procera (pro) is a tall tomato (Solanum lycopersicum) mutant carrying a point mutation in the GRAS region of the gene encoding SlDELLA, a repressor in the gibberellin (GA) signaling pathway. Consistent with the SlDELLA loss of function, pro plants display a GA-constitutive response phenotype, mimicking wild-type plants treated with GA(3). The ovaries from both nonemasculated and emasculated pro flowers had very strong parthenocarpic capacity, associated with enhanced growth of preanthesis ovaries due to more and larger cells. pro parthenocarpy is facultative because seeded fruits were obtained by manual pollination. Most pro pistils had exserted stigmas, thus preventing self-pollination, similar to wild-type pistils treated with GA(3) or auxins. However, Style2.1, a gene responsible for long styles in noncultivated tomato, may not control the enhanced style elongation of pro pistils, because its expression was not higher in pro styles and did not increase upon GA(3) application. Interestingly, a high percentage of pro flowers had meristic alterations, with one additional petal, sepal, stamen, and carpel at each of the four whorls, respectively, thus unveiling a role of SlDELLA in flower organ development. Microarray analysis showed significant changes in the transcriptome of preanthesis pro ovaries compared with the wild type, indicating that the molecular mechanism underlying the parthenocarpic capacity of pro is complex and that it is mainly associated with changes in the expression of genes involved in GA and auxin pathways. Interestingly, it was found that GA activity modulates the expression of cell division and expansion genes and an auxin signaling gene (tomato AUXIN RESPONSE FACTOR7) during fruit-set.

Background: High-throughput molecular approaches for gene expression profiling, such as Serial Analysis of Gene Expression (SAGE), Massively Parallel Signature Sequencing (MPSS) or Sequencing-by-Synthesis (SBS) represent powerful techniques that provide global transcription profiles of different cell types through sequencing of short fragments of transcripts, denominated sequence tags. These techniques have improved our understanding about the relationships between these expression profiles and cellular phenotypes. Despite this, more reliable datasets are still necessary. In this work, we present a web-based tool named S3T: Score System for Sequence Tags, to index sequenced tags in accordance with their reliability. This is made through a series of evaluations based on a defined rule set. S3T allows the identification/selection of tags, considered more reliable for further gene expression analysis. Results: This methodology was applied to a public SAGE dataset. In order to compare data before and after filtering, a hierarchical clustering analysis was performed in samples from the same type of tissue, in distinct biological conditions, using these two datasets. Our results provide evidences suggesting that it is possible to find more congruous clusters after using S3T scoring system. Conclusion: These results substantiate the proposed application to generate more reliable data. This is a significant contribution for determination of global gene expression profiles. In recent decades, all over the world, competition in the electric power sector has deeply changed the way this sector’s agents play their roles. In most countries, electric process deregulation was conducted in stages, beginning with the clients of higher voltage levels and with larger electricity consumption, and later extended to all electrical consumers. The sector liberalization and the operation of competitive electricity markets were expected to lower prices and improve quality of service, leading to greater consumer satisfaction. Transmission and distribution remain noncompetitive business areas, due to the large infrastructure investments required. However, the industry has yet to clearly establish the best business model for transmission in a competitive environment. After generation, the electricity needs to be delivered to the electrical system nodes where demand requires it, taking into consideration transmission constraints and electrical losses. If the amount of power flowing through a certain line is close to or surpasses the safety limits, then cheap but distant generation might have to be replaced by more expensive closer generation to reduce the exceeded power flows. In a congested area, the optimal price of electricity rises to the marginal cost of the local generation or to the level needed to ration demand to the amount of available electricity. Even without congestion, some power will be lost in the transmission system through heat dissipation, so prices reflect that it is more expensive to supply electricity at the far end of a heavily loaded line than close to an electric power generation. Locational marginal pricing (LMP), resulting from bidding competition, represents electrical and economical values at nodes or in areas that may provide economical indicator signals to the market agents. This article proposes a data-mining-based methodology that helps characterize zonal prices in real power transmission networks. To test our methodology, we used an LMP database from the California Independent System Operator for 2009 to identify economical zones. (CAISO is a nonprofit public benefit corporation charged with operating the majority of California’s high-voltage wholesale power grid.) To group the buses into typical classes that represent a set of buses with the approximate LMP value, we used two-step and k-means clustering algorithms. By analyzing the various LMP components, our goal was to extract knowledge to support the ISO in investment and network-expansion planning.

