Heating and cooling temperature jumps (T-jumps) were performed using a newly developed technique to trigger unfolding and refolding of wild-type ribonuclease A and a tryptophan-containing variant (Y115W). From the linear Arrhenius plots of the microscopic folding and unfolding rate constants, activation enthalpy (ΔH#), and activation entropy (ΔS#) were determined to characterize the kinetic transition states (TS) for the unfolding and refolding reactions. The single TS of the wild-type protein was split into three for the Y115W variant. Two of these transition states, TS1 and TS2, characterize a slow kinetic phase, and one, TS3, a fast phase. Heating T-jumps induced protein unfolding via TS2 and TS3; cooling T-jumps induced refolding via TS1 and TS3. The observed speed of the fast phase increased at lower temperature, due to a strongly negative ΔH# of the folding-rate constant. The results are consistent with a path-dependent protein folding/unfolding mechanism. TS1 and TS2 are likely to reflect X-Pro114 isomerization in the folded and unfolded protein, respectively, and TS3 the local conformational change of the β-hairpin comprising Trp115. We compare the characteristics of synthetic European droughts generated by the HiGEM1 coupled climate model run with present day atmospheric composition with observed drought events extracted from the CRU TS3 data set. The results demonstrate consistency in both the rate of drought occurrence and the spatiotemporal structure of the events. Estimates of the probability density functions for event area, duration and severity are shown to be similar with confidence > 90%. Encouragingly, HiGEM is shown to replicate the extreme tails of the observed distributions and thus the most damaging European drought events. The soil moisture state is shown to play an important role in drought development. Once a large-scale drought has been initiated it is found to be 50% more likely to continue if the local soil moisture is below the 40th percentile. In response to increased concentrations of atmospheric CO2, the modelled droughts are found to increase in duration, area and severity. The drought response can be largely attributed to temperature driven changes in relative humidity. 1 HiGEM is based on the latest climate configuration of the Met Office Hadley Centre Unified Model (HadGEM1) with the horizontal resolution increased to 1.25 x 0.83 degrees in longitude and latitude in the atmosphere and 1/3 x 1/3 degrees in the ocean.

Sodium channel toxins from sea anemones are employed as tools for dissecting the biophysical properties of inactivation in voltage-gated sodium channels. Cangitoxin (CGTX) is a peptide containing 48 amino acid residues and was formerly purified from Bunodosoma cangicum. Nevertheless, previous works reporting, the isolation procedures for such peptide from B. cangicum secretions are controversial and may lead to incorrect information. In this paper, we report a simple and rapid procedure, consisting of two chromatographic steps, in order to obtain a CGTX analog directly from sea anemone venom. We also report a substitution of N16D in this peptide sample and the co-elution of an inseparable minor isoform presenting the R14H substitution. Peptides are named as CGTX-II and CGTX-III, and their effects over Nav1.1 channels in patch clamp experiments are demonstrated. (c) 2008 Elsevier Ltd. All rights reserved.

The purpose of this research was to elucidate the mechanism of assembly of retroviruses, specifically of murine leukemia virus, as studied through the treatment of virus-infected cells with interferon and through the use of temperature sensitive (ts) mutants. Our studies have shown a rapid and specific association of Rauscher murine leukemia virus (R-MuLV) precursor polyprotein Pr65('gag) with cytoskeletal elements in infected mouse fibroblasts. The Pr65('gag) associated with Nonidet P-40 (NP40)-insoluble cytoskeletal structures appeared to be subphosphorylated in comparison to NP40-soluble Pr65('gag). The association of Pr65('gag) with skeletal elements could be disrupted by extraction of the cytoskeleton with sodium deoxycholate, an ionic detergent. Both the skeleton-associated Pr65('gag) and its NP40-soluble counterpart were labeled with {('3)H}-palmitate, indicating their probable association with lipids presumably in the plasma membrane. Pr65('gag) molecules bound to skeletal elements in the infected cell appeared to be more stable to proteolytic processing than NP40-soluble Pr65('gag). Our studies with certain ts mutants of murine leukemia virus, defective in virus assembly, including Mo-MuLV ts3 and R-MuLV ts17, ts24, ts25 and ts26, have shown that virions released at 39(DEGREES)C (nonpermissive temperature) had high levels of uncleaved Pr65('gag) relative to that seen in virions released at 33(DEGREES)C (permissive temperature). Examination of cell extracts revealed that Pr54('gag) was more stable to processing at 39(DEGREES)C than at 33(DEGREES)C, whereas the 'env' and glycosylated 'gag' proteins were processed to the same extent at both temperatures. Detergent extraction of pulse-labeled cells to generate an NP40-insoluble cytoskeleton-enriched fraction showed that in ts3-, ts17- and ts24-infected cells, Pr65('gag) accumulated in the cytoskeleton-enriched fraction. In contrast, cells infected with ts25 or ts26 showed no preferential localization of Pr65('gag) in the cytoskeleton in a short pulse, but instead, Pr65('gag) accumulated in both the NP40-soluble and -insoluble fractions during a chase-incubation. The association of Pr65('gag) with cytoskeletal elements in the cell was neither increased nor decreased by blocking virus assembly and release with interferon. Based on these and other results, we have proposed a model for the active role of cytoskeleton-associated Pr65('gag) in retrovirus assembly.^

