Im Rahmen meiner Dissertation habe ich gemäß dem Medizinproduktegesetz ein MR-kompatibles Verabreichungsgerät entwickelt, um Patienten hyperpolarisierte Gas Boli (He-3, Xe-129) als Kontrasgas zur MRT der Lunge zu applizieren. Das Gerät wurde dazu optimiert, die Gase oder Gasmischungen (z. B. HP Gas + N2) in definierten Mengen und zu definierten Zeitpunkten während der Inspiration mit hoher Reproduzierbarkeit und Zuverlässigkeit zu verabreichen, ohne dabei die MR-Qualität zu beeinträchtigen. Aus Sicherheitsgründen und zur späteren Datenanalyse werden die Atemflusskurven der Patienten kontinuierlich angezeigt und aufgezeichnet. Ein Kolbenkompressor ist integriert, um die gesamte gespeicherte He-3-Menge nutzen zu können. Weiterhin ist es möglich, die Polarisation vor Ort zu bestimmen und das abgeatmete He-3 zur späteren Rückgewinnung aufzufangen. Diese Auffangeffizienz konnte durch Untersuchungen mit He-4 (als He-3-Ersatz) gesteigert werden.rnDie ersten MR-Aufnahmen von 10 gesunden Probanden wurden mit diesem Aufbau imrnRahmen einer klinischen Studie durchgeführt. Die Analyse der bestimmten MR-Parameter Signal-Rausch-Verhältnis (SNR), Sauerstoffpartialdruck (pO2), Scheinbarer Diffusionskoeffizient (ADC=apparent diffusion coefficient) sowie die Signalanstiegszeit (rise-time) zeigt eine deutlich bessere Reproduzierbarkeit bei der Verabreichung des He-3 mit dem Applikators anstelle eines Tedlarbags (= kleine heliumdichte Plastiktüte).

Im Rahmen meiner Dissertation wurde ein 3He-Applikationssystem für die in-vivo 3He-Magnetresonanztomographie der Lunge von Ratten entwickelt. Im Gegensatz zu anderen bisher entwickelten MR-kompatiblen Beatmungsgeräten wurde in diesem Applikationssystem erstmals der polarisationserhaltende ³He-Langzeitspeicher im Streufeld des Tomographen integriert. Bei der 3He-Applikation wird das 3He-Gas automatisch aus dem Langzeitspeicher außerhalb des Magneten zum Tier in den Tomographen geleitet. Die Kernspin-Polarisationsverluste, die beim 3He-Transfer zum Tier in den Tomographen auftreten, betragen etwa 1% (rel.) und sind vernachlässigbar gering. Das Beatmungssystem wurde zusammen mit der MRT-Bildgebungssequenz COMSPIRA an mehr als 200 Tieren erfolgreich getestet. Die MRT-Aufnahmen fanden hauptsächlich an einem 0,47 T Niederfeldtomographen bei der Firma Boehringer-Ingelheim Pharma in Biberach statt. Es wurden sowohl morphologische Aufnahmen der Rattenlunge gemacht als auch der „Apparent Diffusion Coefficient“ (ADC) gemessen. Die relative Streuung der ADC-Werte innerhalb einer Gruppe von Tieren ähnlichen Alters und Gewichts betrug dabei 5%. Bei Wiederholungsmessungen an ein und demselben Tier verringerte sich die relative Streuung auf 1%. Diese Werte unterstreichen die hohe Reproduzierbarkeit des Beatmungssystems sowie des gesamten Messverfahrens. Weiterhin wurde ein Verfahren vorgestellt, das den experimentell bestimmten ADC unabhängig von dem Mischungsverhältnis zwischen dem applizierten 3He und der Atemluft in der Lunge macht. Dies kann erreicht werden, indem das hp 3He mit dem chemisch inerten und ungiftigen Gas SF6 in einem definierten Verhältnis gemischt wird und anschließend dem Tier appliziert wird. Zuletzt wurde der ADC von emphysematischen Rattenlungen und von gesunden Rattenlungen gemessen. Die Ergebnisse zeigten einen signifikant kleineren ADC innerhalb der Gruppe der erkrankten Tiere. Since the discovery of the nuclear magnetic resonance (NMR) phenomenon, countless NMR techniques have been developed that are today indispensable tools in physics, chemistry, biology, and medicine. As one of the main obstacles in NMR is its notorious lack of sensitivity, different hyperpolarization (HP) methods have been established to increase signals up to several orders of magnitude. In this work, different aspects of magnetic resonance, using HP noble gases, are studied, hereby combining different disciplines of research. The first part examines new fundamental effects in NMR of HP gases, in theory and experiment. The spin echo phenomenon, which provides the basis of numerous modern experiments, is studied in detail in the gas phase. The changes of the echo signal in terms of amplitude, shape, and position, due to the fast translational motion, are described by an extension of the existing theory and computer simulations. With this knowledge as a prerequisite, the detection of intermolecular double-quantum coherences was accomplished for the first time in the gas phase. The second part of this thesis focuses on the development of a practical method to enhance the dissolution process of HP 129Xe, without loss of polarization or shortening of T1. Two different setups for application in NMR spectroscopy and magnetic resonance imaging (MRI) are presented. The continuous operation allows biological and multidimensional spectroscopy in solutions. Also, first in vitro MRI images with dissolved HP 129Xe as contrast agent were obtained at a clinical scanner.

