Com o objetivo de contribuir com conhecimentos sobre a reprodução em laboratório de ostras nativas do gênero Crassostrea e estudar as possíveis interações entre as espécies cultivadas no Brasil, o presente trabalho avaliou: 1) um método de anestesia e amostragem de tecido gonádico sem sacrifício de animais para a análise do estado de desenvolvimento sexual; 2) a hibridação entre a espécie de ostra do Pacífico, Crassostrea gigas, e as espécies nativas, C. rhizophorae e C. gasar; e 3) o uso de marcadores de DNA mitocondrial e nuclear para a identificação de híbridos. Como resultados, o uso de Cloreto de Magnésio (50 g.L-1), aplicado à água do mar, promoveu o relaxamento muscular e a abertura das valvas nas três espécies de ostras estudadas, permitindo biópsias de tecido gonádico com seringas e agulhas e a determinação do sexo dos animais. Os procedimentos de anestesia e amostragem de tecido não causaram mortalidade nestes indivíduos que apresentaram 100% de sobrevivência após 10 dias. Após o uso do anestésico, também não foram observadas alterações na atividade reprodutiva e na geração de larvas-D de C. gigas. A partir dos cruzamentos recíprocos entre C. gigas, C. rhizophorae e C. gasar, houve sucesso assimétrico na fecundação de oócitos de C. rhizophorae (R) com espermatozoides de C. gigas (G), oócitos de C. gasar (B) com espermatozoides de C. gigas (G) e oócitos de C. rhizophorae (R) com espermatozoides de C. gasar (B). A compatibilidade unidirecional de gametas entre as três espécies resultou na formação de larvas híbridas que apresentaram crescimento similar à espécie materna até sete dias de idade. Após este período, as larvas pararam de crescer e morreram. As análises de marcadores moleculares confirmaram que as progênies RG eram híbridos verdadeiros e continham o DNA de ambas as espécies parentais em seu genoma. A inviabilidade no desenvolvimento de larvas híbridas interespecíficas em laboratório sugere que a incompatibilidade genômica é suficiente para evitar o risco de hibridação natural entre C. gigas e as espécies nativas C. rhizophorae e C. gasar.

本文对我国己发表的牡蛎作了详细的比较解剖学研究。在强蛎亚科(Pycnodonteinae)发现了第三个心耳--右上心耳和与其相连的第三条回心静脉--后静脉,这在双壳类中实属首次报道。第一次提出了牡蛎循环系统的两种类型,附心脏型和无附心脏型。两者之间的主要区别是:前者无肌套血管,其外套血液主要来自环外套动脉和附心脏,而后者由于无附心脏和环外套动脉,其外套血液主要来自肌套血管;前者是通过出鳃静脉和外套静脉分别将鳃前部和后部的血液送回心耳,而后者的外套静脉与外入鳃血管不通,鳃中的血液只能通过出鳃静脉回到心耳。根据外套腔的形态特点,本文将中国的牡蛎分为三种类型六个组。第一种类型,左右侧都具侧水腔,包括一个组;第二种类型,仅右侧具有侧水腔,包括四个组;第三种类型;不具侧水腔,包括一个组。在比较解剖学研究的基础上,文章还讨论了消化系统,神经系统,循环系统等一些主要系统的演化过程,并推测了现生牡蛎各属间的演化关系。提出了一些新的分类依据,发现了一个具有重要演化意义的单行属种,爪蛎属(Talonostrea),猫爪牡蛎(T. talonata)。澄清了目前世界上在舌骨蛎属(Hyotissa)、拟舌骨蛎属(Parahyotissa)、小蛎属(Saccostrea)和巨蛎属(Crassostrea)分类中的混乱。解决了我国牡蛎分类中存在己久的疑难问题,将己发表的20种牡蛎重新鉴定为15种,即舌骨牡蛎(Hyotissa hyotis),复瓦牡蛎(Parahyotissa imbricata)。中华牡蛎(P. sinensis),鸡冠牡蛎(Lopha cristagalli)。薄片牡蛎(Dendostren folium),缘齿牡蛎(D. cre-nulifera),褶牡蛎(Alectryonella plicatula),猫爪牡蛎(T. talonata),长牡蛎(Crassostrea gigas),近江牡(C. rivularis),拟近江牡蛎(Crassostrea sp.),僧帽牡蛎(Saccostrea cucullata),棘刺牡蛎(S. echinata),鹅掌牡蛎(Planostrea pestigris)和密鳞牡蛎(Ostrea denselamellosa)。它们分别隶属于二个科,曲蛎科(Grypheidae)和牡蛎科(Ostreidae);四个亚科,强蛎亚科(Pycnodonteinae)。冠蛎亚科(Pycnodonteinae),巨蛎亚科(Lopheinae)和牡蛎亚科(Crassostreinae)。

