The relationship between the level of cell confluence near the plateau phase of growth and blastocyst yield following somatic cell cloning is not well understood. We examined the effect of distinct cell culture confluence levels on in vitro development of cloned bovine embryos. In vitro-matured bovine oocytes were manually bisected and selected by DNA staining. One or two enucleated hemi-cytoplasts were paired and fused with an adult skin somatic cell. Cultured skin cells from an adult Nellore cow harvested at three distinct culture confluence levels (70-80, 80-90, and > 95%) were used for construction of embryos and hemi-embryos. After activation, structures were cultured in vitro as one embryo (1 x 100%) or as aggregates of two hemi-embryos (2 x 50%) per microwell. Fusion, cleavage and blastocyst rates were compared using the chi(2) test. The fusion rate for hemi-embryos (51.4%) was lower than for embryos (67.6%), with no influence of degree of cell confluence. However, blastocyst rates improved linearly (7.0, 17.5, and 29.4%) with increases in cell confluence. We conclude that degree of cell culture confluence significantly influences subsequent embryo development; use of a cell population in high confluence (> 90%) for nuclear transfer significantly improved blastocyst yield after cloning.

Reproductive failures are still common grounds for complaint by commercial swine producers. Porcine parvovirus (PPV) is associated with different clinical reproductive signs. The aim of the present study was to investigate PPV fetal infection at swine farms having ongoing reproductive performance problems. The presence of virus in fetal tissues was determined by nested-polymerase chain reaction assay directed to the conserved NS1 gene of PPV in aborted fetuses, mummies and stillborns. Fetuses show a high frequency of PPV infection (96.4%; N = 28). In 60.7% of the fetuses, PPV were detected in all tissue samples (lung, heart, thymus, kidney, and spleen). Viral infection differed among fetal tissues, with a higher frequency in the lung and heart (P < 0.05). Fetuses with up to 99 days of gestational age and from younger sows showed a higher frequency of PPV (P < 0.05). No significant difference in the presence of PPV was detected among the three clinical presentations. The results suggest that PPV remains an important pathogenic agent associated with porcine fetal death.

Cell cycle synchronization by serum starvation (SS) induces apoptosis in somatic cells. This side effect of SS is hypothesized to negatively affect the outcome of somatic cell nuclear transfer (SCNT). We determined whether apoptotic fibroblasts affect SCNT yields. Serum-starved, adult, bovine fibroblasts were stained with annexin V-FITC/propidium iodide to allow apoptosis detection by flow cytometry. Positive and negative cells sorted by fluorescence activated cell sorting (FACS) and an unsorted control group were used as nuclear donors for SCNT. Reconstructed embryos were cultured in vitro and transferred to synchronized recipients. Apoptosis had no effect on fusion and cleavage rates; however, it resulted in reductions in blastocyst production and quality measured by apoptotic index. However, reconstructed embryos with apoptotic cells resulted in pregnancy rates similar to that of the control on day 30, and generated one live female calf. In conclusion, we showed that apoptotic cells present in serum-starved cultures negatively affect embryo production after SCNT without compromising full-term development. Further studies will evaluate the ability of the oocyte to reprogram cells in specific phases of apoptosis.

Engineering of fetal tissue has a high potential for the treatment of acute and chronic wounds of the skin in humans as these cells have high expansion capacity under simple culture conditions and one organ donation can produce Master Cell Banks which can fabricate over 900 million biological bandages (9 x 12cm). In a Phase 1 clinical safety study, cases are presented for the treatment of therapy resistant leg ulcers. All eight patients, representing 13 ulcers, tolerated multiple treatments with fetal biological bandages showing no negative secondary effects and repair processes similar to that seen in 3rd degree burns. Differential gene profiling using Affymetrix gene chips (analyzing 12,500 genes) were accomplished on these banked fetal dermal skin cells compared to banked dermal skin cells of an aged donor in order to point to potential indicators of wound healing. Families of genes involved in cell adhesion and extracellular matrix, cell cycle, cellular signaling, development and immune response show significant differences in regulation between banked fetal and those from banked old skin cells: with approximately 47.0% of genes over-expressed in fetal fibroblasts. It is perhaps these differences which contribute to efficient tissue repair seen in the clinic with fetal cell therapy.

