An analytical method for the measurement of five naturally occurring bromophenols of sensory relevance in seafood (barramundi and prawns) is presented. The method combines simultaneous distillation−extraction followed by alkaline back extraction of a hexane extract and subsequent acetylation of the bromophenols. Analysis of the bromophenol acetates was accomplished by headspace solid phase microextraction and gas chromatography−mass spectrometry using selected ion monitoring. The addition of 13C6 bromophenol stable isotope internal standards for each of the five congeners studied permitted the accurate quantitation of 2-bromophenol, 4-bromophenol, 2,6-dibromophenol, 2,4-dibromophenol, and 2,4,6-tribromophenol down to a limit of quantification of 0.05 ng/g of fish flesh. The method indicated acceptable precision and repeatability and excellent linearity over the typical concentration range of these compounds in seafood (0.5−50 ng/g). The analytical method was applied to determine the concentration of bromophenols in a range of farmed and wild barramundi and prawns and was also used to monitor bromophenol uptake in a pilot feeding trial.

采用固相微萃取(SPME)高效液相色谱法(HPLC)同时测定了水中苯酚、4-硝基酚、3-甲基酚、2,4-二氯酚、2,4,6-三氯酚、五氯酚等六种酚类化合物的含量.采用ZORBOX SB-C18柱,以甲醇-1%乙酸水溶液为流动相进行梯度洗脱,流速为1.0 mL/min.紫外检测波长为254、280 nm.六种酚类化合物的检出限为0.31~1.90μg/L,加标回收率为88%~103%.该方法操作简单,能快速、准确地检测水中的酚类化合物.

利用顶空固相微萃取(HSSPME)和气相色谱检测技术,通过对三种不同的SPME萃取头(PDMS,PMPVS,PA)实验条件的优化,并参照液液萃取的方法,对水相中六氯苯,DDT及其代谢产物的萃取效果进行了比较.结果表明,SPME方法比液液萃取方法在检测限和回收率上有很大的提高,其中PDMS对检测六氯苯,DDT及其代谢产物的效果较其它两种萃取头好.

A method for the determination of some pesticide residues in must and wine samples was developed using solid-phase microextraction (SPME) and gas chromatography – electron capture detection (GC/ECD). The procedure only needs dilution as sample pre-treatment and is therefore simple, fast and solvent-free. Eight fungicides (vinclozolin, procymidone, iprodione, penconazole, fenarimol, folpet, nuarimol and hexaconazole), one insecticide (chlorpyriphos) and two acaricides (bromopropylate and tetradifon) can be quantified. Good linearity was observed for all the compounds in the range 5–100 µg/L. The reproducibility of the measurements was found acceptable (with RSD’s below 20%). Detection limits of 11 µg/L, on average, are sufficiently below the proposed maximum residue limits (MRL’s) for these compounds in wine. The analytical method was applied to the determination of these compounds in Portuguese must and wine samples from the Demarcated Region of Alentejo, where any residues could be detected.

A procedure for the determination of seven indicator PCBs in soils and sediments using microwave-assisted extraction (MAE) and headspace solid-phase microextraction (HS-SPME) prior to GC-MS/MS is described. Optimization of the HS-SPME was carried out for the most important parameters such as extraction time, sample volume and temperature. The adopted methodology has reduced consumption of organic solvents and analysis runtime. Under the optimized conditions, the method detection limit ranged from 0.6 to 1 ng/g when 5 g of sample was extracted, the precision on real samples ranged from 4 to 21% and the recovery from 69 to 104%. The proposed method, which included the analysis of a certified reference material in its validation procedure, can be extended to several other PCBs and used in the monitoring of soil or sediments for the presence of PCBs.

Trihalomethanes (THMs) are widely referred and studied as disinfection by-products (DBPs). The THMs that are most commonly detected are chloroform (TCM), bromodichloromethane (BDCM), chlorodibromomethane (CDBM), and bromoform (TBM). Several studies regarding the determination of THMs in swimming pool water and air samples have been published. This paper reviews the most recent work in this field, with a special focus on water and air sampling, sample preparation and analytical determination methods. An experimental study has been developed in order to optimize the headspace solid-phasemicroextraction (HS-SPME) conditions of TCM, BDCM, CDBM and TBM from water samples using a 23 factorial design. An extraction temperature of 45 °C, for 25min, and a desorption time of 5 min were found to be the best conditions. Analysis was performed by gas chromatography with an electron capture detector (GC-ECD). The method was successfully applied to a set of 27 swimming pool water samples collected in the Oporto area (Portugal). TCM was the only THM detected with levels between 4.5 and 406.5 μg L−1. Four of the samples exceeded the guideline value for total THMs in swimming pool water (100 μgL−1) indicated by the Portuguese Health Authority.

