Sjögren s syndrome (SS) is a common autoimmune disease affecting the lacrimal and salivary glands. SS is characterized by a considerable female predominance and a late age of onset, commonly at the time of adreno- and menopause. The levels of the androgen prohormone dehydroepiandrosterone-sulphate (DHEA-S) in the serum are lower in patients with SS than in age- and sex-matched healthy control subjects. The eventual systemic effects of low androgen levels in SS are not currently well understood. Basement membranes (BM) are specialized layers of extracellular matrix and are composed of laminin (LM) and type IV collagen matrix networks. BMs deliver messages to epithelial cells via cellular LM-receptors including integrins (Int) and Lutheran blood group antigen (Lu). The composition of BMs and distribution of LM-receptors in labial salivary glands (LSGs) of normal healthy controls and patients with SS was assessed. LMs have complex and highly regulated distribution in LSGs. LMs seem to have specific tasks in the dynamic regulation of acinar cell function. LM-111 is important for the normal acinar cell differentiation and its expression is diminished in SS. Also LM-211 and -411 seem to have some acinar specific functional tasks in LSGs. LM-311, -332 and -511 seem to have more general structure maintaining and supporting roles in LSGs and are relatively intact also in SS. Ints α3β1, α6β1, α6β4 and Lu seem to supply structural basis for the firm attachment of epithelial cells to the BM in LSGs. The expression of Ints α1β1 and α2β1 differed clearly from other LM-receptors in that they were found almost exclusively around the acini and intercalated duct cells in salivons suggesting some type of acinar cell compartment-specific or dominant function. Expression of these integrins was lower in SS compared to healthy controls suggesting that the LM-111 and -211-to-Int α1β1 and α2β1 interactions are defective in SS and are crucial to the maintenance of the acini in LSGs. DHEA/DHEA-S concentration in serum and locally in saliva of patients with SS seems to have effects on the salivary glands. These effects were first detected using the androgen-dependent CRISP-3 protein, the production and secretion of which were clearly diminished in SS. This might be due to the impaired function of the intracrine DHEA prohormone metabolizing machinery, which fails to successfully convert DHEA into its active metabolites in LSGs. The progenitor epithelial cells from the intercalated ductal area of LSGs migrate to the acinar compartment and then undergo a phenotype change into secretory acinar cells. This migration and phenotype change seem to be regulated by the LM-111-to-Int α1β1/Int α2β1 interactions. Lack of these interactions could be one factor limiting the normal remodelling process. Androgens are effective stimulators of Int α1β1 and α2β1 expression in physiologic concentrations. Addition of DHEA to the culture medium had effective stimulating effect on the Int α1β1 and α2β1 expression and its effect may be deficient in the LSGs of patients with SS.

The aim of this study was twofold- Firstly, to determine the composition of the type IV collagen which are the major components of the basement membrane (BM), in the synovial lining of the rheumatoid arthritis (RA) patient and in the BM in the labial salivary gland of the Sjögrens syndrome (SS) patient. Secondly, this thesis aimed to investigate the role of the BM component laminin α4 and laminin α5 in the migration of neutrophils from the blood vessels thorough the synovial lining layer into synovial fluid and the presence of vWF in the microvasculature of labial salivary gland in SS. Our studies showed that certain α chains type IV collagen are low in RA compared to control synovial linings, while laminin α5 exhibited a pattern of low expression regions at the synovial lining interface towards the joint cavity and fluid. Also, high numbers of macrophage-like lining cells containing MMP-9 were found in the lining. MMP-9 was also found in the synovial fluid. Collagen α1/2 (IV) mRNA was found to be present in high amount compared to the other α(IV) chains and also showed intense labelling in immunohistochemical staining in normal and SS patients. In healthy glands α5(IV) and α6(IV) chains were found to be continuous around ducts but discontinuous around acini. The α5(IV) and α6(IV) mRNAs were present in LSG explants and HSG cell line, while in SS these chains seemed to be absent or appear only in patches around the ductal BM and tended to be absent around acini in immunohistochemical staining, indicating that their synthesis and/or degradation seemed to be locally regulated around acinar cells. The provisional matrix component vWF serves as a marker of vascular damage. Microvasculature in SS showed signs of focal damage which in turn might impair arteriolar feeding, capillary transudation and venular drainage of blood. However, capillary density was not decreased but rather increased, perhaps as a result of angiogenesis compensatory to microvascular damage. Microvascular involvement of LSG may contribute to the pathogenesis of this syndrome. This twofold approach allows us to understand the intricate relation between the ECM components and the immunopathological changes that occur during the pathogenesis of these inflammatory rheumatic disease processes. Also notably this study highlights the importance of maintaining a healthy ECM to prevent the progression or possibly allow reversal of the disease to a considerable level. Furthermore, it can be speculated that a healthy BM could quarantine the inflamed region or in case of cancer cells barricade the movement of malignant cells thereby preventing further spread to the surrounding areas. This understanding can be further applied to design appropriate drugs which act specifically to maintain a proper BM/BM like intercellular matrix composition.

