Cystic fibrosis (CF) lung disease is characterized by infection with Pseudomonas aeruginosa and a sustained accumulation of neutrophils. In this study, we analyzed 1) the expression of MyD88-dependent TLRs on circulating and airway neutrophils in P. aeruginosa-infected CF patients, P. aeruginosa-infected non-CF bronchiectasis patients, and noninfected healthy control subjects and 2) studied the regulation of TLR expression and functionality on neutrophils in vitro. TLR2, TLR4, TLR5, and TLR9 expression was increased on airway neutrophils compared with circulating neutrophils in CF and bronchiectasis patients. On airway neutrophils, TLR5 was the only TLR that was significantly higher expressed in CF patients compared with bronchiectasis patients and healthy controls. Studies using confocal microscopy and flow cytometry revealed that TLR5 was stored intracellularly in neutrophils and was mobilized to the cell surface in a protein synthesis-independent manner through protein kinase C activation or after stimulation with TLR ligands and cytokines characteristic of the CF airway microenvironment. The most potent stimulator of TLR5 expression was the bacterial lipoprotein Pam(3)CSK(4). Ab-blocking experiments revealed that the effect of Pam(3)CSK(4) was mediated through cooperation of TLR1 and TLR2 signaling. TLR5 activation enhanced the phagocytic capacity and the respiratory burst activity of neutrophils, which was mediated, at least partially, via a stimulation of IL-8 production and CXCR1 signaling. This study demonstrates a novel mechanism of TLR regulation in neutrophils and suggests a critical role for TLR5 in neutrophil-P. aeruginosa interactions in CF lung disease.

This study investigates the cyclic changes in innate immunity in the female reproductive tract (FRT) of mice during the estrous cycle. By examining uterine and vaginal tissues and secretions we show that innate immunity varies with the stage of the estrous cycle and site in the FRT. Secretions from the uterine lumen contained cytokines and chemokines that were significantly higher at proestrus and estrus relative to that measured at diestrus. In contrast, analysis of vaginal secretions indicated that only IL-1β and CXCL1/mouse KC changed during the cycle, with highest levels measured at diestrus and estrus. In contrast, vaginal α-defensin 2 and β-defensins 1-4 mRNA levels peaked at proestrus and estrus and are expressed 1-4 logs greater than that seen in the uterus. These studies further indicate that TLR5 and TLR12 in the uterus, and TLR1, TLR2, TLR5 and TLR13 in the vagina varies with stage of the estrous cycle, with some peaking at proestrus/estrus and others at diestrus. Overall, these studies indicate that innate immune parameters in the uterus and vagina are separate and discrete, and regulated precisely during the estrous cycle.