Siberian boreal forests are expected to expand northwards in the course of global warming. However, processes of the treeline ecotone transition, as well astiming and related climate feedbacks are still not understood. Here, we present 'Larix Vegetation Simulator' LAVESI, an individual-based spatially-explicit model that can simulate Larix gmelinii (RUPR.) RUPR. stand dynamics in an attempt to improve our understanding about past and future treeline movements under changing climates. The relevant processes (growth, seed production and dispersal, establishment and mortality) are incorporated and adjusted to observation data mainly gained from the literature. Results of a local sensitivity analysis support the robustness of the model's parameterization by giving relatively small sensitivity values. We tested the model by simulating tree stands under modern climate across the whole Taymyr Peninsula, north-central Siberia (c. 64-80° N; 92-119° E). We find tree densities similar to observed forests in the northern to mid-treeline areas, but densities are overestimated in the southern parts of the simulated region. Finally, from a temperature-forcing experiment, we detect that the responses of tree stands lag the hypothetical warming by several decades, until the end of 21st century. With our simulation experiments we demonstrate that the newly-developed model captures the dynamics of the Siberian latitudinal treeline.

Diagnostic techniques based on PCR have two major problems: false-positive reactions due to contamination with DNA fragments from previous PCRs (amplicons) and false-negative reactions caused by inhibitors that interfere with the PCR. We have improved our previously reported PCR based on the amplification of a fragment of the Mycobacterium tuberculosis complex-specific insertion element IS6110 with respect to both problems. False-positive reactions caused by amplicon contamination were prevented by the use of uracil-N-glycosylase and dUTP instead of dTTP. We selected a new set of primers outside the region spanned by the formerly used primers to avoid false-positive reactions caused by dTTP-containing amplicons still present in the laboratory. With this new primer set, 16 copies of the IS6110 insertion element, the equivalent of two bacteria, could be amplified 10(10) times in 40 cycles, resulting in a mean efficiency of 77% per cycle. To detect the presence of inhibitors of the Taq polymerase, which may cause false-negative reactions, part of each sample was spiked with M. tuberculosis DNA. The DNA purification method using guanidinium thiocyanate and diatoms effectively removed most or all inhibitors of the PCR. However, this was not suitable for blood samples, for which we developed a proteinase K treatment followed by phenol-chloroform extraction. This method permitted detection of 20 M. tuberculosis bacteria per ml of whole blood. Various laboratory procedures were introduced to reduce failure or inhibition of PCR and avoid DNA cross contamination. We have tested 218 different clinical specimens obtained from patients suspected of having tuberculosis. The samples included sputum (n=145), tissue biopsy samples (n=25), cerebrospinal fluid (n=15), blood (n=14), pleural fluid (n=9), feces, (n=7), fluid from fistulae (n=2), and pus from a wound (n=1). The results obtained by PCR were consistent with those obtained with culture, which is the "gold standard." We demonstrate that PCR is a useful technique for the rapid diagnosis of tuberculosis at various sites.

A utilização de revestimentos poliméricos em rolhas de cortiça natural é uma prática recorrente na indústria de produção de rolhas. A sua aplicação tem como principal objetivo a melhoria da qualidade visual das rolhas. Contudo, este tratamento altera também as suas caraterísticas de molhabilidade, pelo que é importante otimizar os processos de revestimento de superfície. O presente trabalho teve como objetivo principal a averiguação da influência de um revestimento colorido e de diferentes tratamentos de superfície no comportamento da rolha em garrafa. Foram utilizadas amostras, cedidas pela Cork Supply Portugal, provenientes de quatro branqueamentos distintos (LVL, CSP, CPI e SIL). Foram estudadas amostras com e sem revestimento colorido e, todas elas, tratadas superficialmente com quatro tipos de tratamento (TS1, TS2, TS3 e TS4), sendo as quantidades de parafina e silicone administradas todas diferentes. Para observar o efeito do revestimento colorido e dos tratamentos de superfície nas rolhas de cortiça, procedeu-se a testes para determinação de capilaridade, inserções e forças de extração, simulando as condições de engarrafamento/extração do cliente, de forma a verificar a conformidade dos resultados. Todos estes testes procederam-se nas instalações da CSP. Foram também efetuados testes de molhabilidade pela determinação de ângulos de contacto para três diferentes solventes, nomeadamente água, formamida e diiodometano, e obtidos os Wetting Envelopes, pelo método de OWRK, de forma a prever a molhabilidade das várias amostras. Os procedimentos experimentais descritos efetuaram-se nos laboratórios da Universidade de Aveiro. Aliando as análises aos vários testes efetuados, concluiu-se que o revestimento colorido tem um possível efeito favorecedor nas propriedades mecânicas das rolhas de cortiça, possui maior recetividade aos agentes lubrificantes e maior molhabilidade. Relativamente aos tratamentos de superfície, o melhor, tendo em conta os fatores de molhabilidade, foi o TS4, proporcionando melhor vedação ao vinho. Já a respeito da lubrificação, verificou-se melhores resultados para os tratamentos TS1 e TS2. A lavação SIL apresenta menor afinidade aos agentes vedantes e a CSP possui maior recetibilidade aos agentes lubrificantes.
