Antirrhinum majus L. and Senecio douglasii DC. are herbaceous perennial ornamental plants used in landscaping. The multiplication of these plants is by seed; however, there are still doubts about the temperature that can provide higher rates and speed of germination. Thus the aim was to study the effect of temperature on seed germination of A. majus and S. douglasii. The study was conducted separately for each species. The experimental design was entirely randomized with six temperature conditions (temperature controlled constant of 20, 25, 30, 35°C and alternating temperatures of 20-30 and 25-35°C with a photoperiod of 12 hours) with four replications of 100 seeds each. Total germination percentage and germination rate were determined. The means were compared by Tukey test at 5%. For A majus the highest germination percentage was observed at 20°C that did not differ statistically from other temperatures. The highest germination rate was obtained at the temperature of 20 and 25°C. For S. douglasii seeds it was observed that the highest germination percentage and germination rate occurred at 20°C. The lower temperature showed the better percentage and germination rate for these species.

This study is the first in the literature to focus attention on the possible immunotoxic effect of integerrimine N-oxide content in the butanolic residue (BR) of Senecio brasiliensis, a poisonous hepatotoxic plant that contains pyrrolizidine alkaloids (PAs). PAs have been reported as a pasture and food contaminant and as herbal medicine used worldwide and are responsible for poisoning events in livestock and human beings. After the plant extraction, BR extracted from Senecio brasiliensis was found to contain approximately 70% integerrimine N-oxide by elemental and spectral analyses ((1)H and (13)C NMR), which was administered to adult male Wistar Hannover rats at doses of 3, 6 and 9 mg/kg for 28 days. Body weight gain, food consumption, lymphoid organs, neutrophil analysis, humoural immune response, cellular immune response and lymphocyte analysis were evaluated. Our study showed that integerrimine N-oxide could promote an impairment in the body weight gain, interference with blood cell counts and a reducing T cell proliferative activity in rats; however, no differences in the neutrophil activities, lymphocytes phenotyping and humoural and cellular immune responses were observed. It is concluded that doses of integerrimine N-oxide here employed did not produce marked immunotoxic effects. (C) 2011 Elsevier Ltd. All rights reserved.

This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.

BACKGROUND AND AIMS: The Senecio hybrid zone on Mt Etna, Sicily, is characterized by steep altitudinal clines in quantitative traits and genetic variation. Such clines are thought to be maintained by a combination of 'endogenous' selection arising from genetic incompatibilities and environment-dependent 'exogenous' selection leading to local adaptation. Here, the hypothesis was tested that local adaptation to the altitudinal temperature gradient contributes to maintaining divergence between the parental species, S. chrysanthemifolius and S. aethnensis. METHODS: Intra- and inter-population crosses were performed between five populations from across the hybrid zone and the germination and early seedling growth of the progeny were assessed. KEY RESULTS: Seedlings from higher-altitude populations germinated better under low temperatures (9-13 °C) than those from lower altitude populations. Seedlings from higher-altitude populations had lower survival rates under warm conditions (25/15 °C) than those from lower altitude populations, but also attained greater biomass. There was no altitudinal variation in growth or survival under cold conditions (15/5 °C). Population-level plasticity increased with altitude. Germination, growth and survival of natural hybrids and experimentally generated F(1)s generally exceeded the worse-performing parent. CONCLUSIONS: Limited evidence was found for endogenous selection against hybrids but relatively clear evidence was found for divergence in seed and seedling traits, which is probably adaptive. The combination of low-temperature germination and faster growth in warm conditions might enable high-altitude S. aethnensis to maximize its growth during a shorter growing season, while the slower growth of S. chrysanthemifolius may be an adaptation to drought stress at low altitudes. This study indicates that temperature gradients are likely to be an important environmental factor generating and maintaining adaptive divergence across the Senecio hybrid zone on Mt Etna.

Senecio pterophorus DC. is an erect, semilignose shrub (fig. 1) which grows up to 2 m high. Is a chamaephyte basipetal branch-shedder (ORSHAN, Plant pheno-morphological studies in Mediterranean type ecosystems: 210-211. 1989) whose new branches appear below inflorescences but not from the lower parts of the plant. Leaves lanceolate. Stems are annual and develop at their apex a great number of small (up to 15 mm in diameter) yellow capitula which are grouped in a corymbose-paniculated inflorescence (HARVEY & SONDER, Flora Capensis: Cape Colony, Caffraria & Port Natal 3: 386.1865).