树突状细胞(dendritic cells, DC)作为机体天然免疫和获得性免疫反应的桥梁和枢纽,发挥着重要的启动和调控作用。随着体外诱导方法的建立和生物学技术的进步,有关DC 的基础生物学研究得到了快速的发展,在诱导方法、个体发生及基因表达和调控等方面,涌现出很多新的、未解的关键问题。同时,随着对粘膜免疫机理研究的深入,DC 在粘膜生态环境中的功能和影响,渐已成为免疫学研究前沿领域中的热点和要点。在本研究中,为了确定DC 体外分化成熟的最短时程,同时为了研究DC 分化成熟相关的基因表达调控,我们建立了快速的DC 体外诱导方法,分析了体外快速诱导 DC 的mi/mRNA 表达谱。此外,在原始分离的女性生殖道共生乳酸杆菌的基础上,以THP-1作为DC 前体细胞的细胞系模型,开展了女性生殖道共生乳酸杆菌刺激活化 THP-1 的研究,希望能够为乳酸杆菌作为生殖道粘膜免疫疫苗的应用提供理论基础。首先,采用外周血单个核细胞(peripheral blood mononuclear cells, PBMC)来源的CD14+细胞为DC 前体,经过GM-CSF 和IL-4 的刺激,1-6 天后得到未成熟DC (immature dendritic cells, iDC),并经成熟因子(TNF-α, IL-1β, IL-6 与PGE2)诱导 1-2 天后,获得成熟DC(mature dendritic cells, mDC)。经过比较和分析,明确了完全分化和成熟各2 天,即“2+2”,为DC 诱导分化的最佳和最短时程,从而证实和建立了DC 体外快速诱导的体系和方法。该方法获得的iDC 与mDC,具有与传统的“6+2” 方法获得的DC 相同的形态与表型,而且,利用该方法获得的DC 总数高于“1+1”, “1+2”与“6+2”的方法,为DC 的生物学研究提供了基础数据。我们进而采用芯片技术,对体外快速分化成熟的DC 进行了mi/mRNA 表达谱分析,确定了DC 不同分化发育阶段特征性的mi/mRNA 表达差异。结果发现,与CD14+ 单核细胞即DC 前体相比,iDC 与mDC 之间具有更加相近的mi/mRNA 表达方式。 miRNA 表达谱分析则表明,不同的miRNA 表达与DC 的不同分化和发育阶段相关。而且,位于同一基因簇内的miRNA,呈现协同表达的情况。特别值得注意的是,本研究发现了在DC 的某些发育阶段特异表达的miRNA,它们在DC 发育过程中的功能,还未得到诠释,它们在DC 某些分化阶段的特异表达,提示了DC 各分化阶段的相关性与特异性。结合mRNA 表达谱分析,我们发现miRNA 的表达与其目的基因的表达在mRNA 水平呈现负相关的特性。同时,免疫相关mRNA 与miRNA 在DC 体外不同发育阶段的表达亦呈现差异,其中,miRNA(如hsa-miR-181a, hsa-miR-223, hsa-miR-155, hsa-miR-146, hsa-miR-106a 与hsa-miR-20a 等)与mRNA(如ALM1 等)参与了特定的与免疫相关的GO(Gene Ontology)与通路(Pathway),提示这些miRNA 与mRNA 可能通过不同的方式调节控制着DC 的体外诱导过程。在有关粘膜生态环境中DC 的分化、成熟及其功能影响的研究中,我们首先通过各种乳酸杆菌鉴定方法的综合应用,确定了6 种原始分离的女性生殖道主要共生乳酸杆菌:发酵乳酸杆菌(L.Fermentum)、约氏乳酸杆菌(L.Johnsonni)、卷曲乳酸杆菌(L.Crispatus)、革氏乳酸杆菌(L.Gasseri)、詹氏乳酸杆菌(L.Jensenii)与德氏乳酸杆菌(L.Delbrueckii )。其中,德氏乳酸杆菌(L.Delbrueckii)和发酵乳酸杆菌(L.Fermentum)具有较高的产H2O2 的能力。在此基础上,我们在与THP-1 的共同培养体系中,将乳酸杆菌对DC 前体的作用和影响进行了比较和研究。结果发现,L.Crispatus 在分离的各原始菌株中,具有最强的刺激THP-1 活化的能力,而且,在相同刺激比例下,L.Crispatus 活菌具有比死菌更强的免疫刺激能力,表现为明显上调THP-1 细胞表面标志CD40、CD80、CD86、 CD1a、CCR6 与CD324 的表达水平,同时可诱导活化THP-1 上调表达Th1 型细胞因子。通过FITC-Dextran 吞噬实验,我们发现,经过L.Crispatus 刺激的THP-1 细胞,其吞噬外来抗原的能力明显下降,但尚未检测到经过活化的THP-1 细胞刺激T 细胞增殖的能力。通过流式细胞术分析的方法,我们检测了TLR1、TLR2、TLR4 与TLR6 在不同的刺激分化阶段的表达水平,结果表明,THP-1 主要通过TLR2 与TLR6 识别女性生殖道L.Crispatus。综上所述,本研究首先通过对DC 体外分化成熟的最短时程的分析,确立了快速诱导DC 的最佳方法,进而利用芯片技术,研究了快速诱导DC 的mi/mRNA 表达谱,揭示了DC 体外分化发育过程中可能的调控途径,为进一步研究DC 的基础生物学提供了恰当的模型和具有指向性的线索。同时,通过与DC 前体THP-1 的共同培养体系,证实了生殖道共生乳酸杆菌的免疫调节作用,为以乳酸杆菌为载体的生殖道粘膜免疫疫苗的研究和应用提供了实验依据

