Há cerca de 20 anos a vanilina vem sendo descrita como uma substância moduladora capaz de inibir eventos relacionados à indução e promoção do processo carcinogênico. Este comportamento associado ao seu consumo elevado despertou o nosso interesse científico - resultando na publicação do primeiro trabalho associando a VA a acréscimos expressivos em eventos recombinacionais mitóticos, acompanhados de decréscimos na freqüência de mutações pontuais e cromossômicas. Entretanto, quando a antimutagênese e a co-recombinogênese foram avaliadas simultaneamente, a ação final da VA refletiu-se não como proteção, mas sim como um efeito potencializador expresso como um aumento de cerca de 200 vezes na genotoxicidade total da MMC. Na procura de respostas adicionais concernentes à ação da VA como moduladora de diferentes espectros de lesões no DNA utilizamos o Teste para Detecção de Mutação e Recombinação Somática em Drosophila melanogaster (SMART) com o intuito de avaliar o comportamento deste flavorizante em relação à genotoxicidade dos agentes químicos: N-methyl-N-nitrosourea (MNU), N-ethyl-N-nitrosourea (ENU), ethylmethanesulphonate (EMS) e bleomicina (BLEO), em dois protocolos de administração do modulador – pós e co-tratamento. Pós-tratamento Os dados obtidos através do sistema de pós-tratamento evidenciaram que a VA não altera a mutagenicidade e a recombinogenicidade do ENU e MNU - o que sugere a não interferência deste flavorizante sobre os mecanismos envolvidos na correção das lesões induzidas por estes alquilantes. Ao contrário, a toxicidade genética do EMS foi significativamente aumentada em valores compreendidos entre 7,79 a 29,79%, representando a expressão final de dois efeitos antagônicos: (i) sinergismo em recombinação mitótica e (ii) proteção em relação à mutagênese. Tais achados sugerem que diferenças entre o espectro dos danos induzidos por estes agentes alquilantes, podem afetar os caminhos de reparação a serem priorizados. Como conseqüência, o efeito potencializador da VA sobre recombinação homóloga (HR) está restrito ao EMS – o único dos agentes alquilantes monofuncionais estudados cujas lesões são processadas, em Drosophila melanogaster, por ambos mecanismos de reparo: excisão de nucleotídeos e pós-replicativo. A VA também causou drásticos incrementos na genotoxicidade da BLEO - 120 a 178% - que estão limitados a aumentos em recombinação, uma vez que não foram observadas alterações na sua potência mutacional. Como a genotoxicidade da BLEO resulta basicamente da indução de quebras duplas corrigidas por mecanismos de reparação dependentes de recombinação - que podem ocorrer tanto entre cromossomos homólogos (HR) como não-homólogos (end joining -NHEJ) – e como o teste SMART privilegia a detecção de recombinação homóloga, os nossos dados indicam que a ação potencializadora de VA em relação a BLEO deve-se especificamente a incrementos em reparo dependente de HR. Ainda relevante é o fato de que estes acréscimos não estão associados a decréscimos em mutação, como anteriormente observado para a MMC.Todos estes dados indicam que a modulação da VA está restrita ao seu efeito sinérgico sobre recombinação somática – promovendo especificamente a recombinação homóloga em células proliferativas de Drosophila. Co-tratamento Através deste procedimento ficou claro que a VA diminui significativamente a toxicidade genética total dos alquilantes MNU e ENU e do agente intercalante bleomicina. Os decréscimos observados tanto para o MNU quanto para o ENU são basicamente atribuídos ao seu papel promotor sobre o processo de detoxificação - que leva a diminuição no número de metilações e etilações induzidas respectivamente pelo MNU e pelo ENU. Adicionalmente, a caracterização da VA como um potente captador de radicais livres, especialmente em função do seu efeito sobre os danos oxidativos induzidos pela BLEO – explica a sua ação desmutagênica em relação a este agente intercalante. Todos estes dados referentes ao efeito modulador da VA não permitem a quantificação da relação risco-benefício do seu consumo, especialmente pela dificuldade prática de se medir o quanto a sua presença concomitante com as genotoxinas – representado por efeito benéfico, via interferência no potencial genotóxico – ou a sua ação após a indução dos danos genéticos, através da promoção de reparo recombinacional e conseqüente aumento em HR, contribuem para a expressão final do seu efeito modulador. Entretanto, o papel fundamental da recombinação homóloga na gênese de inúmeras doenças genéticas, incluindo o câncer, e a preponderante ação recombinogênica da VA são um sinal de alerta.

