The world energy consumption is expected to increase strongly in coming years, because of the emerging economies. Biomass is the only renewable carbon resource that is abundant enough to be used as a source of energy Grape pomace is one of the most abundant agro-industrial residues in the world, being a good biomass resource. The aim of this work is the valorization of grape pomace from white grapes (WWGP) and from red grapes (RWGP), through the extraction of phenolic compounds with antioxidant activity, as well as through the extraction/hydrolysis of carbohydrates, using subcritical water, or hot compressed water (HCW). The main focus of this work is the optimization of the process for WWGP, while for RWGP only one set of parameters were tested. The temperatures used were 170, 190 and 210 °C for WWGP, and 180 °C for RWGP. The water flow rates were 5 and 10 mL/min, and the pressure was always kept at 100 bar. Before performing HCW assays, both residues were characterized, revealing that WWGP is very rich in free sugars (around 40%) essentially glucose and fructose, while RWGP has higher contents of structural sugars, lignin, lipids and protein. For WWGP the best results were achieved at 210 °C and 10 mL/min: higher yield in water soluble compounds (69 wt.%), phenolics extraction (26.2 mg/g) and carbohydrates recovery (49.3 wt.% relative to the existing 57.8%). For RWGP the conditions were not optimized (180 °C and 5 mL/min), and the values of the yield in water soluble compounds (25 wt.%), phenolics extraction (19.5 mg/g) and carbohydrates recovery (11.4 wt.% relative to the existing 33.5%) were much lower. The antioxidant activity of the HCW extracts from each assay was determined, the best result being obtained for WWGP, namely for extracts obtained at 210 °C (EC50=20.8 μg/mL; EC50 = half maximum effective concentration; EC50 = 22.1 μg/mL for RWGP, at 180 ºC).

The development of modern analytical tools plays an important role in quality control. The main purpose of this study was to explore the use of subcritical water as a versatile analytical tool, employed simultaneously as a reagent and solvent, as well as the application of high temperature-high resolution gas chromatography (HT-HRGC) to develop a procedure for the analysis of triacylglycerides and fatty acids in Azadirachta indica A. Juss. (Neem) oil without the need for solvents, chemical reagents, or catalytic agents. The developed method presented satisfactory results and is in agreement with the concepts of Green Analytical Chemistry (GAC).

Alikriittisellä vedellä tarkoitetaan paineistettua vettä, joka on kriittisen lämpötilansa (374 °C) alapuolella nestemäisessä tilassa. Veden tiheys pienenee lämpötilan kasvaessa Veden liuotinominaisuuksia voidaan säädellä lämpötilan avulla. Veden pintajännitys, viskositeetti, tiheys ja polaarisuus pienenevät lämpötilan kasvaessa, ja alikriittisen veden aineominaisuudet muuttuvat lähemmäksi orgaanista liuotinta. Alikriittisen veden dielektrisyysvakion aleneminen johtuu pääasiassa lämpötilan vaikutuksesta ja vain vähän paineen vaikutuksesta. Alikriittistä vettä on käytetty liuottimena uutossa, mutta nyt myös alikriittinen kromatografia on kehittymässä oleva erotusmenetelmä. Työn kokeellisessa osassa kehitettiin kromatografinen laitteisto alikriittiselle vedelle, jolla tutkittiin sokerialkoholien ja sokerien kromatografista erotusta alikriittisen veden avulla. Lisäksi tutkittiin sokerialkoholien, sokereiden ja stationäärifaasien termistä kestävyyttä. Tutkittavina komponentteina olivat sorbitoli, mannitoli, ksylitoli, arabinoosi, mannoosi, ksyloosi, maltoosi ja ramnoosi. Stationäärifaaseina käytettiin makrohuokoista funktionalisoimatonta polystyreenidivinyylibentseenikopolymeeriä, sekä vahvoja ja heikkoja divinyylibentseenillä ristisilloitettuja kationinvaihtohartseja, jotka olivat joko Na+- tai Ca2+-ionimuodoissa. Veden lämpötilan nostaminen vaikuttaa sekä kromatografisen stationäärifaasin tilavuusmuutoksiin että näytekomponenttien ominaisuuksiin. Vahvoilla kationinvaihtimilla havaittiin termisten tilavuusmuutosten riippuvan ionimuodosta: Na+-muotoiset hartsit turpoavat ja Ca2+-muotoiset kutistuvat lämpötilan noustessa. Heikot kationinvaihtimet kutistuvat molemmissa ionimuodoissa, mutta Ca2+-muoto kutistuu Na+-muotoa voimakkaammin. Näytekomponenteista sokerialkoholien havaittiin kestävän paremmin korkeita lämpötiloja kuin sokerien. Sokerialkoholeista kestävimmäksi havaittiin ksylitoli ja sokereista ramnoosi. Tutkittavien komponenttien piikkien havaittiin kapenevan, häntimisen vähenevän, ja piikkien eluoituvan aikaisemmin riippuen käytettävästä stationäärifaasista. Ca2+-muotoisen vahvan kationinvaihtimen kompleksinmuodostuskyky heikkeni lämpötilan kasvaessa. Näytekomponenttien erotus ei kuitenkaan parantunut lämpötilan noustessa tutkituilla stationäärifaaseilla.

