 degrees C. Control consisted of uncoated fruit. On days 0, 14 and 28, samples were taken to perform physicochemical and biochemical analysis [color CIE (L*, h degrees), firmness, soluble solids content (SSC), weight Loss, trolox equivalent antioxidant capacity (TEAC), microbial growth and taste panels]. Results showed that edible coatings of 1% AL were the best to maintain most quality attributes of the commodity through storage at 0.5 degrees C. The incorporation of Cit and Eug into the alginate edible coatings improved the coatings in most cases, AL 1% + Eug 0.20% and AL 1% + Cit 0.15% + Eug 0.10% being those that better preserved sensory and nutritional attributes and reduced microbial spoilage. Thus, these coatings may be useful for improving postharvest quality and storage life of fresh arbutus fruit. (C) 2014 Elsevier B.V. All rights reserved.

The effects of cultivar and orchard on the content of ascorbic acid and flavanone glycosides as well as on the antioxidant activity of citrus juices were studied. Ascorbic acid and flavanone glycosides were determined using HPLC method. For antiradicalar activity evaluation, TAEC and ORAC methods were used. Citrus of 'Valencia late' from Faro and 'D. Joao' presented the highest levels of hesperidin in contrast to the citrus of 'Valencia late' from Silves with 0.4 mg/ml, a value closer to those of Navel oranges 'Navelate' and 'Lanelate'. Citrus of 'Valencia late' from Silves and 'D. Joao' presented the lowest levels of narirutin. 'Ortanique' and 'Encore' cultivars were poorer in ascorbic acid compared with the oranges. When the TEAC method was used for the determination of antioxidant activity, 'Valencia late' from Faro presented the best activity. A good relation was observed between antiradical activity measured through the TEAC method and the amount of ascorbic acids in samples. None relation was found between the levels of flavanones and antiradical activity. When the ORAC method was applied, the best activity was found for 'Navelate' and 'Encore' samples and practically none relation between this method and the amounts of ascorbic acid and flavanones was found.

The purpose of the present work is to determine the antioxidant capacity (AC) of 27 commercial beers. The AC indicates the degree of protection of a certain organism against oxidative damage provoked by reactive oxygen and nitrogen species. Assays were carried out by the following methods: (i) total radical trapping antioxidant parameter (TRAP); (ii) trolox equivalent antioxidant capacity (TEAC); (iii) trolox equivalent antioxidant capacity (DPPH); (iv) ferric-ion reducing antioxidant parameter (FRAP); (v) cupric reducing antioxidant capacity (CUPRAC); (vi) oxygen radical absorbance capacity (ORAC). Ascorbic acid (AA), gallic acid (GA) and trolox (TR) were used as standards. All beers showed antioxidant power, but a wide range of ACs was observed. The effect of several factors upon these differences was studied. Statistical differences were found between ACs of beers of different colours. ORAC method provided always higher experimental ACs, of significant statistical differences to other assays.

The total antioxidant capacity (TAC) of 28 flavoured water samples was assessed by ferric reducing antioxidant potential (FRAP), oxygen radical absorbance capacity (ORAC), trolox equivalent antioxidant capacity (TEAC) and total reactive antioxidant potential (TRAP) methods. It was observed that flavoured waters had higher antioxidant activity than the corresponding natural ones. The observed differences were attributed to flavours, juice and vitamins. Generally, higher TAC contents were obtained on lemon waters and lower values on guava and raspberry flavoured waters. Lower and higher TACs were obtained by TRAP and ORAC method, respectively. Statistical analysis suggested that vitamins and flavours increased the antioxidant content of the commercial waters.

