The studies provided data on the spoilage pattern of Otolithus argenteus during low temperature preservation. Changes in the total volatile bases, hypoxanthine, tyrosine, salt soluble nitrogen, non-protein nitrogen, pH, peroxide value, free fatty acids and thiobarbituric acid number along with organoleptic score have been reported. Organoleptically, fish stored at +20 degree C remained in acceptable condition upto 12 days while for those stored at 0 degree C in ice upto 19 days. Of the various indices tested Hypoxanthine, salt soluble nitrogen and total volatile bases nitrogen, in the order of merit can be used as freshness tests for refrigerated fish.

Fresh sole fish (Cynoglossus macrolepidotus) was quick frozen at -40°C and stored at -18°C. Shelf life was evaluated by following biochemical, bacteriological and organoleptic changes occurring during storage. Rapid decrease was noted in the water extractable nitrogen and salt soluble nitrogen fractions. Samples of frozen sole fish remained in acceptable condition for 20 weeks.

Fish minces from dhoma and lactarius were mixed in the ratio of 40:60 respectively, and quick frozen along with individual minces at -40°C and stored at -20°C. Shelf life was evaluated by following biochemical, physical and organoleptic changes occurred during storage up to 44 weeks. Rapid decreases were noted in water soluble nitrogen and salt soluble nitrogen fractions during the initial period of 16 weeks. A good correlation was observed between changes in salt-soluble nitrogen and organoleptic evaluation. The minces were in good and acceptable condition up to 32 weeks of storage.

The freezing and cold storage characteristics of cuttle fish fillets have been studied. The yield of fillets from cuttle fish was about 35% and the fillet had an average moisture content of 76.85% and fat 0.82% During storage at -20 ± 1°C for 16 months the salt soluble nitrogen of the fillets decreased from 85.1to35.36%, the non-protein nitrogen from 24.61 to 20.84% and alpha amino nitrogen from 252 to 140mg/100g. Initially the fillets were white in colour, showed signs of desiccation by 4 months storage which increased on further storage and the fillets finally became dull white with yellow discolouration inside. The firm and chewy texture of the cooked fillets changed to rubbery even though the product was slightly sweet at the end of that storage period of 16 months.

The biochemical, bacteriological and organoleptic changes in cultured rohu (Labeo rohita) during iced storage have been studied. Non-protein nitrogen decreased and water soluble nitrogen remained almost same during storage in ice. Initially, when the fish was in pre-rigor and rigor conditions, the extractability of protein was low (45 to 50%) which increased after the resolution of the rigor and the decrease in extractability towards the end of storage was insignificant. The total volatile base nitrogen remained steady up to 7 days in ice and showed slight decrease on further storage. During iced storage the bacterial count increased from 10^3/g to 10^5/g by the 11th day of storage. Nearly 80-90% of the total bacterial population in fresh fish was constituted by mesophiles which decreased gradually (decreased to 1% by 13th day of iced storage). Organoleptically the fish was acceptable up to 15 days in ice.

Surimi was prepared from silver carp with an aim to put this underutilized fish for profitable use. The mince prepared was washed twice with chilled water (5°C) using mince to water ratio (w/v) of 1:2 for 5-6 minutes each. After final dewatering to moisture content to about 80%; half the quantity of washed minced meat was mixed with cryoprotectants (4% sorbitol, 4% sucrose and 0.3% sodium tripolyphosphate) to produce surimi. The prepared surimi and the dewatered minced meat were packed in LDPE bags, frozen using a plate freezer and stored at -20°C. Surimi and dewatered minced meat from frozen storage were used as base material for production of fish cakes. These were fried at 160°C for 3 to 4 minutes before serving for organoleptic test. Changes in salt soluble nitrogen, total volatile base nitrogen, non-protein nitrogen, peroxide value and free fatty acid of surimi and dewatered mince were estimated at every ten days interval during the storage period of 3 months. The study has indicated that frozen storage of surimi could be a potential method for effective utilization of silver carp. This surimi when incorporated in fish cakes yielded products which retained the shelf life even up to 90 days of storage.

