A facile and convenient synthesis method has been developed for substituted quinoxalines and 2H-benzo[b][1,4]oxazines from the reactions of alpha-bromoketones with benzene-1,2-diamine and 2-aminophenol, respectively, which were catalyzed by tetrabutyl ammonium bromide (TBAB) in aqueous basic media.

An efficient ring rearrangement metathesis (RRM) approach to the synthesis of benzo[b]quinolizine and benzoindolizine systems from N-propargyl-phenanthridine derivatives is reported. A Hovel use of flash vacuum pyrolysis (FVP) for the Boc-deprotection of acid-sensitive substrates is also disclosed.

Asymmetric heteroatom oxidation of benzo[b]thiophenes to yield the corresponding sulfoxides was catalysed by toluene dioxygenase (TDO), naphthalene dioxygenase (NDO) and styrene monooxygenase (SMO) enzymes present in P. putida mutant and E. coli recombinant whole cells. TDO-catalysed oxidation yielded the relatively unstable benzo[b] thiophene sulfoxide; its dimerization, followed by dehydrogenation, resulted in the isolation of stable tetracyclic sulfoxides as minor products with cis-dihydrodiols being the dominant metabolites. SMO mainly catalysed the formation of enantioenriched benzo[b] thiophene sulfoxide and 2-methyl benzo[b] thiophene sulfoxides which racemized at ambient temperature. The barriers to pyramidal sulfur inversion of 2- and 3-methyl benzo[b] thiophene sulfoxide metabolites, obtained using TDO and NDO as biocatalysts, were found to be ca.: 25-27 kcal mol(-1). The absolute configurations of the benzo[b] thiophene sulfoxides were determined by ECD spectroscopy, X-ray crystallography and stereochemical correlation. A site-directed mutant E. coli strain containing an engineered form of NDO, was found to change the regioselectivity toward preferential oxidation of the thiophene ring rather than the benzene ring.

Enzymatic cis-dihydroxylation of benzo[b]thiophene, benzo[b]furan and several methyl substituted derivatives was found to occur in both the carbocyclic and heterocyclic rings. Relative and absolute configurations and enantiopurities of the resulting dihydrodiols were determined. Hydrogenation of the alkene bond in carbocyclic cis-dihydrodiols and ring-opening epimerization/reduction reactions of heterocyclic cis/trans-dihydrodiols were also studied. The relatively stable heterocyclic dihydrodiols of benzo[b]thiophene and benzo[b]furan showed a strong preference for the trans configuration in aqueous solutions. The 2,3-dihydrodiol metabolite of benzo[b]thiophene was utilized as a precursor in the chemoenzymatic synthesis of the unstable arene oxide, benzo[b]thiophene 2,3-oxide.