The jinjiang oyster Crassostrea rivularis [Gould, 1861. Descriptions of Shells collected in the North Pacific Exploring Expedition under Captains Ringgold and Rodgers. Proc. Boston Soc. Nat. Hist. 8 (April) 33-40] is one of the most important and best-known oysters in China. Based on the color of its flesh, two forms of C rivularis are recognized and referred to as the "white meat" and 11 red meat" oysters. The classification of white and red forms of this species has been a subject of confusion and debate in China. To clarify the taxonomic status of the two forms of C. rivularis, we collected and analyzed oysters from five locations along China's coast using both morphological characters and DNA sequences from mitochondrial 16S rRNA and cytochrome oxidase 1, and the nuclear 28S rRNA genes. Oysters were classified as white or red forms according to their morphological characteristics and then subjected to DNA sequencing. Both morphological and DNA sequence data suggest that the red and white oysters are two separate species. Phylogenetic analysis of DNA sequences obtained in this study and existing sequences of reference species show that the red oyster is the same species as C. ariakensis Wakiya [1929. Japanese food oysters. Jpn. J. Zool. 2, 359-367.], albeit the red oysters from north and south China are genetically distinctive. The white oyster is the same species as a newly described species from Hong Kong, C. hongkongensis Lam and Morton [2003. Mitochondrial DNA and identification of a new species of Crassostrea (Bivalvia: Ostreidae) cultured for centuries in the Pearl River Delta, Hong Kong, China. Aqua. 228, 1-13]. Although the name C. rivularis has seniority over C. ariakensis and C. hongkongensis, the original description of Ostrea rivularis by Gould [1861] does not fit shell characteristics of either the red or the white oysters. We propose that the name of C. rivularis Gould [1861] should be suspended, the red oyster should take the name C. ariakensis, and the white oyster should take the name C. hongkongensis. Genetic markers are needed for rapid and reliable identification of oysters. In this study, we developed multiplex genus- and species-specific PCR markers for the identification of oysters from China. We used the mitochondrial cytochrome oxidase I (COI) and nuclear 28S ribosomal RNA genes for marker development. DNA sequences from different species were obtained from GenBank or by direct sequencing. Sequences were aligned, and genus- and species-specific nucleotides were identified. Primers were designed for genus/species-specific amplification to generate fragments of different sizes. A multiplex set of genus- and species-specific primers from the 28S gene was able to separate C. ariakensis and C. hongkongensis from other species and assign oysters to four genera. A set of species-specific COI primers provided positive identification of all five Crassostrea species from China, C. ariakensis, C. hongkongensis, C. angulata, C. gigas, and C. sikamea in a single PCR. The multiplex PCR assays do not require fluorescence-labeling or post-PCR enzyme digestion, providing a simple, fast and reliable method for the identification of oysters from China.

In an effort to develop genetic markers for oyster identification, we studied length polymorphism in internal transcribed spacers (ITS) between major ribosomal RNA genes in 12 common species of Ostreidae: Crassostrea virginica, C. rhizophorae, C. gigas, C. angulata, C. sikamea, C. ariakensis, C. hongkongensis, Saccostrea echinata, S. glomerata, Ostrea angasi, O. edulis, and O. conchaphila. We designed two pairs of primers and optimized PCR conditions for simultaneous amplification of ITS 1 and ITS2 in a single PCR. Amplification was successful in all 12 species, and PCR products were visualized on high-resolution agarose gels. ITS2 was longer than ITS 1 in all Crassostrea and Saccostrea species, whereas they were about the same size in the three Ostrea species. No intraspecific variation in ITS length was detected. Among species, the length of ITS I and ITS2 was polymorphic and provided unique identification of 8 species or species pairs: C. ariakensis, C. hongkongensis, C. sikamea, O. conchaphila, C. virginica/C. rhizophorae, C. gigas/C. angulata, S. echinata/S. glonzerata, and O. angasi/O. edulis. The ITS assay provides simple, rapid and effective identification of C. ariakensis and several other oyster species. Because the primer sequences are conserved, the ITS assay may be useful in the identification of other bivalve species.

Oysters (Ostreidae) manifest a high degree of phenotypic plasticity, whereby morphology is of limited value for species identification and taxonomy. By using molecular data, the aim was to genetically characterize the species of Crassostrea occurring along the Brazilian coast, and phylogenetically relate these to other Crassostrea from different parts of the world. Sequencing of the partial cytochrome oxidase c subunit I gene (COI), revealed a total of three species of Crassostrea at 16 locations along the Brazilian coast. C. gasar was found from Curuçá (Pará state) to Santos (São Paulo state), and C. rhizophorae from Fortim (Ceará state) to Florianópolis (Santa Catarina state), although small individuals of the latter species were also found at Ajuruteua beach (municipality of Bragança, Pará state). An unidentified Crassostrea species was found only on Canela Island, Bragança. Crassostrea gasar and C. rhizophorae grouped with C. virginica, thereby forming a monophyletic Atlantic group, whereas Crassostrea sp. from Canela Island was shown to be more similar to Indo-Pacific oysters, and either arrived in the Atlantic Ocean before the convergence of the Isthmus of Panama or was accidentally brought to Brazil by ship.