Interactions between tumour cells and extracellular matrix proteins of the tumour microenvironment play crucial roles in cancer progression. So far, however, there are only a few experimental platforms available that allow us to study these interactions systematically in a mechanically defined three-dimensional (3D) context. Here, we have studied the effect of integrin binding motifs found within common extracellular matrix (ECM) proteins on 3D breast (MCF-7) and prostate (PC-3, LNCaP) cancer cell cultures, and co-cultures with endothelial and mesenchymal stromal cells. For this purpose, matrix metalloproteinase-degradable biohybrid poly(ethylene) glycol-heparin hydrogels were decorated with the peptide motifs RGD, GFOGER (collagen I), or IKVAV (laminin-111). Over 14 days, cancer spheroids of 100-200µm formed. While the morphology of poorly invasive MCF-7 and LNCaP cells was not modulated by any of the peptide motifs, the aggressive PC-3 cells exhibited an invasive morphology when cultured in hydrogels comprising IKVAV and GFOGER motifs compared to RGD motifs or nonfunctionalised controls. PC-3 (but not MCF-7 and LNCaP) cell growth and endothelial cell infiltration were also significantly enhanced in IKVAV and GFOGER presenting gels. Taken together, we have established a 3D culture model that allows for dissecting the effect of biochemical cues on processes relevant to early cancer progression. These findings provide a basis for more mechanistic studies that may further advance our understanding of how ECM modulates cancer cell invasion and how to ultimately interfere with this process.

The binding of a 14 kDa beta-galactoside animal lectin to splenocytes has been studied in detail. The binding data show that there are two classes of binding sites on the cells for the lectin: a high-affinity site with a K-a ranging from 1.1 x 10(6) to 5.1 x 10(5) M-1 and a low affinity binding site with a K-a ranging from 7.7 x 10(4) to 3.4 x 10(4) M-1 The number of receptors per cell for the high- and low-affinity sites is 9 +/- 3 x 10(6) and 2.5 +/- 0.5 x 10(6) respectively. The temperature dependence of the K value yielded the thermodynamic parameters. The energetics of this interaction shows that, although this interaction is essentially enthalpically driven (Delta H - 21 kJ lambda mol(-1)) for the high-affinity sites, there is a very favorable entropy contribution to the free energy of this interaction (-T Delta S - 17.5 Jmol(-1)), suggesting that hydrophobic interaction may also be playing a role in this interaction. Lactose brought about a 20% inhibition of this interaction, whereas the glycoprotein asialofetuin brought about a 75 % inhibition, suggesting that complex carbohydrate structures are involved in the binding of galectin-1 to splenocytes, Galectin-1 also mediated the binding and adhesion of splenocytes to the extracellular matrix glycoprotein laminin, suggesting a role for it in cell-matrix interactions. Asthma is a chronic inflammatory disorder of the airways. Remodelling in asthma is defined as the structural changes seen in the airways of asthmatics in comparison to healthy controls. Progressive loss of lung function also seen in asthma might be caused by remodelling. The research aims of this thesis were to investigate inflammation and remodelling in the airways of different types of asthmatics and smokers. The association between inflammation and remodelling was also examined in a mouse model of allergic airway inflammation. Healthy smokers showed increased numbers of macrophages in the BAL with no changes in the inflammatory cells in biopsies. Macrophages seemed to be quite quiescent, since mRNA expression for a wide variety of inflammatory mediators, especially chemokines CCL3, CCL4, CCL5 and CCL20, secreted by macrophages was significantly lower than in healthy non-smokers. Attenuated macrophage activity in the airway lumen may render smokers more susceptible to airway infections and have an impact on the development of other airway pathology. Patients with diisocyanate-induced asthma (DIA) on inhaled corticosteroids (ICS) who still had non-specific bronchial hyperreactivity (NSBHR) at the end of the follow-up showed increased