The present study examined the in vitro and in vivo development of bovine nuclear-transferred embryos. A bovine fetal fibroblast culture was established and used as nucleus donor. Slaughterhouse oocytes were matured in vitro for 18 h before enucleation. Enucleated oocytes were fused with fetal fibroblasts with an electric stimulus and treated with cytochalasin D and cycloheximide for 1 h followed by cycloheximide alone for 4 h. Reconstructed embryos were cultured for 7-9 days and those which developed to blastocysts were transferred to recipient cows. Of 191 enucleated oocytes, 83 (43.5%) were successfully fused and 24 (28.9%) developed to blastocysts. Eighteen freshly cloned blastocysts were transferred to 14 recipients, 5 (27.8%) of which were pregnant on day 35 and 3 (16.7%) on day 90. Of the three cows that reached the third trimester, one recipient died of hydrallantois 2 months before term, one aborted fetus was recovered at 8 months of gestation, and one delivered by cesarian section a healthy cloned calf. Today, the cloned calf is 15 months old and presents normal body development (378 kg) and sexual behavior (libido and semen characteristics).

Cell death by apoptosis is considered to be irreversible. However, reports have indicated that its reversibility is possible if the cells have not yet reached the "point of no return.'' In order to add new information about this topic, we used cells at different moments of apoptotic process as nuclear donors in somatic cell nuclear transfer (SCNT) in order to test if programmed cell death can be reversed. Adult bovine fibroblasts were treated with 10 mu M of staurosporine (STP) for 3 h and analyzed for phosphatidylserine externalization (Annexin assay) and presence of active caspase-9. Annexin-positive (Anx +) and Caspase-9-positive (Casp-9 +) cells were isolated by FACS and immediately transferred into enucleated in vitro matured bovine oocytes. After STP treatment, 89.9% of cells were Anx + (4.6% in control cells; p < 0.01) and 24.9% were Casp-9 + (2.4% in control cells; p < 0.01). Fusion and cleavage were not affected by the use apoptotic cells (p > 0.05). Also, the use of Anx + cells did not affect blastocyst production compared to control (26.4% vs. 22.9%, respectively; p > 0.05). However, blastocyst formation was affected by the use of Casp-9 + cells (12.3%; p < 0.05). These findings contribute to the idea of that apoptosis is reversible only at early stages. Additionally, we hypothesize that the "point of no return'' for apoptosis may be located around activation of Caspase-9.

Animal cloning by nuclear transfer (NT) has made the production of transgenic animals using genetically modified donor cells possible and ensures the presence of the gene construct in the offspring. The identification of transgene insertion sites in donor cells before cloning may avoid the production of animals that carry undesirable characteristics due to positional effects. This article compares blastocyst development and competence to establish pregnancies of bovine cloned embryos reconstructed with lentivirus-mediated transgenic fibroblasts containing either random integration of a transgene (random integration group) or nuclear transfer derived transgenic fibroblasts with known transgene insertion sites submitted to recloning (recloned group). In the random integration group, eGFP-expressing bovine fetal fibroblasts were selected by fluorescence activated cell sorting (FACS) and used as nuclei donor cells for NT. In the recloned group, a fibroblast cell line derived from a transgenic cloned fetus was characterized regarding transgene insertion and submitted to recloning. The recloned group had higher blastocyst production (25.38 vs. 14.42%) and higher percentage of 30-day pregnancies (14.29 vs. 2.56%) when compared to the random integration group. Relative eGFP expression analysis in fibroblasts derived from each cloned embryo revealed more homogeneous expression in the recloned group. In conclusion, the use of cell lines recovered from transgenic fetuses after identification of the transgene integration site allowed for the production of cells and fetuses with stable transgene expression, and recloning may improve transgenic animal yields.