Teollisessa kromatografiassa kolonnia pyritään kuormittamaan mahdollisimman paljon, jotta saataisiin maksimoitua erotetun komponentin määrä aikayksikköä kohden. Tässä työssä kuormitusta tutkittiin nostamalla syöttöliuoksen, synteettisen melassin, näyteväkevyyttä 80-125 ºC:ssa. Eluenttina oli paineistettu kuumaa vesi ja hartsina vahva Na-muotoinen PS-DVB pohjainen vahva kationinvaihtohartsi. Lämpötilaa nostamalla piikit kapenivat ja tulivat symmetrisemmiksi, erotus nopeutui sekä suola erottui usein paremmin sokereista. Syöttöliuoksen kuiva-ainetta lisättiin asteittain 55 p-% saakka, jolloin ei vielä havaittu ongelmia erotuksessa. Lämpötilassa 125 ºC havaittiin erotuksen aikana kuormituksesta riippumatonta sakkaroosin invertoitumista. Vertailtaessa eri stationäärifaaseja havaittiin Na-muotoisen PS-DVB pohjaisen kationinvaihtohartsin erottavan yleensä sokereita, sokerialkoholeja, oligosakkarideja ja betaiinia lähes poikkeuksetta paremmin alhaisilla pitoisuuksilla kuin neutraalihartsi ja Na-muotoinen zeoliitti. Erottuminen ei yleensä parantunut lämpötilaa nostamalla, mutta piikit kapenivat ja erotus nopeutui. Monosakkaridien erotus huononi 125 ºC:ssa kationinvaihtohartsilla. Tutkittaessa terveysvaikutteisten ksylo-oligosakkaridien soveltuvuutta alikriittiseen erotukseen, niiden havaittiin huomattavasti hydrolysoituvan happamissa olosuhteissa koeputkessa 100 ºC:ssa kahdessa tunnissa. Näytteessä olevien epäpuhtauksien havaittiin katalysoineen hydrolyysiä. Hydrolysoituminen oli hitaampaa neutraaleissa olosuhteissa korotetussa lämpötilassa. Tästä voitiin tehdä johtopäätös, että alikriittiset olosuhteet eivät sovi ksylo-oligosakkaridien erotukseen.

