A presente investigação tem como objectivo analisar e descrever as mudanças ocorridas, em particular a transformação narrativa nos pais que frequentaram um grupo de Formação Parental, aplicando para tal, a Grelha de Análise do Processo Terapêutico (GAPT) enquanto instrumento de avaliação da mudança narrativa, ao longo das sessões do programa. A GAPT é constituída por 5 dimensões, que, correspondem aos eixos considerados importantes na transformação narrativa, ou seja na mudança dos sistemas. Nesse sentido foram transcritas e cotadas em painel de peritos, 10 sessões do Programa “Incredible Years”, tendo em conta os 4 subsistemas parentais que o frequentaram. Verificou-se que ocorreram Singularidades, em grande número, desde o início da intervenção e que estas tendiam gradualmente em diversificar-se, flexibilizando a narrativa: no Eixo das Interacções, na Forma como é Narrada a História, na sua Conotação e nos seus Temas Centrais. Em suma a amplificação das Singularidades possibilita a reorganização e reconceptualização coerente das experiências do sujeito, que quanto mais complexa for mais coerente é o seu significado

This thesis addresses the Batch Reinforcement Learning methods in Robotics. This sub-class of Reinforcement Learning has shown promising results and has been the focus of recent research. Three contributions are proposed that aim to extend the state-of-art methods allowing for a faster and more stable learning process, such as required for learning in Robotics. The Q-learning update-rule is widely applied, since it allows to learn without the presence of a model of the environment. However, this update-rule is transition-based and does not take advantage of the underlying episodic structure of collected batch of interactions. The Q-Batch update-rule is proposed in this thesis, to process experiencies along the trajectories collected in the interaction phase. This allows a faster propagation of obtained rewards and penalties, resulting in faster and more robust learning. Non-parametric function approximations are explored, such as Gaussian Processes. This type of approximators allows to encode prior knowledge about the latent function, in the form of kernels, providing a higher level of exibility and accuracy. The application of Gaussian Processes in Batch Reinforcement Learning presented a higher performance in learning tasks than other function approximations used in the literature. Lastly, in order to extract more information from the experiences collected by the agent, model-learning techniques are incorporated to learn the system dynamics. In this way, it is possible to augment the set of collected experiences with experiences generated through planning using the learned models. Experiments were carried out mainly in simulation, with some tests carried out in a physical robotic platform. The obtained results show that the proposed approaches are able to outperform the classical Fitted Q Iteration.

The major aim of this study was to test the hypothesis that the introduction of the Nile tilapia (Oreochromis niloticus) and the enrichment with nutrients (N and P) interact synergistically to change the structure of plankton communities, increase phytoplankton biomass and decrease water transparency of a semi-arid tropical reservoir. One field experiment was performed during five weeks in twenty enclosures (8m3) to where four treatments were randomly allocated: with tilapia addition (T), with nutrients addition (NP), with tilapia and nutrients addition (T+NP) and a control treatment with no tilapia or nutrients addition (C). A two-way repeated measures ANOVA was done to test for time (t), tilapia (T) and nutrient (NP) effects and their interaction on water transparency, total phosphorus, total nitrogen, phytoplankton and zooplankton. The results show that there was no effect of nutrient addition on these variables but significant fish effects on the biomass of total zooplankton, nauplii, rotifers, cladocerans and calanoid copepods, on the biovolume of Bacillariophyta, Zygnemaphyceae and large algae (GALD ≥ 50 μm) and on Secchi depth. In addition, we found significant interaction effects between tilapia and nutrients on Secchi depth and rotifers. Overall, tilapia decreased the biomass of most zooplankton taxa and large algae (diatoms) and decreased the water transparency while nutrient enrichment increased the biomass of zooplankton (rotifers) but only in the absence of tilapia. In conclusion, the influence of fish on the reservoir plankton community and water transparency was greater than that of nutrient loading. This finding suggests that biomanipulation should be a greater priority in the restoration of eutrophic reservoirs in tropical semi-arid regions

