The previous uncertain placement of Lysapsus and Pseudis within the neobatrachians was recently resolved by molecular and morphological studies, which supported them as members of the Hylinae subfamily. Their inter- and intrageneric relationships, however, have long been under debate and no studies shed light on these questions. Aiming to elucidate such questions, this paper used 3.2 kb comprising the mitochondrial genes 12S, tRNA valine, 16S and cytochrome b, and the nuclear exon 1 coding for rhodopsin, to all representatives of both genera (except to two subspecies of Pseudis paradoxa). The results identified three major clades: the clade 1 was composed by Lysapsus species and subspecies; clade 2 included the subspecies of the Pseudis paradoxa (Pseudis paradoxa paradoxa, P. paradoxa platensis and P. paradoxa occidentalis), P. fusca, P. bolbodactyla and P. tocantins, and clade 3 was composed by Pseudis southern Brazil species (Pseudis cardosoi and P. minuta). As closely related taxa we found Pseudis minuta + P. cardosoi; P. tocantins + P. fusca, and the subspecies within each genus. Evidence that Pseudis is not monophyletic with respect to Lysapsus was found and we suggest Lysapsus to be a junior synonym of Pseudis. Based on pair-wise comparison among gene sequences, we also suggest that the subspecies of Pseudis paradoxa and Lysapsus limellum must be considered as full species. Systemic arterial blood pressure and heart rate (f(H)) were measured in unanesthetized, unrestrained larvae and adults of the paradoxical frog, Pseudis paradoxus from São Paulo State in Brazil. Four developmental groups were used, representing the complete transition from aquatic larvae to primarily air-breathing adults. f(H) (49-66 beats/min) was not significantly affected by development, whereas mean arterial blood pressure was strongly affected, being lowest in the stage 37-39 larvae (10 mmHg), intermediate in the stage 44-45 larvae (18 mmHg), and highest in the juveniles and adults (31 and 30 mmHg, respectively). Blood pressure was not significantly correlated with body mass, which was greatest in the youngest larvae and smallest in the juveniles. In the youngest larvae studied (stages 37-39), lung ventilation was infrequent, causing a slight decrease in arterial blood pressure but no change in heart rate. Lung ventilation was more frequent in stages 44-45 larvae and nearly continuous in juveniles and adults floating at the surface. Bradycardia during both forced and voluntary diving was observed in almost every advanced larva, juvenile, and adult but in only one of four young larvae. Developmentally related changes in blood pressure were not complete until metamorphosis, whereas diving bradycardia was present at an earlier stage.

Hylidae is a large family of American, Australopapuan, and temperate Eurasian treefrogs of approximately 870 known species, divided among four subfamilies. Although some groups of Hylidae have been addressed phylogenetically, a comprehensive phylogenetic analysis has never been presented. The first goal of this paper is to review the current state of hylid systematics. We focus on the very large subfamily Hylinae (590 species), evaluate the monophyly of named taxa, and examine the evidential basis of the existing taxonomy. The second objective is to perform a phylogenetic analysis using mostly DNA sequence data in order to (1) test the monophyly of the Hylidae; (2) determine its constituent taxa, with special attention to the genera and species groups which form the subfamily Hylinae, and c) propose a new, monophyletic taxonomy consistent with the hypothesized relationships. We present a phylogenetic analysis of hylid frogs based on 276 terminals, including 228 hylids and 48 outgroup taxa. Included are exemplars of all but 1 of the 41 genera of Hylidae (of all four nominal subfamilies) and 39 of the 41 currently recognized species groups of the species-rich genus Hyla. The included taxa allowed us to test the monophyly of 24 of the 35 nonmonotypic genera and 25 species groups of Hyla. The phylogenetic analysis includes approximately 5100 base pairs from four mitochondrial (12S, tRNA valine, 16S, and cytochrome b) and five nuclear genes (rhodopsin, tyrosinase, RAG-1, seventh in absentia, and 28S), and a small data set from foot musculature. Concurring with previous studies, the present analysis indicates that Hemiphractinae are not related to