Surface flavonoids in nine species of Origanum, representing taxa from all three of the currently recognised subgeneric groups, were examined both by HPLC coupled to diode-array detection and APCI-MS. Many of the flavonoids present were characterised by O-substituent at C-6 (OH, OMe) and/or C-8 (OMe). In total, 25 flavones and flavanones are described in this study, of which 13 are new to the genus and 5,4'-dihydroxy-6,7,3'-trimethoxyflavanone is reported for the first time. Taxa in subgeneric Group A accumulated flavonoids with methoxyl groups at both C-6 and C-4'; however, taxa in subgeneric Group B did not accumulate 4'-methoxylated compounds, and taxa in Group C did not accumulate 6-methoxylated compounds. Purpose: The objective of this study was to evaluate the antioxidant effect of oregano and thyme extracts isolatedly and combinedly applied in soybean oil. Design/methodology/approach: Soybean oil containing 3,000 mg/kg of oregano and thyme oleoresins and the mixture of both, as well as soybean oil containing TBHQ (50 mg/kg) and soybean oil free of antioxidants, were subjected to accelerated oven test (60°C/10 days). Samples were collected every two days and analyzed as to their peroxide and conjugated diene values. Findings: The mixture of oleoresins and consequent increase of concentration were as effective as the antioxidant TBHQ. Practical implications: These studies may prove to be beneficial to the exploitation of natural antioxidant sources for the preservation and/or extension of raw and processed food shelf life. Therefore, they could also be applied in the area of pharmaceuticals for the protection of human life. Originality/value: This study offers information on the use of natural antioxidants as an alternative to the use of synthetic antioxidants, which might be considered toxic. © Emerald Group Publishing Limited.

La productividad del orégano (Origanum vulgare L.) está determinada por la conjunción entre cantidad de biomasa acumulada y contenido de aceite esencial hasta el momento de su cosecha. Numerosos autores han constatado que dicho contenido es máximo al momento de floración pero los procesos que determinan la ocurrencia de la misma son poco claros en esta especie. A través de la prolongación artificial del fotoperíodo, se evaluó la sensibilidad fotoperiódica de dos subespecies tradicionales de orégano (Compacto: Origanum vulgare ssp. vulgare y Criollo: Origanum vulgare ssp. hirtum Ietsw.) y su incidencia en el desarrollo, duración de fases fenológicas y en la dinámica de crecimiento. Se encontró que ambas responden al aumento del fotoperíodo reduciendo la longitud de su ciclo. Ante estas condiciones, las mismas difirieron en la magnitud de su respuesta, siendo la subespecie Criollo más sensible que Compacto. Esto sugiere que el umbral fotoperiódico de inducción a floración es menor en el orégano Criollo que en el orégano Compacto. El acortamiento de la fase de desarrollo vegetativo en ambas subespecies generó menor número de nudos y longitud de ramas finales (disminución más notoria en la subespecie Criollo). El fotoperíodo extendido generó un cambio en el modelo de crecimiento de la longitud de ramas de lineal a lineal con meseta o cuadrático.

The objective of this study was to determine the antioxidant effect of essential oregano and alfavaca oil on the fatty acid profile of palm oil, used in industrial potato chips processing, aiming to reduce the oxidative state of palm oil. Essential oregano oil was obtained from industrial residues and commercial essential oregano oil was also acquired. The antioxidant activity of Oregano’s essential oil was taken by DPPH method. The DPPH analysis of the essential oil of oregano residue resulted in an IC50 of 797.04 mg.mL-1 and the essential oil of commercial oregano was 424.25 mg.mL-1. Regarding the content of total phenols, the essential oil of commercial oregano showed 0.167 ± 0.058 ug EAG.g-1. Based on these results, concentrations of 50, 100 e 150ug.g-1 of commercial oregano essential oil was added to palm oil and the fatty acid profile was determined by gas cromatography. Analysis were done with palm oil without being used in frying processes and without added essential oil, as well as with the different essential oil concentrations submitted at three frying processes at one single day. In the second part of the project, the effect of oreganos’s comercial essential oil compared to alfavacão essential oil, at the concentration of 100ug.g-1 in palm oil, and submitted to three frying processes during eleven cycles, was studied. The lipid profile was similar to that described in the literature, which an increase in the number of frying times caused a polyunsaturated fatty acids decrease and, therefore, a proportional increase in saturated fatty acids. It was noted an increase in the amounts of total saturated fatty acids from 43.36% (control) to 43.60% (palm oil with essential oil after 11 frying cycles at the concentration of 100ug.g-1). This fact proves that during the frying process, there is an increase in the rate of formation of saturated fatty acids. However, the addition of oregano essential oil did not provide significant change in the fatty acids of palm oil used in frying process of potato chips. The addition of oregano essential oil in a concentration of 100 ug./mL-1 until the third frying cycle showed a reduction effect of trans fatty acid formation. Although, during 11 frying cycles it was not noticed the essential oil effect under trans fatty acid formation. It can be suggested that this factor may be correlated to the frying time, which may not have been sufficient for significant formation of saturated and trans compounds.