As 4-quinolonas e as acridonas são duas importantes famílias de compostos heterocíclicos azotados naturais que apresentam uma variedade de importantes aplicações biológicas. As 4-quinolonas têm sido objecto de extensos estudos devido às suas potenciais aplicações como fortes agentes citotóxicos, antimitóticos e anti-plaquetários e como protectores cardiovasculares, mas o seu uso é principalmente como antibióticos de largo espectro. As acridonas são conhecidas por apresentarem importante actividade antiviral, antiparasitária, contra a leishmania e a malária, e anticancerígena. A variedade de importantes aplicações biológicas das 4-quinolonas e das acridonas e a contínua procura pela comunidade científica de novas substâncias com actividades biológicas atractivas destaca estes compostos como alvos interessantes para a preparação de novos derivados e/ou para o desenvolvimento de novos métodos de síntese destas duas famílias de compostos. No primeiro capítulo desta dissertação descreve-se a síntese dos compostos de partida que tiveram de ser previamente preparados para o desenvolvimento das novas rotas de síntese apresentadas nos capítulos seguintes. A 4-cloro-3- formilquinolina foi obtida através da reacção de Vilsmeier-Haack da 2’-aminoacetofenona enquanto que a 3-formilquinolin-4(1H)-ona foi facilmente preparada por hidrólise ácida da anterior. Foi necessário proteger o grupo amina da quinolin-4(1H)-ona para evitar reacções secundárias e foram descritas as reacções de protecção com o grupo metilo, etoxicarbonilo e ptoluenossulfonilo. Os 2,2-dióxidos de 1,3-di-hidrobenzo[c]tiofeno, necessários para o estudo das reacções de Diels-Alder, não se encontram disponíveis comercialmente e a sua síntese é também descrita. No segundo capítulo reporta-se uma nova e eficiente rota de síntese de (Z)- e (E)-3-estirilquinolin-4(1H)-onas a partir da reacção de Wittig da 4-cloro-3- formilquinolina e de 3-formilquinolin-4(1H)-onas com benzilidenotrifenilfosforanos. As (Z)-3-estirilquinolin-4(1H)-onas foram obtidas com elevada diastereoselectividade a partir da reacção das 3-formilquinolin- 4(1H)-onas N-protegidas enquanto que as (E)-3-estirilquinolin-4(1H)-onas foram preparadas através da reacção de Wittig da 4-cloro-3-formilquinolina seguida da hidrólise ácida das respectivas (Z)- e (E)-4-cloro-3-estirilquinolinas obtidas. Ambas as rotas sintéticas são eficientes, independentemente dos substituintes dos benzilidenotrifenilfosforanos. No terceiro capítulo, descreve-se um novo método de síntese de novas benzo[b]acridonas a partir da reacção de Diels-Alder de 3-formilquinolin-4(1H)- onas N-protegidas, que actuam como dienófilos, com dienos altamente reactivos, os orto-benzoquinodimetanos, preparados in situ através da extrusão térmica do dióxido de enxofre dos respectivos 2,2-dióxidos de 1,3-dihidrobenzo[ c]tiofeno. A reacção de cicloadição das 3-formilquinolin-4(1H)-onas N-protegidas com orto-benzoquinodimetanos origina as benzo[b]-1,6,6a,12atetra- hidroacridin-12(7H)-onas esperadas, que são o resultado da referida cicloadição seguida de desformilação in situ, e mostrou ser eficiente apenas quando o grupo amina está derivatizado com um grupo sacador de electrões. A desidrogenação destas benzo[b]-1,6,6a,12a-tetra-hidroacridin-12(7H)-onas em dimetilsulfóxido utilizando uma quantidade catalítica de iodo foi também descrita e originou como produto principal as benzo[b]acridin-12(7H)-onas N-desprotegidas. Todos os compostos novos sintetizados foram caracterizados por diversas técnicas analíticas, especialmente por estudos espectroscópicos de ressonância magnética nuclear (RMN), incluindo espectros de 1H e 13C, bidimensionais de correlação espectroscópica homonuclear e heteronuclear e de efeito nuclear de Overhauser (NOESY). Foram também efectuados, sempre que possível, espectros de massa (EM) e análises elementares ou espectros de massa de alta resolução (EMAR) para todos os compostos novos sintetizados. O tautomerismo da 3-formil- e 3-estirilquinolin-4(1H)-onas e as isomerizações (E) (Z) e rotacional das 3-estirilquinolin-4(1H)-onas e das 4-cloro-3-estirilquinolinas foram estudados através de ressonância magnética nuclear experimental (RMN 1H e 13C) e teórica [B3LYP/6-311++G(d,p)].

A simple and convenient synthesis of 3-methylene-4-aryl-1,3,4,5-tetrahydro-benzo[b][1,4] diazepin-2-ones was accomplished by the SN2 nucleophilic substitution of the acetates of Baylis-Hillman adducts of acrylate with 1,2-phenylenediamines followed by base-mediated intramolecular cyclization. On the other hand similar substrates derived from the Baylis-Hillman adducts of acrylonitrile via Pinner’s reaction leads to 3-arylmethylene-4,5-dihydro-3H-benzo[b][1,4]diazepin-2-ylamines in good yields..

