The effects of the inhalation of Cymbopogon martinii essential oil (EO) and geraniol on Wistar rats were evaluated for biochemical parameters and hepatic oxidative stress. Wistar rats were divided into three groups (n = 8): G1 was control group, treated with saline solution; G2 received geraniol; and G3 received C. martinii EO by inhalation during 30 days. No significant differences were observed in glycemia and triacylglycerol levels; G2 and G3 decreased (P < 0.05) total cholesterol level. There were no differences in serum protein, urea, aspartate aminotransferase activity, and total hepatic protein. Creatinine levels increased in G2 but decreased in G3. Alanine aminotransferase activity and lipid hydroperoxide were higher in G2 than in G3. Catalase and superoxide dismutase activities were higher in G3. C. martinii EO and geraniol increased glutathione peroxidase. Oxidative stress caused by geraniol may have triggered some degree of hepatic toxicity, as verified by the increase in serum creatinine and alanine aminotransferase. Therefore, the beneficial effects of EO on oxidative stress can prevent the toxicity in the liver. This proves possible interactions between geraniol and numerous chemical compounds present in C. martinii EO.

A kind of optical pH sensor was demonstrated that is based on a pH-sensitive fluorescence dye-doped (eosin) cellulose acetate (CA) thin-film modified microstructured polymer optical fiber (MPOF). It was obtained by directly inhaling an eosin-CA-acetic acid mixed solution into array holes in a MPOF and then removing the solvent (acetic acid). The sensing film showed different fluorescence intensities to different pH solutions in a pH range of 2.5-4.5. Furthermore, the pH response range could be tailored through doping a surfactant, hexadecyl trimethyl ammonium bromide (CTAB), in the sensing film. (c) 2007 Optical Society of America.

Cryptococcus neoformans var. grubii (Cng) is the most common cause of fungal meningitis, and its prevalence is highest in sub-Saharan Africa. Patients become infected by inhaling airborne spores or desiccated yeast cells from the environment, where the fungus thrives in avian droppings, trees and soil. To investigate the prevalence and population structure of Cng in southern Africa, we analysed isolates from 77 environmental samples and 64 patients. We detected significant genetic diversity among isolates and strong evidence of geographic structure at the local level. High proportions of isolates with the rare MATa allele were observed in both clinical and environmental isolates; however, the mating-type alleles were unevenly distributed among different subpopulations. Nearly equal proportions of the MATa and MATα mating types were observed among all clinical isolates and in one environmental subpopulation from the eastern part of Botswana. As previously reported, there was evidence of both clonality and recombination in different geographic areas. These results provide a foundation for subsequent genomewide association studies to identify genes and genotypes linked to pathogenicity in humans.

Rationale: Increasing epithelial repair and regeneration may hasten resolution of lung injury in patients with the Acute Respiratory Distress Syndrome (ARDS). In animal models of ARDS, Keratinocyte Growth Factor (KGF) reduces injury and increases epithelial proliferation and repair. The effect of KGF in the human alveolus is unknown.
Objectives: To test whether KGF can attenuate alveolar injury in a human model of ARDS.
Methods: Volunteers were randomized to intravenous KGF (60 μg/kg) or placebo for 3 days, before inhaling 50μg lipopolysaccharide. Six hours later, subjects underwent bronchoalveolar lavage (BAL) to quantify markers of alveolar inflammation and cell-specific injury.
Measurements and Main Results: KGF did not alter leukocyte infiltration or markers of permeability in response to LPS. KGF increased BAL concentrations of Surfactant Protein D (SP-D), MMP-9, IL-1Ra, GM-CSF and CRP. In vitro, BAL fluid from KGF-treated subjects (KGF BAL) inhibited pulmonary fibroblast proliferation, but increased alveolar epithelial proliferation. Active MMP-9 increased alveolar epithelial wound repair. Finally, BAL from the KGF pre-treated group enhanced macrophage phagocytic uptake of apoptotic epithelial cells and bacteria compared with BAL from the placebo-treated group. This effect was blocked by inhibiting activation of the GM-CSF receptor.
Conclusions: KGF treatment increases BAL SP-D, a marker of type II alveolar epithelial cell proliferation in a human model of ALI. Additionally KGF increases alveolar concentrations of the anti-inflammatory cytokine IL-1Ra, and mediators that drive epithelial repair (MMP-9) and enhance macrophage clearance of dead cells and bacteria (GM-CSF).