En este trabajo se describen el área de distribución y el número de poblaciones de la especie sudafricana Senecio pterophorus DC. en Cataluña, única área de la Europa continental de donde hasta ahora se conoce. Esta especie coloniza ambientes perturbados, como lechos de ríos, márgenes de carreteras y áreas forestales perturbadas, siempre que no haya limitación por la luz y la humedad. En los hábitats riparios y ruderales las poblaciones son más estables y densas, con un gran número de individuos de diversas clases de edad, mientras que en ambientes forestales las poblaciones suelen contar con pocos individuos adultos distribuidos de forma dispersa. Senecio pterophorus presenta, además, una acusada plasticidad morfológica en relación con el hábitat, de modo que en ambientes riparios y ruderales los individuos desarrollan un porte esférico, de menor tamaño, y unas hojas más pequeñas que en ambientes forestales, donde el porte de las plantas es más alargado y el tamaño de las hojas mucho mayor. Esta especie, en creciente expansión, debe ser considerada una planta invasora en la Península Ibérica, ya que ha aumentado considerablemente el número de sus poblaciones en un período relativamente corto. Además, se sugiere que S. pterophorus puede llegar a ser una amenaza para la conservación de la diversidad de especies y de hábitats naturales autóctonos, como ocurre en Australia, donde desplaza a las especies nativas e incluso se hibrida con otras especies de Senecio.

Senecio species contain a large variety of secondary metabolites and many of these plants afford pyrrolizidine alkaloids. This paper is a review of the literature, describing 62 pyrrolizidine alkaloids already isolated in 62 of more than 2000 species of Senecio, distributed worldwide. The structure-activity relationships involving their toxicity are also discussed, since some Senecio species used for medicinal purposes are responsible for causing serious adverse effects.

The essential oils of leaves, aerial and underground stems of Senecio crassiflorus var. crassiflorus were extracted by hydrodistillation and their analyses were performed by GC/MS. Qualitative and quantitative differences have been found between the analyzed parts. The main compounds of the essentials oils of the leaves were found to be oxygenated sesquiterpenes, while the oils from the aerial and underground stems showed sesquiterpene hydrocarbons as major constituents. Furthermore, the essential oils were tested against Staphylocccus aureus, Escherichia coli, Pseudomonas aeruginosa and Bacillus cereus. The essential oil of the leaves was shown to be the most active.

Pyrrolizidine alkaloids (PAs) are N-based plant secondary metabolites that function as chemical defenses against vertebrate and invertebrate herbivores. PAs can be highly variable at intraspecific level, both in their absolute and relative concentrations. Changes in the chemical composition of exotic plants when they invade a new environment have been poorly explored. Here we studied the biogeographical variation on PAs in Senecio pterophorus (Asteraceae) in the native region in Eastern South Africa, an expanded region in Western South Africa, and two introduced regions in Australia and Europe. PAs in S. pterophorus were represented by the highly toxic 1,2-unsaturated PAs and the less toxic 1,2-saturated PAs. Our results show a change in the plant chemical composition after invasion. Total PAs concentrations were highest in Australia compared to any other region. Plants from Europe contained the highest relative concentrations of 1,2-saturated PAs. The positive correlation between the chemical and the genetic distances estimated between populations suggests that the chemical profiles in the non-native regions were related to the plant dispersal routes. The decrease in the chemical diversity and the change in the absolute PAs concentrations in S. pterophorus after invasion may have consequences in the interactions between plants and herbivores in the novel habitats.

Successful plant invaders may have specific morphological and physiological traits that promote invasion in a new habitat. The Evolution of Increased Competitive Ability (EICA) hypothesis predicts that plants released from natural enemies in the introduced habitats are more competitive and perform better than plants from the native populations. An increased phenotypic plasticity may also favour invasion because it allows plants to function under a wider range of environments. In this study we used Senecio pterophorus (Asteraceae) to test whether introduced plant populations are 1)more competitive and 2) more plastic compared with the native populations. We conducted a common garden experiment using plants from the native range (South Africa, Eastern Cape), an expanded range (South Africa, Western Cape) and two introduced ranges (Australia and Europe) under different conditions of water availability. Contrary to the EICA and the increased plasticity hypotheses, plants from the invasive and expanded populations grew less and responded less to watering than those from their native range. These results may be caused by a depleted competition as well as the presence of genetic bottlenecks in the newly invaded areas.