Neste trabalho utilizou-se a técnica de HS-SPME/GC/ITMS para o estudo qualitativo dos compostos voláteis (VOC) emitidos pelas folhas de três espécies de Eucalyptus (E.): E. citriodora, E. dunnii e E. saligna tanto em laboratório como in situ, além do estudo da relação entre folhas de eucalipto e rãs Litoria ewingi. Para este fim, foram desenvolvidos métodos análiticos. Os compostos tentativamente como (E,E)-a-farseno, (E)-4,8-dimetil-1,3,7-nonatrieno (DMNT),(E,E)-4,8,12-trimetil-1,3,7,11-tridecatetraeno (TMTT),B-cariofileno,a-cariofileno, gerrmacreno D e (E,E,E)-3,7,11,15-tetrametil-1,3,6,10,14-hexadecapentaeno (TMHP) foram encontrados no headspace de folhas picadas de eucalipto, sendo que os três primeiros também foram nas emissões áreas de E. saligna in situ, não tendo sido encontrados nos óleos voláteis das mesmas árvores, obtidos por hidrodestilação. Nas amostragens in situ, foram observados dois tipos de perfis circadianos nas emissões voláteis, incluindo compostos como cis-, trans-óxido de rosa, trans-B-ocimeno, citronelal, citronelol, entre outros. O comportamento dos compostos citados sugere que os mesmos sejam semioquímicos. Os resultados obtidos com HS-SPME (PDMS/DVB) mostraram que esta é uma ferramenta analítica relativamente simples, que preserva a vida devido ao seu caráter não invasivo, de mínima pertubação do sistema vivo sob amostragem e que, devido a sua rapidez (1 min), permite o acompanhamento de alteraçãoes rápidas provocadas por processos endógenos e/ou exógenos nos seres vivos. O monitoramento da emissão de voláteis de folhas in situ durante 8 a 10 h por vários dias consecutivos, e a extração dos voláteis de rãs vivas sob estresse, submetidas a diferentes condições de dieta e meio ambiente, exemplificam o potencial desta técnica, que abre novos horizontes na investigação de voláteis de seres vivos.

The present study aimed to evaluate the volatiles profile of red mombin (Spondias purpurea) pulp and its powder produced by spray-drying (SD) as an example to show utility of headspace solid-phase microextraction (HS-SPME) in the analysis of parameters such as the quality and stability of fruit products. Volatiles profiles of the pulp were identified by gas chromatography-mass spectrometry (GC-MS), quantified by gas chromatography-flame ionization detector (GC-FID) and compared to the profile of the powder stored at 0, 60 and 120 days in plastic (PP) or laminated packages (LP). The results showed that the technique was able to identify 36 compounds in the red mombin pulp, 17 out of which have been described for the first time in this fruit, showing that red mombin fresh pulp appears to be unique in terms of volatiles composition. However, only 24 compounds were detected in the powder. This decrease is highly correlated (r(2) = 0.99), at least for the majority of compounds, to the degree of volatility of compounds. Furthermore, the powder stored in PP or LP showed no statistical differences in the amounts of its components for a period of 120 days of storage. Finally, this work shows how HS-SPME analysis can be a valuable tool to assess the quality and stability of fruit products.

A simple and sensitive method using solid phase microextraction (SPME) and liquid chromatography (LC) with heated online desorption (SPME-LC) was developed and validated to analyze anticonvulsants (AEDs) in human plasma samples. A heated lab-made interface chamber was used in the desorption procedure, which allowed the transference of the whole extracted sample. The SPME conditions were optimized by applying an experimental design. Important factors are discussed such as fiber coating types, pH, extraction time and desorption conditions. The drugs were analyzed by LC, using a C18 column (150 mm 4.6 mm 5 mm); and 50 mmol L1 , pH ¼ 5.50 ammonium acetate buffer : acetonitrile : methanol (55 : 22 : 23 v/v) as the mobile phase with a flow rate of 0.8 mL min1 . The suggested method presented precision (intra-assay and inter-assay), linearity and limit of quantification (LOQ) all adequate for the therapeutic drug monitoring (TDM) of AEDs in plasma.

Direct immersion SPME-GC-MS-MS was used for the analysis of steroids in water at part-per-trillion(ppt) and lower concentrations. The method was validated and extended to real sample analysis. The method were linear from 0.01 to 5 ng/ml with precision less than 10% relative standard deviation for a steroid mixture at 1 ng/ml. Limit of quantitation and limit of detection was found to be 200- 1200 pg/L and 30-200 pg/L respectively and recoveries ranged from 88-103 %. To understand the extraction efficiency of the fiber, a depletion study was performed. The fiber/ sample partition coefficients for the steroids were determined to be 1.0 x 104 to 1.5 x 104 . The extraction was performed without derivatization or the use of an internal standard. SPMEGC-MS-MS effectively demonstrated ultra-trace level detection of steroids in water.