The host genotype has been proposed to contribute to individually composed bacterial communities in the gut. To provide deeper insight into interactions between gut bacteria and host, we associated germ-free C3H and C57BL/10 mice with intestinal bacteria from a C57BL/10 donor mouse. Analysis of microbiota similarity between the animals with denaturing gradient gel electrophoresis revealed the development of a mouse strain-specific microbiota. Microarray-based gene expression analysis in the colonic mucosa identified 202 genes whose expression differed significantly by a factor of more than 2. Application of bioinformatics tools demonstrated that functional terms including signaling/secretion, lipid degradation/catabolism, guanine nucleotide/guanylate binding and immune response were significantly enriched in differentially expressed genes. We had a closer look at the 56 genes with expression differences of more than 4 and observed a higher expression in C57BL/10 mice of the genes coding for Tlr1 and Ang4 which are involved in the recognition and response to gut bacteria. A higher expression of Pla2g2a was detected in C3H mice. In addition, a number of interferon-inducible genes were higher expressed in C3H than in C57BL/10 mice including Gbp1, Mal, Oasl2, Ifi202b, Rtp4, Ly6g6c, Ifi27l2a, Usp18, Ifit1, Ifi44, and Ly6g indicating that interferons may play an essential role in microbiota regulation. However, genes coding for interferons, their receptors, factors involved in interferon expression regulation or signaling pathways were not differentially expressed between the two mouse strains. Taken together, our study confirms that the host genotype is involved in the establishment of host-specific bacterial communities in the gut. Based on expression differences after colonization with the same bacterial inoculum, we propose that Pla2g2a and interferon-dependent genes may contribute to this phenomenon.

The particular microenvironment of the skeletal muscle can be the site of complex immune reactions. Toll-like receptors (TLRs) mediate inflammatory stimuli from pathogens and endogenous danger signals and link the innate and adaptive immune system. We investigated innate immune responses in human muscle. Analyzing TLR1-9 mRNA in cultured myoblasts and rhabdomyosarcoma cells, we found constitutive expression of TLR3. The TLR3 ligand Poly (I:C), a synthetic analog of dsRNA, and IFN-gamma increased TLR3 levels. TLR3 was mainly localized intracellularly and regulated at the protein level. Poly (I:C) challenge 1) activated nuclear factor-kappaB (NF-kappaB), 2) increased IL-8 release, and 3) up-regulated NKG2D ligands and NK-cell-mediated lysis of muscle cells. We examined muscle biopsy specimens of 6 HIV patients with inclusion body myositis/polymyositis (IBM/PM), 7 cases of sporadic IBM and 9 nonmyopathic controls for TLR3 expression. TLR3 mRNA levels were elevated in biopsy specimens from patients with IBM and HIV-myopathies. Muscle fibers in inflammatory myopathies expressed TLR3 in close proximity of infiltrating mononuclear cells. Taken together, our study suggests an important role of TLR3 in the immunobiology of muscle, and has substantial implications for the understanding of the pathogenesis of inflammatory myopathies or therapeutic interventions like vaccinations or gene transfer.