A bacteriocin-producing strain of Lactobacillus paracasei DPC 4715 was used as an adjunct culture in Cheddar cheese in order to control the growth of “wild” nonstarter lactic acid bacteria. No suppression of growth of the indicator strain was observed in the experimental cheese. The bacteriocin produced by Lactobacillus paracasei DPC 4715 was sensitive to chymosin and cathepsin D and it may have been cleaved by the rennet used for the cheese manufactured or by indigenous milk proteases. A series of studies were performed using various microbial adjuncts to influence cheese ripening. Microbacterium casei DPC 5281, Corynebacterium casei DPC 5293 and Corynebacterium variabile DPC 5305 were added to the cheesemilk at level of 109 cfu/ml resulting in a final concentration of 108 cfu/g in Cheddar cheese. The strains significantly increased the level of pH 4.6-soluble nitrogen, total free amino acids after 60 and 180 d of ripening and some individual free amino acids after 180 d. Yarrowia lipolytica DPC 6266, Yarrowia lipolytica DPC 6268 and Candida intermedia DPC 6271 were used to accelerate the ripening of Cheddar cheese. Strains were grown in YG broth to a final concentration of 107 cfu/ml, microfluidized, freeze-dried and added to the curd during salting at level of 2% w/w. The yeasts positively affected the primary, secondary proteolysis and lipolysis of cheeses and had aminopeptidase, dipeptidase, esterase and 5’ phosphodiestere activities that contributed to accelerate the ripening and improve the flavor of cheese. Hafia alvei was added to Cheddar cheesemilk at levels of 107 cfu/ml and 108 cfu/ml and its contribution during ripening was evaluated. The strain significantly increased the level of pH 4.6-soluble nitrogen, total free amino-acids, and some individual free amino-acids of Cheddar cheese, whereas no differences in the urea-polyacrylamide gel electrophoresis (urea-PAGE) electrophoretograms of the cheeses were detected. Hafia alvei also significantly increased the level of some biogenic amines. A low-fat Cheddar cheese was made with Bifidobacterium animalis subsp. lactis, strain BB-12® at level of 108 cfu/ml, as a probiotic adjunct culture and Hi-Maize® 260 (resistant high amylose maize starch) at level of 2% and 4% w/v, as a prebiotic fiber which also played the role of fat replacer. Bifidobacterium BB-12 decreased by 1 log cycle after 60 d of ripening and remained steady at level of ~107 cfu/g during ripening. The Young’s modulus also increased proportionally with increasing levels of Hi-maize. Hencky strain at fracture decreased over ripening and increased with increasing in fat replacer. A cheese based medium (CBM) was developed with the purpose of mimicking the cheese environment at an early ripening stage. The strains grown in CBM showed aminopeptidase activity against Gly-, Arg-, Pro- and Phe-p-nitroanalide, whereas, when grown in MRS they were active against all the substrates tested. Both Lb. danicus strains grown in MRS and in CBM had aminotransferase activity towards aromatic amino acids (Phe and Trp) and also branched-chain amino acids (Leu and Val). Esterase activity was expressed against p-nitrophenyl-acetate (C2), pnitrophenyl- butyrate (C4) and p-nitrophenyl-palmitate (C16) and was significantly higher in CBM than in MRS.