Trisomy 21 is the most frequent genetic cause of cognitive impairment. To assess the perturbations of gene expression in trisomy 21, and to eliminate the noise of genomic variability, we studied the transcriptome of fetal fibroblasts from a pair of monozygotic twins discordant for trisomy 21. Here we show that the differential expression between the twins is organized in domains along all chromosomes that are either upregulated or downregulated. These gene expression dysregulation domains (GEDDs) can be defined by the expression level of their gene content, and are well conserved in induced pluripotent stem cells derived from the twins' fibroblasts. Comparison of the transcriptome of the Ts65Dn mouse model of Down's syndrome and normal littermate mouse fibroblasts also showed GEDDs along the mouse chromosomes that were syntenic in human. The GEDDs correlate with the lamina-associated (LADs) and replication domains of mammalian cells. The overall position of LADs was not altered in trisomic cells; however, the H3K4me3 profile of the trisomic fibroblasts was modified and accurately followed the GEDD pattern. These results indicate that the nuclear compartments of trisomic cells undergo modifications of the chromatin environment influencing the overall transcriptome, and that GEDDs may therefore contribute to some trisomy 21 phenotypes.

La nature a développé diverses stratégies afin d’assurer le commencement de la vie dans des conditions d’homoplasmie, c’est-à-dire des conditions telles que les cellules sont dotées du même ADN mitochondrial. Toutefois, des nouveaux haplotypes de l’acide désoxyribonucléique mitochondrial (ADNmt) peuvent apparaitre et croître de plusieurs façons tout au long de la durée d’une vie menant à l’hétéroplasmie. Par exemple, l’hétéroplasmie de l’ADNmt peut être créée artificiellement par des technologies reproductives assistées, ainsi que naturellement par le processus de vieillissement. De ce fait, la thèse de ce doctorat fut divisée en deux principaux objectifs. Le premier étant celui d’analyser les changements survenus dans l’hétéroplasmie de l’ADNmt produit par le transfert nucléaire des cellules somatiques (SCNT) lors du développement de l’embryon jusqu’au fœtus et aux tissus adultes de bovins clonés. En ce qui concerne le second objectif, il s’agit d’analyser les changements survenus dans l’hétéroplasmie de l’ADNmt causés par le vieillissement dans une cellule somatique adulte et dans des tissus germinaux durant l’ovogénèse, ainsi qu’au début de l’embryogenèse et dans la procédure de culture in vitro sur des souris. Dans la première série d’expériences sur des bovins, des fibroblastes fœtaux transportant une mutation d’ADNmt (insertion de 66 pb) furent fusionnés avec des ovocytes receveurs transportant l’ADNmt du type sauvage. La présence d’ADNmt venant de la cellule donneuse a été analysée à différents stades de développement, soit sur des embryons âgés de 17 jours (n=17), des fœtus âgés de 40 jours (n=3), des fœtus âgés de 60 jours (n=3), un fœtus âgé de 240 jours et 3 clones post-nataux âgés de 18 à 24 mois. Chaque individu s’est avéré être hétéroplasmique et 99 % (103/104) des échantillons de tissus analysés étaient également hétéroplasmiques. Cependant, l’ovaire venant du fœtus de 240 jours fut le seul à être homoplasmique pour l’ADNmt de l’ovocyte receveur. Dans la plupart des échantillons analysés (95,2 %, soit 99/104) la moyenne d’hétéroplasmie était de 1,46 %. Par contre, un fœtus âgé de 40 jours a présenté un niveau élevé d’hétéroplasmie (20,9 %), indiquant ainsi que des évènements rares d’augmentation de l’ADNmt des cellules donneuses peuvent survenir. Étant donné que la majorité des clones SCNT montrait de l’hétéroplasmie de l’ADNmt à des proportions comparables à celles des cellules donneuses au moment de la reconstruction de l’embryon, on a pu conclure que l’hétéroplasmie produite par des techniques de transfert nucléaire utilisant des cellules somatiques est due à une ségrégation neutre de l’ADNmt. Dans la seconde série d’expériences sur des souris, des femelles de différents âges, c.à.d. jeunes (0 – 8 mois), moyennes (8 – 16 mois) et vieilles (16 – 24 mois), ont été synchronisées (gonadotrophines) et sacrifiées dans le but d’obtenir des ovocytes au stade de vésicule germinal, et des ovocytes au stade métaphase-II produits in vivo et in vitro. De plus, des embryons in vivo et in vitro au stade de deux-cellules et des embryons au stade de blastocystes ont été obtenus de femelles jeunes. Différents tissus somatiques, venant de femelles des trois stades d’âge ont été obtenus : cerveau, foie, muscle et du cumulus ovocytaire. De plus, l’effet du vieillissement a été mesuré selon la fertilité de la femelle. En effet, les effets sur l’hétéroplasmie du vieillissement, du stade de développement et de la culture in vitro ont été mesurés dans des ovocytes et dans des embryons. Les effets du vieillissement sur les mitochondries ont été mesurés par rapport au nombre total de copies de l’ADNmt, au pourcentage des délétions communes et sur l’expression de trois gènes : Ndufs4, Mt-nd2 and Mt-nd4. Il a été possible d’observer que la fertilité des femelles dans la colonie de souris diminuait avec l’âge. En fait, le vieillissement affectait l’ADNmt dans les tissus somatiques, cependant il n’avait pas d’effet sur le cumulus, les ovocytes et les embryons. Le nombre de délétions de l’ADNmt augmentait pendant la reprise de la méiose et celui-ci diminuait au début du développement embryonnaire. La culture in vitro n’affectait pas la quantité d’ADNmt dans la plupart des tissus germinaux. Puisque nous n’avons pas trouvé d’effet de l’âge dans la majorité des paramètres mitochondriaux analysés dans les ovocytes et les embryons, il est suggéré que la délétion commune de l’ADNmt dans les tissus germinaux est davantage reliée au statut cellulaire de la production d’énergie qu’au processus de vieillissement. Deux sources différentes de mutations de l’ADNmt produites dans les ovocytes normaux ou reconstitués ont produit différents résultats d’hétéroplasmie au début de l’embryogénèse. Chez les bovins, l’hétéroplasmie artificielle impliquant une petite insertion (66 pb) dans la région non codante (D-loop) de l’ADNmt a été vraisemblablement non nocive pour l’embryon, tolérant la persistance de l’ADNmt étranger pendant les différents stades du développement des clones. Chez les souris, l’hétéroplasmie naturelle produite par une grande délétion (4974 pb délétion commune) dans la région codante de l’ADNmt a été vraisemblablement nocive pour l’embryon et par conséquent éliminée pour assurer l’homoplasmie au début du développement embryonnaire.