the other three hylid subfamilies. It is therefore removed from the family and tentatively considered a subfamily of the paraphyletic Leptodactylidae. Hylidae is now restricted to Hylinae, Pelodryadinae, and Phyllomedusinae. Our results support a sister-group relationship between Pelodryadinae and Phyllomedusinae, which together form the sister taxon of Hylinae. Agalychnis, Phyllomedusa, Litoria, Hyla, Osteocephalus, Phrynohyas, Ptychohyla, Scinax, Smilisca, and Trachycephalus are not monophyletic. Within Hyla, the H. albomarginata, H. albopunctata, H. arborea, H. boons, H. cinerea, H. eximia, H. geographica, H. granosa, H. microcephala, H. miotympanum, H. tuberculosa, and H. versicolor groups are also demonstrably nonmonophyletic. Hylinae is composed of four major clades. The first of these includes the Andean stream-breeding Hyla, Aplastodiscus, all Gladiator Frogs, and a Tepuian clade. The second clade is composed of the 30-chromosome Hyla, Lysapsus, Pseudis, Scarthyla, Scinax (including the H. uruguaya group), Sphaenorhynchus, and Xenohyla. The third major clade is composed of Nyctimantis, Phrynohyas, Phyllodytes, and all South American/West Indian casque-headed frogs: Aparasphenodon, Argenteohyla, Corythomantis, Osteocephalus, Osteopilus, Tepuihyla, and Trachycephalus. The fourth major clade is composed of most of the Middle American/Holarctic species groups of Hyla and the genera Acris, Anotheca, Duellmanohyla, Plectrohyla, Pseudacris, Ptychohyla, Pternohyla, Smilisca, and Triprion. A new monophyletic taxonomy mirroring these results is presented where Hylinae is divided into four tribes. Of the species currently in Hyla, 297 of the 353 species are placed in 15 genera; of these, 4 are currently recognized, 4 are resurrected names, and 7 are new. Hyla is restricted to H. femoralis and the H. arborea, H. cinerea, H. eximia, and H. versicolor groups, whose contents are redefined. Phrynohyas is placed in the synonymy of Trachycephalus, and Pternohyla is placed in the synonymy of Smilisca. The genus Dendropsophus is resurrected to include all former species of Hyla known or suspected to have 30 chromosomes. Exerodonta is resurrected to include the former Hyla sumichrasti group and some members of the former H. miotympanum group. Hyloscirtus is resurrected for the former Hyla armata, H. bogotensis, and H. larinopygion groups. Hypsiboas is resurrected to include several species groups - many of them redefined here - of Gladiator Frogs. The former Hyla albofrenata and H. albosignata complexes of the H. albomarginata group are included in Aplastodiscus. New generic names are erected for (1) Agalychnis calcarifer and A. craspedopus; (2) Osteocephalus langsdorffii; the (3) Hyla aromatica, (4) H. bromeliacia, (5) H. godmani, (6) H. mixomaculata, (7) H. taeniopus, (8) and H. tuberculosa groups; (9) the clade composed of the H. pictipes and H. pseudopuma groups; and (10) a clade composed of the H. circumdata, H. claresignata, H. martinsi, and H. pseudopseudis groups. The objective of this work was to make a comparative analysis of germ cell organization at different stages of cellular differentiation in adult males of Dendropsophus nanus (Boulenger, 1889), Pseudis limellum (Cope, 1862), P. paradoxa (Linnaeus, 1758), and Scinax acuminatus (Cope, 1862), belonging to the family Hylidae; and Leptodactylus chaquensis (Cei, 1950) and L. podicipinus (Cope, 1862), belonging to the family Leptodactylidae, collected in the Pantanal and in Serra da Bodoquena, Mato Grosso do Sul State, Brazil. The testes were removed and fixed, dehydrated in a graded series of ethanol and embedded in methacrylate glycol resin (Historesin Leica®). The sections were stained by 1% toluidine blue and observed under light microscope. It was detected that all individual of the Hylidae family show, throughout the year, the presence of all germ cell types of spermatogenesis. However, all Leptodactylidae family individuals only show the presence of all germ cell types during the rainy season. The variations of characteristics in seminiferous epithelium organization, as well as the evident difference in the amount of spermatozoa inside the tubules, are evidence that the anurans in this work show different forms of spermatogenesis development throughout the year: the cycle is continuous for the Hylidae family, and discontinuous with explosive release of spermatozoa for the Leptodactylidae family.