Antifungal compounds produced by Lactic acid bacteria (LAB) metabolites can be natural and reliable alternative for reducing fungal infections pre- and post-harvest with a multitude of additional advantages for cereal-base products. Toxigenic and spoilage fungi are responsible for numerous diseases and economic losses. This thesis includes an overview of the impact fungi have on aspects of the cereal food chain. The applicability of LAB in plant protection and cereal industry is discussed in detail. Specific case studies include Fusarium head blight, and the impact of fungi in the malting and baking industry. The impact of Fusarium culmorum infected raw barley on the final malt quality was part of the investigation. In vitro infected barley grains were fully characterized. The study showed that the germinative energy of infected barley grains decreased by 45% and grains accumulated 199 μg.kg-1 of deoxynivalenol (DON). Barley grains were subsequently malted and fully characterized. Fungal biomass increased during all stages of malting. Infected malt accumulated 8-times its DON concentration during malting. Infected malt grains revealed extreme structural changes due to proteolytic, (hemi)-cellulolytic and starch degrading activity of the fungi, this led to increased friability and fragmentation. Infected grains also had higher protease and β-glucanase activities, lower amylase activity, a greater proportion of free amino and soluble nitrogen, and a lower β-glucan content. Malt loss was over 27% higher in infected malt when compared to the control. The protein compositional changes and respective enzymatic activity of infected barley and respective malt were characterized using a wide range of methods. F. culmorum infected barley grains showed an increase in proteolytic activity and protein extractability. Several metabolic proteins decreased and increased at different rates during infection and malting, showing a complex F. culmorum infection interdependence. In vitro F. culmorum infected malt was used to produce lager beer to investigate changes caused by the fungi during the brewing processes and their effect on beer quality attributes. It was found, that the wort containing infected malt had a lower pH, a higher FAN, higher β-glucan and a 45% increase in the purging rate, and led to premature yeast flocculation. The beer produced with infected malt (IB) had also a significantly different amino acid profile. IB flavour characterization revealed a higher concentration of esters, fusel alcohols, fatty acids, ketones, and dimethylsulfide, and in particular, acetaldehyde, when compared to the control. IB had a greater proportion of Strecker aldehydes and Maillard products contributing to an increased beer staling character. IB resulted in a 67% darker colour with a trend to better foam stability. It was also found that 78% of the accumulated mycotoxin deoxynivalenol in the malt was transferred into beer. A LAB cell-freesupernatant (cfs), produced in wort-base substrate, was investigated for its ability to inhibit Fusarium growth during malting. Wort was a suitable substrate for LAB exhibiting antifungal activity. Lactobacillus amylovorus DSM19280 inhibited 104 spores.mL-1 for 7 days, after 120 h of fermentation, while Lactobacillus reuteri R29 inhibited 105 spores.mL-1 for 7 days, after 48 h of fermentation. Both LAB cfs had significant different organic acid profiles. Acid-base antifungal compounds were identified and, phenyllactic, hydroxy-phenyllactic, and benzoic acids were present in higher concentrations when compared to the control. A 3 °P wort substrate inoculated With L. reuteri R29 (cfs) was applied in malting and successfully inhibited Fusarium growth by 23%, and mycotoxin DON by 80%. Malt attributes resulted in highly modified grains, lower pH, higher colouration, and higher extract yield. The implementation of selected LAB producing antifungal compounds can be used successfully in the malting process to reduce mould growth and mycotoxin production.

Total biochemical composition of freshwater major carps, Labeo rohita -(r9hu); Catla catla (catla); Cirrltinus tnrigala (mrigal), in relation to size was evaluated . The water soluble nitrogen fraction constituted about 21% of the tota l nitro gen . Salt so luble fraction constituted 55 - 60% of the total nitrogen. Non-protein nitrogen represented 12% of the total nit rogen in all the cases except mri gal of small size (9.36%). The insoluble connec tive tissu e contributed to 2-3 per cent of tota l nitrogen . The monounsaturated fatty acid s (MUFA) formed 31-39% of the total fatty acids and 60-68% of the MUFA in freshwat er carps was CI 8:1. The esse ntial amino acids contributed to 41 to 51% of the total amino acid s in freshwater major carps. Arom atic amino acid content was sligh tly higher compared to marine fishe s while the proportion of proline was less. Varia tions in compos ition in rela tion to size are discu ssed . Agricultural systems with conventional tillage and intensive use of agrochemicals, especially those on high slopes and with shallow soils, have the potential to release pollutants. This study aimed at evaluating the soil, water and nutrient lost via agricultural runoff in large plots (small catchments) under conventional and organic farming of vegetables as well as under forest (control) system in a Cambisol in the Campestre catchment. Samples of runoff were collected biweekly for one year through a Coshocton wheel. The soil and water losses from the conventional farming were 218 and 6 times higher, respectively, than forest. Under organic farming the soil and water losses were 12 and 4 times higher, respectively, than forest. However the soil losses (0.5 to 114 kg ha^(−1) year^(−1)) are considered low in agronomy but environmentally represent a potential source of surface water contamination by runoff associated pollutants. The concentrations and losses of all forms of phosphorus (P) were higher in the conventional system (9.5, 0.9 and 0.3 mg L^(−1) of total P for conventional, organic and forest systems, respectively), while the organic system had the highest concentrations and losses of soluble nitrogen (4.7, 38.6 and 0.4 mg L^(−1) of NO_3-N, respectively). The percentage of bioavailable P was proportionally higher in the organic system (91% of total P lost was as bioavailable P), indicating greater potential for pollution in the short term.