Polymeeriadsorbentteja valmistetaan silloittamalla styreeniä, akrylaattia tai fenoliformaldehydiä. Useimmiten ristisilloittajana toimii divinyylibentseeni. Polymeeriadsorbenteissa ei itsessään ole ioninvaihtoryhmiä, joten ne sopivat ionittomien ja heikosti ionisoitujen aineiden adsorptioon. Usein polymeeriadsorbentteja käytetään vaihtoehtona aktiivihiilelle eri sovelluksissa. Työn kirjallisuusosassa on katsaus polymeeriadsorbenttien sovelluksiin lähinnä elintarviketeollisuudessa. Lisäksi siinä selvitetään polymeeriadsorbenttien rakennetta ja synteesimenetelmiä. Kokeellisessa osassa tutkittiin valittujen styreeni- ja akrylaattipohjaisten polymeeriadsorbenttien soveltuvuutta kromatografisen erotuksen stationaarifaasiksi. Kromatografia-ajoissa käytettiin eluenttina vettä, jonka lämpötila oli pääasiassa joko 75 tai 125 °C. Jälkimmäisessä lämpötilassa vesi on paineistettua neste, jota kutsutaan myös alikriittiseksi vedeksi. Malliaineina oli eri sokereita, aminohappoja sekä bentsoehappoa ja bentsyylialkoholia. Kromatografisen soveltuvuuden lisäksi selvitettiin adsorbenttien termistä kestävyyttä ja rakennetta. Termisesti polymeeriadsorbentit kestivät hyvin lämpötiloja 125 °C:eseen saakka. Polymeeriadsorbenteilla, joilla on suuri ominaispinta-ala, on myös suuri adsorptiokapasiteetti. Styreenipohjaiset adsorbentit erottivat kaikkia tutkittuja malliaineita akrylaattipohjaisia paremmin. Jotkut adsorbentit eivät erottaneet mitään tutkituista yhdisteistä. Lämpötilan nostaminen kavensi piikkejä ja nopeutti malliaineiden retentoitumista, mutta ei parantanut erottumista.

Objectives. Evaluate the flexural strength (sigma) and subcritical crack growth (SCG) under cyclic loading of glass-infiltrated alumina-based (IA, In-Ceram Alumina) and zirconia-reinforced (IZ, In-Ceram Zirconia) ceramics, testing the hypothesis that wet environment influences the SCG of both ceramics when submitted to cyclic loading.Methods. Bar-shaped specimens of IA (n = 45) and IZ ( n = 45) were fabricated and loaded in three-point bending (3P) in 37 degrees C artificial saliva (IA(3P) and IZ(3P)) and cyclic fatigued (F) in dry (D) and wet (W) conditions (IA(FD), IA(FW), IZ(FD), IZ(FW)). The initial sigma and the number of cycles to fracture were obtained from 3P and F tests, respectively. Data was examined using Weibull statistics. The SCG behavior was described in terms of crack velocity as a function of maximum stress intensity factor (K(Imax)).Results. The Weibull moduli (m = 8) were similar for both ceramics. The characteristic strength (sigma(0)) of IA and IZ was and 466 MPa 550 MPa, respectively. The wet environment significantly increased the SCG of IZ, whereas a less evident effect was observed for IA. In general, both ceramics were prone to SCG, with crack propagation occurring at K(I) as low as 43-48% of their critical K(I). The highest sigma of IZ should lead to longer lifetimes for similar loading conditions.Significance. Water combined with cyclic loading causes pronounced SCG in IZ and IA materials. The lifetime of dental restorations based on these ceramics is expected to increase by reducing their direct exposure to wet conditions and/or by using high content zirconia ceramics with higher strength. Adsorption and regeneration of ion exchange resins were studied using a subcritical solution of a CO2-H2O mixture and a fixed bed column. The commercial Amberlite IRC-50/IRC-86 cation exchange resins and Amberlite IRA-67 anion exchange resin were tested for heavy metals (Pb, Cu, Cd) adsorption from a solution with different initial metal concentrations at different temperatures. After adsorption, the loaded resins were regenerated with water and carbon dioxide at different temperatures and a pressure of 25 MPa. The efficiency of the IRC-50 resin was lower than that of the IRC-86 resin for the adsorption of metals like Cd, Cu and Pb. Results obtained for desorption of these metals indicated that the process could be used for Cd and in principle for Cu. Sorption of metal ions depended strongly on feed concentration. Mathematical modeling of the metal desorption process was carried out successfully as an extraction process. For this purpose, the VTII Model, which is applied to extraction from solids using supercritical solvents, was used in this work.