Embora exista uma grande diversidade de complementos cromossômicos em Leptodactylidae (2n = 18 a 2n = 26) e Hylidae (2n = 20 a 2n = 32), a elevada fragmentação de dados limita o acesso a informações sobre as origens e os mecanismos responsáveis por esta diversidade. Isto provavelmente tem influenciado que os dados citogenéticos tenham sido principalmente utilizados na caracterização do status de espécies mais do que incluídos amplamente em análises filogenéticas. Este trabalho aborda, por meio de dados citogenéticos, aspectos evolutivos de três grandes grupos de anuros de ampla distribuição na região Neotropical. O gênero Leptodactylus é agrupado com Hydrolaetare, Paratelmatobius e Scythrophrys na família Leptodactylidae. Os antecedentes cromossômicos neste gênero indicam variações nos números diplóides de 2n = 18 a 2n = 26, assim como variações nos números fundamentais (número de braços autossômicos, NF) e nas posições das Regiões Organizadoras do Nucléolo (NOR). Os resultados das análises de 26 espécies de Leptodactylus empregando diversas técnicas representa, provavelmente, a análise citogenética mais inclusiva realizada no gênero Leptodactylus até o momento, e os resultados constituem um marco para a proposição de hipóteses consistentes de evolução cromossômica no gênero. A tribo Lophyiohylini agrupa atualmente 81 espécies distribuídas em 10 gêneros. A informação citogenética é escassa e restrita apenas a 12 espécies. São aqui apresentados comparativamente dados citogenéticos em espécies dos gêneros Argenteohyla, Itapotihyla, Phyllodytes, Trachycephalus e Osteocephalus. Os resultados indicam que, com exceção de O. buckleyi (2n = 26; NF = 50) e P. edelmoi (2n = 22; NF = 44), todas as demais espécies analisadas coincidem com os dados citogenéticos disponíveis, que indicam um 2n = 24 (NF = 48) na maioria das espécies cariotipadas, com NOR e constrições secundarias (CS) localizadas no par 11. Entretanto, em Phyllodytes edelmoi e Argentohyla siemersi pederseni, essas regiões localizam-se nos pares 2 e 5, respectivamente. Blocos heterocromáticos foram associados às CS adicionais (sítios frágeis) em Osteocephalus, mas não em Trachycephalus. Dados citogenéticos nos gêneros Nyctimantis e Tepuihyla, assim como técnicas com maior poder de resolução e estudos mais inclusivos, são necessários para compreender melhor a evolução cromossômica da tribo. A tribo Dendropsophini atualmente agrupa os gêneros Scinax, Pseudis, Scarthyla, Sphaenorhynchus, Xenohyla e Dendropsophus. Os dados citogenéticos registrados em todos os gêneros revelaram uma elevada diversidade cariotípica com grandes variações nos números diplóides (2n = 22 em Scarthyla; 2n = 24 em Scinax e Xenohyla; 2n = 24, 24 +1-2B e 26 em Sphaenorhynchus; 2n = 24 e 28 em Pseudis; e, 2n = 30 em Dendropsophus). O 2n = 24 observado em X. truncata indica que o 2n = 30constitui uma sinapomorfia do gênero Dendropsophus. A localização das NOR no par 7 é uma característica compartilhada por espécies dos gêneros Scarthyla, Xenohyla, Pseudis e Sphaenorhynchus, com algumas exceções nos dois últimos (P. caraya e S. carneus). Entretanto, o gênero Dendropsophus exibe uma interessante diversidade em relação a número e localização das NOR. Por outro lado, a distribuição de heterocromatina apresentou padrões variáveis, particularmente gênero Pseudis. Embora exista uma excepcional variação cromossômica neste grupo, a informação fragmentária em alguns gêneros dificulta a formulação de hipóteses consistentes sobre o papel dos cromossomos na evolução do grupo.