La sang de porc és un subproducte comestible que es genera als escorxadors industrials durant el procés d'obtenció de la canal. Aquest subproducte es caracteritza per presentar una elevada càrrega contaminant i, degut a l'elevat volum que es genera, és necessari trobar estratègies que permetin la seva revaloració i aprofitament, a la vegada que disminuïm la contaminació ambiental i les despeses que es deriven del seu processament abans de l'abocament. La fracció cel·lular (FC) constitueix el 40 % de la sang de porc i conté principalment l'hemoglobina (Hb), que representa al voltant del 90 % del contingut en proteïna d'aquesta fracció (un 35 % aproximadament). L'elevat percentatge en proteïna i en ferro, i les seves bones propietats funcionals fan que l'aprofitament d'aquest subproducte com a primera matèria o ingredient de la indústria alimentària sigui una alternativa molt útil a l'hora de reduir les despeses de la indústria càrnia, sempre que es resolguin els problemes de l'enfosquiment i dels sabors estranys que pot conferir la FC quan s'addiciona a productes alimentaris. Una altra possible utilització de la FC és aprofitar les propietats colorants de l'Hb o del grup hemo, com a colorant d'origen natural en diversos productes alimentaris. Els objectius del present treball eren, en primer lloc, determinar les millors condicions d'aplicació del procés de conservació de la FC mitjançant la deshidratació per atomització i caracteritzar físico-químicament i microbiològica el concentrat d'Hb en pols. En segon lloc, avaluar l'eficàcia de diferents additius antioxidants i/o segrestants del ferro per prevenir l'enfosquiment que pateix la FC durant la deshidratació. En tercer lloc, aplicar tractaments d'altes pressions hidrostàtiques com a procés d'higienització i avaluar els efectes d'aquest tractament sobre la microbiota contaminant, el color i les propietats funcionals de la FC. Finalment, desenvolupar un procés d'obtenció d'hidrolitzats proteics descolorats a partir de l'Hb amb la finalitat d'utilitzar-los com a ingredients nutricionals i/o funcionals. La millor temperatura de deshidratació per atomització de la FC hemolitzada era 140ºC. La FC en pols presentava un contingut en humitat del 5,3 % i un percentatge de solubilitat proteica del 96 %. La deshidratació per atomització induïa canvis en l'estructura nativa de l'Hb i, per tant, un cert grau de desnaturalització que pot conduir a una disminució de les seves propietats funcionals. L'extracte sec de la FC en pols estava composat per un 94,6 % de proteïna, un 3 % de sals minerals i un 0,7 % de greix. Els valors CIE L*a*b* del color de la FC en pols eren força constants i reflectien el color vermell marró fosc d'aquesta, a causa de l'oxidació del ferro hèmic que es produeix durant la deshidratació. La càrrega contaminant de la FC fresca de la sang de porc era força elevada i el tractament d'hemòlisi amb ultrasons i la centrifugació posterior no produïen una reducció significativa de la microbiota contaminant, obtenint un producte amb uns recomptes microbiològics de l'ordre de 106 ufc·mL-1. La deshidratació per atomització produïa una disminució d'una unitat logarítmica dels recomptes totals de la FC hemolitzada. Tanmateix, el producte en pols encara reflectia l'elevada contaminació de la primera matèria, fet que condiciona negativament la seva utilització com a ingredient alimentari, a no ser que es millorin les condicions de recollida de la sang a l'escorxador o que aquesta o la FC es sotmeti a algun tractament d'higienització prèviament a la deshidratació. Les isotermes de sorció a 20ºC de la FC en pols tenien forma sigmoïdal i