La industria de la producción de camarón es una de las industrias acuícolas que se encuentra en más crecimiento en la actualidad. Los estudios para encontrar marcadores genéticos son muy efectivos para la mejora de sus propiedades y de gran interés para los productores de camarón. En este trabajo se utilizaron seis individuos de una población de Litopenaeus vannamei, donde se encontraron cuatro polimorfismos de nucleótido único (SNPs) en el gen 5HT1R (5-hidroxitriptamina receptor1) y un SNP en el gen STAT (transductor de señal y activador de la transcripción). Sin embargo, el polimorfismo en el gen STAT resultó ser homocigoto en una población diferente utilizada para análisis de asociación. Los presentes análisis revelaron que el alelo C, en dos polimorfismos SNP (C109T y C395G) del gen 5HT1R, tiende a estar asociado con el aumento del peso corporal. Consideramos que hay necesidad de hacer nuevos estudios utilizando una muestra más amplia y diversa de la población en cuestión.

The marine bioprocessing industry offers great potential to utilize byproducts for fish meal replacement in aquafeeds. Jumbo squid is an important fishery commodity in Mexico, but only the mantle is marketed. Head, fins, guts and tentacles are discarded in spite of being protein-rich byproducts. This study evaluated the use of two jumbo squid byproduct hydrolysates obtained by acid-enzymatic hydrolysis (AEH) and by autohydrolysis (AH) as ingredients in practical diets for shrimp. The hydrolysates were included at levels of 2.5 and 5.0% of the diet dry weight in four practical diets, including a control diet without hydrolysate. Shrimp growth and survival were not significantly affected by the dietary treatments. Postharvest quality of abdominal muscle was evaluated in terms of proximate composition and sensory evaluation. Significantly higher crude protein was observed in the muscle of shrimp fed the highest hydrolysate levels, AH 5% (204.8 g kg- 1) or AEH 5% (201.3 g kg- 1). Sensory analysis of cooked muscle showed significant differences for all variables evaluated: color, odor, flavor, and firmness. It was concluded that Jumbo squid byproducts can be successfully processed by autohydrolysis or acid-enzymatic hydrolysis, and that up to 5.0% of the hydrolysates can be incorporated into shrimp diets without affecting growth or survival.

The immunostimulatory effect of an alkali insoluble glucan extracted from marine yeast isolate Candida sake S165 was tested in Fenneropenaeus indicus. Post larvae (PL) of F. indicus, fed glucan incorporated diet at varying concentrations (0.05, 0.1, 0.2, 0.3, 0.4 g glucan/100 g feed) for 21 days were challenged orally with white spot syndrome virus (WSSV). Maximum survival was observed in PL fed the 0.2% glucan incorporated diet. Subsequently the feed incorporated with 0.2% glucan was fed to F. indicus post larvae at different feeding intervals, i.e. daily, once every two days, once every five days, once every seven days and once every ten days. After 40 days, the prawns were challenged orally with WSSV and post challenge survival was recorded. Shrimp feed containing 0.2% glucan when administered once every seven days gave maximum survival. This was supported by haematological data obtained from adult F. indicus, i.e. total haemocyte count, phenoloxidase activity and nitroblue tetrazolium reduction (NBT). The present observation confirms the importance of dose and frequency of administration of immunostimulants in shrimp health management

E¡ect of an extraction method on the structure of glucan and its immunostimulatory response in Fenneropenaeus indicus was investigated. Here we extracted alkali insoluble glucan (AIG) and alkali soluble glucan (ASG) from a ¢lamentous fungi Acremonium diospyri following alkali^acid hydrolysis and the sodium hypochlorite oxidation and dimethyl sulphoxide extraction method respectively. Structural analysis showed that 85% of glucan in AIG was a (1 !3)-b-D-glucan and it increased the prophenoloxidase and reactive oxygen intermediate activity when administered to F. indicus. On the other hand, ASG, which contained 93% (1 !3)-a-glucan, did not induce signi¢cant immune response in shrimp. Here we report that the di¡erence in immunostimulatory potential between AIG and ASG is due to the di¡erence in the percentage of (1 !3)-b-D-glucans present in each preparation, which varies with the method of extraction employed. Also our observations suggest that glucan can be used as a potential immunostimulant to shrimp, provided it contains (1 !3)-b-D-glucan as the major fraction.

A virus, tentatively identified as reo-like, occurred concurrently with experimentally-induced Baculovirus penaei (BP) infection in cultured white shrimp larvae Penaeus vannamei. Each shrimp with a reo-like viral infection also had a BP infection, but not all BP-infected shrimp had a reo-like infection. Both viruses occurred in the same tissues and occasionally withln the same cell. The reolike virus developed in epithelial cells of the anterior midgut and in reserve- and fibrillar-cells of the hepatopancreas. The paraspherical and non-enveloped reo-like virions (ca. 50 nm diam.) occurred as unordered aggregates in the cell cytoplasm. Their etiology has not been determined. Reo-like virions may have been introduced along with the BP virus, or, were latent and only manifested due to stress induced by the more pathogenic BP virus.