Metalworking fluid-associated hypersensitivity pneumonitis (MWF-HP) is a pulmonary disease caused by inhaling microorganisms present in the metalworking fluids used in the industrial sector. Mycobacterium immunogenum is the main etiological agent. Among the clinical, radiological and biological tools used for diagnosis, serological tests are important. The aim of this study was to identify immunogenic proteins in M. immunogenum and to use recombinant antigens for serological diagnosis of MWF-HP. Immunogenic proteins were detected by two-dimensional Western blot and candidate proteins were identified by mass spectrometry. Recombinant antigens were expressed in Escherichia coli and tested by enzyme-linked immunosorbent assay (ELISA) with the sera of 14 subjects with MWF-HP and 12 asymptomatic controls exposed to M. immunogenum. From the 350 spots visualized by two-dimensional gel electrophoresis with M. immunogenum extract, 6 immunogenic proteins were selected to be expressed as recombinant antigens. Acyl-CoA dehydrogenase antigen allowed for the best discrimination of MWF-HP cases against controls with an area under the receiver operating characteristics (ROC) curve of 0.930 (95% CI=0.820-1), a sensitivity of 100% and a specificity of 83% for the optimum threshold. Other recombinant antigens correspond to acyl-CoA dehydrogenase FadE, cytosol aminopeptidase, dihydrolipoyl dehydrogenase, serine hydroxymethyltransferase and superoxide dismutase. This is the first time that recombinant antigens have been used for the serodiagnosis of hypersensitivity pneumonitis. The availability of recombinant antigens makes it possible to develop standardized serological tests which in turn could simplify diagnosis, thus making it less invasive.

The emergence of electronic cigarettes (e-cigs) has given cannabis smokers a new method of inhaling cannabinoids. E-cigs differ from traditional marijuana cigarettes in several respects. First, it is assumed that vaporizing cannabinoids at lower temperatures is safer because it produces smaller amounts of toxic substances than the hot combustion of a marijuana cigarette. Recreational cannabis users can discretely "vape" deodorized cannabis extracts with minimal annoyance to the people around them and less chance of detection. There are nevertheless several drawbacks worth mentioning: although manufacturing commercial (or homemade) cannabinoid-enriched electronic liquids (e-liquids) requires lengthy, complex processing, some are readily on the Internet despite their lack of quality control, expiry date, and conditions of preservation and, above all, any toxicological and clinical assessment. Besides these safety problems, the regulatory situation surrounding e-liquids is often unclear. More simply ground cannabis flowering heads or concentrated, oily THC extracts (such as butane honey oil or BHO) can be vaped in specially designed, pen-sized marijuana vaporizers. Analysis of a commercial e-liquid rich in cannabidiol showed that it contained a smaller dose of active ingredient than advertised; testing our laboratory-made, purified BHO, however, confirmed that it could be vaped in an e-cig to deliver a psychoactive dose of THC. The health consequences specific to vaping these cannabis preparations remain largely unknown and speculative due to the absence of comprehensive, robust scientific studies. The most significant health concerns involve the vaping of cannabinoids by children and teenagers. E-cigs could provide an alternative gateway to cannabis use for young people. Furthermore, vaping cannabinoids could lead to environmental and passive contamination.

