The impacts of germanic acid Ge(OH)(4) on the growth of Psuedonitzschia pungens f. multiseries and on the production of the algal toxin, domoic acid, were studied. The results showed that germanic acid in the range of concentrations could inhibit the growth of the algal cells and the inhibition was enhancing with the concentrations of germanic acid increasing. Germanic acid also could inhibit the production of the algal toxin, domoic acid in cells and the inhibition reached up to 100% at Ge/Si = 35. Based on the results, the mechanism was discussed.

研究胶州湾浮游植物的物种组成与时空分布的特点,对于了解该湾生态系统的现状与历史变化趋势以及生态系统对自然条件变化和人类干扰的响应具有重要意义。 本文根据2004年每月一次采集的浮游植物样品,分析了胶州湾浮游植物的物种组成和优势种的时空分布情况。调查发现浮游植物共142种,分属于6门53属,其中硅藻门40属113种,占总物种数的79.6%;甲藻门10属24种,占总物种数的16.9%;其它为金藻门1属2种,裸藻1属1种,绿藻1属2种。浮游植物丰度周年波动范围为11.12-14602.39×104cells/m3,全年平均为1857.55×104cells/m3。全年丰度最高的藻为环纹劳德藻(Lauderia annulata),出现在2月份,而全年的丰度最高值也出现在2月份。在胶州湾中,硅藻所占比例最大,平均为97.44%,最高为99.95%,最低为82.55%。浮游植物的丰度的周年变化:有两个高峰,分别出现在2月份和10月份,是典型的温带海域双高峰的分布形式。 胶州湾浮游植物的优势种分析,采用Kikvidze等(2002)提出的确定优势种数量的计算方法,再依据各物种的优势度排序最终确定优势种。其中在多样性最低的6月和10月,优势种数目均为1种,分别为丹麦细柱藻(Leptocylindrus danicus)和中肋骨条藻(Skeletonema costatum)。而在多样性指数较低的1月、2月,优势种数目分别为2和3;而在物种多样性比较高的5月、7月、11月,由于分布相对比较分散,所以优势种数量较多。这说明这个方法可以较好地完成对胶州湾浮游植物群落分析时确定优势种的目标。优势种出现频率较多的种类为中肋骨条藻(1、3、4、5、8、10、11、12月)、洛氏角毛藻(Chaetoceros lorenzianus)(7、8、9、11、12月)、尖刺拟菱形藻(Pseudonitzschia pungens)(2、3、5、7月)、加拉星杆藻(Asteronella kariana)(1、2、3、5月)、密联角毛藻(Chaetoceros densus)(5、11、12月)、扭鞘藻(Streptothece thamesis)(7、8、9月)、夜光藻(Noctiluca scintillancs)(4、5、7月)、奇异菱形藻(Nitzschia paradoxa)(5、11、12月),对这些种的时空分布进行了分析。 对2004年胶州湾的浮游植物数据进行分层聚类分析后,发现可以将胶州湾划分为三个海区:湾南与湾外海区、湾中西部海区、湾东海区。

The ability to detect harmful algal bloom (HAB) species and their toxins in real- or near real-time is a critical need for researchers studying HAB/toxin dynamics, as well as for coastal resource managers charged with monitoring bloom populations in order to mitigate their wide ranging impacts. The Environmental Sample Processor (ESP), a robotic electromechanical/fluidic system, was developed for the autonomous, subsurface application of molecular diagnostic tests and has successfully detected several HAB species using DNA probe arrays during field deployments. Since toxin production and thus the potential for public health and ecosystem effects varies considerably in natural phytoplankton populations, the concurrent detection of HAB species and their toxins onboard the ESP is essential. We describe herein the development of methods for extracting the algal toxin domoic acid (DA) from Pseudonitzschia cells (extraction efficiency >90%) and testing of samples using a competitive ELISA onboard the ESP. The assay detection limit is in the low ng/mL range (in extract), which corresponds to low ng/L levels of DA in seawater for a 0.5 L sample volume acquired by the ESP. We also report the first in situ detection of both a HAB organism (i.e., Pseudo-nitzschia) and its toxin, domoic acid, via the sequential (within 2-3 h) conduct of species- and toxin-specific assays during ESP deployments in Monterey Bay, CA, USA. Efforts are now underway to further refine the assay and conduct additional calibration exercises with the aim of obtaining more reliable, accurate estimates of bloom toxicity and thus their potential impacts. El Crucero de Evaluación de Recursos Pelágicos 0209-11 de los BIC’s Humboldt, Olaya y SNP-2 abarcó de Paita a IIo; se colectó 158 muestras de fitoplancton, 126 para análisis semi cuantitativo y 32 para análisis cuantitativos. El promedio de biomasa planctónica fue 1,01 mL.m-3, con núcleos de alta concentración al norte (5–6 y 7-8°S) dentro de las 30 mn. El fitoplancton fue predominante en el 37% del área evaluada con dominancia de Coscinodiscus perforatus, C. excentricus, Actinocyclus sp., Thalassiosira rotula, Pseudonitzschia pungens, Detonula confervacea, Pleurosigma sp. y Ditylum brightwellii, asociadas a especies de fase temprana en sucesión (Chaetoceros spp.). Las mayores concentraciones celulares (500x103 cel.L-1) se localizaron en los perfiles de Atico con abundancia de fitoflagelados y en Paita de diatomeas. Ceratium breve, indicador de AES se localizó al norte de los 8°S y por fuera de las 100 mn; y un pequeño núcleo frente a Huacho y Chancay por el desplazamiento de esa masa de agua. Ceratium praelongum, indicador de ASS presentó un ligero acercamiento en Callao a 30 mn y en San Juan a 60 mn. Protoperidinium obtusum, indicador de ACF amplió su distribución debido a la intensificación de los afloramientos.

