This report presents a new extraction method of the dinophysistoxins (DTXs), confirmed by quantification using high-performance liquid chromatography coupled to mass spectrometry with an ion trap and electro spray interface (HPLC/ESI/MS2). The method originality consists on the adaptation of DTXs basic extraction procedure (liquid/ liquid) to a solid phase extraction (SPE) via a robotic station: ASPEC XLi The parameters of the automatization procedure were optimized to obtain the best DTXs recovery rate. These improvements were loaded with digestive gland mussel homogenat realized on a silica cartridge SPE, activated in hexane/chloroform (50:50), washed with hexane/chloroform (50:50) and extracted by an elution gradient (chloroform methanol (65:35) and methanol (100%)). This method was validated according to two normative referentials (linearity, detection quantification limits and accuracyâ¦) : - The Guide of the Pharmacy industry: Analytical Validation, report of the commission SFSTP 1992 (French Corporation of the Sciences and Technical Pharmaceutical), - - The Procedure of validation of an alternative method in compare to a reference method. (AFNOR, 1998. NF V 03-110). Comparison with the classical liquid/liquid extraction and the automated method present clear advantages. In an analytical method the extraction is generally considered to be the most labor-intensive and error-prone step. This new procedure allowed us to increase throughput, to improve the reproducibility and to reduce the error risks due to the individual manual treatments.

En la actualidad, los acuicultores del mundo tienen la necesidad de disponer de cepas de rotíferos debidamente determinadas y caracterizadas, para poder optimizar el cultivo en masa de estos y evitar las posibles mortalidades repentinas en los sistemas de cultivo. En este contexto, es necesario también identificar las cepas peruanas e introducidas que están vienen siendo utilizadas para acuicultura en el Perú. Por lo que en el presente trabajo se realizó la caracterización morfométrica de las cepas peruanas, Tumbes (TP), Puerto Viejo 2 (PVL2), Ventanilla (VL) y de 2 cepas introducidas cepa FONDEPES (CF) y Universidad Agraria "La Molina" (CA). EI estudio morfométrico se realizó con rotíferos de la misma cohorte, los cuales fueron cultivados bajo el diseño del jardín común. Como cepas patrón se utilizaron 3 cepas de las especies de rotíferos del complejo Brachionus plicatilis (Brachionus plicatilis s. s. (cepa L 1), B. rotundiformis (cepa SS2) y B. ibericus (cepa SM2). La caracterización morfométrica se realizó mediante el análisis discriminante por pasos, utilizando para ello 9 caracteres morfométricos, y como confirmación se realizaron 6 análisis de varianza entre las proporciones de los caracteres medidos.

INTRODUCTION: Congenital hypogonadotropic hypogonadism (CHH) is a rare, genetic, reproductive endocrine disorder characterized by absent puberty and infertility. Limited information is available on the psychosocial impact of CHH and psychosexual development in these patients. AIM: The aim of this study was to determine the impact of CHH on psychosexual development in men on long-term treatment. METHODS: A sequential mixed methods explanatory design was used. First, an online survey (quantitative) was used to quantify the frequency of psychosexual problems among CHH men. Second, patient focus groups (qualitative) were conducted to explore survey findings in detail and develop a working model to guide potential nursing and interdisciplinary interventions. MAIN OUTCOME MEASURES: Patient characteristics, frequency of body shame, difficulty with intimate relationships, and never having been sexually active were assessed. Additionally, we collected subjective patient-reported outcomes regarding the impact of CHH on psychological/emotional well-being, intimate relationships, and sexual activity. RESULTS: A total of 101 CHH men on long-term treatment (>1 year) were included for the analysis of the online survey (mean age 37&#8201;±&#8201;11 years, range 19-66, median 36). Half (52/101, 51%) of the men had been seen at a specialized academic center and 37/101 (37%) reported having had fertility-inducing treatment. A high percentage of CHH men experience psychosexual problems including difficulty with intimate relationships (70%) and body image concerns/body shame (94/101, 93%), and the percentage of men never having been sexually active is five times the rate in a reference group (26% vs. 5.4%, P&#8201;<&#8201;0.001). Focus groups revealed persisting body shame and low self-esteem despite long-term treatment that has lasting impact on psychosexual functioning. CONCLUSIONS: CHH men frequently experience psychosexual problems that pose barriers to intimate relationships and initiating sexual activity. These lingering effects cause significant distress and are not ameliorated by long-term treatment. This study aimed to evaluate the durability of adhesion between acrylic teeth and denture base acrylic resin. The base surfaces of 24 acrylic teeth were flatted and submitted to 4 surface treatment methods: SM1 (control): No SM; SM2: application of a methyl methacrylate-based bonding agent (Vitacol); SM3: air abrasion with 30-μm silicone oxide plus silane; SM4: SM3 plus SM2. A heat-polymerized acrylic resin was applied to the teeth. Thereafter, bar specimens were produced for the microtensile test at dry and thermocyled conditions (60 days water storage followed by 12,000 cycles). The results showed that bond strength was significantly affected by the SM (P < .0001) (SM4 = SM2 > SM3 > SM1) and storage regimens (P < .0001) (dry > thermocycled). The methyl methacrylate-based adhesive showed the highest bond strength.