expression of TNF-α, IL-6 and IL-15 mRNA in BAL cells compared to those without NSBHR. In addition to being markers for poor prognosis and possible slight glucocorticoid resistance, these cytokines might aid in guiding the treatment of DIA. The increase in the thickness of tenascin-C layer in the bronchial basement membrane (BM) was much less than usually seen in other types of asthma, which might not make tenascin-C a good marker for DIA. OVA-induced tenascin-C expression in the lung was attenuated in STAT4-/- mice with impaired Th1-type immunity compared to WT mice. Interestingly, STAT6-/- mice with impaired Th2-type immunity showed tenascin-C expression levels similar to those of WT mice. The clearest difference between these two knockout strains in response to OVA was that STAT4-/- mice exhibited no upregulation of IFN-γ and TNF-α mRNA expression. Thus, tenascin-C expression was unexpectedly more related to Th1 type reactions. In vitro studies confirmed the results. Human fibroblasts stimulated by TNF-α and IFN-γ showed increased expression of tenascin-C. Patients with newly diagnosed asthma showed increased expression of laminin α2 in the bronchial BM in comparison to patients with asthma symptoms only and healthy controls. Both patients with asthma and those with only asthma symptoms showed increased expression of the laminin β2 chain in comparison to controls. Thus, laminin α2 expression differentiated patients with clinical asthma from patients with symptoms only. Furthermore, the expression of laminin α2 and β2 was associated with NSBHR, linking very specific remodelling events to clinical findings.

Sporothrix schenckii é um fungo dimórfico e agente etiológico da esporotricose, uma micose profunda que apresenta diferentes manifestações clínicas. As diversas manifestações clínicas desta e de outras doenças infecciosas podem ser relacionadas ao status imune do hospedeiro, a fatores de virulência do patógeno ou a diferentes genótipos. Dados anteriores do nosso grupo demonstram que diferenças na expressão de adesinas do S. schenckii para fibronectina estão diretamente relacionadas à virulência de diferentes cepas. Neste trabalho visamos avaliar caracteres morfológicos bioquímicos e genotípicos de doze isolados de geofílicos, zoofílicos e antropofílicos de S. schenckii, de diferentes origens geográficas, que apresentam diferentes graus de virulência. Foi analisada a morfologia das formas de micélio e levedura de cada isolado. Foi observado que a fase de micélio dos isolados estudados apresentaram morfologia típica com hifas finas e septadas, com conídios obovóides ou ovóides alongados. As leveduras apresentaram pleomorfismo típico da espécie, com células variando do formato ovóide ao alongado. Verificamos ainda a expressão de adesinas para fibronectina e laminina, do antígeno gp70 e, o padrão de bandas antigênicas reconhecidas por anticorpos IgG presentes em soro de pacientes com esporotricose ou de camundongos infectados. Para isso, foram extraídas proteínas de superfície da forma de levedura de cada isolada, sendo os extratos ensaiados por Western blot. Nestes ensaios observamos que os isolados mais virulentos de S. schenckii expressavam mais adesinas para fibronectina e laminina. A presença da gp70 foi detectada em dez dos doze isolados, sendo que apenas os isolados zoofílicos não expressam esta glicoproteína. O padrão antigênico foi variável entre os isolados, não havendo clara relação com a origem e/ou distribuição geográfica. Os dados fenotípicos foram confrontados com dados genotípicos. Para isso, sequenciamos os loci da calmodulina (CAL) e do Internal Transcribed Spacer 1/2 (ITS 1/2) a fim de averiguar se haviam diferenças genotípicas entre os isolados estudados. As análises do sequenciamento do loci CAL e ITS, contudo, apontam a divisão dos isolados em duas espécies filogenéticas, S. schenckii e S. brasiliensis não correlacionada com a distribuição geográfica dos mesmos. Nosso estudo reforça a hipótese de haver uma correlação entre virulência e expressão de adesinas, porém, sem qualquer relação entre a distribuição geográfica dos isolados zoofilicos, antropofílicos ou geofílicos, bem como dos genótipos encontrados.