Com a evolução da Internet houve uma consequente transformação da forma como os resultados são apresentados e das técnicas de visualização e apresentação de conteúdos. A presente dissertação é sobre visualização com nuvens de tags e foi a conjugação de diversos factores que originou a investigação sobre este tema. Uma nuvem de tags (em inglês: tag clouds) é uma concepção visual de um conjunto de tags isoladas com uma representação ilustrativa da sua importância, ou seja visualmente hierarquizadas e em que cada tag se hiperliga a um ou mais sítios Web ou documentos. São extremamente úteis para a navegação ou para a descoberta de informação genérica, podendo requerer menor carga cognitiva do utilizador durante consultas e pesquisas se alguns aspectos forem considerados na sua construção. Nesta dissertação discutem-se alguns dos factores visuais que podem contribuir para que as nuvens de tags sejam ferramentas efectivas e intuitivas para os utilizadores. O conceito de folksonomia está relacionado com nuvens de tags, permitindo categorizar as definições atribuídas a conteúdos recorrendo a palavras-chave (tags). Neste documento são ainda exploradas as suas vantagens e desvantagens. Discutem-se ainda alguns modos de visualização e apresentação de nuvens de tags, desde o tipo de ordenação ao algoritmo de geração da nuvem de tags. Aborda-se ainda o impacto do tipo de fonte e a utilização de cores monocromáticas ou coloridas que estas possam ter na visualização da nuvem. A própria tag também é categorizada e explorada ao nível do seu posicionamento na nuvem e a percepção que causa no utilizador. Sobre a visualização de nuvens de tags, refere-se ainda que as conclusões obtidas foram utilizadas num repositório de objectos educativos.

This work project was conducted under a Direct Research internship (DRI) that consists on an individual dissertation established on a given organization. DRI has a problem solving format to an empirical question to be addressed, «Which country has the highest potential for the next step of XY internationalization process? ». In order to achieve the project’s purpose, it was conducted a scanning process using a top-down approach over an initial list of nine countries given by XY. To do so it was developed an international scanning framework based on different domains and weights that allowed to achieve the top two countries with highest potential. After an in depth analysis over the final set, it was recommended Switzerland as the best country to make the next step of XY internationalization in Europe.

Plant membrane compartments and trafficking pathways are highly complex, and are often distinct from those of animals and fungi. Progress has been made in defining trafficking in plants using transient expression systems. However, many processes require a precise understanding of plant membrane trafficking in a developmental context, and in diverse, specialized cell types. These include defense responses to pathogens, regulation of transporter accumulation in plant nutrition or polar auxin transport in development. In all of these cases a central role is played by the endosomal membrane system, which, however, is the most divergent and ill-defined aspect of plant cell compartmentation. We have designed a new vector series, and have generated a large number of stably transformed plants expressing membrane protein fusions to spectrally distinct, fluorescent tags. We selected lines with distinct subcellular localization patterns, and stable, non-toxic expression. We demonstrate the power of this multicolor 'Wave' marker set for rapid, combinatorial analysis of plant cell membrane compartments, both in live-imaging and immunoelectron microscopy. Among other findings, our systematic co-localization analysis revealed that a class of plant Rab1-homologs has a much more extended localization than was previously assumed, and also localizes to trans-Golgi/endosomal compartments. Constructs that can be transformed into any genetic background or species, as well as seeds from transgenic Arabidopsis plants, will be freely available, and will promote rapid progress in diverse areas of plant cell biology.

Es defineix l'expansió general d'operadors com una combinació lineal de projectors i s'exposa la seva aplicació generalitzada al càlcul d'integrals moleculars. Com a exemple numèric, es fa l'aplicació al càlcul d'integrals de repulsió electrònica entre quatre funcions de tipus s centrades en punts diferents, i es mostren tant resultats del càlcul com la definició d'escalat respecte a un valor de referència, que facilitarà el procés d'optimització de l'expansió per uns paràmetres arbitraris. The distribution limits of Crocidura russula (Hermann, 1780) and C. leucodon (Hermann, 1780) were investigated during an interval of 25 years in the bottom of the Rhone valley above Lake Geneva, Switzerland (total data set: 105 spatio-temporal occurrences, 1137 shrews). In 1975, the contact zone between the two species was situated in the region of Martigny. In 1999/2000, new sampling revealed three results: (1) The contact zone showed an upward shift of about 25 km. (2) In the expanded range of C. russula, the resident species has totally disappeared (confirmed by owl pellets analysis). (3) This demonstrates a dominance of C, russula over C. leucodon. Three hypotheses which may explain the range expansion of C. russula were evaluated: (1) habitat modification favouring linear dispersal due to the construction of a highway; (2) temporal event favoured by climate fluctuations, or (3) ongoing postglacial colonisation of Europe. Hypothesis 1 was rejected, because the progression of the shrews anticipated the construction. Hypothesis 3 received only weak support because range limits of C. russula in the region of Nice have been stable for thousands of years. Therefore hypothesis 2, admitting that ongoing climate change has facilitated range expansion, is the most probable.