Deep inelastic neutron scattering measurements on liquid 3He-4He mixtures in the normal phase have been performed on the VESUVIO spectrometer at the ISIS pulsed neutron source at exchanged wave vectors of about q≃120.0Å-1. The neutron Compton profiles J(y) of the mixtures were measured along the T=1.96K isotherm for 3He concentrations, x, ranging from 0.1 to 1.0 at saturated vapor pressures. Values of kinetic energies 〈T〉 of 3He and 4He atoms as a function of x, 〈T〉(x), were extracted from the second moment of J(y). The present determinations of 〈T〉(x) confirm previous experimental findings for both isotopes and, in the case of 3He, a substantial disagreement with theory is found. In particular 〈T〉(x) for the 3He atoms is found to be independent of concentration yielding a value 〈T〉3(x=0.1)≃12K, much lower than the value suggested by the most recent theoretical estimates of approximately 19 K.

Reactions produced by the He3 bombardment of the He3 have been investigated for bombarding energies from 1 to 20 MeV using a tandem Van de Graaff accelerator. Proton spectra from the three-body reaction He3(He3, 2p)He4 have been measured with a counter telescope at 13 angles for 9 bombarding energies between 3 and 18 MeV. The results are compared with a model for the reaction which includes a strong p-He4 final-state interaction. Alpha-particle spectra have been obtained at 12 and 18 MeV for forward angles with a magnetic spectrometer. These spectra indicate a strongly forward-peaked mechanism involving the 1S0 p-p interaction in addition to the p-He4 interaction. Measurements of p-He4 and p-p coincidence spectra at 10 MeV confirm these features of the reaction mechanism. Deuteron spectra from the reaction of He3(He3, d)pHe3 have been measured at 18 MeV. A triton spectrum from the reaction He3(He3, t)3p at 20 MeV and 40 is interpreted in terms of a sequential decay through an excited state of the alpha particle at 20.0 MeV. No effects are observed which would indicate an interaction in the residual (3p) system. Below 3 MeV the He3(He3, 2p)He4 reaction mechanism is observed to be changing and further measurements are suggested in view of the importance of this reaction in stellar interiors.

The (He3, n) reactions on B11, N15, O16, and O18 targets have been studied using a pulsed-beam time-of-flight spectrometer. Special emphasis was placed upon the determination of the excitation energies and properties of states with T = 1 (in Ne18), T = 3/2 (in N13 and F17) and T = 2 (in Ne20). The identification of the T = 3/2 and T = 2 levels is based on the structure of these states as revealed by intensities and shapes of angular distributions. The reactions are interpreted in terms of double stripping theory. Angular distributions have been compared with plane and distorted wave stripping theories. Results for the four reactions are summarized below:

1) O16 (He3, n). The reaction has been studied at incident energies up to 13.5 MeV and two previously unreported levels in Ne18 were observed at Ex = 4.55 ± .015 MeV (Γ = 70 ± 30 keV) and Ex = 5.14 ± .018 MeV (Γ = 100 ± 40 keV).

2) B11 (He3, n). The reaction has been studied at incident energies up to 13.5 MeV. Three T = 3/2 levels in N13 have been identified at Ex = 15.068 ± .008 MeV (Γ ˂ 15 keV), Ex = 18.44 ± .04, and Ex 18.98 ± .02 MeV (Γ = 40 ± 20 keV).

3) N15 (He3, n). The reaction has been studied at incident energies up to 11.88 MeV. T = 3/2 levels in F17 have been identified at Ex = 11.195 ± .007 MeV (Γ ˂ 20 keV), Ex = 12.540 ± .010 MeV (Γ ˂ 25 keV), and Ex = 13.095 ± .009 MeV (Γ ˂ 25 keV).