A fibrose hepática é o resultado de uma resposta cicatrizante frente a repetidas lesões no fígado, e é caracterizada pelo acúmulo excessivo de proteínas da matriz extracelular (MEC) no parênquima hepático, incluindo colágeno, fibronectina, elastina, laminina e proteoglicanos, com a participação de diferentes populações celulares do fígado. As principais células responsáveis pela síntese de proteínas da MEC na fibrose hepática são as células estreladas hepáticas ativadas e os miofibroblastos, que surgem após estímulo inflamatório e são caracterizadas pela expressão de alfa-actina de músculo liso (α-SMA). Sabe-se que durante a progressão da fibrose hepática, ocorre a morte de hepatócitos e sua substituição por células fibrogênicas α-SMA+. A apoptose dessas células fibrogênicas é de grande relevância para a regressão da fibrose e regeneração hepática. Nos últimos anos, a terapia com células tronco de medula óssea tem sido utilizada para estimular a regeneração hepática em diferentes modelos experimentais e protocolos clínicos. A fração mononuclear da medula óssea adulta possui duas populações de células-tronco importantes no tratamento de diversas doenças hepáticas: células-tronco hematopoiéticas e células-tronco mesenquimais. O objetivo deste estudo foi analisar a expressão de α-SMA e o processo de apoptose de células hepáticas durante a fibrose hepática induzida por ligadura do ducto biliar (LDB) e após o transplante de células mononucleares de medula óssea (CMMO). Os fígados foram coletados de ratos dos seguintes grupos: normal, 14 dias de LDB, 21 dias de LDB e animais que receberam CMMO após 14 dias de LDB, e foram analisados após 7 dias (totalizando 21 dias de LDB). Para quantificar a expressão de α-SMA por células fibrogênicas nos grupos experimentais, foi realizada imunoperoxidase para α-SMA, seguida de morfometria no programa Image Pro Plus. Para analisar a apoptose nas células hepáticas, foi realizada imunoperoxidase e Western Blotting (WB) para caspase-3 (proteína apoptótica) e imunofluorescência com dupla-marcação para caspase-3 e α-SMA, seguida de observação em microscópio confocal. Os resultados da quantificação de α-SMA por morfometria mostraram que a expressão de α-SMA aumentou significativamente 14 e 21 dias após a LDB. Entretanto, essa expressão diminuiu significativamente no grupo tratado com CMMO, que apresentou parênquima hepático mais preservado em relação ao grupo com 21 dias de LDB. Os resultados de imunoperoxidase, WB e microscopia confocal para expressão de caspase-3 demonstraram que essa proteína diminuiu nos animais fibróticos com 14 e 21 dias de LDB com relação ao grupo normal, e estava significativamente elevada no grupo tratado com CMMO. A análise por microscopia confocal demonstrou que algumas células coexpressaram α-SMA e caspase-3 nos animais tratados com CMMO, sugerindo a morte de células fibrogênicas e remodelamento do parênquima hepático.

A medula óssea adulta possui duas populações de células-tronco importantes no tratamento de diversas doenças hepáticas: células-tronco hematopoiéticas (CTHs) e células-tronco mesenquimais. A regeneração do fígado após a hepatectomia é um processo complexo que requer a proliferação de todas as células hepáticas. Fatores de crescimento, citocinas e componentes da matriz extracelular são elementos-chave nesse processo. As lamininas são uma família de proteínas de matriz extracelular, com funções adesivas e quimiotáticas pelo recrutamento de integrinas e outros receptores de superfície celular. No fígado normal, a laminina é expressa nas veias porta e centrolobular. O objetivo desse estudo foi investigar a expressão de laminina durante a regeneração hepática induzida por hepatectomia parcial e após o transplante de células mononucleares de medula óssea. As células mononucleares de medula óssea foram obtidas dos fêmures e tíbias de ratos, isoladas, marcadas com DAPI e injetadas pela veia porta em ratos recém-hepatectomizados. Os fígados foram coletados 15 minutos, 1 dia e 3 dias após a hepatectomia e o transplante de células de medula óssea e congelados. Os cortes foram imunomarcados com anticorpos primários anti-CD34 e anti-laminina de rato e observados em microscópio confocal de varredura a laser. Os resultados mostraram que 15 minutos após a hepatectomia parcial, as células-tronco hematopoiéticas CD34+ transplantadas foram encontradas em contato com a laminina localizada nas veias porta e centrolobular, indicando que a laminina poderia participar na adesão inicial das células-tronco a esses vasos logo após o seu transplante. Além disso, 1 e 3 dias após a hepatectomia, as células mononucleares de medula óssea transplantadas foram observadas nos sinusóides hepáticos expressando laminina. Esses resultados sugerem que a laminina pode ser um componente da matriz extracelular importante para a adesão e enxerto de células de medula óssea no fígado após uma lesão. Nós também analisamos a expressão de osteopontina (OPN) em células de medula óssea e CTHs. Os resultados por microscopia confocal demonstraram que a maioria das células mononucleares de medula óssea recém-isoladas expressa quantidades variáveis de OPN. Além disso, algumas CTHs CD34+ também expressam OPN. Após 1 e 4 dias de cultura, observamos uma diminuição de células expressando CD34, e um aumento na expressão de OPN pelas células mononucleares de medula óssea.

