Clusterin is a stress-activated, cytoprotective chaperone that confers broad-spectrum treatment resistance in cancer. However, the molecular mechanisms mediating CLU transcription following anticancer treatment stress remain incompletely defined. We report that Y-box binding protein-1 (YB-1) directly binds to CLU promoter regions to transcriptionally regulate clusterin expression. In response to endoplasmic reticulum stress inducers, including paclitaxel, YB-1 is translocated to the nucleus to transactivate clusterin. Furthermore, higher levels of activated YB-1 and clusterin are seen in taxane-resistant, compared with parental, prostate cancer cells. Knockdown of either YB-1 or clusterin sensitized prostate cancer cells to paclitaxel, whereas their overexpression increased resistance to taxane. Clusterin overexpression rescued cells from increased paclitaxel-induced apoptosis following YB-1 knockdown; in contrast, however, YB-1 overexpression did not rescue cells from increased paclitaxel-induced apoptosis following clusterin knockdown. Collectively, these data indicate that YB-1 transactivation of clusterin in response to stress is a critical mediator of paclitaxel resistance in prostate cancer. Mol Cancer Res; 9(12); 1755–66.

Reactivation of androgen receptor signalling is one of the hallmarks of prostate cancer progression to the terminal castrate resistant stage. A better understanding of mechanisms driving this adaptive response is essential for the development of innovative intervention strategies that effectively delay or halt prostate cancer progression. The Y-box binding protein 1 (YB-1) has been found to be closely associated with prostate cancer progression. By characterising its role in the adaptive process leading to castrate resistance, we aim to promote YB-1 as a novel therapeutic target in advanced prostate cancer.

This work reports on the study of the nanophosphor. Y2O2S:Er(2%),Yb(1%) obtained from polymeric resin to be evaluated as fluorescent label with Suitable features to conjugate with bio-molecules for bioassay up-converting phosphor technology (UPT) application A conjugation protocol between bovine serum albumin (BSA) and the aminofunctionalized nanophosphor containing or not spherical silica was established UV-vis results indicated an effective conjugation between nanophosphor particles and the protein up-conversion measurements under 980 nm excitation performed for samples before and after aminofunctionalization showed that nanophosphor particles luminescence features keep unchanged in all cases All results suggest that the adapted protocol is feasible to provide a nanoparticle-protein effective conjugation preserving nanophosphor optical features The presence of spherical silica can be considered advantageous to increase conjugation efficiency Therefore. the developed procedure is applicable for future conjugations between the chosen nanophosphor and the streptavidin protein chat takes part in the well known self-recognition system avidin-biotin. We have previously shown that Y box-binding protein-1 (YB-1) binds preferentially to cisplatin-modified Y box sequences. Based on structural and biochemical data, we predicted that this protein binds single-stranded nucleic acids. In the present study we confirmed the prediction and also discovered some unexpected functional features of YB-1. We found that the cold shock domain of the protein is necessary but not sufficient for double-stranded DNA binding while the C-tail domain interacts with both single-stranded DNA and RNA independently of the cold shock domain. In an in vitro translation system the C-tail domain of the protein inhibited translation but the cold shock domain did not. Both in vitro pull-down and in vivo co-immunoprecipitation assays revealed that YB-1 can form a homodimer. Deletion analysis mapped the C-tail domain of the protein as the region of homodimerization. We also characterized an intrinsic 3′→5′ DNA exonuclease activity of the protein. The region between residues 51 and 205 of its 324-amino acid extent is required for full exonuclease activity. Our findings suggest that YB-1 functions in regulating DNA/RNA transactions and that these actions involve different domains.

基于不同稀土氯化物在高温与AlCl3 生成气态配合物LnAlnCl3n + 3 的热力学行为不同 ,利用化学气相传输法研究了二元相邻混合重稀土氧化物Ho2 O3 Er2 O3 、Er2 O3 Tm2 O3 、Tm2 O3 Yb2 O3 和Yb2 O3 Lu2 O3 的分离特性。结果发现 ,Ho、Er、Tm、Yb和Lu的氯化物大量沉积在 80 0~ 10 5 0K的中、低温区。温度愈高 ,原子序数愈小的稀土氯化物优先沉积 ,温度愈低 ,原子序数愈大的稀土氯化物优先沉积。最大分离系数 :SFEr/Ho=1 4 1,SFEr/Tm=1 6 0 ,SFTm/Yb=1 91,SFLu/Yb=1 37。

在Mg-9Al-1Zn合金的基础上添加不同质量分数的Yb,制备了4种合金。通过X射线衍射、金相显微镜、扫描电镜及能谱分析仪等手段分析了Yb的添加对AZ91D显微组织和力学性能的影响。结果表明:Yb的加入,显著改变了AZ91D合金的铸态组织,细化了β-Mg17Al12相,且β-Mg17Al12脆性相由连续网状分布逐渐变为离散状,同时晶界上出现了块状的Al11Yb3和点状Al6Mn4Yb化合物。当Yb含量为1.0%时,该合金具有最佳的综合力学性能,抗拉强度达到194 MPa,比AZ91D提高22%,伸长率和硬度也随着Yb含量的增加而提高。研究表明,重稀土元素Yb在镁合金中显示出与轻稀土更相近的作用效果。

