A transtirretina (TTR) é uma proteína plasmática constituída por quatro subunidades idênticas de aproximadamente 14KDa e de massa molecular de 55 KDa (Blake et al., 1978). A TTR é responsável pelo transporte de tiroxina (T4) (Andrea et al., 1980) e retinol (vitamina A), neste último tipo de transporte através da ligação à proteina de ligação ao retinol (RBP) (Kanai et al., 1968). É sintetizada principalmente pelo fígado e secretada para o sangue (Murakami et al., 1987) e também sintetizada pelas células epiteliais do plexo coróide e secretada para o líquido cefaloraquidiano (LCR) (Aleshire et al., 1983). Existem outros locais que expressam TTR mas em menor quantidade, nomeadamente: a retina do olho (Martone et al., 1988), o pâncreas (Kato et al., 1985), o saco vitelino visceral (Soprano et al., 1986) o intestino (Loughna et al., 1995); o estômago, coração, músculo e baço (Soprano et al., 1985). A TTR é uma proteína, do ponto de vista filogenético, extremamente conservada o que já de si é um indicador da sua importância biológica (Richardson, 2009) O objectivo deste trabalho foi avaliar a expressão de transtirretina ao longo do sistema gastrointestinal do murganho, nos seguintes órgãos esófago, estômago, duodeno, cólon e também bexiga, com cerca de 3 meses de idade. O segundo objectivo foi identificar as células responsáveis por essa expressão, nos órgãos em estudo. Foi possível verificar que apenas o estômago apresenta valores de expressão normalizada de TTR diferente de zero, expressão essa muito inferior à do fígado, tal como se esperava. Por imunohistoquímica/imunofluorescência foi possível determinar que as células que expressam TTR são pouco abundantes e estão presentes na região glandular do estômago do murganho e também do humano. Para além disto, verificou-se que a TTR co-localiza com somatostatina e que as células que sintetizam TTR correspondem às células D, responsáveis pela secreção de somatostatina