Purpose Retinal ganglion cells (RGCs) are exposed to injury in a variety of optic nerve diseases including glaucoma. However, not all cells respond in the same way to damage and the capacity of individual RGCs to survive or regenerate is variable. In order to elucidate factors that may be important for RGC survival and regeneration we have focussed on the extracellular matrix (ECM) and RGC integrin expression. Our specific questions were: (1) Do adult RGCs express particular sets of integrins in vitro and in vivo? (2) Can the nature of the ECM influence the expression of different integrins? (3) Can the nature of the ECM affect the survival of the cells and the length or branching complexity of their neurites? Methods Primary RGC cultures from adult rat retina were placed on glass coverslips treated with different substrates: Poly-L-Lysine (PL), or PL plus laminin (L), collagen I (CI), collagen IV (CIV) or fibronectin (F). After 10 days in culture, we performed double immunostaining with an antibody against beta III-Tubulin to identify the RGCs, and antibodies against the integrin subunits: alpha V, alpha 1, alpha 3, alpha 5, beta 1 or beta 3. The number of adhering and surviving cells, the number and length of the neurites and the expression of the integrin subunits on the different substrates were analysed. Results PL and L were associated with the greatest survival of RGCs while CI provided the least favourable conditions. The type of substrate affected the number and length of neurites. L stimulated the longest growth. We found at least three different types of RGCs in terms of their capacity to regenerate and extend neurites. The different combinations of integrins expressed by the cells growing on different substrata suggest that RGCs expressed predominantly alpha 1 beta 1 or alpha 3 beta 1 on L, alpha 1 beta 1 on CI and CIV, and alpha 5 beta 3 on F. The activity of the integrins was demonstrated by the phosphorylation of focal adhesion kinase (FAK). Conclusions Adult rat RGCs can survive and grow in the presence of different ECM tested. Further studies should be done to elucidate the different molecular characteristics of the RGCs subtypes in order to understand the possible different sensitivity of different RGCs to damage in diseases like glaucoma in which not all RGCs die at the same time.

A fibrose hepática é o resultado de uma lesão crônica, com a ativação de células inflamatórias e fibrogênicas no fígado, as quais levam a um acúmulo excessivo de proteínas de matriz extracelular (MEC). Essas alterações resultam na morte de células do fígado, com desorganização e perda da função do parênquima hepático. A cirrose é o estágio avançado da fibrose, e culmina na falência hepática, uma condição potencialmente fatal cujo único tratamento efetivo é o transplante de fígado, o qual é limitado pela disponibilidade de órgãos. Na busca por terapias alternativas visando a regeneração hepática, o transplante de células mononucleares de medula óssea (CMMO) mostrou resultados benéficos e promissores em modelos animais e alguns protocolos clínicos. Entre essas células, estão as células-tronco hematopoiéticas e mesenquimais, que apresentam potencial regenerativo e modulador da resposta inflamatória. Este estudo pretendeu avançar na compreensão dos mecanismos pelos quais as CMMO podem ajudar na regeneração hepática. Ratos com fibrose hepática induzida por ligadura do ducto biliar (LDB) foram transplantados com CMMO e comparados com ratos com fibrose sem transplante e ratos normais. Parâmetros hepáticos como componentes da MEC (colágeno total, colágenos tipos I e IV, laminina, metaloproteinases de matriz MMPs), componentes celulares (células fibrogênicas, células de Kupffer e colangiócitos) e enzimas hepáticas foram analisados por microscopia de luz, microscopia confocal, western blotting e espectrofotometria. Os resultados mostraram que o transplante de CMMO contribui para a regeneração hepática de maneira global, (a) diminuindo o acúmulo de colágeno e laminina; (b) aumentando a produção de MMPs que favorecem o remodelamento da MEC, principalmente por células de Kupffer; (c) normalizando a quantidade de colangiócitos e diminuindo a quantidade de células fibrogênicas; e (d) normalizando os níveis sanguíneos das enzimas hepáticas. Portanto, nós sugerimos que as CMMO podem ajudar na regeneração hepática através de mecanismos parácrinos e se diferenciando em células de Kupffer, contribuindo para a secreção de fatores antiinflamatórios e anti-fibrogênicos no fígado com fibrose.