una histèresi estreta i llarga. L'equació GAB és un bon model matemàtic per ajustar les dades de sorció obtingudes experimentalment i determinar la isoterma d'adsorció de la FC deshidratada per atomització. El percentatge d'humitat de la FC deshidratada a 140ºC es corresponia a un valor d'aw a 20 ºC d'aproximadament el 0,16. Tenint en compte que estava per sota dels valors d'aw corresponents a la capa monomolecular, es pot garantir la conservació a temperatura ambient del producte, sempre que s'envasi en recipients tancats que no permetin l'entrada d'humitat de l'exterior. De l'estudi de la possible estabilització del color de la FC deshidratada per atomització mitjançant l'addició d'antioxidants i/o segrestants de ferro, es va observar que només l'àcid ascòrbic, la glucosa, l'àcid nicotínic i la nicotinamida, tenien efectes positius sobre el color del producte en pols. L'ascòrbic i la glucosa no milloraven la conservació del color de l'Hb però disminuïen l'enfosquiment que es produeix durant la deshidratació, amb la qual cosa es pot obtenir un producte en pols de color marró més clar. L'addició de dextrina o L-cisteïna no disminuïa l'enfosquiment ni evitava el canvi de color de l'Hb. L'àcid nicotínic i la nicotinamida protegien el color de l'Hb durant el procés de deshidratació i l'emmagatzematge de la FC en pols. Les millors condicions d'aplicació del tractament amb altes pressions hidrostàtiques (HHP) sobre la FC eren 400 MPa, a 20ºC, durant 15 minuts, perquè produïen una millora significativa de la qualitat microbiològica, no afectaven negativament al color, no comprometien gaire la solubilitat proteica l'Hb i, malgrat que produïen un augment de la viscositat, la FC romania fluida després del tractament. Aquest tractament permetia una reducció de la microbiota contaminant de la FC d'entre 2 i 3 unitats logarítmiques. L'aplicació de l'alta pressió i la posterior deshidratació per atomització permetien obtenir un producte en pols amb recomptes totals de l'ordre de 2,8 unitats logarítmiques. El color de la FC pressuritzada en pols era igual que el de la FC control deshidratada, perquè ambdues mostres presentaven la mateixa susceptibilitat a l'oxidació del grup hemo produïda per la deshidratació. L'alta pressió incrementava la susceptibilitat de l'Hb als efectes desnaturalitzants de la deshidratació, fonamentalment a pH 7 (PIE), ja que es va observar una disminució de la solubilitat proteica a pH neutre després dels 2 processos tecnològics. La FC en pols presentava una màxima capacitat escumant al PIE de l'Hb. L'aplicació del tractament HHP produïa una disminució de la capacitat escumant de la FC en pols, però no tenia efectes negatius sobre l'estabilitat de l'escuma formada. Tampoc es van observar efectes negatius del tractament HHP sobre l'activitat emulsionant de l'Hb. La màxima activitat emulsionant de l'Hb s'aconseguia amb una concentració de FC en pols de l'1,5 % a pH 7 i de l'1 % a pH 4,5. Les pastes obtingudes per escalfament de la FC presentaven característiques molt diferenciades depenent del pH. A pH neutre es formaven unes pastes dures i consistents, mentre que a pH àcid les pastes eren poc consistents, molt adhesives i més elàstiques que les anteriors. Aquestes tenien una capacitat de retenció d'aigua molt superior que les de pH 7, en les quals l'aigua quedava retinguda per capil·laritat. La textura i capacitat de retenció d'aigua de les pastes tampoc eren afectades pel tractament HHP. El tractament HHP incrementava l'activitat de la Tripsina sobre l'Hb quan el substrat i l'enzim es tractaven conjuntament i afavoria el procés d'obtenció d'hidrolitzats descolorats a