Resumen: En la población docente, el uso de la voz como principal herramienta del trabajo por su esfuerzo constante y prolongado constituye el principal factor de riesgo para padecer trastornos de la voz, concomitante con otros factores que inciden en la presentación de estas patologías. Objetivo: Determinar la prevalencia de los trastornos en la voz y sus factores asociados de la población docente de dos instituciones de educación superior (IES) en Colombia y los factores de riesgo asociados. Materiales y métodos: Se realizó un estudio descriptivo de tipo transversal, en el que participaron 149 docentes de dos IES (instituciones de educación superior) de dos ciudades de Colombia. Se aplicó un cuestionario sustentado en el Modelo de Técnicas Foniátricas para Prevenir la Disfonía en Profesores de Secundaria en Portugal y validado por la escuela de Medicina y Ciencias de la Salud, programa de fonoaudiología de la Universidad del Rosario con el trabajo denominado “La Voz como herramienta de trabajo” para Colombia; el cual indagaba los factores riesgo asociados al uso de la voz en el quehacer docente. Se realizó un análisis descriptivo de las variables cualitativas con frecuencias absolutas y porcentajes, medidas de tendencia central, promedio y mediana, de dispersión y desviación estándar. La información recolectada fue analizada con el software SPSS.PASW 18. La correlación entre las variables dependientes y las independientes, se realizaron con el coeficiente no-paramétrico de Spearman y la asociación con la prueba Ji-cuadrado de Pearson o el test exacto de Fisher (valores esperados < 5). Se evaluaron las distribuciones de las variables ordinales de las variables de problemas de la voz con el género, con la prueba no-paramétrica exacta de Mann Whitney. Las pruebas estadísticas se evaluaron a un nivel de significancia del 5% (p<0.05). Resultados: El promedio de edad fue de 36±11.7 años, con un rango que varió entre 25 y 68 años. El género más frecuente fue el masculino con 57,0% de participación. Los datos presentados se dan como resultado de las respuestas a tres preguntas indicativas de los trastornos de voz, las cuales no fueron respondidas por la totalidad de los docentes, por esta razón los datos se presentan así t= número total de personas que respondieron la pregunta y n= número de personas que respondieron afirmativamente. La prevalencia de alteración de voz fue del 63,4% (t=82; n=52), de disminución en el volumen de su voz de 57,3% (t=122; n=70) y cansancio laríngeo en el 76,8% (t=121; n=93). El cansancio laríngeo fue significativamente mayor en los hombres que en las mujeres (p=0.026, Test Mann Whitney exacto a una cola), con algunos hábitos de voz como el realizar esfuerzos en el cuello al hablar (rs = 0,461; p< 0,001) y con la dificultad para inhalar o exhalar aire al hablar (rs = 0,368; p<0.001), con el ruido de fondo (rs =0,361, p<0,001), al permanecer en ambientes contaminados con polvo y humedad (rs=0,311; p=0.001), con sufrir de estrés representado por dolor cervical en espalda y hombros (rs=0,349, p<0.001), y con los problemas familiares (rs= 0,397; p<0.001). Al analizar con la disminución del volumen de voz durante la práctica docente, las correlaciones más significativas mostradas fue estadísticamente mayor entre los hombres que en las mujeres (p=0.006, Test Mann Whitney exacto a una cola), con el esfuerzo cervical para hablar (rs =:0,596, p=<,001), y con el presentar dificultad para inhalar/exhalar aire al usar la voz (rs =0,508, p<,001), con la exposición a ruido de fondo (rs=0,204, p=0,030), ambientes contaminados con polvo y humedad (rs:=305, p<,001), con el estrés (rs =0,316, p=0,001), con los problemas familiares (rs =0,560, p<,001) y en las mujeres con la alteración de la voz en presencia de la menstruación (rs =0,751, p=0,000). Por último, se correlacionó con las alteraciones de voz durante la exposición laboral en la que se mostró mayor correlación en las mujeres que en los hombres (p=0.001, Test Mann Whitney exacto a una cola). Se mostró correlación directa con el no realizar pausas al hablar (rs=0,337, p<0,001), con toser para aclarar la voz (rs =0,349, p=0,001), y con el uso de la voz grave (rs =0,975; p=,004) ,con la presencia de ruido de fondo mientras se dicta clase (rs =0,370, p=0,001) con el permanecer en ambientes contaminados con polvo y humedad (rs =0,253,p=,023), con los síntomas cervicales asociados al estrés (rs=0,323, p=0,003), e inversa con los cambios en tono y volumen de la voz en presencia de menstruación en las mujeres (rs=-0,414,p=,017). Conclusiones: Con los hallazgos encontrados en este estudio es importante reconocer que en las características demográficas, se encontró que el género masculino refirió mayor cantidad de problemas de voz. Para las características laborales la antigüedad ocupacional es factor principal que incide en los problemas de voz; en el análisis de las características demográfico– laborales en las variables analizadas, el grupo etario que reporto cansancio laríngeo se encontraba entre los 36 a 45 años de edad, y los que referían un tiempo de exposición ocupacional mayor a 6 y menor a 15 años. En cuanto a los factores ambientales se identificó que la asociación más significativa está relacionada con estar sometido a cambios bruscos de temperatura, estar expuesto a ruido de fondo durante su labor y finalmente a permanecer en ambientes contaminados. Por ultimo al analizar las alteraciones de la voz con las características personales de los docentes, se observó que el estrés y los problemas familiares inciden de forma significativa en estas patologías. Con estos resultados se recomienda establecer un sistema de vigilancia epidemiológica en las IES, donde se capacite integralmente a los docentes en cuanto a los factores de riesgo personales y de hábitos de voz, además de supervisar y adecuar los factores ambientales que inciden en la presentación de trastornos de voz para evitar la presentación de estas patologías.