A interleucina 13 (IL-13) tem sido apontada como um dos principais mediadores em processos de ativação de fibroblastos e indução de fibrose pulmonar, sendo, portanto, considerada como um alvo terapêutico importante. A silicose é uma doença pulmonar inflamatória crônica, de caráter ocupacional, caracterizada por uma intensa resposta fibrótica e granulomatosa. Com base nestas observações, tivemos por objetivo investigar o potencial efeito da administração da imunotoxina IL-13-PE38QQR (IL-13PE) sobre o modelo de silicose em camundongos. Camundongos Swiss-Webster foram anestesiados e instilados intranasalmente com partículas de sílica (10 mg), sendo a administração da IL-13PE (200ng/dia) realizada por via intranasal, uma vez ao dia em dias alternados no período entre 21 a 27 dias após a provocação. Analisamos o componente inflamatório, a deposição de colágeno e a área de granuloma avaliados através de técnicas clássicas de histologia, incluindo coloração com H&E e Picrus-sirius, ou ainda a quantificação do conteúdo de colágeno por Sircol. Os componentes de matriz extracelular fibronectina e laminina foram avaliados através de imunohistoquímica. Citocinas e quimiocinas foram quantificadas por sistema de ELISA. As medidas de função pulmonar e resposta de hiperreatividade foram realizadas através do sistema de pletismografia de corpo inteiro invasiva. Verificamos que o tratamento curativo com a IL-13PE inibiu de forma acentuada o comprometimento da função pulmonar nos camundongos silicóticos, incluindo tanto aumento da resistência como da elastância, assim como a resposta de hiperratividade das vias aéreas ao agente broncoconstrictor metacolina. De forma coerente, os animais silicóticos quando submetidos ao tratamento com IL-13PE apresentaram marcada redução do componente inflamatório pulmonar e da resposta fibrótica, atestado pela diminuição na produção de colágeno, laminina e fibronectina e redução importante da área de granuloma. De forma semelhante, as citocinas (TNF-α e TGF-) e quimiocinas (MIP-1α, MIP-2, TARC, IP-10, MDC) detectadas em quantidade aumentada no pulmão de animais silicóticos foram reduzidas pelo tratamento com a IL-13PE. Em conclusão, nossos resultados mostram que a administração curativa da IL-13PE foi capaz de inibir os componentes inflamatórios e fibróticos da fase crônica do quadro silicótico em camundongos, o que se refletiu de forma clara na melhora da função pulmonar. Em conjunto, nossos achados indicam que a utilização da IL13PE parece constituir uma abordagem terapêutica extremamente promissora para aplicação em casos de doenças crônicas de natureza fibrótica como a silicose.