Several hormonal and physiological changes that occur during pregnancy directly affect gastrointestinal motility (GI), however, very little is known about the relationship between the electrical and mechanical activities of the stomach and its implications. The aim of this study was to compare the female sex hormones, with profiles of gastrointestinal motility using AC biosusceptometry (BAC) in pregnant rats. Seven female Wistar rats (weighing 250-350 g) were used and all procedures were approved by the local Ethics Committee of animal. Laparotomy was performed on anesthetized rats and a magnetic marker (3.5 mm in diameter and 3.0 mm height) was implanted in the distal stomach. After 10 days of recuperation, the mice were put to mate until confirmation of pregnancy. Fed animals were anesthetized (mg pentobarbital 30 / kg) and kept supine for recording the signal with a sensor 30 min BAC placed on the abdominal surface on days 0, 7, 14 and 20 of gestation. Statistical analysis was performed by Student's t test statistical Newman-Keuls and a P value of .05 was considered statistically significant. The frequency of contraction of the stomach was 74 mHz ± 3, 67 ± 7 mHz, 59 mHz ± 6 (p <0.05 vs day -1) and 71 ± 5 mHz on days -0, -7, -14, and -20, respectively. The values of 44 ± 15 pg / mL and 31 ± 9 pg / ml 25 ± 6 pg / ml, 22 ± 9 pg / ml on days -0, -7, -14 and -20, respectively, obtained by 17ß- estradiol were not statistically significant. The values of progesterone were 13 ± 6 pg / ml at 54 ± 15 pg / ml, 127 ± 42 pg / ml (p <0.05 vs day-1), 22 ± 13 pg / mL on day -0, - 7, -14 and -20, respectively. A linear correlation (Pearson's test) between the lower frequency gastric and increased levels of progesterone was 80% (p <0.0000001). The study of gastrointestinal tract (GIT) functions is necessary due to the increasing number of pathologies associated with it. Directly influencing the quality of life, the gastrointestinal tract provides a number of parameters that, when analyzed, allow us to describe its dysfunctions. Thus, many techniques can be combined to obtain these properties related to the GIT. However, these techniques are often invasive, require surgery, catheter insertion, or to build a temporal model of these functions, require the sacrifice of animals in a series of data collection. The technique used in this study has the advantage of having a low operating cost, being free of ionizing radiation, non-invasive and is known as biosusceptometry AC (BAC), used to evaluate the properties of the GI tract by monitoring the position and concentration of materials magnetically marked. The sensor consists of two pairs of coils, one reference and one for detection. A fixed base line separates the sensing and reference coils, and also functions as support for the instrumentation. It is also important to note that the detection coils are arranged in a first order (subtraction) gradiometric way. The objective of this study was to analyze the effects of gastrectomy in gastric emptying and gastrointestinal transit time of solid food in rats using a BAC system associated with magnetic markers. To realize this study was constructed a dedicated BAC sensor, built to analyze these GIT properties. Data acquisition was obtained by aligning the magnetic sensor with the stomach and colon of the animal at pre-determined intervals. Thus, when approaching the magnetic material of the sensor, the balance created between the two sides of the sensor is broken. This imbalance can be measured, digitized and acquired. The gastrointestinal tract is the main route of nutrients absorption and drugs delivery. Is important to know the parameters related to the tract, like gastric emptying and gastrointestinal transit, in order to better understand the behavior of different kind of meals or drugs passing through the GIT. Many techniques are used to study these parameters, such as manometry, scintigraphy, phenol red, activated charcoal and carbon-13 reading. However, these methods use radiation, are invasive and require animal sacrifice. As an alternative proposal, the Alternate Current Biosusceptometry (ACB), a magnetic technique, has proved to be effective for these studies with small animals, in a noninvasive way, low cost, radiation free and avoiding the animal death. Associating the ACB to magnetic micro or nanoparticles used as tracers, it is possible to observe the meal behavior inside of the GIT. Focusing meanly on liquid meals digestion, this paper had the objective to evaluate the efficiency of the ACB technique in gastric emptying and gastrointestinal transit evaluation of liquid meals in rats. To perform the experiments, magnetic nanoparticles (ferrite, MgFe2O4) were used on a 1,5 ml solution introduced by gavage on similar weight and age rats. The sensor made by 2 pairs of coils, capable of generating and detecting magnetic fields, creates a field on the interest place and when this field is in contact with the marked meal, it changes, resulting on a variation of the measured voltage. The voltage variation is analyzed and is obtained a particle concentration on the interest region. The results showed that is possible to apply the ACB technique on the GIT evaluation of liquid particles digestion, gastric emptying and meal cecum arrival time curves were obtained and from that, is possible to observe a pattern of gastrointestinal transit. Al llarg dels últims anys l’òxid nítric (NO) i el monòxid de carboni (CO) han estat investigats com dos neurotransmissors gasosos del tracte gastrointestinal. Algunes dades demostren que el sulfur d’hidrogen (H2S) podria ser el tercer neurotransmissor gasos a nivell central i perifèric. Objectiu: valorar si el H2S compleix els requisits per ser considerat com el tercer neurotransmissor gasos a nivell del sistema nerviós entèric. Mètodes: la motilitat espontània de preparacions de múscul llis circular de mostres de còlon sigma humà i còlon mig de rata es van determinar in vitro mitjançant la tècnica de banys d’òrgans. El hidrogensulfur de sodi (NaHS) va ser utilitzat com donador de H2S. Resultats: 1- El NaHS va inhibir de forma concentració depenent la motilitat espontània en presència de bloqueig neural amb Tetrodotoxina (TTX 1 μM) (IC50 101±18 μM rata n=10; 329±86 μM humà n=18). 2- Aquesta relaxació es va veure reduïda per ODQ 10 μM, un inhibidor de la guanilat ciclasa, en còlon mig de rata (n=5). En humà a més de ser reduïda per ODQ 10μM (n=6), també ho va ser per SQ 22,536 100 μM (n=10), un inhibidor de l’adenilat ciclasa, i per L-NNA 1 mM (n=8), un inhibidor de la sintesi de NO. 3- En còlon de rata la incubació amb D,L-Propargilglicina 2 mM (PAG), un inhibidor de la síntesi de H2S, va incrementar la motilitat tant en situació control (10,05±2,06 vs 17,46±3,37 g•min-1 AUC, P&0,001, n=10) com en presència de TTX 1μM (14,45±7,28 vs 21,67±10,05 g•min-1 AUC, P&0,01, n=7). Conclusió: el H2S compleix part dels criteris per ser considerat un neurotransmissor gasos inhibitori en el tracte gastrointestinal: la inhibició de la seva síntesi incrementa la motilitat intestinal i la seva addició exògena produeix relaxació de la musculatura llisa intestinal.

Endoscopy of the gastrointestinal tract is a specialized minimally invasive diagnostic method and with both diagnostic and therapeutic possibilities. It is divided into two major groups, the rigid and flexible endoscopy. The first most widely used for visualization of body cavities that do not have a light, like the chest, for example, and the second for those with light as the gastrointestinal tract. In Brazil there are few professionals, veterinary hospitals or clinics that offer this type of service. Thus, endoscopy is an area of expertise of the veterinarian who is in huge expansion and has shown great applicability with excellent results in both clinical medicine and surgical small animals

This work presents the development of a graphical interface to the Lock-in Amplifier, which is used in physiological studies on the motility of the gastrointestinal tract in rats and signal processing. With a simple and low cost instrumentation, the resources offered by the virtual interface of LabVIEW software allows the creation of commands similar to the actual instrument that, through communication via standard serial port, transmits data between a PC and peripheral device performing specific and particular needs in the amplifier. Created for the lock-in amplifier model SR830 Stanford Research Systems, the remote manipulation gives the user greater accessibility in the process of configuration and calibration. And, since the software is installed, there is the advantage of eliminating the need of purchase new devices to upgrade the system. The commands created were made to perform six basic modifications that are used in routine of the Biomagnetism Laboratory.