The influence of the basis set size and the correlation energy in the static electrical properties of the CO molecule is assessed. In particular, we have studied both the nuclear relaxation and the vibrational contributions to the static molecular electrical properties, the vibrational Stark effect (VSE) and the vibrational intensity effect (VIE). From a mathematical point of view, when a static and uniform electric field is applied to a molecule, the energy of this system can be expressed in terms of a double power series with respect to the bond length and to the field strength. From the power series expansion of the potential energy, field-dependent expressions for the equilibrium geometry, for the potential energy and for the force constant are obtained. The nuclear relaxation and vibrational contributions to the molecular electrical properties are analyzed in terms of the derivatives of the electronic molecular properties. In general, the results presented show that accurate inclusion of the correlation energy and large basis sets are needed to calculate the molecular electrical properties and their derivatives with respect to either nuclear displacements or/and field strength. With respect to experimental data, the calculated power series coefficients are overestimated by the SCF, CISD, and QCISD methods. On the contrary, perturbation methods (MP2 and MP4) tend to underestimate them. In average and using the 6-311 + G(3df) basis set and for the CO molecule, the nuclear relaxation and the vibrational contributions to the molecular electrical properties amount to 11.7%, 3.3%, and 69.7% of the purely electronic μ, α, and β values, respectively

La diversification des résultats de recherche (DRR) vise à sélectionner divers documents à partir des résultats de recherche afin de couvrir autant d’intentions que possible. Dans les approches existantes, on suppose que les résultats initiaux sont suffisamment diversifiés et couvrent bien les aspects de la requête. Or, on observe souvent que les résultats initiaux n’arrivent pas à couvrir certains aspects. Dans cette thèse, nous proposons une nouvelle approche de DRR qui consiste à diversifier l’expansion de requête (DER) afin d’avoir une meilleure couverture des aspects. Les termes d’expansion sont sélectionnés à partir d’une ou de plusieurs ressource(s) suivant le principe de pertinence marginale maximale. Dans notre première contribution, nous proposons une méthode pour DER au niveau des termes où la similarité entre les termes est mesurée superficiellement à l’aide des ressources. Quand plusieurs ressources sont utilisées pour DER, elles ont été uniformément combinées dans la littérature, ce qui permet d’ignorer la contribution individuelle de chaque ressource par rapport à la requête. Dans la seconde contribution de cette thèse, nous proposons une nouvelle méthode de pondération de ressources selon la requête. Notre méthode utilise un ensemble de caractéristiques qui sont intégrées à un modèle de régression linéaire, et génère à partir de chaque ressource un nombre de termes d’expansion proportionnellement au poids de cette ressource. Les méthodes proposées pour DER se concentrent sur l’élimination de la redondance entre les termes d’expansion sans se soucier si les termes sélectionnés couvrent effectivement les différents aspects de la requête. Pour pallier à cet inconvénient, nous introduisons dans la troisième contribution de cette thèse une nouvelle méthode pour DER au niveau des aspects. Notre méthode est entraînée de façon supervisée selon le principe que les termes reliés doivent correspondre au même aspect. Cette méthode permet de sélectionner des termes d’expansion à un niveau sémantique latent afin de couvrir autant que possible différents aspects de la requête. De plus, cette méthode autorise l’intégration de plusieurs ressources afin de suggérer des termes d’expansion, et supporte l’intégration de plusieurs contraintes telles que la contrainte de dispersion. Nous évaluons nos méthodes à l’aide des données de ClueWeb09B et de trois collections de requêtes de TRECWeb track et montrons l’utilité de nos approches par rapport aux méthodes existantes.