The aseptate gregarine Paraophloidina scolecoides n. sp. (Eugregarinorida: Lecudinidae) heavily infected the midgut of cultured larval and postlarval specimens of Penaeus vannamei from a commercial 'seed-production' facility in Texas, USA. It is morphologically similar to P. korotneffi and P. vibiliae, but it can be distinguished from them and from other members of the genus by having gamonts associated exclusively by lateral syzygy. Shrimp acquired the infection at the facility; nauplii did not show any evidence of infection, but protozoea, mysis, and postlarval shrimp had a prevalence and intensity of infection ranging from 56 to 80 % and 10 to >50 parasites, respectively. Infected shrimp removed from the facility to aquaria at another location lost their gamont infection within 7 days When voided from the gut, the gregarine disintegrated in seawater. Results suggest that P. vannamei is an accidental host, although a survey of representative members of the invertebrate fauna from the environment associated with the facility failed to discover other hosts. No link was established between infection and either the broodstock or the water or detritus from the nursery or broodstock tanks.

The relationship between energy reserves of the penaeid shrimp Penaeus vannamei and Baculovirus penaei, or BP, were investigated in a series of experiments using mysis stage or early postlarval shrimp. Pre-exposure and post-exposure levels of protein and triacylgycerol (TAG) were determined. The effect of pre-exposure protein and TAG levels on susceptibility to BP infections was also investigated by starving a group of shrimp immediately prior to BP exposure. There was no consistent relationship between either pre-exposure or post-exposure protein levels and the percent of shrimp developing patent BP infections. There was, however, a significant positive correlation between TAG levels immediately prior to viral exposure and prevalence of infection 72 h later. Experimental reduction of TAG reserves prior to BP exposure delayed the development of a patent infection. In some, but not all, experiments there was a significant reduction in TAG levels of infected compared with uninfected shrimp 72 h post-exposure. The effect of patent BP infections on host TAG levels was subordinate to fluctuations in TAG content associated with the ontogeny of the hepatopancreas. Results of this study support histological observations that shrimp lipid levels can be altered by baculovirus infections. Furthermore, high levels of energy reserves in the form of TAG are associated with increased susceptibility to BP infection in larval and postlarval shrimp.

A white shrimp from Galveston, Texas, is the first reported case of a crustacean internally infected by a diatom. Even though more than one species occurred in debris on and between gill filaments, only individuals of Amphora sp. occurred within gills. To determine if a related diatom would easily reproduce within the shrimp and cause. a host response similar to that observed, we injected cultured specimens of A. coffaeformis into white shrimp. Under the experimental conditions, individuals of that species did not divide, but they elicited an extensive melanistic host-response.

Aquafeed production faces global issues related to availability of feed ingredients. Feed manufacturers require greater flexibility in order to develop nutritional and cost-effective formulations that take into account nutrient content and availability of ingredients. The search for appropriate ingredients requires detailed screening of their potential nutritional value and variability at the industrial level. In vitro digestion of feedstuffs by enzymes extracted from the target species has been correlated with apparent protein digestibility (APD) in fish and shrimp species. The present study verified the relationship between APD and in vitro degree of protein hydrolysis (DH) with Litopenaeus vannamei hepatopancreas enzymes in several different ingredients (n = 26): blood meals, casein, corn gluten meal, crab meal, distiller`s dried grains with solubles, feather meal, fish meals, gelatin, krill meals, poultry by-product meal, soybean meals, squid meals and wheat gluten. The relationship between APD and DH was further verified in diets formulated with these ingredients at 30% inclusion into a reference diet. APD was determined in vivo (30.1 +/- 0.5 degrees C, 32.2 +/- 0.4%.) with juvenile L vannamei (9 to 12 g) after placement of test ingredients into a reference diet (35 g kg(-1) CP: 8.03 g kg(-1) lipid; 2.01 kcal g(-1)) with chromic oxide as the inert marker. In vitro DH was assessed in ingredients and diets with standardized hepatopancreas enzymes extracted from pond-reared shrimp. The DH of ingredients was determined under different assay conditions to check for the most suitable in vitro protocol for APD prediction: different batches of enzyme extracts (HPf5 or HPf6), temperatures (25 or 30 degrees C) and enzyme activity (azocasein): crude protein ratios (4 U: 80 mg CP or 4 U: 40 mg CP). DH was not affected by ingredient proximate composition. APD was significantly correlated to DH in regressions considering either ingredients or diets. The relationships between APD and DH of the ingredients could be suitably adjusted to a Rational Function (y = (a + bx)/(1 + cx + dx2), n = 26. Best in vitro APD predictions were obtained at 25 degrees C, 4 U: 80 mg CP both for ingredients (R(2) = 0.86: P = 0.001) and test diets (R(2) = 0.96; P = 0.007). The regression model including all 26 ingredients generated higher prediction residuals (i.e., predicted APD - determined APD) for corn gluten meal, feather meal. poultry by-product meal and krill flour. The remaining test ingredients presented mean prediction residuals of 3.5 points. A model including only ingredients with APD>80% showed higher prediction precision (R(2) = 0.98: P = 0.000004; n = 20) with average residual of 1.8 points. Predictive models including only ingredients from the same origin (e.g., marine-based, R(2) = 0.98; P = 0.033) also displayed low residuals. Since in vitro techniques have been usually validated through regressions against in vivo APD, the DH predictive capacity may depend on the consistency of the in vivo methodology. Regressions between APD and DH suggested a close relationship between peptide bond breakage by hepatopancreas digestive proteases and the apparent nitrogen assimilation in shrimp, and this may be a useful tool to provide rapid nutritional information. Dry matter, energy, crude protein and amino acid apparent digestibility coefficients (ADCs) were determined in white shrimp juveniles for six wheat products: hard red winter whole grain meal (HWG), Rayon whole grain meal (RWG), Durum whole grain meal (DWG), hard red winter clear flour (HCF), mixed wheat 2nd clear flour (MCF) and semolina (S). The test diets included 30% of the test ingredients and 70% of a ground commercial diet supplemented with 1% chromic oxide and 1% sodium alginate. Amino acid contents in the ingredients, diets and feces were analyzed, and digestibility was determined by difference in order to minimize the impact of endogenous amino acid losses; crude protein and amino acids ADCswere adjusted for dietary preprandial losses in seawater. In general, nutrients digestibility was far higher in the wheat products than in the fish meal-based reference diet. Drymatter and crude proteinADCswere not statistically different amongwheat products (from84 to 96% and from88 to 107% respectively). Energy ADCs were significantly higher for clear flours (96% for HCF and MCF) than forwhole grainmeals and S (from83 to 86%). Total amino acids (TAA) and essentialamino acids (EAA) ADCs, once adjusted for preprandial leaching fromthe experimental diets, ranged from81 to 89% and from 58 to 81% respectively, and were statistically comparable among wheat products. Low Thr ADCs appear as a common feature of the amino acids digestibility profiles for whole grain meals, clear flours, or semolina.