Introducción: La exposición en minas subterráneas a altos niveles de polvo de carbón está relacionada con patologías pulmonares. Objetivo: Determinar la prevalencia de neumoconiosis, medidas de higiene y seguridad industrial y su relación con niveles ambientales de carbón en trabajadores de minas de socavón en Cundinamarca. Materiales y Métodos: Estudio de corte transversal, en 215 trabajadores seleccionados mediante muestreo probabilístico estratificado con asignación proporcional. Se realizaron monitoreos ambientales, radiografías de tórax y encuestas con variables sociodemográficas y laborales. Se emplearon medidas de tendencia central y dispersión y la prueba de independencia ji-cuadrado de Pearson o pruebas exactas, con el fin de establecer las asociaciones. Resultados: El 99,5% de la población perteneció al género masculino, el 36,7% tenía entre 41-50 años, con un promedio de años de trabajo de 21,70 ± 9,99. La prevalencia de neumoconiosis fue de 42,3% y la mediana de la concentración de polvo de carbón bituminoso fue de 2,329670 mg/m3. El índice de riesgo de polvo de carbón presentó diferencias significativas en las categorías de bajo (p=0,0001) y medio (p=0,0186) con la prevalencia de neumoconiosis. El 84,2% reporto no usar mascarilla. No se presentan diferencias entre los niveles de carbón (p=0,194) con la prevalencia de neumoconiosis. Conclusiones: Se encontró una prevalencia de neumoconiosis de 42,3% en Cundinamarca. Se requiere contar con medidas de higiene y seguridad industrial efectivas para controlar el riesgo al que están expuestos los mineros de carbón por la inhalación de polvo de carbón.

Aromatherapy uses essential oils (EOs) for several medical purposes, including relaxation. The association between the use of aromas and a decrease in anxiety could be a valuable instrument in managing anxiety in an ever increasing anxiogenic daily life style. Linalool is a monoterpene commonly found as the major volatile component of EOs in several aromatic plant species. Adding to previously reported sedative effects of inhaled linalool, the aim of this study was to investigate the effects of inhaled linalool on anxiety, aggressiveness and social interaction in mice. Additionally, we investigated the effects of inhaled linalool on the acquisition phase of a step-down memory task in mice. Inhaled linalool showed anxiolytic properties in the light/dark test, increased social interaction and decreased aggressive behavior; impaired memory was only seen the higher dose of linalool. These results strengthen the suggestion that inhaling linalool rich essential oils can be useful as a mean to attain relaxation and counteract anxiety. (C) 2009 Elsevier GmbH. All rights reserved.