灵长类胚胎干细胞(ES 细胞)不仅能为研究生殖发育生物学基础理论提供良好的 模型,而且可为细胞替代治疗提供大量的供体细胞,因此具有重要的研究价值。当前 灵长类ES 细胞研究还有很多问题需要解决,如分离建立更多的胚胎干细胞系,优化培 养体系,提高ES 细胞定向分化为特定细胞的比例,研究ES 细胞自我更新和分化的机 制等。本文一方面概括了灵长类ES 细胞的研究进展,另一方面并对制备抗体,免疫外 科手术法分离灵长类胚胎内细胞团,建立猕猴ES 细胞的无饲养层、无血清培养体系和 诱导猕猴ES 细胞分化成高纯度的O2A 神经胶质前体细胞进行了研究。主要结论如下: 1)分别以猕猴脾脏淋巴细胞和人外周血单个核细胞作为免疫原,免疫日本大耳白兔, 得到免疫血清。在补体介导的细胞毒作用下,兔抗人和兔抗猕猴免疫血清可以裂解人 和猕猴囊胚滋养层细胞,从而分离出内细胞团,用于分离培养人和猕猴胚胎干细胞。2) 猕猴ES 细胞在以层粘连蛋白(laminin)为胞外基质,含转化生长因子beta1(TGFβ1) 的无血清培养基(SFM)中可以稳定的增殖至少22 代,保持不分化,并具有分化成三 个胚层细胞的能力。进一步的研究发现去除TGFβ1 后,猕猴ES 细胞出现分化,整合 素表达降低,推测TGFβ1 可能通过促进猕猴ES 细胞整合素的表达,加强其与胞外基 质的相互作用,从而维持ES 细胞的自我更新。然而猕猴ES 细胞不能在纤粘连蛋白 (fibronectin)和明胶上生长。3)无饲养层、无血清培养体系中长期培养的猕猴ES 细 胞,分化出拟胚体,14 天的拟胚体在血清中分化培养一周后,在含碱性成纤维生长因 子bFGF、表皮生长因子EGF 和胰岛素+转铁蛋白+亚硒酸钠ITS 的培养基中培养, 获得97%的O2A 胶质前体细胞,得到的O2A 细胞能够稳定增殖,并且可以自发分化 为II 型星型胶质细胞和少突胶质细胞。本实验的结果有助于猕猴ES 细胞分离建系和培 养系统的优化、推动猕猴ES 细胞自我更新和诱导为神经胶质细胞机制的研究,便于建 立ES 细胞替代治疗的猕猴模型,从而为人类ES 细胞的临床疾病治疗提供参考。

血栓栓塞性疾病严重影响人类的健康,是导致死亡率最高的病因之一。在过去 的几十年中,已经开发了针对血液凝固、血小板激活和聚集、血栓溶解的不同步骤而 起作用的基础和临床药物,但是许多抗血栓药物经常被包括低血压、出血等系统性的 副作用所限制,因此,需要开发新型的更具潜力和特异性的抗血栓药物。 吸血节肢动物与宿主相互作用的分子机制研究是目前国际研究的热点之一,通过 该类研究可以发现大量的具有药用前景的先导分子和提供节肢动物控制策略。我们通 过生物化学、分子生物学以及药理学研究手段首次从中国特有的姚虻(Tabanus yao Macquart)唾液腺发现了同时具有水解纤维蛋白原和抑制血小板聚集的双功能的 丝氨酸蛋白酶Tablysin、纤维蛋白水解酶TY6和腺苷三磷酸双磷酸酶Tabapy等3个家 族的抗血栓活性成分。并研究了这3类因子在牛虻唾液腺中的分子多样性、它们的结 构基础及发挥抗血栓功能的作用机制,通过实验动物模型研究体内外抗血栓能力,深 入解析了牛虻作为传统抗血栓中药的物质基础,为发掘具有良好抗血栓功能的新型候 选药物分子,为传统活血化瘀中药牛虻的现代化开发提供了广阔的思路和打下了坚实 的基础。 研究表明,姚虻纤维蛋白原水解酶Tablysin为一种单链蛋白。其表观分子量为 27kDa。此酶为一种丝氨酸蛋白酶家族的新成员,其活性不受金属离子螯合剂EDTA 和还原剂DTT的影响。血浆蛋白酶抑制剂Aprotinin,α-macroglobulin能部分抑制该 酶活性。此酶适应的pH范围较广(pH 2.4-10.0),但对热变化不具很好的耐受性。 Edman降解法进行氨基酸测序分析发现,此酶的N-末端有封闭现象,后经Q-TOF质 谱分析,确定此酶的部分氨基酸序列。此外,我们成功构建了具有完整性的姚虻唾液 腺cDNA文库,获得2×106 pfu个重组子,进一步的筛选了Tablysin的编码基因。 对此酶的溶栓机制进行分析发现,其具有直接水解纤维蛋白原的活性,不具有纤 溶酶原激活剂的活性。此酶能明显地水解纤维蛋白原的α-链,而对β-链的水解活性较 低,不能水解γ链(α > β > γ)。此酶不能有效地水解纤维蛋白,不会水解层粘连蛋白 (Laminin)和纤维粘连蛋白(Fibronectin)等基质蛋白。不具有出血活性。进 一步探讨此酶的作用机制,发现其能以剂量依赖的方式抑制ADP诱导的血小板聚 集。利用流式细胞仪检测到该酶能与血小板膜糖蛋白受体GPIIb / IIIa结合,因此推测其可能是通过抑制血小板和纤维蛋白原结合的作用机制来抑制血小板 的聚集。 我们通过将目的基因连接到pET-32a+载体,并在大肠杆菌表达体系中得到 了高效表达。体外活性检测表明重组蛋白质与天然产物活性相当,用其检测了 其对角叉菜胶致小鼠尾静脉血栓模型体内血栓形成的影响, 实验结果表明 Tablysin具有明显的抗静脉血栓作用。该工作对有潜力开发成为新型单组分抗血栓 药物的姚虻活性蛋白进行了较为系统的理化性质研究,为该分子的进一步开发和研究 奠定了基础。 我们从姚虻唾液腺中分离纯化到一组具有水解纤维蛋白原α-链活性的酶, 命名为TY6家族。对TY6的生化性质进行研究,表明此酶为一种单链的丝氨酸蛋白 酶,其表观分子量为27 kDa,其活性不受金属离子螯合剂EDTA的影响,但能被PMSF 所抑制。Molish反应显示为阴性,说明此酶不是一种糖蛋白。从姚虻唾液腺cDNA 文库中克隆得到其编码序列,发现该酶家族的编码基因在一级结构上表现出丰 富的多样性。通过Western blotting检测到其能与牛虻叮咬后过敏患者血清特异性IgE 结合。推测该基因的多样性是吸血昆虫适应吸血寄生生活而采取的进化策略。 该工作为牛虻唾液腺抗血栓功能基因的继续筛选和分析奠定了基础,并为牛虻的生物 防治提供了思路和对策。 我们从姚虻唾液腺中分离纯化到一个具有水解ADP活性的酶(Apyrase), 命名为Tabapy。活性检测发现Tabapy具有明显的水解ADP的活性,并能以剂量 依赖的方式抑制ADP诱导的血小板聚集。通过Western blotting检测到Tabapy能与 牛虻叮咬后过敏患者血清特异性IgE结合。用PCR方法从姚虻唾液腺cDNA文库中 克隆得到编码序列,经BLAST分析表明,该酶与来源于斑虻唾液腺的血小板聚 集抑制剂Chrysoptin具有90%的序列相似性,并经过Q-TOF分析确证,有5个肽段 的氨基酸残基与该cDNA序列推导的多肽链匹配。该工作为研究吸血节肢动物 吸血生理及进一步研究Apyrase的结构和功能打下了基础。