Domoinsäure ist ein von mehreren Arten mariner Kieselalgen der Gattung Pseudonitzschia produziertes Toxin, welches während einer Algenblüte in Molluscen wie z.B. der Miesmuschel Mytilus sp. akkumuliert werden kann. Beim Verzehr solch kontaminierter Muscheln können sowohl beim Menschen als auch bei Tieren erhebliche Vergiftungserscheinungen auftreten, die von Übelkeit, Kopfschmerzen und Orientierungsstörungen bis hin zum Verlust des Kurzzeitgedächtnisses (daher auch als amnesic shellfish poisoning bekannt) reichen und in einigen Fällen tödlich enden. rnDie heute gängigen Methoden zur Detektion von Domoinsäure in Muschelgewebe wie Flüssigkeitschromatographie und Maus-Bioassay sind zeit- und kostenintensiv bzw. in Anbetracht einer Verbesserung des Tierschutzes aus ethischer Sicht nicht zu vertreten. Immunologische Testsysteme stellen eine erstrebenswerte Alternative dar, da sie sich durch eine vergleichsweise einfache Handhabung, hohe Selektivität und Reproduzierbarkeit auszeichnen.rnDas Ziel der vorliegenden Arbeit war es, ein solches immunologisches Testsystem zur Detektion von Domoinsäure zu entwickeln. Hierfür wurden zunächst Antikörper gegen Domoinsäure gewonnen, wofür das Toxin wiederum als erstes über die Carbodiimid-Methode an das Trägerprotein keyhole limpet hemocyanin (KLH) gekoppelt wurde, um eine Immunantwort auslösen zu können. Kaninchen und Mäuse wurden mit KLH-DO-Konjugaten nach vorgegebenen Immunisierungsschemata immunisiert. Nach vier Blutabnahmen zeigte das polyklonale Kaninchenantiserum eine ausreichend hohe Sensitivität zum Antigen; das nachfolgende Detektionssystem wurde mit Hilfe dieses polyklonalen Antikörpers aufgebaut. Zwar ist es gegen Ende der Arbeit auch gelungen, einen spezifischen monoklonalen Antikörper aus der Maus zu gewinnen, jedoch konnte dieser aus zeitlichen Gründen nicht mehr im Detektionssystem etabliert werden, was durchaus wünschenswert gewesen wäre. rnWeiterhin wurde Domoinsäure im Zuge der Entwicklung eines neuartigen Testsystems an die Trägerproteine Ovalbumin, Trypsininhibitor und Casein sowie an Biotin konjugiert. Die Kopplungserfolge wurden im ELISA, Western Blot bzw. Dot Blot nachgewiesen. Die Ovalbumin-gekoppelte sowie die biotinylierte Domoinsäure dienten im Folgenden als die zu messenden Größen in den Detektionsassays- die in einer zu untersuchenden Probe vorhandende, kompetitierende Domoinsäure wurde somit indirekt nachgewiesen. rnDer zulässige Höchstwert für Domoinsäure liegt bei 20 µg/g Muschelgewebe. Sowohl mit Biotin-DO als auch mit OVA-DO als den zu messenden Größen waren Domoinsäurekonzentrationen unterhalb dieses Grenzwertes nachweisbar; allerdings erwies sich der Aufbau mit Biotin-DO um das ca. 20-fache empfindlicher als jener mit OVA-DO. rnDie in dieser Arbeit präsentierten Ergebnisse könnten als Grundlage zur Etablierung eines kommerzialisierbaren immunologischen Testsystems zur Detektion von Domoinsäure und anderen Biotoxinen dienen. Nach erfolgreicher Validierung wäre ein solches Testsystem in seiner Handhabung einfacher als die gängige Flüssigkeitschromatographie und besser reproduzierbar als der Maus-Bioassay.rn