Background: The in vitro production (IVP) of embryos by in vitro fertilization or cloning procedures has been known to cause epigenetic changes in the conceptus that in turn are associated with abnormalities in pre- and postnatal development. Handmade cloning (HMC) procedures and the culture of zona-free embryos in individual microwells provide excellent tools for studies in developmental biology, since embryo development and cell allocation patterns can be evaluated under a wide range of embryo reconstruction arrangements and in in vitro embryo culture conditions. As disturbances in embryonic cell allocation after in vitro embryo manipulations and unusual in vivo conditions during the first third of pregnancy appear to be associated with large offspring, embryo aggregation procedures may allow a compensation for epigenetic defects between aggregated embryos or even may influence more favorable cell allocation in embryonic lineages, favoring subsequent development. Thus, the aim of this study was to evaluate in vitro embryo developmental potential and the pattern of cell allocation in blastocysts developed after the aggregation of handmade cloned embryos produced using syngeneic wild type and/or transgenic somatic cells. Materials, Methods & Results: In vitro-matured bovine cumulus-oocyte complexes (COC) were manually bisected after cumulus and zona pellucida removal; then, two enucleated hemi-oocytes were paired and fused with either a wild type (WT) or a GFP-expressing (GFP) fetal skin cell at the 11th and 19th passages, respectively. Following chemical activation, reconstructed cloned embryos and zona-free parthenote embryos were in vitro-cultured in microwells, for 7 days, either individually (1 x 100%) or after the aggregation of two structures (2 x 100%) per microwell, as follows: (G1) one WT cloned embryo; (G2) two aggregated WT embryos; (G3) one GFP cloned embryo; (G4) two aggregated GFP embryos; (G5) aggregation of a WT embryo and a GFP embryo; (G6) one parthenote embryo; or (G7) two aggregated parthenote embryos. Fusion (clones), cleavage (Day 2), and blastocyst (Day 7) rates, and embryonic cell allocation were compared by the. 2 or Fisher tests. Total cell number (TCN) in blastocysts was analyzed by the Student's test (P < 0.05). Fusion and cleavage rates, and cell allocation were similar between groups. On a per WOW basis, development to the blastocyst stage was similar between groups, except for lower rates of development seen in G3. However, when based on number of embryos per group (one or two), blastocyst development was higher in G1 than all other groups, which were similar between one another. Cloned GFP embryos had lower in vitro development to the blastocyst stage than WT embryos, which had more TCN than parthenote or aggregated chimeric WT/GFP embryos. Aggregated GFP embryos had fewer cells than the other embryo groups. Discussion: The in vitro development of GFP cloned embryos was lower than WT embryos, with no effects on cell allocation in resulting blastocysts. Differences in blastocyst rate between groups were likely due to lower GFP-expressing cell viability, as GFP donor cells were at high population cell doublings when used for cloning. On a per embryo basis, embryo aggregation on Day 1 resulted in blastocyst development similar to non-aggregated embryos on Day 7, with no differences in cell proportion between groups. The use of GFP-expressing cells was proven a promising strategy for the study of cell allocation during embryo development, which may assist in the elucidation of mechanisms of abnormalities after in vitro embryo manipulations, leading to the development of improved protocols for the in vitro production (IVP) of bovine embryos.