This study examined the developmental toxicity of the polycyclic aromatic hydrocarbons (PAHs) 11H-benzo(b)fluorene (BBF) and 4-azapyrene (AP) in comparison to the known teratogen retene. Developmental toxicity assays were performed in zebrafish embryos exposed for 120 h. BBF and retene induced a similar dioxin-like phenotype, whereas AP showed distinct effects, particularly craniofacial malformations. Microarray analysis revealed that for BBF and retene, drug metabolism pathways were induced, which were confirmed by subsequent studies of cyp1a gene expression. For AP, microarray analysis revealed the regulation of genes involved in retinoid metabolism and hematological functions. Studies with a panel of CALUX((R)) bioassays to screen for endocrine disrupting activity of the compounds also revealed novel antagonistic effects of BBF and retene on androgen and progesterone receptors. Classification analysis revealed distinct gene expression profiles for both individual and combined PAH exposure. This study highlights the potential health risk of non priority PAHs.

Background The population exposed to potentially hazardous substances through inappropriate and unsafe management practices related to disposal and recycling of end-of-life electrical and electronic equipment, collectively known as e-waste, is increasing. We aimed to summarise the evidence for the association between such exposures and adverse health outcomes. Methods We systematically searched five electronic databases (PubMed, Embase, Web of Science, PsycNET, and CINAHL) for studies assessing the association between exposure to e-waste and outcomes related to mental health and neurodevelopment, physical health, education, and violence and criminal behaviour, from Jan 1, 1965, to Dec 17, 2012, and yielded 2274 records. Of the 165 full-text articles assessed for eligibility, we excluded a further 142, resulting in the inclusion of 23 published epidemiological studies that met the predetermined criteria. All studies were from southeast China. We assessed evidence of a causal association between exposure to e-waste and health outcomes within the Bradford Hill framework. Findings We recorded plausible outcomes associated with exposure to e-waste including change in thyroid function, changes in cellular expression and function, adverse neonatal outcomes, changes in temperament and behaviour, and decreased lung function. Boys aged 8–9 years living in an e-waste recycling town had a lower forced vital capacity than did those living in a control town. Significant negative correlations between blood chromium concentrations and forced vital capacity in children aged 11 and 13 years were also reported. Findings from most studies showed increases in spontaneous abortions, stillbirths, and premature births, and reduced birthweights and birth lengths associated with exposure to e-waste. People living in e-waste recycling towns or working in e-waste recycling had evidence of greater DNA damage than did those living in control towns. Studies of the effects of exposure to e-waste on thyroid function were not consistent. One study related exposure to e-waste and waste electrical and electronic equipment to educational outcomes. Interpretation Although data suggest that exposure to e-waste is harmful to health, more well designed epidemiological investigations in vulnerable populations, especially pregnant women and children, are needed to confirm these associations. Variations in the concentrations and microheterotrophic degradation rates of selected Polycyclic Aromatic Hydrocarbons (PAH) in the water column of the Tamar Estuary were investigated in relation to the major environmental variables. Concentrations of individual PAH varied typically between i and 50 ng l−1 Based on their observed environmental behaviour the PAH appeared divisible into two groupings: (1) low molecular weight PAH incorporating naphthalene, phenanthrene and anthracence and (a) the larger molecular weight homologues (fluoranthene, pyrene, chrysene, benz(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, benzo(a)-pyrene). Group 1 PAH showed a complex distribution throughout the estuary with no significant correlations with either salinity or suspended particulates. Based on their relatively low particle affinity and high water solubilities and vapour pressures, volatilization is proposed as an important process in determining their fate. Microheterotrophic turnover times of naphthalene varied between x and 30 days, and were independent of suspended solids with maximum degradation rates located in the central and urban regions of the Estuary. When compared with the flushing times for the Tamar (3–5 days), it is probable that heterotrophic activity is important in the removal of naphthalene (and possibly the other Group 1 PAH) from the estuarine environment. In contrast Group 2 PAH concentrations exhibited highly significant correlations with suspended particulates. Highest concentrations occurred at the turbidity maximum, with a secondary concentration maximum localized to the industrialized portion of the estuary and associated with anthropogenic inputs. Laboratory degradation studies of benzo(a)pyrene in water samples taken from the estuary showed turnover times for the compound of between 2000 and 9000 days. Degradation rates correlated positively with suspended solids. The high particulate affinity and microbial refractivity of Group 2 PAH indicate sediment burial as the principal tate of these PAH in the Tamar Estuary. Estuarine sediments contained typically 50–1500 ng g−1 dry weight of individual PAH which were comparable to the levels of Group 2 PAH associated with the suspended particulates. Highest concentrations occurred at the riverine end of the estuary resulting from unresolved inputs in the catchment. Subsequent dilution by less polluted marine sediments together with slow degradation results in a seaward trend of decreasing concentrations. However, there is a secondary maximum of PAH superimposed on this trend which is associated with urban Plymouth.