L'interaction entre le système immunitaire et le métabolisme du fer est bien illustrée par l'anémie des maladies chroniques (ACD), qui est fréquemment rencontrée dans les infections chroniques, l'inflammation et le cancer. La majorité des modifications dans les paramètres du fer observées dans l’ACD tient compte des modifications de l’homéostasie du fer, avec la délocalisation du métal de la circulation et les sites de l'érythropoïèse au compartiment de stockage dans les macrophages. Les mécanismes de la réponse hyposidérémique impliquent des cytokines, notamment TNF-alpha et IL-6, qui régulent les niveaux de plusieurs gènes du métabolisme du fer, y compris les transporteurs de fer et de l'hepcidine, un régulateur négatif de l’absorption du fer, ce qui entraîne l'inhibition de l'exportation du fer à travers la ferroportine 1 (FPN1) au niveau de l'intestin et les macrophages. Des études antérieures ont montré que l'IL-6 induit l’expression d’hepcidine dans les hépatocytes, mais il y a très peu de données concernant la façon par laquelle l'hepcidine et la FPN1 sont régulées dans les macrophages. Récemment, nous avons constaté que l'induction de l'hepcidine dans le foie par le lipopolysaccharide (LPS) dépend de la voie de signalisation médiée par le récepteur Toll-like 4 (TLR4). Le but de ce travail est d’identifier les ligands des TLRs capables d'induire l'hepcidine dans les macrophages et de déterminer l’exigence des TLRs dans l’induction de l’hepcidine et le développement d’hyposidérémie. En plus, nous voulons étudier l’effet de l’inflammation causée par les ligands des TLRs sur le taux de fer sérique, la production des cytokines et l'expression de l’hepcidine et de la ferroportine. D’autre part nous voulons étudier l’effet du taux du fer sur la production d’IL-6 macrophagique en réponse à la stimulation par le TLR4. D'abord, pour identifier les ligands des TLRs capables d'induire l'hepcidine dans les macrophages, nous avons traité les macrophages RAW 264.7 et les macrophages péritonéaux de souris (MPMs) avec différents ligands TLRs et on a mesuré l’expression de l'hepcidine par qRT-PCR. Nous avons observé que Pam3CSK4 (Pam), un ligand de TLR2/1; LPS, un ligand de TLR-4 et FSL1 un ligand de TLR2/6 induisent l’expression de l'hepcidine dans les cellules RAW 264.7 et les MPMs, contrairement au polyinosinic: polycytidylic acid (Poly I: C), un ligand de TLR3. De plus, LPS était capable de réprimer l’expression de la ferroportine dans les cellules RAW 264.7. Afin de mieux définir la nécessité des TLRs pour assurer cette expression, nous avons utilisé les souris TLR-2 knock-out et on a établi que l'expression de l'hepcidine dans les macrophages par LPS, Pam ou FSL1 est dépendante du TLR2. En accord avec les expériences in vitro, les études effectuées in vivo ont montré que LPS réprime l’expression de la ferroportine, ainsi que PolyI:C n’est pas capable de stimuler l'expression d'hepcidine hépatique, par contre il était efficace pour déclencher une hyposidérémie. Ensuite, on voulait déterminer la voie de signalisation utilisée dans l’induction de l’hepcidine dans les macrophages. Comme il y deux voies majeures connues pour la signalisation des TLRs : une dépendante et l’autre indépendante de la protéine MyD88, on a étudié l’expression de l’hepcidine dans les MPMs isolés des souris MyD88-/- et nous avons constaté que l'absence de signalisation MyD88 abolit l'induction de l'hepcidine déclenchée par Pam, LPS et FSL1. D’autre part, la stimulation avec du LPS induisait in vivo la production d’IL-6 et de TNF-alpha, et la stimulation d’IL-6 était renforcée in vitro par la présence du fer. Ces observations indiquent que l’expression de HAMP (Hepcidin Antimicrobial Peptide) dans les macrophages peut être régulée par différents TLRs, ce qui suggère que la production d'hepcidine macrophagique fait partie d'une réponse immunitaire activées par les TLRs.

Toll-like receptors (TLR) recognize microbial and viral patterns and activate dendritic cells (DC). TLR distribution among human DC subsets is heterogeneous: plasmacytoid DC (PDC) express TLR1, 7 and 9, while other DC types do not express TLR9 but express other TLR. Here, we report that mRNA for most TLR is expressed at similar levels by murine splenic DC sub-types, including PDC, but that TLR3 is preferentially expressed by CD8α+ DC while TLR5 and TLR7 are selectively absent from the same subset. Consistent with the latter, TLR7 ligand activates CD8α– DC and PDC, but not CD8α+ DC as measured by survival ex vivo, up-regulation of surface markers and production of IL-12p40. These data suggest that the dichotomy in TLR expression between plasmacytoid and non-plasmacytoid DC is not conserved between species. However, lack of TLR7 expression could restrict the involvement of CD8α+ DC in recognition of certain mouse pathogens.