Die Kontrolle der produktiven Cytomegalovirus- (CMV) Infektion ist von der effizienten Rekonstitution antiviraler CD8 T-Zellen abhängig. Dies führt jedoch nicht zur vollständigen Eliminierung des viralen Genoms aus den Zielorganen, sondern das Virus verbleibt in einem nicht-replikativen Zustand: der Latenz. Es ist bekannt, dass während der Latenz nur ein geringer Anteil latenter mCMV-Genome in der Lunge die Major Immediate Early (MIE) Gene ie1 und ie2 exprimiert, die Latenz aber dennoch bestehen bleibt, weil das differentielle Splicing des primären IE1/3-Transkripts zum Transaktivator-Transkript IE3 nicht erfolgt. Damit war neben der Initiation der IE-Genexpression am MIE-Promotor-Enhancer das IE1/3-Splicing als zweiter molekularer Latenz-Kontrollpunkt identifiziert. Parallel zur Latenz-assoziierten IE1-Genexpression sind in der Lunge aktivierte CD62L-low CD8 T-Zellen mit Spezifität für das immundominante IE1-Peptid 168-YPHFMPTNL-176 angereichert. Dies legte die Hypothese nahe, dass neben der molekularen Kontrolle der Latenz auch eine immunologische Kontrolle, beispielsweise durch IE1-Epitop-spezifische CD8 T-Zellen besteht. Zur Evaluierung dieser Hypothese wurde in der vorliegenden Arbeit mittels BAC-Mutagenese erstmals ein rekombinantes mCMV generiert, in dem das IE1-Peptid durch Punktmutation der C-terminalen MHC-Ankeraminosäure L176A zerstört ist. Dazu musste zunächst die Technik der BAC-Mutagenese herpesviraler Genome (in Anlehnung an die publizierten Arbeiten von Messerle et al., 1997; Borst et al., 1999, 2004; Wagner et al., 1999) in der Arbeitsgruppe etabliert werden. Neben der Funktionsverlust-Mutante (mCMV-IE1-L176A) wurden zur Kontrolle zwei Revertanten (mCMV-IE1-A176L und mCMV-IE1-A176L*) generiert. In letzterer, als Wobble-Revertante bezeichnet, wird wieder die authentische MHC-Ankeraminosäure L eingesetzt, es verbleibt aber ein singulärer Nukleotidaustausch A->T in der Wobble-Position des Codons als Marker zur Unterscheidung zum WT-mCMV zurück. Der immunologische Phänotyp der Funktionsverlust-Mutante, also die funktionelle Auslöschung des antigenen IE1-Peptids im Priming einer CD8 T-Zell-Antwort, entsprach der Erwartung. Entsprechend konnte nach Infektion mit der Funktionsverlust-Mutante keine Reaktivität gegen das IE1-Peptid nachgewiesen werden. In den Revertanten hingegen war die Erkennung des IE1-Peptids wieder hergestellt. Die Ergebnisse dieser Arbeit zeigen weiter, dass die Funktionsverlust-Mutante sowie die Revertanten ohne signifikante Beeinflussung in vitro in permissiven Fibroblasten und in vivo in verschiedenen Geweben replizieren. Wie aktuelle Daten nach Knochenmarktransplantation und Infektion mit der Funktionsverlust-Mutante im Vergleich zu den Revertanten zeigen, ist die Frequenz Latenz-assoziierter IE1-Transkriptionsereignisse bei der Funktionsverlust-Mutante signifikant erhöht. Damit konnte erstmalig der Beweis für eine Kontrolle der Latenz-assoziierten IE1-Genexpression durch IE1-Epitop-spezifische CD8 T-Zellen und damit für eine Präsentation des IE1-Peptids während der Latenz erbracht werden.