Objectives. Verify the influence of different filler distributions on the subcritical crack growth (SCG) susceptibility, Weibull parameters (m and sigma(0)) and longevity estimated by the strength-probability-time (SPT) diagram of experimental resin composites. Methods. Four composites were prepared, each one containing 59 vol% of glass powder with different filler sizes (d(50) = 0.5; 0.9; 1.2 and 1.9 mu m) and distributions. Granulometric analyses of glass powders were done by a laser diffraction particle size analyzer (Sald-7001, Shimadzu, USA). SCG parameters (n and sigma(f0)) were determined by dynamic fatigue (10(-2) to 10(2) MPa/s) using a biaxial flexural device (12 x 1.2 mm; n = 10). Twenty extra specimens of each composite were tested at 10(0) MPa/s to determine m and sigma(0). Specimens were stored in water at 37 degrees C for 24 h. Fracture surfaces were analyzed under SEM. Results. In general, the composites with broader filler distribution (C0.5 and C1.9) presented better results in terms of SCG susceptibility and longevity. C0.5 and C1.9 presented higher n values (respectively, 31.2 +/- 6.2(a) and 34.7 +/- 7.4(a)). C1.2 (166.42 +/- 0.01(a)) showed the highest and C0.5 (158.40 +/- 0.02(d)) the lowest sigma(f0) value (in MPa). Weibull parameters did not vary significantly (m: 6.6 to 10.6 and sigma(0): 170.6 to 176.4 MPa). Predicted reductions in failure stress (P-f = 5%) for a lifetime of 10 years were approximately 45% for C0.5 and C1.9 and 65% for C0.9 and C1.2. Crack propagation occurred through the polymeric matrix around the fillers and all the fracture surfaces showed brittle fracture features. Significance. Composites with broader granulometric distribution showed higher resistance to SCG and, consequently, higher longevity in vitro. (C) 2012 Academy of Dental Materials. Published by Elsevier Ltd. The micellization of a homologous series of zwitterionic surfactants, a group of sulfobetaines, was studied using isothermal titration calorimetry (ITC) in the temperature range from 15 to 65 °C. The increase in both temperature and the alkyl chain length leads to more negative values of ΔGmic(0) , favoring the micellization. The entropic term (ΔSmic(0)) is predominant at lower temperatures, and above ca. 55-65 °C, the enthalpic term (ΔHmic(0)) becomes prevalent, figuring a jointly driven process as the temperature increases. The interaction of these sulfobetaines with different polymers was also studied by ITC. Among the polymers studied, only two induced the formation of micellar aggregates at lower surfactant concentration: poly(acrylic acid), PAA, probably due to the formation of hydrogen bonds between the carboxylic group of the polymer and the sulfonate group of the surfactant, and poly(sodium 4-styrenesulfonate), PSS, probably due to the incorporation of the hydrophobic styrene group into the micelles. The prevalence of the hydrophobic and not the electrostatic contributions to the interaction between sulfobetaine and PSS was confirmed by an increased interaction enthalpy in the presence of electrolytes (NaCl) and by the observation of a significant temperature dependence, the latter consistent with the proposed removal of hydrophobic groups from water.

In this work the archaea and eubacteria community of a hypersaline produced water from the Campos Basin that had been transported and discharged to an onshore storage facility was evaluated by 16S recombinant RNA (rRNA) gene sequence analysis. The produced water had a hypersaline salt content of 10 (w/v), had a carbon oxygen demand (COD) of 4,300 mg/l and contains phenol and other aromatic compounds. The high salt and COD content and the presence of toxic phenolic compounds present a problem for conventional discharge to open seawater. In previous studies, we demonstrated that the COD and phenolic content could be largely removed under aerobic conditions, without dilution, by either addition of phenol degrading Haloarchaea or the addition of nutrients alone. In this study our goal was to characterize the microbial community to gain further insight into the persistence of reservoir community members in the produced water and the potential for bioremediation of COD and toxic contaminants. Members of the archaea community were consistent with previously identified communities from mesothermic reservoirs. All identified archaea were located within the phylum Euryarchaeota, with 98 % being identified as methanogens while 2 % could not be affiliated with any known genus. Of the identified archaea, 37 % were identified as members of the strictly carbon-dioxide-reducing genus Methanoplanus and 59 % as members of the acetoclastic genus Methanosaeta. No Haloarchaea were detected, consistent with the need to add these organisms for COD and aromatic removal. Marinobacter and Halomonas dominated the eubacterial community. The presence of these genera is consistent with the ability to stimulate COD and aromatic removal with nutrient addition. In addition, anaerobic members of the phyla Thermotogae, Firmicutes, and unclassified eubacteria were identified and may represent reservoir organisms associated with the conversion hydrocarbons to methane.