Detectar los adolescentes de entre 12 y 16 años con alta capacidad intelectual o talento específico; estudiar la situación socio-educativa y las situaciones concretas de los adolescentes superdotados; y proponer un diseño de modelos de intervención para la mejora del rendimiento académico y para evitar el fracaso escolar. 11.531 niños entre 12 y 16,17 años matriculados en centros de la Comunidad de Madrid. Primero se analizan los estudios y teorías de los aspectos cognitivos, emocionales y sociales de la adolescencia y la superdotación; la orientación vocacional y profesional de los adolescentes dotados y su bajo rendimiento académico. Después el estudio empírico describe la situación de estos jóvenes en la Comunidad de Madrid. La primera fase de la experiencia consiste en la detección de adolescentes con alta capacidad intelectual a través de pruebas colectivas y nominaciones de profesores. Y la segunda es el estudio individual de características cognitivas y personales, de la situación socio-educativa y de necesidades; y estudio específico de las situaciones de las adolescentes superdotadas. Se utiliza el muestreo aleatorio estratificado y el muestreo incidental. La recogida de datos para el estudio de la inteligencia se realiza mediante Factor G de Cattell, Test de Aptitudes escolares y Escala de inteligencia Wechsler; para el estudio de la personalidad el Inventario Multifásico de Personalidad de Minnesota y Cuestionario de Observación de la Conducta Espontánea; para los datos pedagógicos Test de estrategias CEA y rendimiento académico; Test de creatividad TEAC; y cuestionarios sobre datos del centro, para el profesorado y para los padres. Se han detectado 123 alumnos superdotados que alcanzan el corte estipulado de coeficiente intelectual en el Factor G igual o superior a 130 y en la prueba TEA igual o superior a 125. Supone un 1,75 por ciento de la población, con 37 mujeres, un 30 por ciento, y 86 hombres, un 70 por ciento. En cuanto a las condiciones familiares, están por debajo de la media en divorcios o monoparentales, y en su mayoría son familias de dos hijos. Las áreas de conocimiento preferidas coinciden con aquellas donde tienen rendimientos académicos altos o muy altos, aunque un tercio están indecisos en su vocación, y las Humanidades son las preferidas. Algo menos de la mitad de los alumnos tienen alto o muy alto rendimiento, y el fracaso escolar alcanza a un tercio, lo que no se puede considerar alarmante ya que la media está en torno al 30 por ciento. Y el 14 por ciento sufre trastornos de conducta leves o transitorios, y no muestran problemas de disciplina . Los chicos prefieren la compañía de adultos o niños mayores. No existe un prototipo de familia de niños superdotados y es importante conocer los problemas de éstas para prevenir y orientar. Un gran número de familias manifiestan que sus hijos se relacionan bien y están muy orgullosos de sus logros. Los padres creen que la comunidad no tiene los recursos para atender las necesidades educativas de sus hijos. El hijo varón es el que más retos presenta para la familia lo que muestra dudas acerca de la educación, expectativas y atención prestada a las niñas de alta capacidad. En resumen, menos del 25 por ciento tienen los problemas descritos en la literatura. Al menos un 30 por ciento de los padres necesitan orientación en la vida familiar y social, y más del 40 por ciento en el área escolar. Para solucionarlo se propone una escuela de padres llevada a práctica en el cursos 99-00 con resultados altamente satisfactorios.

1. Soy isoflavones have been extensively studied because of their possible health-promoting effects. Genistein and daidzein, the major isoflavone aglycones, have received most attention; however, they undergo extensive metabolism in the gut and liver, which might affect their biological properties. 2. The antioxidant activity, free radical-scavenging properties and selected cellular effects of the isoflavone metabolites equol, 8-hydroxydaidzein, O-desmethylangiolensin, and 1,3,5 trihydroxybenzene were investigated in comparison with their parent aglycones, genistein and daidzein. 3. Electron spin resonance spectroscopy indicated that 8-hydroxydaidzein was the most potent scavenger of hydroxyl and superoxide anion radicals. Isoflavone metabolites also exhibited higher antioxidant activity than parent compounds in standard antioxidant (FRAP and TEAC) assays. However, for the suppression of nitric oxide production by activated macrophages, genistein showed the highest potency, followed by equol and daidzein. 4. The metabolism of isoflavones affects their free radical scavenging and antioxidant properties, and their cellular activity, but the effects are complex.