partir de la FC, la qual cosa permetia assolir el mateix grau de descoloració amb una dosi d'enzim inferior. El tractament d'hidròlisi de la FC amb la utilització combinada de Tripsina seguida d'un tractament amb Pepsina permetia l'obtenció d'un hidrolitzat proteic d'Hb descolorat i hidrolitzava completament la globina, donant lloc a 2 pèptids de 10,8 i 7,4 KDa. Val a dir que també produïa un 60 80 % de nitrogen soluble en TCA, constituït fonamentalment per pèptids petits i aminoàcids lliures. Els hidrolitzats trípsics i pèpsics d'Hb, obtinguts a partir de FC no pressuritzada i deshidratats per atomització a 180ºC, eren de color blanc i tenien un contingut en humitat del 4,7 %, un 84,2 % de proteïna i 9,7 % de sals minerals. El procés d'hidròlisi permetia una reducció considerable de la contaminació de la FC, obtenint un producte en pols amb uns recomptes totals de l'ordre de 102-103 ufc·g-1. Pel que fa a la funcionalitat dels hidrolitzats d'Hb deshidratats per atomització, aquests presentaven una elevada solubilitat proteica a pH 5 i 7 i romanien solubles després d'un escalfament a 80ºC durant 30 min. Tanmateix, aquesta hidròlisi afectava molt negativament la capacitat de mantenir escumes estables i l'activitat emulsionant.

A whey salts mixture was used as a partial substitute for sodium chloride to provide a modified Na:K ratio (1:3.4) in the manufacture of white salted cheese using ultrafiltration. Reduction of chymosin addition from 20 to 8 mu L kg(-1) of cheese was also investigated. Variation of salt and chymosin levels did not result in any significant differences in composition and physicochemical properties. The rates of proteolysis in terms of water-soluble nitrogen (WSN) and nitrogen soluble in 12% trichloroacetic acid (TCA-SN) were affected by chymosin levels but not by salt treatment. Urea-PAGE electrophoretic analysis of caseins from the cheeses manufactured using three levels of chymosin and two salt types showed that the hydrolysis of alpha(s1)-casein was higher than for beta-caseins but the differences between the cheeses were not significant (P > 0.05). The chymosin level did not have a significant effect (P > 0.05) on hardness and fracturability, suggesting that any variation in hardness due to the initial hydrolysis was being confounded by other variables. Cheeses including the whey salts product were harder and more fracturable (P < 0.01) than the cheese treated with NaCl only. Both hardness and fracturability values decreased (P < 0.05) over the maturation period. This study demonstrated that both chymosin and salt-in-moisture (SM) were important factors for proteolysis in the manufacture of ultrafiltrated white-salted cheese, with significant effects on water-soluble nitrogen and nitrogen soluble in trichloroacetic acid. In contrast, the levels of free amino acids were not significantly affected by chymosin and salt treatments. The cheeses made using high levels of chymosin with low SM had lower levels of residual α(s1)- and β-casein at the end of ripening. On texture profile analysis, the hardness and fracturability of the cheeses significantly increased with SM and decreased during ripening. Increases in chymosin significantly contributed to the overall weakening of the structure throughout ripening. Bitter flavour was detected after 12 weeks in the cheese made with the higher chymosin level and lower SM, which could be the result of accumulation of γ-casein fractions. The sensory data indicated that the hedonic responses for low chymosin with low SM cheeses were good and acceptable in flavour, which may be due to the moderate levels of proteolysis products.