In response to pathogen recognition by Toll-like receptors (TLRs) on their cell surface, macrophages release lipid mediators and cytokines that are widely distributed throughout the body and play essential roles in host responses. Granulocyte macrophage colony-stimulating factor (GM-CSF) is important for the immune response during infections to improve the clearance of microorganisms. In this study, we examined the release of mediators in response to TLR2 ligands by bone marrow-derived macrophages (BMDMs) primed with GM-CSF. We demonstrated that when stimulated with TLR2 ligands, non-primed BMDMs preferentially produced PGE(2) in greater amounts than LTB4. However, GM-CSF priming shifted the release of lipid mediators by BMDMs, resulting in a significant decrease of PGE(2) production in response to the same stimuli. The decrease of PGE(2) production from primed BMDMs was accompanied by a decrease in PGE-synthase mRNA expression and an increase in TNF-alpha and nitric oxide (NO) production. Moreover, some GM-CSF effects were potentiated by the addition of IFN-gamma. Using a variety of TLR2 ligands, we established that PGE(2) release by GM-CSF-primed BMDMs was dependent on TLR2 co-receptors (TLR1, TLR6), CD14, MyD88 and the nuclear translocation of NF kappa B but was not dependent on peroxisome proliferator-activated receptor-gamma (PPAR-gamma) activation. Indeed, GM-CSF priming enhanced TLR2, TLR4 and MyD88 mRNA expression and phospho-I kappa B alpha formation. These findings demonstrate that GM-CSF drives BMDMs to present a profile relevant to the host during infections.

Conflicting findings about the association between leprosy and TLR1 variants N248S and I602S have been reported. Here, we performed case-control and family based studies, followed by replication in 2 case-control populations from Brazil, involving 3162 individuals. Results indicated an association between TLR1 248S and leprosy in the case-control study (SS genotype odds ratio [OR], 1.81; P = .004) and the family based study (z = 2.02; P = .05). This association was consistently replicated in other populations (combined OR, 1.51; P < .001), corroborating the finding that 248S is a susceptibility factor for leprosy. Additionally, we demonstrated that peripheral blood mononuclear cells (PBMCs) carrying 248S produce a lower tumor necrosis factor/interleukin-10 ratio when stimulated with Mycobacterium leprae but not with lipopolysaccharide or PAM3cysK4. The same effect was observed after infection of PBMCs with the Moreau strain of bacillus Calmette-Guerin but not after infection with other strains. Finally, molecular dynamics simulations indicated that the Toll-like receptor 1 structure containing 248S amino acid is different from the structure containing 248N. Our results suggest that TLR1 248S is associated with an increased risk for leprosy, consistent with its hypoimmune regulatory function.

Infection with bacteria such as Chlamydia pneumonia, Helicobacter pylori or Porphyromonas gingivalis may be triggering the secretion of inflammatory cytokines that leads to atherogenesis. The mechanisms by which the innate immune recognition of these pathogens could lead to atherosclerosis remain unclear. In this study, using human vascular endothelial cells or HEK-293 cells engineered to express pattern-recognition receptors (PRRs), we set out to determine Toll-like receptors (TLRs) and functionally associated PRRs involved in the innate recognition of and response to lipopolysaccharide (LPS) from H. pylori or P. gingivalis. Using siRNA interference or recombinant expression of cooperating PRRs, we show that H. pylori and P. gingivalis LPS-induced cell activation is mediated through TLR2. Human vascular endothelial cell activation was found to be lipid raft-dependent and to require the formation of heterotypic receptor complexes comprising of TLR2, TLR1, CD36 and CD11b/CD18. In addition, we report that LPS from these bacterial strains are able to antagonize TLR4. This antagonistic activity of H. pylori or P. gingivalis LPS, as well as their TLR2 activation capability may be associated with their ability to contribute to atherosclerosis.