Hantaviruses belong to the Bunyaviridae family, which consists of vector-borne viruses. These viruses can provoke two infection types: hemorrhagic fever with renal syndrome (HFRS) - which occurs in the Old World - and hantavirus cardiopulmonary syndrome (HCPS) - an emergent zoonosis that can be found in many countries of the western hemisphere. Rodents are hantavirus reservoirs and each species seems to host a different virus type. Humans acquire the infection by inhaling contaminated aerosol particles eliminated by infected animals. The factors involved in the emergence of hantavirus infections in the human population include ecological modifications and changes in human activities. The most important risk factor is contact between man and rodents, as a result of agricultural, forestry or military activities. Rodent control remains the primary strategy for preventing hantavirus diseases, including via health education and hygienic habits.

We described recently that systemic hypoxia provokes vasoconstriction in heart failure (HF) patients. We hypothesized that either the exaggerated muscle sympathetic nerve activity and/or endothelial dysfunction mediate the blunted vasodilatation during hypoxia in HF patients. Twenty-seven HF patients and 23 age-matched controls were studied. Muscle sympathetic nerve activity was assessed by microneurography and forearm blood flow (FBF) by venous occlusion plethysmography. Peripheral chemoreflex control was evaluated through the inhaling of a hypoxic gas mixture (10% O-2 and 90% N-2). Basal muscle sympathetic nerve activity was greater and basal FBF was lower in HF patients versus controls. During hypoxia, muscle sympathetic nerve activity responses were greater in HF patients, and forearm vasodilatation in HF was blunted versus controls. Phentolamine increased FBF responses in both groups, but the increase was lower in HF patients. Phentolamine and N-G-monomethyl-L-arginine infusion did not change FBF responses in HF but markedly blunted the vasodilatation in controls. FBF responses to hypoxia in the presence of vitamin C were unchanged and remained lower in HF patients versus controls. In conclusion, muscle vasoconstriction in response to hypoxia in HF patients is attributed to exaggerated reflex sympathetic nerve activation and blunted endothelial function (NO activity). Gli impianti di incenerimento di rifiuti solidi suscitano preoccupazione nella popolazione per i possibili effetti avversi associati all’esposizione. Gli effetti delle polveri sottili (PM2.5), generate dai processi di combustione, sulla salute umana includono l’insorgenza di patologie a carico del sistema respiratorio e cardiovascolare e l’aumento della mortalità per malattie polmonari e probabilmente cancro al polmone. Lo scopo della tesi è quello di valutare il profilo tossicologico e cancerogeno del particolato atmosferico in prossimità dell’inceneritore di Bologna rispetto alle aree adiacenti mediante l’utilizzo di test alternativi alle metodologie in vivo, come il test di trasformazione cellulare e approcci di tossicogenomica (soprattutto trascrittomica) oltre alla valutazione della variazione del rischio cancerogeno indotto dall’esposizione di PM2.5 in diversi siti (massima ricaduta, controllo, fondo urbano e fondo rurale) e in differenti periodi di campionamento (estate 2008 e inverno 2009). Gli estratti di PM2.5 relativi alla stagione invernale sono risultati più tossici rispetto ai campioni estivi, che inducono tossicità soprattutto alle alte dosi. Per i campioni invernali il numero medio di colonie di cellule BALB/c 3T3 A31-1-1 risulta ridotto in modo significativo anche per le dosi più basse saggiate indipendentemente dal sito di provenienza. Tutti i campioni analizzati sono risultati negativi nel test di trasformazione cellulare in vitro. L’analisi dell’espressione genica delle cellule BALB/c 3T3 A31-1-1, in seguito all’esposizione agli estratti di PM2.5, ha mostrato un effetto stagionale evidente. Relativamente ai campioni invernali è stato evidenziato un maggior effetto tossico da parte del sito di controllo rispetto alla massima ricaduta, poiché nel sito di controllo risultano attivati marcatori di morte cellulare per apoptosi. La valutazione del rischio cancerogeno in tutti i siti valutati non mostra situazioni preoccupanti legate alla predizione di eccessi di rischio di tumori imputabili all’attività dell’inceneritore in quanto le stime di rischio non eccedono mai il valore limite riportato in letteratura.
