Although restriction enzymes are widely distributed in nature, many bacterial genera are yet to be explored for the presence of this important class of enzymes. We have purified and characterized a new type II restriction endonuclease, OfoI from a nonheterocyst cyanobacterium Oscillatoria foreaui. The recognition sequence has been determined by primer extension analysis. The purified enzyme OfoI recognizes and cleaves the palindromic hexanucleotide 5'-Cdown arrowYCGRG-3', generating 5'-protruding ends.

During 1994, weekly spot-sampling of open water sites on Loch Leven took place from 16th March onwards. Very little difference between spot-sampling sites was observed from 16th March to 5th July. Throughout April and May there was a slow increase in levels of chlorophyll-a, followed by a rapid increase through June, reaching a maximum of 230 micrograms per litre. On 8th July there was a very rapid rise in water temperature, with a corresponding increase in dissolved oxygen and pH. At 0915 hours on 9th July there was a drop in all three variables. These changes were coincident with a marked reduction in chlorophyll-a values falling to 70 micrograms per litre on 12th July. On 16th July evidence of a resurgence of photosynthesis was observed.

Sequence of rDNA intergenic spacer region (ISR) from a waterbloom cyanobacterial species Oscillatoria sp, was determined and analyzed. The results of sequence comparison showed that the spacer had a high level sequence divergence, suggesting the sequence may be a target sequence for developing cyanobacteria genus- and species-specific oligonucleotide probes. In addition, a 20bp sequence of rDNA ISR was found highly conserved in all species of cyanobacteria, which was not found in other eubacteria. This conserved sequence within a variable region indicates that it might be a functional oligonucleotide in the processing of the rRNA precursor.

The production of certain odorous metabolites is an undesirable attribute of cyanobacteria (blue-green algae) growth in aquaculture ponds [e.g., channel catfish(Ictalurus punctatus)] and in drinking water reservoirs. The most common odorous compounds encountered in catfish aquaculture are geosmin (trans-1,10-dimethyltrans-9-decalol) and 2-methylisoborneol(exo-1,2,7,7-tetramethylbicyclo[2.2.1]heptan-2-ol). These compounds are also frequently encountered worldwide in reservoirs and aqueducts used for municipal drinking water systems(Schrader et al. 2002). In this study, several algicides were evaluated using a rapid bioassay to determine their effectiveness in controlling the MIB-producing cyanobacterium Oscillatoria perornata from a west Mississippi catfish pond and the MIBproducing Pseudanabaena sp. (strain LW397) from Lake Whitehurst, Virginia, used as a city water supply reservoir. The cyanobacterium Oscillatoria agardhii , not a MIB-producer, and the green alga Selenastrum capricornutum , found in catfish ponds in the southeastern United States, were included in the bioassay to help determine potential broad-spectrum toxicity of the commercial products. This study documents the relative tolerance of the common, weedy mat-forming green algae Hydrodictyon , Oedogonium , Pithophora , Rhizoclonium , and Spirogyra to copper. In addition, the copper tolerance of the cyanobacterial (blue-green algal) mat-forming Oscillatoria was assessed.

In an effort to evaluate the production potential of an artificial impoundment, the phytoplankton of the Shen reservoir was sampled from November 1981 to June 1982 at three stations during three periods of distinct seasonal hydrographic characteristics. The samples were subsampled and quantified. Most of the phytoplankton were identified to the species level. There were in all 53 species comprising Chlorophyceae contributing 36.70% with species of Volvox, Pediastrum, Closterium, Staurodesmus and Ankistrodesmus as dominant species in this group. The Cyanophyceae contributed 30.00% with species of Microcystis, Nostoc , and Oscillatoria as the dominant species. An analysis of temporal and spatial changes in composition and abundance of the various groups showed that these were influenced by water temperature, sampling period and station. Based on the trophic status of the most abundant species, the composition of the phytoplankton is indicative of a tropical reservoir with a moderate productivity for fish culture

River Kubanni, a major tributary of River Galma, receives both organic and inorganic wastes through run-offs and seepage from residential and agricultural areas of Tundun-Wada, Zaria. Water and phytoplankton samples were collected once a month from three stations on a stretch of the river, for eight months (February, 1994-0ctober, 1994). The physico-chemical parameters and phytoplankton composition were determined and correlated to one another. The distribution and composition of phytoplankton species are affected by variations through fluctuations in environmental variables such as temperature, velocity, transparency, pH, dissolved Oxygen, total alkalinity, total hardness, electrical conductivity and total dissolved matter. Highest dissolved oxygen concentration in February coincided with the minimum water temperature due to the cool harmattan winds. Low alkalinity resulted in low phytoplankton productivity while a rise in total dissolved matter resulted in increase in electrical conductivity and high phytoplankton productivity. The presence of Oscillatoria sp and Euglena sp in station 2 and 3 are indicative of organic pollution in these stations. However, the river stretch is suitable for fish production with respect to water hardness and pH