The extracellular matrix (ECM) of the human intervertebral disc is rich in molecules that interact with cells through integrin-mediated attachments. Porcine nucleus pulposus (NP) cells have been shown to interact with laminin (LM) isoforms LM-111 and LM-511 through select integrins that regulate biosynthesis and cell attachment. Since human NP cells lose many phenotypic characteristics with age, attachment and interaction with the ECM may be altered. Expression of LM-binding integrins was quantified for human NP cells using flow cytometry. The cell-ECM attachment mechanism was determined by quantifying cell attachment to LM-111, LM-511, or type II collagen after functionally blocking specific integrin subunits. Human NP cells express integrins β1, α3, and α5, with over 70% of cells positive for each subunit. Blocking subunit β1 inhibited NP cell attachment to all substrates. Blocking subunits α1, α2, α3, and α5 simultaneously, but not individually, inhibits NP cell attachment to laminins. While integrin α6β1 mediated porcine NP cell attachment to LM-111, we found integrins α3, α5, and β1 instead contributed to human NP cell attachment. These findings identify integrin subunits that may mediate interactions with the ECM for human NP cells and could be used to promote cell attachment, survival, and biosynthesis in cell-based therapeutics.

A large percentage of the population may be expected to experience painful symptoms or disability associated with intervertebral disc (IVD) degeneration - a condition characterized by diminished integrity of tissue components. Great interest exists in the use of autologous or allogeneic cells delivered to the degenerated IVD to promote matrix regeneration. Induced pluripotent stem cells (iPSCs), derived from a patient's own somatic cells, have demonstrated their capacity to differentiate into various cell types although their potential to differentiate into an IVD cell has not yet been demonstrated. The overall objective of this study was to assess the possibility of generating iPSC-derived nucleus pulposus (NP) cells in a mouse model, a cell population that is entirely derived from notochord. This study employed magnetic activated cell sorting (MACS) to isolate a CD24(+) iPSC subpopulation. Notochordal cell-related gene expression was analyzed in this CD24(+) cell fraction via real time RT-PCR. CD24(+) iPSCs were then cultured in a laminin-rich culture system for up to 28 days, and the mouse NP phenotype was assessed by immunostaining. This study also focused on producing a more conducive environment for NP differentiation of mouse iPSCs with addition of low oxygen tension and notochordal cell conditioned medium (NCCM) to the culture platform. iPSCs were evaluated for an ability to adopt an NP-like phenotype through a combination of immunostaining and biochemical assays. Results demonstrated that a CD24(+) fraction of mouse iPSCs could be retrieved and differentiated into a population that could synthesize matrix components similar to that in native NP. Likewise, the addition of a hypoxic environment and NCCM induced a similar phenotypic result. In conclusion, this study suggests that mouse iPSCs have the potential to differentiate into NP-like cells and suggests the possibility that they may be used as a novel cell source for cellular therapy in the IVD.
