Determination of anabolic steroids often requires the use of elaborate techniques such as gas chromatography - mass spectrometry (GC-MS), gas liquid chromatography (GLC) and more recently high performance liquid chromatography (HPLC). Most of these methods employ derivatisation techniques prior to detection which makes them tedious and relatively time consuming. Other methods demand a great deal of skill. A simple and rapid analytical method, based on differential pulse polarography at a dropping mercury electrode has been developed for the determination of various anabolic steroids in a range of commercially available pharmaceutical preparations. Detailed investigation of the electrochemical behaviour of these steroids was made in order to elucidate the electrode processes involved, in addition to optimising the method. Several other analytical methods such as GC-MS, NMR, ultraviolet (UV), infrared (IR) spectroscopy were also used to confirm the products of the chemical and electrochemical reactions. Possible reactions are suggested. Various extraction procedures were examined for separation of selected steroids from the oil-based or pill matrix and their suitability for polarographic determination is discussed.

The differential pulse polarographic behaviour of cinnamic acid was studied in acetate and phosphate buffer solutions (pH 3.5-7.5). The reduction mechanism is discussed. The drug can be determined at pH 5.0 over the concentration range 5 X 10(-5)-1 X 10(-3) mol l(-1). The effect of tetraalkylammonium salts on the electroanalytical determination of cinnamic acid was investigated, the direct determination of the drug (0.7-5.5 mu g ml(-1)) in urine samples diluted with acetate buffer (pH 5.0) can be effected in the presence of 1 x 10(-3) mol l(-1) cetyldimethylethylammonium bromide solution. The detection limit was found to be 0.1 mu g ml(-1). The relative standard deviation from six determinations at the 5.5 mu g ml(-1) level was 1%.

Clotrimazole was shown to react at room temperature in Britton Robinson buffer pH 2 with the reactive dye Procion Red HE-3B. The product exhibited a differential pulse polarographic peak at -0.38 V, which was well separated from the peaks of the reactive dye at -0.08, -0.80 and -0.95 V, and this allowed the indirect determination of clotrimazole in the presence of excess of the reactive dye. The method has been applied satisfactorily to the determination of clotrimazole in pharmaceutical formulations, calibration graphs are rectilinear up to at least 40 mug ml(-1). The detection limit was calculated to be 2.6 mug ml(-1) (3 sigma). (C) 2002 Elsevier B.V. B.V. All rights reserved.

Ceftazidime shows two main polarographic reduction peaks at pH 4.0, that at -0.45 V owing to reduction of the C=N bond in the methylethoxyimino group and that at -1.00 V owing to the reductive elimination of pyridine: the first peak is particularly suitable for the determination of ceftazidime. Ceftazidime can also be determined indirectly using the tensammetric peak at -0.60 V (in Britton-Robinson buffer pH 9.5) of pyridine liberated on hydrolysis. Ceftazidime can be determined in urine using the direct method only after Cls solid phase extraction, but it can be determined directly in the urine by hydrolysing it and using the pyridine peak. A novel voltammetric method for simultaneous determination of the glucocorticoid residues prednisone, prednisolone, and dexamethasone was developed. All three compounds were reduced at a mercury electrode in a Britton-Robinson buffer (pH 3.78), and well-defined voltammetric waves were observed. However, the voltammograms of these three compounds overlapped seriously and showed nonlinear character, and thus, it was difficult to analyze the compounds individually in their mixtures. In this work, two chemometrics methods, principal component regression (PCR) and partial least squares (PLS), were applied to resolve the overlapped voltammograms, and the calibration models were established for simultaneous determination of these compounds. Under the optimum experimental conditions, the limits of detection (LOD) were 5.6, 8.3, and 16.8 µg l-1 for prednisone, prednisolone, and dexamethasone, respectively. The proposed method was also applied for the determination of these glucocorticoid residues in the rabbit plasma and human urine samples with satisfactory results.

A differential pulse polarographic (DPP) method based on the adsorption catalytic current in a medium containing chlorate and 8-hydroxyquinoline (oxine) is suggested for the determination of molybdenum(VI). Experimental conditions such as pH and the composition of supporting electrolyte have been optimized to get a linear calibration graph at trace levels of Mo(VI). The sensitivity for molybdenum can be considerably enhanced by this method. The influence of possible interferences on the catalytic current has been investigated. The sensitivity of the method is compared with those obtained for other DPP methods for molybdenum. A detection limit of 1.0 x 10(-8) mol/L has been found.

Here we investigated the analytical performances of the bismuth-modified zeolite doped carbon paste electrode (BiF-ZDCPE) for trace Cd and Pb analysis. The characteristics of bismuth-modified electrodes were improved greatly via addition of synthetic zeolite into carbon paste. To obtain high reproducibility and sensitivity, optimum experimental conditions for bismuth deposition Were Studied.

A cathodically pretreated boron-doped diamond electrode was used for the simultaneous anodic determination of ascorbic acid (AA) and caffeine (CAF) by differential pulse voltammetry Linear calibration curves (r = 0 999) were obtained from 1 9 x 10(-5) to 2 I x 10(-4) mol L(-1) for AA and from 9 7 x 10(-6) to 1 1 x 10-4 mol L(-1) for CAF. with detection limits of 19 wool L(-1) and 7 0 mu nol L(-1). respectively This method was successfully applied for the determination of AA and CAF in pharmaceutical formulations. with results equal to those obtained using a HPLC reference method