Pack ice around Svalbard was sampled during the expedition ARK XIX/1 of RV "Polarstern" (March-April 2003) in order to determine environmental conditions, species composition and abundances of sea-ice algae and heterotrophic protists during late winter. As compared to other seasons, species diversity of algae (total 40 taxa) was not low, but abundances (5,000-448,000 cells/l) were lower by one to two orders of magnitude. Layers of high algal abundances were observed both at the bottom and in the ice interior. Inorganic nutrient concentrations (NO2, NO3, PO4, Si(OH)4) within the ice were mostly higher than during other seasons, and enriched compared to seawater by enrichment indices of 1.6-24.6 (corrected for losses through the desalination process). Thus, the survival of algae in Arctic pack ice was not limited by nutrients at the beginning of the productive season. Based on less-detailed physical data, light was considered as the most probable factor controlling the onset of the spring ice-algal bloom in the lower part of the ice, while low temperatures and salinities inhibit algal growth in the upper part of the ice at the end of the winter. Incorporation of ice algae probably took place during the entire freezing period. Possible overwintering strategies during the dark period, such as facultative heterotrophy, energy reserves, and resting spores are discussed.

Feeding activity, selective grazing and the potential grazing impact of two dominant grazers of the Polar Frontal Zone, Calanus simillimus and Rhincalanus gigas, and of copepods < 2 mm were investigated with incubation experiments in the course of an iron fertilized diatom bloom in November 2000. All grazers were already actively feeding in the low chlorophyll waters prior to the onset of the bloom. C. simillimus maintained constant clearance rates and fed predominantly on diatoms. R. gigas and the small copepods strongly increased clearance and ingestion of diatoms in response to their enhanced availability. All grazers preyed on microzooplankton, most steadily on ciliates, confirming the view that pure herbivory appears to be the exception rather than the rule in copepod feeding. The grazers exhibited differences in feeding behavior based on selectivity indices. C. simillimus and R. gigas showed prey switching from dinoflagellates to diatoms in response to the phytoplankton bloom. All grazers most efficiently grazed on large diatoms leading to differences in daily losses for large and small species, e.g. Corethron sp. or Thalassionema nitzschioides. Species-specific diatom mortality rates due to grazing suggest that the high feeding activity of C. simillimus prior to and during the bloom played a role in shaping diatom population dynamics

While the isolated responses of marine phytoplankton to climate warming and to ocean acidification have been studies intensively, studies on the combined effect of both aspects of Global Change are still scarce. Therefore, we performed a mesocosm experiment with a factorial combination of temperature (9 and 15°C) and pCO2 (560 ppm and 1400 ppm) with a natural autumn plankton community from the western Baltic Sea. Temporal trajectories of total biomass and of the biomass of the most important higher taxa followed similar patterns in all treatments. When averaging over the entire time course, phytoplankton biomass decreased with warming and increased with CO2 under warm conditions. The contribution of the two dominant higher phytoplankton taxa (diatoms and cryptophytes) and of the 4 most important species (3 diatoms, 1 cryptophyte) did not respond to the experimental treatments. Taxonomic composition of phytoplankton showed only responses at the level of subdominant and rare species. Phytoplankton cell sizes increased with CO2 addition and decreased with warming. Both effects were stronger for larger species. Warming effects were stronger than CO2 effects and tended to counteract each other. Phytoplankton communities without calcifying species and exposed to short-term variation of COO2 seem to be rather resistant to ocean acidification.

Domoic acid (DA) is a naturally occurring cyanotoxin, which upon ingestion, is responsible for amnesic shellfish poisoning (ASP) in both humans and animals. Produced by the marine diatom, Pseudonitzschia, DA is accumulated by a number of marine organisms including shellfish, clams and mussels which upon consumption can lead to headaches, nausea and seizures. Possessing a variety of functional groups the structure of DA contains three carboxyl groups, a pyrrole ring and a potent conjugated diene region allowing for binding to glutamate receptors in the dorsal hippocampus of the brain causing the described detrimental effects. Although limitations have been placed regarding the amount of DA that may be contained in seafood no limitations have been placed on the amount present in drinking water. Natural degradation of the toxin may occur through reactive oxygen species such as the hydroxyl radical and singlet oxygen at the conjugated diene region. In this work the photooxidation of DA via singlet oxygen has been studied using sorbic acid as a model compound. The three major reaction pathways observed during the photooxdiation process for both acids include 2 + 4 cycloaddition to produce endoperoxides , 2 + 2 reaction to afford aldehydes and ketones or an ene reaction to generate hydroperoxides. Under similar reaction conditions for SA and DA, the endoperoxide has been seen to be the major product for photoxidation of SA while the hydroperoxide has been seen to be the dominant product during photooxidation of DA.