Caffeine has already been used as an indicator of anthropogenic impacts, especially the ones related to the disposal of sewage in water bodies. In this work, the presence of caffeine has been correlated with the estrogenic activity of water samples measured using the BLYES assay. After testing 96 surface water samples, it was concluded that caffeine can be used to prioritize samples to be tested for estrogenic activity in water quality programs evaluating emerging contaminants with endocrine disruptor activity.

Biofilm formation on reverse osmosis (RO) systems represents a drawback in the application of this technology by different industries, including oil refineries. In RO systems the feed water maybe a source of microbial contamination and thus contributes for the formation of biofilm and consequent biofouling. In this study the planktonic culturable bacterial community was characterized from a feed water of a RO system and their capacities were evaluated to form biofilm in vitro. Bacterial motility and biofilm control were also analysed using phages. As results, diverse Protobacteria, Actinobacteria and Bacteroidetes were identified. Alphaproteobacteria was the predominant group and Brevundimonas, Pseudomonas and Mycobacterium the most abundant genera. Among the 30 isolates, 11 showed at least one type of motility and 11 were classified as good biofilm formers. Additionally, the influence of non-specific bacteriophage in the bacterial biofilms formed in vitro was investigated by action of phages enzymes or phage infection. The vB_AspP-UFV1 (Podoviridae) interfered in biofilm formation of most tested bacteria and may represent a good alternative in biofilm control. These findings provide important information about the bacterial community from the feed water of a RO system that may be used for the development of strategies for biofilm prevention and control in such systems.

The biochemical responses of the enzymatic antioxidant system of a drought-tolerant cultivar (IACSP 94-2094) and a commercial cultivar in Brazil (IACSP 95-5000) grown under two levels of soil water restriction (70% and 30% Soil Available Water Content) were investigated. IACSP 94-2094 exhibited one additional active superoxide dismutase (Cu/Zn-SOD VI) isoenzyme in comparison to IACSP 95-5000, possibly contributing to the heightened response of IACSP 94-2094 to the induced stress. The total glutathione reductase (GR) activity increased substantially in IACSP 94-2094 under conditions of severe water stress; however, the appearance of a new GR isoenzyme and the disappearance of another isoenzyme were found not to be related to the stress response because the cultivars from both treatment groups (control and water restrictions) exhibited identical changes. Catalase (CAT) activity seems to have a more direct role in H2O2 detoxification under water stress condition and the shift in isoenzymes in the tolerant cultivar might have contributed to this response, which may be dependent upon the location where the excessive H2O2 is being produced under stress. The improved performance of IACSP 94-2094 under drought stress was associated with a more efficient antioxidant system response, particularly under conditions of mild stress.

Different types of water bodies, including lakes, streams, and coastal marine waters, are often susceptible to fecal contamination from a range of point and nonpoint sources, and have been evaluated using fecal indicator microorganisms. The most commonly used fecal indicator is Escherichia coli, but traditional cultivation methods do not allow discrimination of the source of pollution. The use of triplex PCR offers an approach that is fast and inexpensive, and here enabled the identification of phylogroups. The phylogenetic distribution of E. coli subgroups isolated from water samples revealed higher frequencies of subgroups A1 and B23 in rivers impacted by human pollution sources, while subgroups D1 and D2 were associated with pristine sites, and subgroup B1 with domesticated animal sources, suggesting their use as a first screening for pollution source identification. A simple classification is also proposed based on phylogenetic subgroup distribution using the w-clique metric, enabling differentiation of polluted and unpolluted sites.