The aim of this work is the treatment of produced water from oil by using electrochemical technology. Produced water is a major waste generated during the process of exploration and production in the oil industry. Several approaches are being studied aiming at the treatment of this effluent; among them can be cited the biological process and chemical treatments such as advanced oxidation process and electrochemical treatments (electrooxidation, electroflotation, electrocoagulation, electrocoagulation). This work studies the application of electrochemical technology in the treatment of the synthetic produced water effluent through the action of the electron, in order to remove or transform the toxic and harmful substances from the environment by redox reactions in less toxic substances. For this reason, we used a synthetic wastewater, containing a mixture H2SO4 0,5M and 16 HPAs, which are: naphthalene, acenaphthylene, acenaphthene, fluorene, phenanthrene, anthracene, fluoranthene, pyrene, benzo (a) anthracene, chrysene, benzo(b)fluoranthene, benzo(k) fluoranthene, benzo(a)pyrene, indeno(1,2,3-cd)pyrene, dibenzo(a, h)anthracene, benzo(g, h, i)perylene. Bulk electrochemical oxidation experiments were performed using a batch electrochemical reactor containing a pair of parallel electrodes, coupled with a power supply using a magnetic stirrer for favoring the transfer mass control. As anodic material was used, a Dimensionally Stable Anode (DSA) of Ti/Pt, while as cathode was used a Ti electrode. Several samples were collected at specific times and after that, the analysis of these samples were carried out by using Gas Chromatography Coupled to Mass Spectrometry (GC - MS) in order to determine the percentage of removal. The results showed that it was possible to achieve the removal of HPAs about 80% (in some cases, more than 80%). In addition, as an indicator of the economic feasibility of electrochemical treatment the energy consumption was analyzed for each hour of electrolysis, and based on the value kWh charged by ANEEL, the costs were estimated. Thus, the treatment costs of this research were quite attractive

In Brazil, sugarcane fields are often burned to facilitate manual harvesting, and this burning causes environmental pollution from the large amounts of soot released into the atmosphere. This material contains numerous organic compounds such as PAHs. In this study, the concentrations of PAHs in two particulate-matter fractions (PM2.5 and PM10) in the city of Araraquara (SE Brazil, with around 200,000 inhabitants and surrounded by sugarcane plantations) were determined during the sugarcane harvest (HV) and non-harvest (NHV) seasons in 2008 and 2009. The sampling strategy included four campaigns, with 60 samples in the NHV season and 220 samples in the HV season. The PM2.5 and PM10 fractions were collected using a dichotomous sampler (10 L min(-1), 24 h) with Teflon (TM) filters. The filter sets were extracted (ultrasonic bath with hexane/acetone (1:1 v/v)) and analyzed by HPLC/Fluorescence. The median concentration for total PAHs (PM2.5 in 2009) was 0.99 ng m(-3) (NHV) and 3.3 ng m(-3) (HV). In the HV season, the total concentration of carcinogenic PAHs (benz(a)anthracene, benzo(b)fluoranthene, benzo(k)fluoranthene, and benzo(a)pyrene) was 5 times higher than in the NHV season. B(a)P median concentrations were 0.017 ng m(-3) and 0.12 ng m(-3) for the NHV and HV seasons, respectively. The potential cancer risk associated with exposure through inhalation of these compounds was estimated based on the benzo[a]pyrene toxic equivalence (BaPeq), where the overall toxicity of a PAR mixture is defined by the concentration of each compound multiplied by its relative toxic equivalence factor (TEF). BaPeq median (2008 and 2009 years) ranged between 0.65 and 1.0 ng m(-3) and 1.2-1.4 ng m(-3) for the NHV and HV seasons, respectively. Considering that the maximum permissible BaPeq in ambient air is 1 ng m(-3), related to the increased carcinogenic risk, our data suggest that the level of human exposure to PAHs in cities surrounded by sugarcane crops where the burning process is used is cause for concern. The incomplete combustion of biomass is one of the most important sources of emissions of organic compounds into the atmosphere, like polycyclic aromatic hydrocarbons (PAHs) which show genotoxic activity. Since environmental samples generally contain interferents and trace amounts of PAHs of interest, concentration and clean-up procedures are usually required prior to the final chromatographic analysis. This paper discusses the performance of Sep-Pak cartridges (silica gel and RP18) on clean-up of sugar cane soot extract. The best results were obtained with a silica Sep-Pak cartridge. The recoveries ranged from 79% (benzo[b]fluoranthene) to 113% (benzo[e]pyrene). (C) 2000 Elsevier B.V. B.V. All rights reserved.