A zona subgranular (ZSG) do giro denteado (GD) de mamíferos adultos é conhecida por produzir constantemente novos neurônios. A busca por novas moléculas que possam modular a formação de novas células neurais são bastante atuais. Visto que a Amazônia é conhecida mundialmente pela sua biodiversidade, com um potencial pouco explorado de fármacos naturais derivados de plantas medicinais típicas da região. O trabalho buscou investigar o efeito neurogênico do extrato aquoso (EA) da Physalis angulata e da substância purificada Fisalina D sobre as células-tronco do GD do hipocampo de camundongos adultos. Os camundongos machos (BALB/c), 6 a 8 semanas de idade foram divididos em quatro grupos experimentais: controle e tratados com EA ou substância purificada. Os animais receberam diferentes doses do extrato (0,1; 1 e 5 mg/Kg) e/ou substância purificada (5mg/Kg) ou salina (grupo controle), 5 horas depois uma única dose de 5-Bromodeoxiuridina (BrdU) [50mg/kg]. Em seguida, os animais foram sacrificados 24 horas ou 7 dias após a administração do BrdU. Os cérebros foram coletados e cortes coronais do hipocampo (40 μm) foram realizados para contagem das células BrdU-positivas no GD hipocampal. Para avaliação estatística realizamos análise de variância (ANOVA) das médias amostrais seguida pelo pós-teste t de Student. O EA promoveu um aumento significativo do número de células BrdU positivas no GD dos grupos tratados em relação ao grupo controle [Controle, 92±24 (n=9); 0,1mg/Kg, 160±22 (n=4); 1mg/Kg, 310±5 (n=4); 5mg/Kg, 501±24 (n=3)] nos animais sacrificados 24 horas após administração do BrdU. Quando os animais foram sacrificados 7 dias após administração do BrdU, o número de células BrdU+ no GD também foi maior no grupo tratado em relação ao controle [Controle, 107±7 (n=4); 5mg/Kg, 145±23 (n=4)]. Usando a substância purificada, Fisalina D, também observamos um aumento do número de células BrdU+ no GD do grupo tratado com a droga em relação ao grupo controle [Controle, 92±24 (n=9); Fisalina D, 5mg/Kg, 316±37 (n=3)]. Este resultado sugere que o EA e a sustância purificada, na dose de 5 mg/Kg, estimulam a proliferação de células BrdU-positivas na ZSG do GD do hipocampo de camundongos adultos.