Nos últimos anos, notou-se aumento da incidência de carcinoma espinocelular de orofaringe (CECOF) associado ao HPV. Sabe-se que CECOF associado ao HPV apresenta melhor prognóstico do que CECOF não infectado por HPV. Inúmeros estudos em carcinoma cervical demonstram alterações de TLRs, isto provavelmente devido às associações das oncoproteínas E6 e E7 com estes receptores. Em humanos, existem 10 TLRs identificados, os quais colaboram na resposta imune contra bactérias, fungos e vírus, bem como colaboram na promoção ou regressão do tumor. Esta influência do TLR na carcinogênese tem sido alvo de inúmeros estudos devido à ligação entre inflamação e o câncer. O presente trabalho teve como objetivo verificar diferenças na expressão e função de receptores Toll-like em carcinoma espinocelular de orofaringe (CECOF). Para tal, foram utilizados trinta e sete espécimes diagnosticados como CECOF e a expressão imuno-histoquímica das proteínas p16 e TLR4 analisadas. Duas linhagens de CECOF HPV16 + e duas CECOF HPV-. foram utilizadas para análise da expressão de TLR1-10, IL-6 e IL-8, por qPCR. A detecção dos principais TLRs (TLR1, TLR2, TLR6 e TLR4) foi feita por citometria de fluxo. Para ativação da via de sinalização de TLR2, e posterior análise da expressão de IL6 e IL8, as células foram estimuladas com peptidoglicano. Para verificar a expressão e função de TLR4, as células foram estimuladas com LPS e LPS UP para posterior análise de IL-6 e IL-8, por ELISA. Os resultados demonstraram diferenças na expressão gênica de TLR1 e TLR6 entre as linhagens HPV- e o grupo HPV+ e diferenças na expressão proteica de TLR9. TLR2 apresentou aumento da expressão proteica em todas as linhagens e demonstra desencadeamento da resposta imune, com secreção de IL6 e IL8 nas linhagens HPV- (SCC72 e SCC89) e em uma das linhagens HPV+ (SCC2). Interessantemente, TLR4 não apresentou diferenças significativas na expressão gênica e proteica. Entretanto, as linhagens HPV+ não demonstraram resposta pró-inflamatória mesmo quando estimuladas com LPS e LPS ultra puro, agonista específico de TLR4. Assim, este trabalho contribui para estabelecer o perfil da expressão dos receptores Toll-like em linhagens celulares de CECOF HPV- e HPV+, e aponta para alterações ocorridas na via de sinalização mediada por TLR4. Além disso, nossos resultados abrem portas para futuros estudos na avaliação de alterações causadas no sistema imune inato pelo HPV, em carcinomas espinocelulares de orofaringe.

Unlike fish and amphibians, mammals do not regenerate retinal neurons throughout life. However, neurogenic potential may be conserved in adult mammal retina and it is necessary to identify the factors that regulate retinal progenitor cells (RPC) proliferative capacity to scope their therapeutic potential. Müller cells can be progenitors for retinal neuronal cells and can play an essential role in the restoration of visual function after retinal injury. Some members of the Toll-like receptor (TLR) family, TLR2, TLR3 and TLR4, are related to progenitor cells proliferation. Müller cells are important in retinal regeneration and stable cell lines are useful for the study of retinal stem cell biology. Our purpose was to obtain a Müller-derived cell line with progenitor characteristics and potential interest in regeneration processes. We obtained and characterized a murine Müller-derived cell line (MU-PH1), which proliferates indefinitely in vitro. Our results show that (i) MU-PH1 cells expresses the Müller cell markers Vimentin, S-100, glutamine synthetase and the progenitor and stem cell markers Nestin, Abcg2, Ascl1, α-tubulin and β-III-tubulin, whereas lacks the expression of CRALBP, GFAP, Chx10, Pax6 and Notch1 markers; (ii) MU-PH1 cell line stably express the photoreceptor markers recoverin, transducin, rhodopsin, blue and red/green opsins and also melanopsin; (iii) the presence of opsins was confirmed by the recording of intracellular free calcium levels during light stimulation; (iv) MU-PH1 cell line also expresses the melatonin MT1 and MT2 receptors; (v) MU-PH1 cells express TLR1, 2, 4 and 6 mRNA; (vi) MU-PH1 express TLR2 at cell surface level; (vii) Candida albicans increases TLR2 and TLR6 mRNA expression; (viii) C. albicans or TLR selective agonists (Pam(3)CysSK(4), LPS) did not elicit morphological changes nor TNF-α secretion; (ix) C. albicans and Pam(3)CysSK(4) augmented MU-PH1 neurospheres formation in a statistically significant manner. Our results indicate that MU-PH1 cell line could be of great interest both as a photoreceptor model and in retinal regeneration approaches and that TLR2 may also play a role in retinal cell proliferation.
