Schistosomiasis is one of the world`s greatly neglected tropical diseases, and its control is largely dependent on a single drug, praziquantel. Here, we report the in vitro effect of piplartine, an amide isolated from Piper tuberculatum (Piperaceae), on Schistosoma mansoni adult worms. A piplartine concentration of 15.8 mu M reduced the motor activity of worms and caused their death within 24 h in a RPMI 1640 medium. Similarly, the highest sub-lethal concentration of piplartine (6.3 mu M) caused a 75% reduction in egg production in spite of coupling. Additionally, piplartine induced morphological changes on the tegument, and a quantitative analysis carried out by confocal microscopy revealed an extensive tegumental destruction and damage in the tubercles. This damage was dose-dependent in the range of 15.8-630.2 mu M. At doses higher than 157.6 mu M, piplartine induced morphological changes in the oral and ventral sucker regions of the worms. It is the first time that the schistosomicidal activity has been reported for piplartine. Published by Elsevier Inc.

Piplartine (PPTN) is an alkaloid amide found in Piper species that presents different activities. PPTN determination in rat plasma is necessary to better understand its biological effects. The aim of this study was to develop a sensitive LC-MS/MS method for the determination of PPTN in rat plasma. The performance criteria for linearity, sensitivity, precision, accuracy, recovery, and stability have been assessed and were within the recommended guidelines. The validated method proved to be suitable in a pilot study of PPTN kinetic disposition in rat plasma after a single intraperitoneal dose, and represents an appropriate tool to further pharmacokinetic studies.

Schistosomiasis is one of the most important parasitic infections in humans that occur in many tropical and subtropical countries. Currently, the control of schistosomiasis rests with a single drug, praziquantel, which is effective against adult worms but not the larval stages. Recent studies have shown that piplartine, an amide isolated from plants of the genus Piper (Piperaceae), reveals interesting antischistosomal properties against Schistosoma mansoni adult worms. Here, we report the in vitro antischistosomal activity of piplartine on S. mansoni schistosomula of different ages (3 h old and 1, 3, 5, and 7 days old), and examine alterations on the tegumental surface of worms by means of confocal laser scanning microscopy. Piplartine at a concentration of 7.5 mu M caused the death of all schistosomula within 120 h. The lethal effect occurred in a dose-dependent manner and was also dependent on the age of the parasite. Microscopy observation revealed extensive tegumental destruction, including blebbing, granularity, and a shorter body length. This report provides the first evidence that piplartine is able to kill schistosomula of different ages and reinforce that piplartine is a promising compound that could be used for the development of new schistosomicidal agent. A rapid, sensitive and reliable reverse-phase HPLC method was used for the quantitative determination of the anti-fungal and insecticide amides, dihydropiplartine (1), piplartine (2), Delta(alpha,beta)-dihydropiperine (3) and pellitorine (4) in plants in natura, in plantlets in vitro and ex vitro, and in callus of Piper tuberculatum. Well-resolved peaks were obtained with good detection response and linearity in the range of 15.0-3000 mug/mL. The plants in natura contained compounds 1-4, the plantlets ex vitro and in vitro accumulated compounds 1-2 and 1-4, respectively, while only amide 4 was found in callus. O estudo químico das folhas e dos frutos de P. richardiaefolium resultou no isolamento de oito lignanas, sendo duas lignanas furofurânicas (sesamina e kobusina), quatro lignanas dibenzilbutirolactônicas (hinokinina, kusunokinina, arctigenina e haplomirfolina), duas lignanas dibenzilbutirolactólicas (cubebina e 3,4- dimetoxi-3,4-desmetilenodioxicubebina), dois cinamatos de bornila (ferulato de bornila e cumarato de bornila) e na identificação de duas amidas (piplartina e diidropiplartina). Das folhas de P. richardiaefolium foi extraído e analisado o óleo volátil. As estruturas das substâncias isoladas foram identificadas através de métodos espectroscópicos (RMN de 1H e de 13C e espectrometria de massas). O estudo de análise de componentes principais (PCA) das espécies Piper (P. truncatum - k 616, P. richardiaefolium - k 290, P. richardiaefolium - k 350, P. richardiaefolium - k 593, P. truncatum - k 597, P. pseudopotifolium - k 598, P. richardiaefolium - k 854, P. richardiaefolium - k 610, P. truncatum - k 112, P. pseudopotifolium - k 211 e P. cernuum - k 137) permitiu agrupar as espécies em dois grandes grupos e quatro subgrupos em relação à similaridade entre elas. Ligninas do caule de seis espécies de Piper foram extraídas utilizando o método de degradação de Klason e método de Bjorkman, e analisadas por métodos espectroscópicos (IV, RMN de 1H e de 13C). O método de degradação por oxidação por nitrobenzeno foi o escolhido para determinar a relação entre os monolignóis siringila e guaiacila. Os principais metabólitos das espécies estudadas foram comparados com os tipos de ligninas das mesmas espécies e os resultados sugeriram uma independência entre as vias biossintéticas de ligninas e lignanas.