High-nutrient tropical carbonate systems are known to produce sediments that, in terms of skeletal composition, are reminiscent of their extra-tropical counterparts. Such carbonate systems and associated carbonate grain assemblages in the tropics are rare in the present-day world. Nonetheless, it is crucial to gain a better understanding of those ecosystems, including their drivers and players because such settings potentially represent models for ancient depositional systems as well as for predicted future environmental conditions. One of the modern occurrences of eutrophic tropical carbonate systems is the northern Mauritanian Shelf. The marine environment is characterized by an eastern boundary upwelling system that pushes cool and nutrient-rich intermediate waters onto a wide epicontinental platform (Golfe d'Arguin) where the waters warm up to tropical temperatures. The resulting facies is mixed carbonate-siliciclastic with a dominant foramol association grading into bimol and barnamol grain assemblages in the shallowest areas forming the Banc d'Arguin. Besides this cool water-related heterozoan association, the carbonate sediment is characterized by tropical molluskan species, while chlorozoan biota (e.g., corals and algal symbiont-bearing foraminifers) are entirely absent. We here present a first comprehensive facies analysis of this model example of eutrophic tropical carbonates. Furthermore, we reconstruct the loci of carbonate production and provide a conclusive depositional model of the Banc d'Arguin that received little attention to date due to its poorly accessible nature.

Ecological and taxonomic study of the mollusk-rich fauna of the Golfe d'Arguin, North Mauritania, investigates the various environmental influences affecting this tropical shelf. The upwelling of nutrient-rich waters leads to a highly productive environment under tropical conditions. The resulting mixed carbonate-siliciclastic sediment contains a large portion of calcareous components produced by heterotrophic organisms-e.g., mollusks, foraminifers, worms, barnacles-that are reworked on the open shelf. On the basis of mollusk assemblages, six taphocoenoses are defined, all being characterized by a mixed fauna of tropical (e.g., Tellina densestriata), subtropical (e.g., Macoma cumana) and temperate (e.g., Spisula subtruncata) species. Differences between the assemblages are related to the medium-grain size ranging from mud to gravel-that results from local hydrodynamic conditions and water depth. Among carbonate grains, Donax burnupi shells are very abundant in the swell-exposed, northern part of the Golfe d'Arguin and reflect the tropical to subtropical, high-energy, and high-nutrient waters. Mollusk assemblages are demonstrated to be a sensitive tool for deciphering complex environmental conditions in sedimentary archives.

Digenean parasites of marine bivalves are relatively poorly known, particularly in Australia. We surveyed 2256 bivalve individuals ( 47 species, 17 families) from Queensland marine waters incorporating south-east Queensland, Heron Island ( southern Great Barrier Reef) and Lizard Island ( northern Great Barrier Reef). Infections of trematode species from three families, Bucephalidae, Gorgoderidae and Monorchiidae, were found. Overall prevalence of infection was 2.3%. The Bucephalidae was the most commonly found family; 11 species were found in Tellinidae, Ostreidae, Isognomonidae and Spondylidae - the latter two previously unknown as hosts for bucephalids. A single gorgoderid infection was found in a venerid, Lioconcha castrensis. Five species of monorchiids were found from Tellinidae and Lucinidae. All infections are new host/parasite records. No infections were found in 35 of the 47 bivalve species sampled. The generally low prevalence of infection by digeneans of bivalves suggests that it is unlikely that any of the species reported here are seriously damaging to bivalve populations in these waters. We deduce that, at best, we have some life-cycle information but no actual identifications for 10% of the species of trematodes that infect bivalves of Queensland marine waters.

The malacocultura, particularly oyster farming, appears on the world stage as one of the most viable alternatives to fishing decline and supply of fresh product. In Brazil, the development of mollusc cultivationis through the genus oyster cultivation Crassostrea, among them Crassostrea rhizophorae (Guilding, 1828), known for oyster-the-swamp, one of the main species of farmed bivalves in the state of Pará. This so it aimed to characterize the biomorphometrics relations, estimate the Shape Stabilization Index (IEF) of the shell and the yield of edible meat C. rhizophorae grown in an Amazonian coast, state of Pará, northern Brazil. When all is sampled 1,028 individuals, in April 2016, measuring the external morphometric measures (length, width and height) and total and visceral biomass. The results obtained are C. rhizophorae with (1) excellent biomorphometrics relationships among both external measures, the measures of the shell and biomass generating equations that satisfy morphometric pet species, (2) yield of edible meat 15% of the total biomass and variation in the shell along its development to adulthood, with a tendency to stabilize the reach 60mm in length.