ABSTRACT: In Guajará bay, mouth of the Amazon River, is located the capital of Pará state (Belém) and its metropolitan area. Exists in this area an intense boat traffic, as well as transportation and sale of fuels in floating docks and activities related to the storage and transportation of oil in Miramar Petrochemical Terminal (TEMIR). Small spills and discharges of oil in water can serve as sources of pollution by polycyclic aromatic hydrocarbons (PAHs). PAHs are organic compounds generated by incomplete combustion of organic matter (OM) and are among the contaminants of most interest in environmental studies due to their mutagenic and carcinogenic potential. One way to detect and evaluate the impact of PAHs in an environment is using biomonitors, however the qualitative and quantitative analysis in sediments are most widely used. This study aimed to evaluate, by Gas Chromatography/Mass Spectrometry (GC/MS), the 16 PAHs considered as priority by the Environmental Protection Agency of the United States in sediments and benthic organisms (Namalicastys abiuma) of TEMIR. Field expeditions were carried out in December 2012, March, May and June 2013 representing the dried, wet (two samples) and dried seasons successively. Fine sediments levels dominated in both sampling periods and the %Fines in Guajará bays has a direct relationship to the %OM. In relation to the 16 PAHs studied, 10 of them were detected in the wet season sediments samples and 8 in the dried season. Even with a low diversity of aromatic compounds, sediment samples of the dried season presented greater ΣHPAs (1.351,43 ng g-1) than the sediments of the wet season (263,99 ng g-1), which can be related to the increased hydrodynamic in Guajará bay this last period. Correlation analysis indicated that ΣHPAs not seem to be influenced by %Fine and %OM. Benzo(a)pyrene accounted for 87% of ΣHPAs during the wet season, the other PAHs showed percentage ≤ 3%. During the other period stood out: pyrene (18% of ΣHPAs) fluoranthene (16%), the chrysene, benzo(b)fluoranthene (15%) and benzo(a)pyrene (11%). The use of the geochemical reason to interpret likely sources of PAHs indicated that there is a predominance of aromatics from pyrolytic origin in TEMIR. The activities that may be responsible for the PAHs levels observed in sediments and benthic organisms in TEMIR are the discharge of untreated domestic sewage and the emissions of particulates from cars and small/medium boats. The ΣHPAs seems to influence the density of polychaetes because a reduction of about 50% in the number of organisms was observed during the dry season when there was the greater amount of ΣHPAs in their body structure. Among the 16 PAHs studied, 11 were detected in the polychaetes during the dry season (ΣHPAsb>MEDb> = 848,71 ng g-1) and 10 in the wet season (ΣHPAsb>MEDb> = 141,85 ng g-1). Were highlighted during the dried season: indeno(1,2,3-c,d)pyrene (47%) and pyrene (23%). While in the wet season: pyrene (23%), chrysene (17%), fluorene (17%) and fluoranthene (13%). The %Rec obtained was >>100% indicating a matrix effect and reducing the quantitative accuracy of results. The use of the polychaete N. abiuma as biomonitor of pollution by PAHs in estuaries not seem to be effective when the sample period is limited, because collecting it requires a great effort to obtain a small amount of mass and it still results in a matrix effect in the chromatographic analysis (method %Rec >> 100%) that may not be corrected due to the restricted availability of sample mass. Studies with a larger number of samples, enabling depth statistical analysis, and covering other collection points in Guajará bay are required to prove mathematically that has been stated in this dissertation.