con el objetivo de evaluar la eficacia antibacteriana in vitro de dos formulaciones de un colutorio de aceite esencial de orégano sobre Streptococcus mutans (S. mutans) el principal agente causal de la placa bacteriana dental, para determinar dicha actividad, primero se realizó la determinación de la concentración inhibitoria mínima (CIM) del aceite esencial de orégano (AEO) sobre S. mutans mediante el método de microdilución en caldo y se obtuvo una CIM de 0.0025 %, esa concentración o una mayor podría ser útil en una forma farmacéutica con propósito antibacteriano. se realizó la determinación de la aptitud de los métodos de recuento microbiano que permitió indicar la metodología adecuada para evaluar la biocarga microbiana en los colutorios. Esta prueba indicó que el polisorbato 80 al 1% es un agente neutralizante idóneo para recuperar el S. mutans si se encuentra presente en los colutorios de AEO al 0.05 % y 0.10 %, y la mezcla de tiosulfato de sodio al 0.3 %, lecitina al 0.5% y polisorbato 80 al 1 % es la indicada para neutralizar las propiedades antimicrobianas del colutorio de clorhexidina al 0.12 % ylograr una recuperación adecuada del microorganismo en estudio cuando estuviera presente. Finalmente, se realizó la determinación de la eficacia antibacteriana de los colutorios de AEO, el vehículo de estos y un colutorio comercial de clorhexidina sobre S. mutans, los resultados reflejaron que el porcentaje máximo alcanzado de reducción para los colutorios de AEO al 0.05% y 0.10 % fue de 48.280 % a los 15 minutos y de 78.535 % en 1 minuto respectivamente, dichas reducciones fueron mayores que la que presento el vehículo, la cual fue de 29.292 % a los 15 minutos. El colutorio de clorhexidina al 0.12 % fue el que produjo la máxima reducción del microorganismo de estudio, la cual fue de99.996% en 6 minutos. Todos los ensayos se realizaron en el laboratorio del Centro de Investigación y Desarrollo en Salud, durante el periodo de julio a septiembre de 2015. Se concluyó que, ningún colutorio alcanzó el 99.999 % de reducción especificado por la AOAC para germicidas y sanitizantes, se observaron reducciones que sugieren buenas perspectivas de desarrollo para este tipo de formulaciones que incluyen activos de origen natural, por lo tanto se recomienda la necesidad de realizar más estudios sobre la composición y diferentes actividades del aceite esencial de orégano que pueden aprovecharse en futuras formulaciones.

Purpose: To determine the heavy metal and trace element composition of the powdered aerial parts of Origanum sipyleum L. and its water extract. Methods: The heavy metal and trace elements content of the powdered plant material and 2 % aqueous extract were evaluated by x-ray fluorescence spectroscopy with silicon drift detector SDD at a resolution of 145 eV and 10,000 pulses. The process conditions were 0.1 g sample weight, process time of 300 s at a voltage of 25 kV and 50 kV, and at a current of 0.5 and 1.0 mA under helium atmosphere. Results: The major elements, K, Ca and Na, known as macronutrients, constituted 11990, 10490 and 970 ppm of the powdered drug and 8910, 2991 and 810 ppm of the water extract, respectively. Among other constituents, arsenic, lead and uranium levels were < 1, 2.1 and < 3 ppm, respectively, in the powdered material while in the aqueous extract, the levels were < 1, < 2 and 200 ppm, respectively. Conclusion: O. sipyleum is a potential source of macro- and micronutrients from which useful food additives and health supplements can be derived.

A microplate assay was modified for the detection of antimicrobial activity in plant extracts. The aim was to develop an in vitro assay that could rapidly screen plant extracts to provide quantitative data on inhibition of microbial growth. A spectrophotometric assay using a microplate with serial dilutions of the plant extract and the bacteria was developed. Two bacteria, Staphylococcus aureus and Escherichia coli, were used for this study. Essential oils, oregano (Origanum vulgare) and lemon myrtle (Backhousia citriodora), and three active components carvacrol, thymol and citral were evaluated. The reproducibility of the assay was high, with correlation coefficients (r aureus and E. coli between 0.9321 and 0.9816. Similarly, r and 0.9814. This assay could also be used to measure antimicrobial activity in plant extracts which vary in pH and color.