Desde o seu surgimento, as favelas no Rio de Janeiro foram, ao longo dos anos, foco de diferentes políticas públicas desenvolvidas pelos governantes. Primeiramente a ênfase dessas políticas foi pautada na questão da remoção das favelas, pois eram consideradas verdadeiras aberrações que necessitavam ser erradicadas. Com o passar dos anos nota-se uma grande mudança. O discurso de valorização da urbanização das favelas ganha força e o remocionismo parece ficar um pouco esquecido. Entretanto, a partir do ano de 2009, vemos que a agenda pública passa a dar grande ênfase ao fomento do empreendedorismo nas favelas, já que este é visto como um importante fator de geração de riquezas e combate da pobreza. Para isso, foram criados diferentes projetos por parte do poder público e da iniciativa privada que visam fomentar a iniciativa empreendedora nas favelas. Entretanto, como será exposto através de algumas entrevistas que foram realizadas com empreendedoras da favela do Cantagalo, estes projetos apresentam algumas barreiras e desafios que precisam ser vencidos para que possam servir à finalidade para a qual foram criados e para que sejam mais efetivos.

Human multipotent mesenchymal stromal cells (MSCs), also known as mesenchymal stem cells, have become an important and attractive therapeutic tool since they are easily isolated and cultured, have in vitro expansion potential, substantial plasticity and secrete bioactive molecules that exert trophic effects. The human umbilical cord as a cell source for cell therapy will help to avoid several ethical, political, religious and technical issues. One of the main issues with SC lines from different sources, mainly those of embryonic origin, is the possibility of chromosomal alterations and genomic instability during in vitro expansion. Cells isolated from one umbilical cord exhibited a rare balanced paracentric inversion, likely a cytogenetic constitutional alteration, karyotype: 46,XY,inv(3)(p13p25~26). Important genes related to cancer predisposition and others involved in DNA repair are located in 3p25~26. Titanium is an excellent biomaterial for bone-implant integration; however, the use can result in the generation of particulate debris that can accumulate in the tissues adjacent to the prosthesis, in the local bone marrow, in the lymph nodes, liver and spleen. Subsequently may elicit important biological responses that aren´t well studied. In this work, we have studied the genetic stability of MSC isolated from the umbilical cord vein during in vitro expansion, after the cryopreservation, and under different concentrations and time of exposition to titanium microparticles. Cells were isolated, in vitro expanded, demonstrated capacity for osteogenic, adipogenic and chondrogenic differentiation and were evaluated using flow cytometry, so they met the minimum requirements for characterization as MSCs. The cells were expanded under different concentrations and time of exposition to titanium microparticles. The genetic stability of MSCs was assessed by cytogenetic analysis, fluorescence in situ hybridization (FISH) and analysis of micronucleus and other nuclear alterations (CBMN). The cells were able to internalize the titanium microparticles, but MSCs preserve their morphology, differentiation capacity and surface marker expression profiles. Furthermore, there was an increase in the genomic instability after long time of in vitro expansion, and this instability was greater when cells were exposed to high doses of titanium microparticles that induced oxidative stress. It is necessary always assess the risks/ benefits of using titanium in tissue therapy involving MSCs, considering the biosafety of the use of bone regeneration using titanium and MSCs. Even without using titanium, it is important that the therapeutic use of such cells is based on analyzes that ensure quality, security and cellular stability, with the standardization of quality control programs appropriate. In conclusion, it is suggested that cytogenetic analysis, FISH analysis and the micronucleus and other nuclear alterations are carried out in CTMH before implanting in a patient