A helmintofauna tem papel central no ecossistema, pois afeta a ecologia, evolução, crescimento e regulação da população de hospedeiros e, consequentemente, toda a biodiversidade. Neste estudo foram selecionados anuros pertencentes a uma guilda, de acordo com o modo reprodutivo e sítio de vocalização, e foi estudada a supracomunidade de parasitas associadas a estes anfíbios. Em janeiro de 2009 foram coletados 60 anuros no Pantanal Sul- Matogrossense, sub-região Miranda-Abobral no município de Corumbá, MS. As espécies de anuros coletadas foram Rhinella granulosa, R. schineideri, Pseudis paradoxa, P. limellum, Pleurodema fuscomaculata e Physalaemus albonotatus. Os anuros foram necropsiados e analisados quanto à presença de helmintos. Dezesseis espécies de helmintos foram identificadas em 50 indivíduos hospedeiros (prevalência total = 83,3%). Dentre os parasitas foram encontradas três espécies de trematódeos (Catadiscus marinholutzi, Catadiscus propinquus e Glypthelmins palmipedis), uma espécie de cestoda, Cylindrotaenia americana, dez espécies de nematóides (Aplectana sp., Contracecum sp.,Cosmocerca podicipinus, Cosmocercidae, Falcaustra mascula, Physaloptera sp., Ochoterenella sp., Oswaldocruzia subauricularis, Physalopteroides venancioi sp. e Rhabdias sp.) e cistos de acantocéfalo. Os trematódeos foram encontrados apenas em P. paradoxa e P. albonotatus, enquanto C. americana foi encontrada apenas em P. fuscomaculata. As espécies Aplectana sp. e Falcaustra máscula foi encontrada apenas em R. schineideri. O único indivíduo encontrado em P. limellum foi da espécie Physalopteroides venancoi. As suprapopulações de parasitas se mostraram mais agregadas em R. granulosa e P. paradoxa, onde seus hábitos de vida e hábitos alimentares podem influenciar na comunidade parasitária. The amphibian antimicrobial peptide pseudin-2 is a peptide derived from the skin of the South-American frog Pseudis paradoxa (Olson et al., 2001). This peptide possesses tremendous potential as a therapeutic lead since it has been shown to possess both antimicrobial as well insulin-releasing properties (Olson et al., 2001; Abdel-Wahab et al., 2008). This study aimed to develop pseudin-2’s potential by understanding and improving its properties as an antimicrobial agent. The structure-function relationships of pseudin-2 were explored using a combination of in-vitro and in-silico techniques, with an aim to predict how the structure of the peptide may be altered in order to improve its efficacy. A library of pseudin-2 mutants was generated by randomizing codons at positions 10, 14 and 18 of a synthetic gene, using NNK saturation mutagenesis. Analysis of these novel peptides broadly confirmed, in line with literature precedent, that anti-microbial activity increases with increased positive charge. Specifically, 2 positively-charged residues at positions 10 and 14 and a hydrophobic at position 18 are preferred. However, substitution at position 14 with some polar, non-charged residues also created peptides with antimicrobial activity. Interestingly, the pseudin-2 analogue [10-E, 14-Q, 18-L] which is identical to pseudin-2, except that the residues at positions 10 and 14 are switched, showed no anti-microbial activity at all. Molecular dynamics simulations of pseudin-2 showed that the peptide possesses two equilibrium structures in a membrane environment: a linear and a kinked a-helix which both embed into the membrane at an angle. Biophysical characterization using circular dichroism spectroscopy confirmed that the peptide is helical within the membrane environment whilst linear dichroism established that the peptide has no defined orientation within the membrane. Collectively, these data indicate that Pseudin-2 exerts its antimicrobial activity via the carpet model.