The synergistic extraction of rare earths (La, Nd, Gd, Y and Yb) with a mixture of 2-ethylhexyl 2-ethylhexylphosphonate (EHEHPA) (HA) and trialkylphosphine oxide (Cyanex 923) (B) from a hydrochloride medium was investigated. The mixed system significantly enhances the extraction efficiency for lighter lanthanides and the synergistic enhancement coefficients for La (4.52), Nd (3.35), Gd (2.08), Y (1.31) and Yb (1.08) decrease with decreasing ionic radius of the rare earths. The extraction equilibrium of La, Nd and Gd indicate that La and Nd were extracted as MA(3)(.)B, whereas Gd was extracted as Gd(OH)A(2)(HA)(2)B-.. The equilibrium constants, thermodynamic functions such as Delta G, Delta H and Delta S and formation constants of the extracted species were determined. The stripping properties were also studied.

Fused in sarcoma (FUS), also called translocated in liposarcoma (TLS), is a ubiquitously expressed DNA/RNA binding protein belonging to the TET family and predominantly localized in the nucleus. FUS is proposed to be involved in various RNA metabolic pathways including transcription regulation, nucleo-cytosolic RNA transport, microRNA processing or pre-mRNA splicing [1]. Mutations in the FUS gene were identified in patients with familial amyotrophic lateral sclerosis (ALS) type 6 and sporadic ALS [2, 3]. ALS, also termed Lou Gehrig's disease, is a fatal adult-onset neurodegenerative disease affecting upper and lower motor neurons in the brain and spinal cord. There is increasing evidence supporting the hypothesis that FUS might play an important role in pre-mRNA splicing regulation. Several splicing factors were identified to associate with FUS including hnRNPA2 and C1/C2 [4], Y-box binding protein 1 (YB-1) [5] and serine arginine (SR) proteins (SC35 and TASR) [6]. Additionally, FUS was identified as a constituent of human spliceosomal complexes [1]. Our recent results indicate that FUS has increased affinity for certain but not all snRNPs of the minor and major spliceosome. Furthermore, in vitro studies revealed that FUS directly interacts with a factor specific for one of those snRNPs. These findings might uncover the molecular mechanism by which FUS regulates splicing and could explain previously observed effects of FUS on the splicing of the adenovirus E1A minigene [7] and changes in splicing caused by ALS associated FUS mutations. O Complexo Rio Capivari (CRC) é constituído por ortognaisses migmatíticos de composições graníticas a tonalíticas e anfibolitos subordinados (magmas toleíticos) em lascas tectônicas no Terreno Embu. As composições dos gnaisses do CRC são predominantemente cálcio-alcalinas a álcali-cálcicas. Idades U-Pb em núcleos de zircão com zoneamento oscilatório indicam cristalização magmática dos protólitos em três períodos principais 2.4, 2.2-2.1 e 2.0 Ga. Idades metamórficas foram reconhecidas em bordas de zircão totalmente escuras nas imagens de catodoluminescência e variam entre 620-590 Ma. A suíte sideriana (2.4 Ga) apresenta caráter juvenil, como evidenciado pelos valores positivos de \'\'épsilon\'\'IND.Nd\' (+3.8) e \'\'épsilon\'\'IND.Hf\' (+0.3 a +4.8) e pela ausência de núcleos de zircão herdado, comumente encontrados em rochas que sofreram retrabalhamento crustal. A suíte de idades riacianas (2.2-2.1 Ga) apresenta idades modelos TDM arqueanas (2.6-3.3 Ga), valores negativos de \'\'épsilon\'\'IND.Nd\' (-12.0 a -4.0) e negativos a levemente positivos de \'\'épsilon\'\'IND.Hf\' (-7.8 a +0.5). Portanto, tais rochas derivam de retrabalhamento de reservatórios crustais antigos. A suíte de idade orosiriana (2.0 Ga) apresenta fontes mais antigas e retrabalhadas com valores altamente negativos de \'\'épsilon\'\'IND.Nd\' (-10.4) e \'\'épsilon\'\'IND.Hf\' (-1.2 a -13.6), sugerindo prolongada residência crustal com idades modelo \'T IND.DM\' e \'T IND.Hf\' >3.3 Ga. As assinaturas de elementos traços em rocha total e a química de zircão sugerem fontes máficas para o gnaisse sideriano. Reservatórios de crosta média, mas de profundidades variáveis, parecem ser a principal fonte dos gnaisses riacianos e orosirianos. Análises em diagramas tectônicos discriminantes baseados em elementos traços de rocha total com elevadas razões \'La/Yb IND.(N)\' (>10), Nb/Yb (>2) e Th/Yb (>1), somados aos valores de \'Y IND.2\'\'O IND.3\' (<3000 ppm), U/Yb (>0.5) e Nb/Yb (0.01-0.10) da química de zircão, sugerem que ambas as suítes de idades foram geradas em ambientes de arco magmático continental, mas com um gap de 200-300 Ma entre o gnaisse sideriano e os gnaisses riacianos sem dados ou informações geológicas. Perfis multielementos (elementos traços) comparativos entre representação de amostras típicas de arco continental associado à subducção de crosta oceânica (margem andina) e amostras de arcos de ilha (Ilhas Mariana) confirmam afinidade com ambiente de arco continental para o CRC, associado à subducção de placa oceânica, principalmente para o gnaisse sideriano. Apesar de pouco representativo, devido ao número de amostras (n=1), uma acresção juvenil em 2.4 Ga colabora para uma dinâmica contínua da evolução da crosta continental. O papel desempenhado pelo CRC na evolução geral do Terreno Embu permanece enigmático. Os dados isotópicos de \'\'épsilon\'\'IND.Nd(590)\' e \'ANTPOT.87 Sr\'/\'ANTPOT.88 Sr IND.(i)\' do CRC (-27.3 a -19.7 e 0.704 a 0.722, respectivamente) indicam evolução temporal não compatível com o requerido para as fontes dos granitos ediacaranos do Terreno Embu, que exigem a participação de reservatórios mais primitivos (\'\'épsilon\'\'IND.Nd(590)\' -13 a -7) e empobrecidos em Rb (\'ANTPOT.87 Sr\'/\'ANTPOT.88 Sr IND.(i)\' \'\'QUASE IGUAL A\' 0,710).