The objective of this study was to verify application of two methodologies: substrate moistened with herbicide solution (SM) and immersion of seeds in herbicide solution (IH) for detecting soybean seeds genetically modified. For this, non-transgenic and transgenic soybean seeds, harvested in the 2008/2009 crop seasons were used. The treatments with substrate moistened were: SM1) 0.03% herbicide solution, at 25 ºC, with evaluation in the sixth day (hs -0.03% -25 ºC, 6th d); SM2) HS -0.03% -35 ºC, 5th d; SM3) HS -0.03% - 40 ºC, 5th d; and SM4) hs -0.06% -5 ºC, 5th d. In the methodology of immersion of seeds the following treatments were performed: IH1) seed immersion in a 0.6% herbicide solution, at 25 ºC, for 1 h, (si -0.06% -25 ºC, 1 h; IH2) si -0.06% - 35 ºC, 30 min.; IH3) si -0.06% -40 ºC, 30 min.; IH4) si -0.12% -35 ºC, 30 min.; and IH5) si -0.12% -40 ºC, 30 min. Bioassays allow detecting soybean seeds tolerant to glyphosate herbicide within five days. The seeds of non-genetically modified and genetically modified soybean cultivars may be easily distinguished through the treatments SM2 and SM4 of the moistened substrate methodology; and treatments IH3, IH4, and IH5 of seed immersion methodology. Both methodologies are easily feasible, practical, and applicable in seed analysis laboratories, once do not require special equipments.

The spark plasma sintering (SPS) technique, by using a compacting pressure of 50 MPa, was used to consolidate pre-reacted powders of Bi1.65Pb0.35Sr2Ca2Cu3O10+delta (Bi-2223). The influence of the consolidation temperature, T-D, on the structural and electrical properties has been investigated and compared with those of a reference sample synthesized by the traditional solid-state reaction method and subjected to the same compacting pressure. From the X-ray diffraction patterns, performed in both powder and pellet samples, we have found that the dominant phase is the Bi-2223 in all samples but traces of the Bi2Sr2CaCu2O8+x (Bi-2212) were identified. Their relative density were similar to 85% of the theoretical density and the temperature dependence of the electrical resistivity, rho(T), indicated that increasing T-D results in samples with low oxygen content because the SPS is performed in vacuum. Features of the rho(T) data, as the occurrence of normal-state semiconductor-like behavior of rho(T) and the double resistive superconducting transition, are consistent with samples comprised of grains with shell-core morphology in which the shell is oxygen deficient. The SPS samples also exhibited superconducting critical current density at 77 K, J(c)(77K), between 2 and 10A/cm(2), values much smaller than similar to 22A/cm(2) measured in the reference sample. Reoxygenation of the SPS samples, post-annealed in air at different temperatures and times, was found to improve their microstructural and transport properties. Besides the suppression of the Bragg peaks belonging to the Bi-2212 phase, the superconducting properties of the post-annealed samples and particularly J(c)(77K) were comparable or better than those corresponding to the reference sample. Post-annealed samples at 750 degrees C for 5min exhibited J(c)(77K) similar to 130A/cm(2) even when uniaxially pressed at only 50 MPa. (C) 2012 American Institute of Physics. We have conducted magnetoresistance measurements rho(T,H) in applied magnetic fields up to 18 T in Bi1.65Pb0.35Sr2Ca2Cu3O10+delta ceramic samples which were subjected to different uniaxial compacting pressures. The anisotropic upper critical fields H (c2)(T) were extracted from the rho(T,H) data, yielding and the out-of-plane superconducting coherence length xi (c) (0)similar to 3 . We have also estimated and xi (ab) (0) similar to 90 . In addition to this, a flux-line-lattice (FLL) melting temperature T (m) has been identified as a second peak in the derivative of the magnetoresistance d rho/dT data close to the superconducting transition temperature. An H (m) vs. T phase diagram was constructed and the FLL boundary lines were found to obey a temperature dependence H (m) ae(T (c) /T-1) (alpha) , where alpha similar to 2 for the sample subjected to the higher compacting pressure. A reasonable value of the Lindemann parameter c (L) similar to 0.29 has been found for all samples studied.