Worldwide, families Carangidae and Rachycentridae represent one of the groups most important commercial fish, used for food, and great potential for marine aquaculture. However, the genetic bases that can underpin the future cultivation of these species, cytogenetic between these aspects are very weak. The chromosomal patterns have provided basic data for the exploration of biotechnological processes aimed at handling chromosomal genetic improvement, such as induction of polyploidy, androgenesis and ginogenesis, as well as obtaining monosex stocks and interspecific hybridizations. This paper presents a comprehensive cytogenetic survey in 10 species, seven of the family Carangidae and the monotypic family Rachycentridae. Classical cytogenetic analysis and in situ mapping of multigene sequences were employed, and additionally for the genus Selene and morphotypes of Caranx lugubris, comparisons were made using geometric morphometrics. In general, conservative species exhibit a marked chromosome number (2n=48). Although present in large part, different karyotypic form, retain many characteristics typical of chromosomal Order Perciformes, the high number of elements monobrachyal, Ag-NORs/18S rDNA sites and heterochromatin simply reduced, preferably centromeric. The main mechanisms involved in karyotypic diversification are the pericentric inversions, with secondary action of centric fusions. In addition to physical mapping and chromosome detail for the species are presented and discussed patterns of intra-and interspecific diversity, cytotaxonomic markers. Human mesenchymal stem cells (MSC) are powerful sources for cell therapy in regenerative medicine. The long time cultivation can result in replicative senescence or can be related to the emergence of chromosomal alterations responsible for the acquisition of tumorigenesis features in vitro. In this study, for the first time, the expression profile of MSC with a paracentric chromosomal inversion (MSC/inv) was compared to normal karyotype (MSC/n) in early and late passages. Furthermore, we compared the transcriptome of each MSC in early passages with late passages. MSC used in this study were obtained from the umbilical vein of three donors, two MSC/n and one MSC/inv. After their cryopreservation, they have been expanded in vitro until reached senescence. Total RNA was extracted using the RNeasy mini kit (Qiagen) and marked with the GeneChip ® 3 IVT Express Kit (Affymetrix Inc.). Subsequently, the fragmented aRNA was hybridized on the microarranjo Affymetrix Human Genome U133 Plus 2.0 arrays (Affymetrix Inc.). The statistical analysis of differential gene expression was performed between groups MSC by the Partek Genomic Suite software, version 6.4 (Partek Inc.). Was considered statistically significant differences in expression to p-value Bonferroni correction ˂.01. Only signals with fold change ˃ 3.0 were included in the list of differentially expressed. Differences in gene expression data obtained from microarrays were confirmed by Real Time RT-PCR. For the interpretation of biological expression data were used: IPA (Ingenuity Systems) for analysis enrichment functions, the STRING 9.0 for construction of network interactions; Cytoscape 2.8 to the network visualization and analysis bottlenecks with the aid of the GraphPad Prism 5.0 software. BiNGO Cytoscape pluggin was used to access overrepresentation of Gene Ontology categories in Biological Networks. The comparison between senescent and young at each group of MSC has shown that there is a difference in the expression parttern, being higher in the senescent MSC/inv group. The results also showed difference in expression profiles between the MSC/inv versus MSC/n, being greater when they are senescent. New networks were identified for genes related to the response of two of MSC over cultivation time. Were also identified genes that can coordinate functional categories over represented at networks, such as CXCL12, SFRP1, xvi EGF, SPP1, MMP1 e THBS1. The biological interpretation of these data suggests that the population of MSC/inv has different constitutional characteristics, related to their potential for differentiation, proliferation and response to stimuli, responsible for a distinct process of replicative senescence in MSC/inv compared to MSC/n. The genes identified in this study are candidates for biomarkers of cellular senescence in MSC, but their functional relevance in this process should be evaluated in additional in vitro and/or in vivo assays