To study the phase relations in the Bi-2212 and Yb2O3 system, Bi2Sr2Ca1-xYbxCu 2Oy thick films are prepared by partial melt processing via an intermediate reaction between Bi-2212 and Yb2O3. When Bi-2212 and Yb2O3 are partially melted and then slowly cooled, solid solutions of Bi2Sr2Ca 1-xYbxCu2Oy, form by reactions between liquid and solid phases which contain Yb. Following these reactions, Ca is partially replaced in Bi-2212 matrix and participates in the formation of secondary phases, such as Bi-free, (Ca, Sr)Ox and CaO. Variation of the Bi-2212-Yb2O3 ratios and processing parameters changes the balance between the phases and leads to different Yb:Ca ratios in the Bi-2212 matrix of processed thick films. When the partial melting process is optimized for each sample to minimize the growth of secondary phases, x = 0.42-0.46 for the samples prepared at pO2 = 0.01 atm, x = 0.24-0.29 for the samples prepared at pO2 = 0.21 atm, x = 0.18-0.23 for the samples prepared at pO2 = 0.99 atm are obtained regardless to the starting compositions. It is found that superconducting properties of Bi 2Sr2Ca1-xYbxCu2O y thick films strongly depend on the processing conditions, because the conditions result in different Yb content in the Bi-2212 matrix and the volume fraction of the secondary phases. The highest Tc(0) of 77, 90 and 91 K were obtained for the samples processed at 0.01, 0.21 and 0.99 atm of O2, respectively.

We apply our technique of using a Rb-stabilized ring-cavity resonator to measure the frequencies of various spectral components in the 555.8-nm 1S0-->3P1 line of Yb. We determine the isotope shifts with 60 kHz precision, which is an order-of-magnitude improvement over the best previous measurement on this line. There are two overlapping transitions, 171Yb(1/2-->3/2) and 173Yb(5/2-->3/2), which we resolve by applying a magnetic field. We thus obtain the hyperfine constants in the 3P1 state of the odd isotopes with a significantly improved precision. Knowledge of isotope shifts and hyperfine structure should prove useful for high-precision calculations in Yb necessary to interpret ongoing experiments testing parity and time-reversal symmetry violation in the laws of physics.

We investigate the laser actions of 5at.% Yb:Gd2xY2(1-x)SiO5 (Yb:GYSO; x = 0.1) crystals with different cutting directions, parallel and vertical to the growth axis. Our results show that the cutting direction of the sample plays an astonished role in the laser operation. The sample cut vertically to the growth axis possesses the favourable lasing characteristics. Its output power reaches 3.13W at 1060nm with a slope efficiency of 44.68% when the absorbed pump power is 8.9 W. In contrast, the sample cut parallel reaches only 1.65 W at 1044 nm with a slope elLiciency of 33.76% with absorbed pump power of 7.99 W. The absorption and emission spectra of the two samples are examined and the merit factor M is calculated. Our analysis is in agreement well with the experimental results. The wavelength tuning range of the superior sample covers from 1013.68 nm to 1084.82 nm.