Objective. The relationship of multipotent mesenchymal stromal cells (MSC) with pericytes and fibroblasts has not been established thus far, although they share many markers of primitive marrow stromal cells and the osteogenic, adipogenic, and chondrogenic differentiation potentials. Materials and Methods. We compared MSCs from adult or fetal tissues, MSC differentiated in vitro, fibroblasts and cultures of retinal pericytes obtained either by separation with anti-CD146 or adhesion. The characterizations included morphological, immunophenotypic, gene-expression profile, and differentiation potential. Results. Osteogenic, adipocytic, and chondrocytic differentiation was demonstrated for MSC, retinal perivascular cells, and fibroblasts. Cell morphology and the phenotypes defined by 22 markers were very similar. Analysis of the global gene expression obtained by serial analysis of gene expression for 17 libraries and by reverse transcription polymerase chain reaction of 39 selected genes from 31 different cell cultures, revealed similarities among MSC, retinal perivascular cells, and hepatic stellate cells. Despite this overall similarity, there was a heterogeneous expression of genes related to angiogenesis, in MSC derived from veins, artery, perivascular cells, and fibroblasts. Evaluation of typical pericyte and MSC transcripts, such as NG2, CD146, CD271, and CD140B on CD146 selected perivascular cells and MSC by real-time polymerase chain reaction confirm the relationship between these two cell types. Furthermore, the inverse correlation between fibroblast-specific protein-1 and CD146 transcripts observed on pericytes, MSC, and fibroblasts highlight their potential use as markers of this differentiation pathway. Conclusion. Our results indicate that human MSC and pericytes are similar cells located in the wall of the vasculature, where they function as cell sources for repair and tissue maintenance, whereas fibroblasts are more differentiated cells with more restricted differentiation potential. This study investigated the effect of porcine follicular fluid (PFF) and dibutyryl cyclic adenosine monophosphate (dbcAMP) during in vitro maturation (IVM) of porcine oocytes on meiotic maturation, fertilization and embryo development, and compared the effect of supplementing the embryo culture media with PFF or foetal bovine serum (FBS) on embryo development. Oocytes from pre-pubertal gilts were IVM for 44 h, and parthenogenetically activated or in vitro-fertilized. Embryos were cultured in porcine zygote medium (PZM3) for 7 days. Cleavage and blastocyst rates were evaluated at 48 h and 7 days of culture. The supplementation of the IVM medium with 25% PFF and 1 mm dbcAMP for the first 22 h resulted in more (p < 0.05) embryos developing to the blastocyst stage as compared with the inclusion of dbcAMP alone. The dbcAMP + PFF combination increased (p < 0.05) the average number of nuclei per blastocyst as compared with either of these components alone or in its absence. A synergistic effect of dbcAMP + PFF during IVM was also reflected in the capacity of oocytes to regulate sperm penetration and prevent polyspermy, as twice as many oocytes from the control group were penetrated by more than one sperm as compared with those matured in the presence of both dbcAMP and PFF. The supplementation of PZM3 with 10% FBS from days 5 to 7 of culture significantly improved the total cell quantity in embryos derived either from control or dbcAMP + PFF matured oocytes. There was no effect on the total cell quantity when FBS was replaced by the same concentration of PFF. These studies showed that dbcAMP, PFF and FBS can improve both the quantity (57.3% vs 41.5%) and quality (74.8 vs 33.3 nuclei) of porcine blastocysts derived from oocytes recovered of pre-pubertal gilts.