Perciformes are dominant in the marine environment, characterized as the largest and most diverse fish group. Some families, as Gerreidae, popularly known as silver jennies, carapebas, or mojarras have a high economic potential to marine fish farming, natural explotation and game fishing. Genetic information of these species are of fundamental importance for their management and production. Despite exist over 13,000 marine fish species described, only 2% were cytogenetically analyzed and less than 1% have some reproductive characteristics known. Induced breeding, cytogenetic characterization and cryopreservation of gametes, represent important areas in applied fish studies. In this project cytogenetic analyzes were performed to acess genetic aspects of Gerreidae species, distributed in coastal and estuarine regions of Northeast Brazil. Different methods for identifying chromosomal regions were employed using conventional techniques (Ag-NORs, C-banding), staining with base-specific fluorochromes (DAPI-CMA3), and physical mapping of ribosomal genes 18S and 5S rDNA, through hybridization in situ with fluorescent probes (FISH). The six species analyzed showed remarkable chromosome conservatism. The 18S and 5S ribosomal genes when analyzed in phylogenetic perspective demonstrate varied evolutionary dynamics, suggesting ocurrence of stasis process in some groups and greater dynamism in others. Double FISH with 18S and 5S probes showed both how efficient cytotaxonomic markers in the homogeneous karyotypes of this group of species. The karyotypic pattern identified in addition to the evolutionary aspects of karyotype, are suggestive of existence of low potential of post-zygotic barrier, prompting further research to prospect for artificial interspecific hybridization of these species of commercial importance

Leukemia is a heterogeneous group of hematologic malignancies that result from partial or total transformation of the blast cells. The Acute Lymphoblastic Leukemia (ALL) is the most common malignancy in childhood, especially in male, Caucasian children younger than 14 years. Several criteria are adopted to classify ALL, including the cell morphology, cytochemistry, immunophenotyping and cytogenetic analysis. Cytogenetic studies allow a more detailed analysis to detect chromosomal abnormalities of leukemic cells. These modifications will determine the diagnosis, classification, stage characterization, remission assessment and prognosis. In this study were evaluated 30 patients, aged from four months to seventeen years, of both sexes and various ethnicities. The age distribution showed that 67% of patients had between one and ten years (with mean age of XX years old), the most prevalent ethnic was Caucasian (50%) and 57% were males. According to immunophenotype, 93% of patients had B-cells progenitor ALL and 7% early lineage T. Considering the total studied population, the most frequent medical findings were lymphadenopathy (37%), hepatomegaly (77%) and splenomegaly (70%), where one patient could present more than one of these medical findings. Regarding the CBC, the majority of patients had hemoglobin below 10 g / dl (73%), leukocyte count less than 10.000/μL (60%) and platelet count below 150.000/μL (83%). Chromosomal abnormalities were observed in 64% of all patients, where hyperdiploidy was the most common numerical change (67%), followed by hypodiploid (33%). All these data are in agreement with the literature. Moreover, complexes structural and/or number changes not yet described in literature were observed, which indicated poor prognosis. Finally, we concluded that this study demonstrated the importance of cytogenetic study in the diagnosis and identification of prognostic factors in pediatric patients with ALL in Rio Grande do Norte. The results obtained in this study are extremely useful and emphasizes that surveys of this nature must be conducted more frequently in our state

A proposta deste trabalho é a construção artesanal de um telescópio refletor do tipo Newtoniano relacionando conteúdos abordados na disciplina de óptica, como a formação de imagens em lentes e espelhos, aberrações esféricas e cromáticas, interferência e difração, com cada fase do processo de construção. Com o aparelho construído é possível abordar a fotografia lunar e planetária com alta resolução e sua utilização por alunos do curso de Licenciatura em Física já demonstra ser um grande incentivo à contemplação do céu e à compreensão de muitos fenômenos físicos, com ocorrências de eclipses, formação das marés, estações do ano, etc, que geralmente são pouco abordados nas escolas de Ensino Fundamental e Médio. de acordo com diagnóstico realizado pelos alunos de graduação em algumas escolas do município de Bauru, verificou-se que a única abordagem de astronomia é apenas em relação ao sistema solar, especificamente órbitas planetárias, nas disciplinas de Física ou Geografia. Além do telescópio, a utilização de animações produzidas por alunos de graduação também pode ser considerada como uma ferramenta eficiente no ensino de astronomia, principalmente para alunos de Ensino Fundamental.