Six pyrrolizidine alkaloids (PAs) (two saturated macrocyclic, three unsaturated macrocyclic and one unsaturated seco-macrocyclic) were isolated from native Peruvian Senecio species. The structures of these alkaloids were established by a complete NMR spectroscopic analysis, chemical transformations and comparison of their NMR data with those published for similar alkaloids. Three PAs were then tested for antifungal activity against Fusarium moniliforme, F. (Sheldon), F. oxysporum fs. lycopersici (Scheldt) and F. solani (Mart), no significant activity being observed.

Cases of seneciosis in horses occurring in four farms in the state of Santa Catarina and in another in the state of Rio Grande do Sul, southern Brazil, are reported. S. brasiliensis or S. oxyphyllus or both were detected in four of the five properties. Five horses (one on each property) were necropsied, and tissues for histopathological examination were collected from four horses. Neurological signs, such as depression, ataxia, aimeless walking, circling, head pressing, faulty prehension of food, dysphagia and blindness were consistently observed. Other signs included inappetence, loss of weight, colic, subcutaneous edema, icterus and photodermatitis. At necropsy the livers were firmer and darker than normal and had accentuation of lobular pattern. Edema of the mesentery and ascites were observed in one horse. Main histopathological changes consisted of hepatic chiefly periportal fibrosis, hepatomegalocytosis and biliary hyperplasia. Marked cholestasis and morphological evidence of hepatic encephalopathy were seen respectively in the liver and brain of one of the horses.

Descreve-se a ocorrência de um surto de intoxicação espontânea por Senecio brasiliensis em ovinos em um estabelecimento do município de Mata, Estado do Rio Grande do Sul, em meados de janeiro de 1997. De um total de 94 ovinos, 51 (54,25%) animais adoeceram e 50 (53,2%) morreram. Esse rebanho permaneceu durante aproximadamente 7 meses (de junho de 1996 a janeiro de 1997) em piquetes de pastagem nativa onde havia grande quantidade de S. brasiliensis. O quadro clínico manifestado pelos animais afetados consistia em fotossensibilização, emagrecimento progressivo, apatia, fraqueza, perturbações neurológicas como depressão, andar a esmo e desequilibrado, icterícia e hemoglobinúria. Houve melhora das lesões de pele naqueles ovinos que desenvolveram fotossensibilização hepatógena depois que foram retirados do sol. As principais lesões macroscópicas observadas em 9 dos 10 ovinos necropsiados incluíam fígado diminuído de tamanho, firme, difusamente marrom amarelado ou esverdeado, com quantidades variáveis de nódulos de 1-3 mm de diâmetro, bem circunscritos, salientes na cápsula, amarelados, distribuídos aleatoriamente por todo o parênquima. A vesícula biliar estava repleta e preenchida por bile verde escura e espessa. Havia também derrames cavitários (hidropericárdio e ascite). Crise hemolítica aguda fatal associada à intoxicação crônica hepatógena por cobre foi observada em cinco ovinos. Além das lesões hepáticas macroscópicas já mencionadas, foi observada icterícia generalizada da carcaça, rins tumefeitos, friáveis, difusamente escurecidos ou com fino pontilhado enegrecido; a urina era marrom escura (hemoglobinúria). As principais lesões microscópicas foram observadas no fígado e consistiam em hepatomegalocitose, proliferação de ductos biliares (hiperplasia ductal) e fibrose periportal moderada acompanhada de infiltrado inflamatório mononuclear. Macrófagos carregados de pigmento acastanhado formavam aglomerados nas tríades portais ou estavam dispersos entre os hepatócitos remanescentes. O material armazenado no citoplasma desses macrófagos correspondia a ceróide e cobre, positivo nas técnicas de PAS e rodanina, respectivamente. Nos rins de cinco animais, havia nefrose hemoglobinúrica caracterizada por degeneração e necrose do epitélio tubular, presença de hemoglobina e hemossiderina no citoplasma das células epiteliais dos túbulos contorcidos e cilindros de hemoglobina na luz tubular. Evidência morfológica de encefalopatia hepática incluía degeneração esponjosa (status spongiosus) da substância branca do encéfalo. Achados ultra-estruturais no fígado incluíam graus variáveis de degeneração hepatocelular caracterizada pelo acúmulo de numerosas gotas lipídicas no citoplasma das células hepáticas e presença de lisossomos carregados de material eletrodenso que, na maioria dos casos, correspondia à lipofuscina-ceróide. Adicionalmente, havia discreta dilatação do retículo endoplasmático rugoso e moderada hiperplasia do retículo endoplasmático liso em algumas regiões do citoplasma dos hepatócitos. No epitélio dos túbulos contorcidos proximais do rim foi observado edema intracelular e diversas alterações mitocondriais de caráter degenerativo que incluíam tumefação, desorganização e ruptura das cristas, matriz finamente granular, acúmulo de gotículas de gordura e ruptura das membranas em alguns casos. Lisossomos contendo material fortemente eletrodenso foram observados em muitas células tubulares renais. O exame laboratorial de fragmentos de fígado e rim dos ovinos afetados revelou níveis elevados de cobre que variaram respectivamente de 369 ppm a 1248 ppm e 152 ppm a 687 ppm com base na matéria seca. O diagnóstico de intoxicação por Senecio brasiliensis baseou-se em dados epidemiológicos, clínicos, de necropsia, histopatológicos e laboratoriais.