Two feeding trials were carried out to evaluate the effect of diets containing corn or peanut grains naturally contaminated with aflatoxins on the growth, feed intake, survival, and histological response of the white shrimp Litopenaeus vannamei. In trial 1, four experimental diets were formulated to contain 0, 500, 1000, and 2000 g kg–1 of total aflatoxins (TA) and fed to L. vannamei juveniles for 28 days. In trial 2, six experimental diets were formulated to contain 0, 10, 20, 40, 60, and 120 g kg–1 TA and fed to L. vannamei juveniles for 64 days. Feed intake and weight gain were significantly affected by the presence of aflatoxins from naturally contaminated grains. Feed conversion rate increased significantly from a level of inclusion of 60 g kg–1. Survival was significantly reduced only for shrimp fed diets supplemented with 1000 and 2000 g kg–1 TA. Shrimp exposed to higher aflatoxin inclusion levels presented significantly lower lipid vacuole levels in R-cells (12–28%), lower B-cell activity, and lower mitotic E-cell activity. Tubular epithelial atrophy increased from the inclusion level of 20 g kg–1. Hepatopancreatocyte sloughing was significantly higher in shrimp fed diets supplemented with 1000 and 2000 g kg–1 TA. It is worth noting that shrimp fed 40 g kg–1 TA presented a high hepatopancreatocyte sloughing coefficient. Based on these results we conclude that the presence of aflatoxins, even at low levels, reduces feed intake and weight gain, and alters the cells of the hepatopancreas. RESUMEN. Se llevaron a cabo dos bioensayos para evaluar el efecto de granos de maíz y maní contaminados naturalmente con aflatoxinas sobre el crecimiento, consumo de alimento, supervivencia y daños histológicos de juveniles del camarón blanco Litopenaeus vannamei. Para el bioensayo 1, se formularon cuatro dietas con 0, 500, 1000 y 2000 g kg–1 de aflatoxinas totales (AT) y se proporcionaron a juveniles de L. vannamei durante 28 días. Para el bioensayo 2, se formularon seis dietas con 0, 10, 20, 40, 60 y 120 g kg–1 AT y se proporcionaron a juveniles de L. vannamei durante 64 días. El consumo de alimento fue significativamente afectado por la presencia de aflatoxinas. La tasa de conversión alimenticia incrementó significativamente a partir de un nivel de inclusión de 60 g kg–1. La supervivencia fue significativamente reducida solamente en los camarones que fueron alimentados con las dietas suplementadas con 1000 y 2000 g kg–1 AT. Los camarones expuestos a los niveles de inclusión altos presentaron un menor nivel de vacuolas lipídicas en las células R (12–28%), y una menor actividad de las células B y de la actividad mitótica de las células E. La atrofia de los túbulos del epitelio se incrementó a partir de un nivel de inclusión de 20 g kg–1. La descamación de las células del hepatopáncreas fue significativamente mayor en camarones alimentados con las dietas suplementadas con 1000 y 2000 g kg–1 AT, mientras que para las dosis bajas no se observaron diferencias significativas, aunque en camarones alimentados a partir de 40 g kg–1 AT se observa un coeficiente de descamación alto. Con base en los resultados, se concluye que la presencia de aflatoxinas, incluso a niveles bajos, reduce el consumo de alimento y el aumento de peso y altera las células del hepatopáncreas.