Visceral leishmaniasis (VL) is a zoonotic disease characterized by infection of mononuclear phagocytes by Leishmania chagasi. The primary vector is Lutzomyia longipalpis and the dog is the main domestic reservoir. The control and current treatment of dogs using synthetic drugs have not shown effectiveness in reducing the incidence of disease in man. In attempt to find new compounds with leishmanicidal action, plant secondary metabolites have been studied in search of treatments of VL. This study aimed to evaluate the leishmanicidal activity of Musa paradisiaca (banana tree) and Spondias mombin (cajazeira) chemical constituents on promastigotes and amastigotes of L. chagasi. Phytochemical analysis by column chromatography was performed on ethanol extracts of two plants and fractions were isolated. Thin layer chromatography was used to compare the fractions and for isolation the substances to be used in vitro tests. The in vitro tests on promastigotes of L chagasi used the MTT colorimetric method and the method of ELISA in situ was used against amastigotes besides the cytotoxicity in RAW 264.7 cells. Of the eight fractions tested, Sm1 and Sm2 from S. mombin had no action against promastigotes, but had good activity against amastigotes. The fractions Mp1 e Mp4 of M. paradisiaca were very cytotoxic to RAW 264.7 cells. The best result was obtained with the fraction Sm3 from S. mombin with IC50 of 11.26 mu g/ml against promastigotes and amastigotes of 0.27 mu g/ml. The fraction Sm3 characterized as tannic acid showed the best results against both forms of Leishmania being a good candidate for evaluation in in vivo tests. (C) 2012 Published by Elsevier B.V.

We have performed Monte Carlo and molecular dynamics simulations of suspensions of monodisperse, hard ellipsoids of revolution. Hard-particle models play a key role in statistical mechanics. They are conceptually and computationally simple, and they offer insight into systems in which particle shape is important, including atomic, molecular, colloidal, and granular systems. In the high density phase diagram of prolate hard ellipsoids we have found a new crystal, which is more stable than the stretched FCC structure proposed previously . The new phase, SM2, has a simple monoclinic unit cell containing a basis of two ellipsoids with unequal orientations. The angle of inclination is very soft for length-to-width (aspect) ratio l/w=3, while the other angles are not. A symmetric state of the unit cell exists, related to the densest-known packings of ellipsoids; it is not always the stable one. Our results remove the stretched FCC structure for aspect ratio l/w=3 from the phase diagram of hard, uni-axial ellipsoids. We provide evidence that this holds between aspect ratios 3 and 6, and possibly beyond. Finally, ellipsoids in SM2 at l/w=1.55 exhibit end-over-end flipping, warranting studies of the cross-over to where this dynamics is not possible. Secondly, we studied the dynamics of nearly spherical ellipsoids. In equilibrium, they show a first-order transition from an isotropic phase to a rotator phase, where positions are crystalline but orientations are free. When over-compressing the isotropic phase into the rotator regime, we observed super-Arrhenius slowing down of diffusion and relaxation, and signatures of the cage effect. These features of glassy dynamics are sufficiently strong that asymptotic scaling laws of the Mode-Coupling Theory of the glass transition (MCT) could be tested, and were found to apply. We found strong coupling of positional and orientational degrees of freedom, leading to a common value for the MCT glass-transition volume fraction. Flipping modes were not slowed down significantly. We demonstrated that the results are independent of simulation method, as predicted by MCT. Further, we determined that even intra-cage motion is cooperative. We confirmed the presence of dynamical heterogeneities associated with the cage effect. The transit between cages was seen to occur on short time scales, compared to the time spent in cages; but the transit was shown not to involve displacements distinguishable in character from intra-cage motion. The presence of glassy dynamics was predicted by molecular MCT (MMCT). However, as MMCT disregards crystallization, a test by simulation was required. Glassy dynamics is unusual in monodisperse systems. Crystallization typically intervenes unless polydispersity, network-forming bonds or other asymmetries are introduced. We argue that particle anisometry acts as a sufficient source of disorder to prevent crystallization. This sheds new light on the question of which ingredients are required for glass formation.

Comparison of the crystal structure of a transition state analogue that was used to raise catalytic antibodies for the benzoyl ester hydrolysis of cocaine with structures calculated by ab initio, semiempirical, and solvation semiempirical methods reveals that modeling of solvation is crucial for replicating the crystal structure geometry. Both SM3 and SM2 calculations, starting from the crystal structure TSA I, converged on structures similar to the crystal structure. The 3-21G(*)/HF, 6-31G*/HF, PM3, and AM1 calculations converged on structures similar to each other, but these gas-phase structures were significantly extended relative to the condensed phase structures. Two transition states for the hydrolysis of the benzoyl ester of cocaine were located with the SM3 method. The gas phase calculations failed to locate reasonable transition state structures for this reaction. These results imply that accurate modeling of the potential energy surfaces for the hydrolysis of cocaine requires solvation methods.