O objetivo principal desse trabalho foi determinar a fenologia de Senecio brasiliensis, S. oxyphyllus, S. heterotrichius e S. selloi, e relacioná-la com a epidemiologia da intoxicação em bovinos, na região sul do Rio Grande do Sul. O estudo fenológico foi feito durante dois anos nos municípios de Bagé e Capão do Leão. As leituras foram mensais durante o período vegetativo e quinzenais no período reprodutivo das espécies, para observação desde sua emergência até dispersão de sementes, avaliando-se o vigor, e relacionando essas variáveis com fatores ambientais. Os resultados permitiram concluir que durante todo o ano há emergência de plantas de Senecio spp, desde que haja condições ambientais favoráveis, como umidade e luz, e as fenofases vegetativas são praticamente constantes durante todo o ciclo da planta. Fatores ambientais desfavoráveis como o déficit hídrico, o manejo do solo e o dano de insetos, associados ou não, podem alterar o ciclo das plantas e serem determinantes para a sua permanência no ambiente. A maioria dos exemplares, das quatro espécies, comportou-se como anual e monocárpica. A espécie mais persistente no ambiente foi S. heterotrichius (15% das plantas persistiram durante os dois anos de estudo), seguida de S. selloi (2,8%) e S. brasiliensis (0,9%). S. oxyphyllus não permaneceu no ambiente por mais de um ano.

Lesões hepáticas causadas pela intoxicação por Senecio spp em bovinos são progressivas e mortes podem ocorrer vários meses após a ingestão da planta. Testes laboratoriais da função hepática nem sempre são indicadores confiáveis de animais subclinicamente afetados. A biópsia hepática pode ser indicada para identificar bovinos com lesões hepáticas, mas sem sinais clínicos, e pode ter também valor prognóstico, uma vez que se acredita que as lesões hepáticas evoluam para causar insuficiência hepática e morte. Tais bovinos poderiam ser identificados pela biópsia hepática e enviados para o abate antes de desenvolverem os sinais clínicos, minimizando assim os prejuízos. Este estudo visou avaliar a biópsia hepática como um método de diagnóstico e prognóstico em casos de intoxi-cação por Senecio spp. em bovinos. Um surto de intoxicação por Senecio brasiliensis foi diagnosticado em bezerros de leite que haviam ingerido feno contaminado por 5-10% dessa planta tóxica. Biópsia hepática com agulha de Menghini por abordagem transtorácica foi realizada em 135 bezerros que ingeriram feno contaminado. Os bezerros biopsiados foram acompanhados por um período de 26 meses após a biópsia. Dezessete bezerros biopsiados tinham lesões típicas da intoxicação por Senecio spp (bezerros positivos) e 118 tinham fígados histologicamente normais (bezerros negativos). As lesões hepáticas dos bovinos positivos incluíam fibrose, hepatomegalocitose e hiperplasia de ductos biliares. Quinze dos 17 bezerros positivos morreram com sinais clínicos típicos de intoxicação por Senecio spp. 17-149 dias após a biópsia; 13 desses foram necropsiados e apresentavam lesões macro e microscópicas típicas de intoxicação por Senecio spp. Dois bovinos positivos mantinham-se clinicamente normais ao final do período de observação pós-biópsia. O valor prognóstico (sensibilidae) do teste foi considerado alto, uma vez que 88,23% dos bezerros positivos morreram. A especificidade do teste foi considerada bastante alta (99.16%), uma vez que apenas um dos 118 bovinos negativos na biópsia hepática morreu durante o período de observação pós-biópsia. Em nenhum bovino foi observado qualquer efeito negativo relacionado à técnica da biópsia hepática.