Mesenchymal stem cells (MSCs) are known as a population of multi-potential cells able to proliferate and differentiate into multiple mesodermal tissues including bone, cartilage, muscle, ligament, tendon, fat and stroma. Several applications of the study of EC can be emphasized the therapeutic techniques such as guided bone regeneration by implantation of EC in the affected site, without the need for bone grafts, using titanium as a vehicle. The process of cryopreservation is essential for the maintenance of cell cultures, since the cell line is frozen, it can be maintained in liquid nitrogen for an indefinite period and then thawed for therapeutic or experimental purposes. The aim of this study was to isolate a population of MSCs derived from the subendothelium of the umbilical vein human (MSCs-SUVH) to assess cytogenetic analysis by the possibility of appearance of chromosomal changes in two different situations: MSCs-SUVH regarding the process of cryopreservation and MSCs-SUVH grown on the surface of titanium. Flow cytometry analysis revealed that, this cell population was positive for the markers CD29, CD73 and CD90, but there was no expression of hematopoietic lineage markers, such as CD14, CD34 and CD45 and demonstrated capacity for osteogenic differentiation. The chromosomes obtained from the primary culture of MSCs-SUVH were analyzed by GTW banding technique, and results are described as guidelines to ISCN 2005. There was not the emergence of clonal chromosomal changes in the MSCs-SUVH in different situations analyzed. However one of the strings presented a balanced paracentric inversion, probably a cytogenetic constitutional alterations, which was present before and after the experimental situations that the MSCs-SUVH was submitted

The Anguiliformes is constituted by 15 families, 141 sorts and 737 species. In this group eight families possess at least one karyotyped species, where a prevalence of karyotypes with 2n=38 is evidenced chromosomes and high NF, apparently basal for the Anguiliformes. The only family who shows a different karyotypic pattern from the others is the Muraenidae family. In this, of the eight species already described, all of them present 2n=42 chromosomes. Despite the dimension of this Order, few species present karyotypics descriptions. In the present work, a species of Ophichthidae, Myrichthys ocellatus (2n=38, 8m+14sm+10st+6a, NF=70) and three species of Muraenidae, Enchelycore nigricans (2n=42, 6m+8sm+12st+16a, NF=68), Gymnothorax miliaris (2n=42, 14m+18sm+10st, NF=84), Gymnothorax vicinus (2n=42, 8m+6sm+28a, NF=56) and Muraena pavonina (2n=42, 6m+4sm+32a, NF=52), collected in the coast of the Rio Grande do Norte state, Saint Peter and Paul Rocks and in the coast of Bahia state were analyzed. Mitotics chromosomes had been gotten through mitotic stimulation with yeasts. Among the analyzed species, it is observed the presence of characteristic large metacentric chromosomic pairs (≅10µm). As for the structural standard, heterochromatics regions in these species in centromeric position of the majority of the chromosomic pairs and simple ribosomal sites had been evidenced. For the Ophichthidae family, the gotten data corroborate the hypothesis of karyotypic diversification mediated by the occurrence of pericentrics inversions and robertsonians rearrangements, while in the Muraenidae, the identification of larger chromosomic values (2n=42), suggests derived karyotypes, possibly caused by possible chromosomic fissions

Foram realizados estudos citogenéticos e morfológicos em nove populações locais de Astyanax scabripinnis paranae. Todas as populações apresentaram 2n = 50 cromossomos e conspícuas diferenças envolvendo a morfologia dos cromossomos, número e posição das NORs e quantidade e localização dos blocos de heterocromatina constitutiva. O estudo quantitativo dos dados citogenéticos, usando a análise das variáveis canônicas (CVA), mostraram que oito populações apresentaram diferentes constituições cariotípicas. Análises morfológicas baseadas em nove caracteres morfométricos e dois merísticos foram eficazes no estabelecimento de uma clara identificação de cinco populações. Análises comparativas dos dados citogenéticos e morfológicos sugerem que as modificações cromossômicas e morfológicas ocorreram em diferentes taxas, sendo as cromossômicas mais rápidas do que as morfológicas. A despeito da similaridade morfológica encontrada entre algumas populações, diferenças cromossômicas foram detectadas entre todas as populações estudadas.