The lamina-associated polypeptide 1 (LAP1) is a type II transmembrane protein of the inner nuclear membrane encoded by the human gene TOR1AIP1. LAP1 is involved in maintaining the nuclear envelope structure and appears be involved in the positioning of lamins and chromatin. In the nuclear envelope, LAP1 is suggested to exist as a complex with A-type and B-type lamins, torsins and emerin. The presence of such complexes suggests that LAP1 may cooperate functionally with these proteins in tissues where they play a critical role. Therefore, the identification of LAP1 binding partners and the signalling pathways where LAP1 participates, is crucial for a better understanding of LAP1 functions. The work described in this thesis addresses novel human LAP1 associated proteins found through bioinformatic tools. Public databases allowed for the discovery of the LAP1 interactome, which was manually curated, identifying several functionally relevant proteins. Subsequently, the integration of multiple bioinformatic tools established novel functions to LAP1 such as DNA damage response and telomere association. In conjunction, bioinformatic results also reinforced the association of LAP1 with mitosis, and the already identified role of LAP1 in nuclear morphology. Interestingly, this association of LAP1 with the regulation of the nuclear envelope structure and mitosis progression, shares functional elements with spermatogenesis. Therefore, this work additionally described the localization of LAP1 and some of its interactors throughout the spermatogenic cycle, in mouse and human testis. The results established that the activity of LAP1 during the mouse spermatogenic cycle is most evident from stage VIII until the end of spermiogenesis, which is characteristic of manchette development. Concomitantly, some LAP1 interactors studied in this work share a similar localization, namely, PP1γ2, Lamin B1 and Lamin A/C. The results obtained from the study of LAP1 throughout different periods of the male reproductive system attributed potential new biological functions to LAP1. Thereby, this work can be the foundation of future studies regarding LAP1 and the regulation of multiple cellular processes and disease conditions.

Artificial lakes must differ from natural lakes in important structural and functional aspects that need to be understood so that these ecosystems can be properly managed. The aim of this work was to test the hypothesis that the artificial lakes (impoundments) in the semi-arid region of the Rio Grande do Norte State are more eutrophic and turbid and have different trophic structure when compared to the natural coastal lakes that occur in the humid eastern coast of the State. To test this hypothesis, 10 natural lakes and 8 artificial lakes with about 100 ha were sampled between September and November 2005 for the determination of some limnological variables and the abundance of the main fish species, which were grouped in three trophic guilds: facultative piscivores, facultative planktivores and omnivores. The results show that the artificial lakes had significantly higher concentrations of total nitrogen, total phosphorus, chlorophyll a , total and volatile suspended solids than the natural lakes. Results also show that the values of pH, total alkalinity, electric conductivity, turbidity as well as the coefficient of vertical attenuation of light were significantly higher in the artificial lakes than in the natural lakes. In the artificial lakes, the abundance of facultative planktivores was significantly higher, while the abundance of facultative piscivores significantly lower than in the natural lakes. There was no significant difference in the abundance of omnivorous fish between the two types of lakes. These results suggest that the increase in turbidity together with the other changes in the water quality of the artificial lakes, modifies the trophic structure of the fish communities reducing the importance of piscivores and the length of the food chains

Fertilization is a multistep and complex process culminating in the merge of gamete membranes, cytoplasmic unity and fusion of genome. CD81 is a tetraspanin protein that participates in sperm-oocyte interaction, being present at the oocyte surface. CD81 has also been implicated in other biological processes, however its specific function and molecular mechanisms of action remain to be elucidated. The interaction between CD81 and its binding partner proteins may underlie the CD81 involvement in a variety of cellular processes and modulate CD81/interactors specific functions. Interestingly, in a Yeast two Hybrid system previously performed in our lab, CD81 has emerged as a putative interactor of the Amyloid Precursor Protein (APP). In the work here described, bioinformatics analyses of CD81 interacting proteins were performed and the retrieved information used to construct a protein-protein interaction network, as well as to perform Gene Ontology enrichment analyses. CD81 expression was further evaluated in CHO, GC-1 and SH-SY5Y cell lines, and in human sperm cells. Additionally, its subcellular localization was analyzed in sperm cells and in the neuronal-like SH-SY5Y cell line. Subsequently, coimmunoprecipitation assays were performed in CHO and SH-SY5Y cells to attempt to prove the physical interaction between CD81 and APP. A functional interaction between these two proteins was accessed thought the analyses of the effects of CD81 overexpression on APP levels. A co-localization analysis of CD81 and some interactors proteins retrieved from the bioinformatics analyses, such as APP, AKT1 and cytoskeleton-related proteins, was also performed in sperm cells and in SH-SY5Y cells. The effects of CD81 in cytoskeleton remodeling was evaluated in SH-SY5Y cells through monitoring the effects of CD81 overexpression in actin and tubulin levels, and analyzing the colocalization between overexpressed CD81 and F-actin. Our results showed that CD81 is expressed in all cell lines tested, and also provided the first evidence of the presence of CD81 in human sperm cells. CD81 immunoreactivity was predominantly detected in the sperm head, including the acrosome membrane, and in the midpiece, where it co-localized with APP, as well as in the post-acrosomal region. Furthermore, CD81 co-localizes with APP in the plasma membrane and in cellular projections in SH-SY5Y cells, where CD81 overexpression has an influence on APP levels, also visible in CHO cells. The analysis of CD81 interacting proteins such as AKT1 and cytoskeletonrelated proteins showed that CD81 is involved in a variety of pathways that may underlie cytoskeleton remodeling events, related to processes such as sperm motility, cell migration and neuritogenesis. These results deepen our understanding on the functions of CD81 and some of its interactors in sperm and neuronal cells.