The effects of laser phototherapy on the release of growth factors by human gingival fibroblasts were studied in vitro. Cells from a primary culture were irradiated twice (6 h interval), with continuous diode laser [gallium-aluminum-arsenium (GaAlAs), 780 nm, or indium-gallium-aluminum-phosphide (InGaAlP),_660 nm] in punctual and contact mode, 40 mW, spot size 0.042 cm(2), 3 J/cm(2) and 5 J/cm(2) (3 s and 5 s, respectively). Positive [10% fetal bovine serum (FBS)] and negative (1%FBS) controls were not irradiated. Production of keratinocyte growth factor (KGF) and basic fibroblast growth factor (bFGF) was quantified by enzyme-linked immunosorbent assay (ELISA). The data were statistically compared by analysis of variance (ANOVA) followed by Tukey`s test (P a parts per thousand currency signaEuro parts per thousand 0.05). The characterization of the cell line indicated a mesenchymal nature. KGF release was similar in all groups, while that of bFGF was significantly greater (1.49-times) in groups treated with infra-red laser. It was concluded that increased production of bFGF could be one of the mechanisms by which infra-red laser stimulates wound healing.

A new culture model was developed to study the role of proliferation and apoptosis in the etiology of keloids. Fibroblasts were isolated from the superficial, central, and basal regions of six different keloid lesions by using Dulbecco's Modified Eagle Medium containing 10% fetal calf serum as a culture medium. The growth behavior of each fibroblast fraction was examined in short-term and long-term cultures, and the percentage of apoptotic cells was assessed by in situ end labeling of fragmented DNA. The fibroblasts obtained from the superficial and basal regions of keloid tissue showed population doubling times and saturation densities that were similar to those of age-matched normal fibroblasts. In contrast, the fibroblasts from the center of the keloid lesions showed significantly reduced doubling times (25.9 +/- 6.3 hours versus 43.5 +/- 6.3 hours for normal fibroblasts) and reached higher cell densities. In long-term culture, central keloid fibroblasts formed a stratified three-dimensional structure, contracted the self-produced extracellular matrix, and gave rise to nodular cell aggregates, mimicking the formation of keloid tissue. Apoptotic cells were detected in both normal and keloid-derived fibroblasts, but their numbers were twofold higher in normal cells compared with all keloid fibroblasts. To examine whether apoptosis mediates the therapeutic effect of ionizing radiation on keloids, the cells were exposed to gamma rays at a dose of 8 Gy. Under these conditions, a twofold increase in the population of apoptotic cells was detected. These results indicate that the balance between proliferation and apoptosis is impaired in keloid fibroblasts, which could be responsible for the formation of keloid tumors. The results also suggest that keloids contain at least two different fibroblast fractions that vary in growth behavior and extracellular matrix metabolism.