4) O18 (He3, n). The reaction has been studied at incident energies up to 9.0 MeV. The excitation energy of the lowest T = 2 level in Ne20 has been found to be 16.730 ± .006 MeV (Γ ˂ 20 keV).

Angular distributions of the transitions leading to the above higher isospin states are well described by double stripping theory. Analog correspondences are established by comparing the present results with recent studies (t, p) and (He3, p) reactions on the same targets.

Using a transport model coupled with a phase-space coalescence afterburner, we study the triton-He-3 (t-He-3) ratio with both relative and differential transverse flows in semicentral Sn-132 + Sn-124 reactions at a beam energy of 400 MeV/nucleon. The neutron-proton ratios with relative and differential flows are also discussed as a reference. We find that similar to the neutron-proton pairs, the t-He-3 pairs also carry interesting information regarding the density dependence of the nuclear symmetry energy. Moreover, the nuclear symmetry energy affects more strongly the t-He-3 relative and differential flows than the pi(-)/pi(+) ratio in the same reaction. The t-He-3 relative flow can be used as a particularly powerful probe of the high-density behavior of the nuclear symmetry energy.

The single charge transfer process in He-3(2+)+He-4 collisions is investigated using the quantum-mechanical molecular-orbital close-coupling method, in which the adiabatic potentials and radial couplings are calculated by using the ab initio multireference single- and double-excitation configuration interaction methods. The differential cross sections for the single charge transfer are presented at the laboratorial energies E = 6 keV and 10 keV for the projectile He-3(2+). Comparison with the existing data shows that the present results are better in agreement with the experimental measurements than other calculations in the dominant small angle scattering, which is attributed to the accurate calculations of the adiabatic potentials and the radial couplings.

Glucose-dependent insulinotropic polypeptide (gastric inhibitory polypeptide [GIP]) is an important incretin hormone secreted by endocrine K-cells in response to nutrient ingestion. In this study, we investigated the effects of chemical ablation of GIP receptor (GIP-R) action on aspects of obesity-related diabetes using a stable and specific GIP-R antagonist, (Pro3)GIP. Young adult ob/ob mice received once-daily intraperitoneal injections of saline vehicle or (Pro3)GIP over an 11-day period. Nonfasting plasma glucose levels and the overall glycemic excursion (area under the curve) to a glucose load were significantly reduced (1.6-fold; P <0.05) in (Pro3)GIP-treated mice compared with controls. GIP-R ablation also significantly lowered overall plasma glucose (1.4-fold; P <0.05) and insulin (1.5-fold; P <0.05) responses to feeding. These changes were associated with significantly enhanced (1.6-fold; P <0.05) insulin sensitivity in the (Pro3)GIP-treated group. Daily injection of (Pro3)GIP reduced pancreatic insulin content (1.3-fold; P <0.05) and partially corrected the obesity-related islet hypertrophy and ß-cell hyperplasia of ob/ob mice. These comprehensive beneficial effects of (Pro3)GIP were reversed 9 days after cessation of treatment and were independent of food intake and body weight, which were unchanged. These studies highlight a role for GIP in obesity-related glucose intolerance and emphasize the potential of specific GIP-R antagonists as a new class of drugs for the alleviation of insulin resistance and treatment of type 2 diabetes.

Mechanical ventilation is the major cause of iatrogenic lung damage in intensive care units. Although inflammation is known to be involved in ventilator-induced lung injury (VILI), several aspects of this process are still unknown. Pentraxin 3 (PTX3) is an acute phase protein with important regulatory functions in inflammation which has been found elevated in patients with acute respiratory distress syndrome. This study aimed at investigating the direct effect of PTX3 production in the pathogenesis of VILI. Genetically modified mice deficient and that over express murine Ptx3 gene were subjected to high tidal volume ventilation (V-T = 45 mL/kg, PEEPzero). Morphological changes and time required for 50% increase in respiratory system elastance were evaluated. Gene expression profile in the lungs was also investigated in earlier times in Ptx3-overexpressing mice. Ptx3 knockout and wild-type mice developed same lung injury degree in similar times (156 +/- 42 min and 148 +/- 41 min, respectively: p = 0.8173). However, Ptx3 overexpression led to a faster development of VILI in Ptx3-overexpressing mice (77 +/- 29 min vs 118 +/- 41 min, p = 0.0225) which also displayed a faster kinetics of Il1b expression and elevated Ptx3, Cxcl1 and Ccl2 transcripts levels in comparison with wild-type mice assessed by quantitative real-time polymerase chain reaction. Ptx3 deficiency did not impacted the time for VILI induced by high tidal volume ventilation but Ptx3-overexpression increased inflammatory response and reflected in a faster VILI development.