O trabalho pretende questionar se as necessidades da gestão da floresta e as interacções com o ambiente, justificam o repensar do conceito de propriedade privada, na definição de políticas florestais concretizadoras duma «mundividência ambiental». Estrutura-se em quatro partes: primeiro, procura-se dar uma perspectiva do direito de propriedade privada, a sua perenidade na nossa cultura, sobretudo na cultura jurídica; segundo, analisa-se a dinâmica da propriedade privada da floresta nos últimos séculos, e as soluções de políticas florestais- destacando-se a criação das zonas de intervenção florestal (ZIF); terceiro, estuda-se em concreto o regime de certificação florestal, como forma específica de gestão florestal; quarto, sublinham-se novos desafios ambientais à gestão das florestas, designadamente a identificação dos problemas que emergem em resultado das alterações climáticas, destacando-se o sequestro de carbono e a criação do mercado de carbono. Este estudo não é estanque, faz parte de um mesmo questionamento: é o ambiente um valor para a humanidade ou é um valor em si mesmo? ABSTRACT: The work intends to question if the private property of the forest, toward the necessities of management and interactions with the environment, justifies the rethink of the concept, in forest politics definition, producer of an interactive view of the environment. lt is structured in four parts: first, it intends to give a perspective of the right of private property, its longevity in our culture, the importance of legal quarrel; second, the dynamics of the private property of the forest, in the latest centuries, and the solutions of the implemented forest politics - being distinguished the creation of zones of forest intervention, the ZIF; third, the forest management ­ forest certification; fourth, the environmental source, the identification of problems that emerge in result of climatic alterations, being distinguished the kidnap of carbon and the creation of the carbon market. This study is not tight; it is part of the same reflection: of being the environment a value for the humanity or a value in itself exactly.

A transição da adolescência para a idade adulta é uma fase crucial do desenvolvimento de qualquer jovem, na qual são colocados vários desafios em relação ao seu futuro. Os adolescentes com deficiência enfrentam desafios adicionais: quando comparados com os seus pares, têm mais dificuldades em encontrar um percurso de vida adulta, por exemplo no emprego ou no prosseguimento de estudos. Por outro lado, a baixa participação social é um dos fatores que mais afeta a qualidade de vida. Percebendo-se que o processo de transição é multifacetado, as seis investigações apresentadas neste trabalho colocam o enfoque no próprio indivíduo, mas também em contextos importantes para a transição como a família, através da seguinte sequência de estudos: (1) dois estudos preliminares que caracterizam adolescentes e jovens com deficiência no que se refere a comportamentos ligados à saúde, estilos de vida e respetivas interacções com a satisfação com a vida; (2) dois estudos qualitativos que confirmam a problemática da transição e aferem os fatores críticos envolvidos, através das percepções de pais e jovens; e (3) dois estudos que fazem a análise do comportamento destes factores numa amostra de adolescentes e jovens com deficiência. Os resultados revelam um padrão de saúde e sintomático mais negativo nos jovens com deficiência quando comparados com os seus pares, e salientam a importância da relação com os outros significativos, em especial os pais. Os vários estudos mostram ainda que os contextos de vida devem proporcionar suportes adequados e atempados à transição e apelam para a importância do desenvolvimento de aptidões críticas, tais como a promoção de competências de autodeterminação, autoeficácia, e promoção da resiliência. Os resultados dos estudos são discutidos de forma integrada. São também feitas recomendações futuras desde a perspectiva individual até à das políticas e planeamento. Finalmente, é proposto um modelo do planeamento e desenvolvimento da transição que pretende ser orientador para todos os atores envolvidos no processo.

Over the last few years, football entered in a period of accelerated access to large amount of match analysis data. Social networks have been adopted to reveal the structure and organization of the web of interactions, such as the players passing distribution tendencies. In this study we investigated the influence of ball possession characteristics in the competitive success of Spanish La Liga teams. The sample was composed by OPTA passing distribution raw data (n=269,055 passes) obtained from 380 matches involving all the 20 teams of the 2012/2013 season. Then, we generated 760 adjacency matrixes and their corresponding social networks using Node XL software. For each network we calculated three team performance measures to evaluate ball possession tendencies: graph density, average clustering and passing intensity. Three levels of competitive success were determined using two-step cluster analysis based on two input variables: the total points scored by each team and the scored per conceded goals ratio. Our analyses revealed significant differences between competitive performances on all the three team performance measures (p < .001). Bottom-ranked teams had less number of connected players (graph density) and triangulations (average clustering) than intermediate and top-ranked teams. However, all the three clusters diverged in terms of passing intensity, with top-ranked teams having higher number of passes per possession time, than intermediate and bottom-ranked teams. Finally, similarities and dissimilarities in team signatures of play between the 20 teams were displayed using Cohen’s effect size. In sum, findings suggest the competitive performance was influenced by the density and connectivity of the teams, mainly due to the way teams use their possession time to give intensity to their game.
