The Norwegian spring spawning (NSS) herring is an ecologically important fish stock in the Norwegian Sea, and with a catch volume exceeding one million tons a year it is also economically important and a valuable food source. In order to provide a baseline of the levels of contaminants in this fish stock, the levels of organohalogen compounds were determined in 800 individual herring sampled at 29 positions in the Norwegian Sea and off the coast of Norway. Due to seasonal migration, the herring were sampled where they were located during the different seasons. Concentrations of dioxins and dioxin-like PCBs, non-dioxin-like PCBs (PCB7) and PBDEs were determined in fillet samples of individual herring, and found to be relatively low, with means (min-max) of 0.77 (0.24-3.5) ngTEQ/kg wet weight (ww), 5.0 (1.4-24) µg/kg ww and 0.47 (0.091-3.1) µg/kg ww, respectively. The concentrations varied throughout the year due to the feeding- and spawning cycle: Starved, pre-spawning herring caught off the Norwegian coast in January-February had the highest levels and those caught in the Norwegian Sea in April-June, after further starvation and spawning, had the lowest levels. These results show that the concentrations of organohalogen compounds in NSS herring are relatively low and closely tied to their physiological condition, and that in the future regular monitoring of NSS herring should be made in the spawning areas off the Norwegian coast in late winter.

The active migration of neurons from their sites of origin to their final destinations requires the unidirectional translocation of the nuclei and somatic cytoplasm within the growing leading processes. To explore the cellular machinery underlying this translocation, we determined the polarity of microtubules situated within the leading and trailing processes of migrating cerebellar granule cells in situ. Our analysis reveals that the newly assembled positive ends of the microtubules in the leading process uniformly face the growing tip, while their disintegrating negative ends face the nucleus. In the trailing process, by contrast, microtubule arrays are of mixed polarity. We suggest that the dynamics of slow polymerization in combination with fast disintegration of oriented microtubules create "push" and "pull" forces that contribute to the piston-like saltatory displacement of the nucleus and cytoplasm within the membrane cylinder of the leading process of the migrating neuron.

Background: Prostate tumor heterogeneity is a major factor in disease management. Heterogeneity could be due to multiple cancer cell types with distinct gene expression. Of clinical importance is the so-called cancer stem cell type. Cell type-specific transcriptomes are used to examine lineage relationship among cancer cell types and their expression similarity to normal cell types including stem/progenitor cells. Methods: Transcriptomes were determined by Affymetrix DNA array analysis for the following cell types. Putative prostate progenitor cell populations were characterized and isolated by expression of the membrane transporter ABCG2. Stem cells were represented by embryonic stem and embryonal carcinoma cells. The cancer cell types were Gleason pattern 3 (glandular histomorphology) and pattern 4 (aglandular) sorted from primary tumors, cultured prostate cancer cell lines originally established from metastatic lesions, xenografts LuCaP 35 (adenocarcinoma phenotype) and LuCaP 49 (neuroendocrine/small cell carcinoma) grown in mice. No detectable gene expression differences were detected among serial passages of the LuCaP xenografts. Results: Based on transcriptomes, the different cancer cell types could be clustered into a luminal-like grouping and a non-luminal-like (also not basal-like) grouping. The non-luminal-like types showed expression more similar to that of stem/progenitor cells than the luminal-like types. However, none showed expression of stem cell genes known to maintain stemness. Conclusions: Non-luminal-like types are all representatives of aggressive disease, and this could be attributed to the similarity in overall gene expression to stem and progenitor cell types.

The problem of spectra formation in hydrodynamic approach to A + A collisions is considered within the Boltzmann equations. It is shown analytically and illustrated by numerical calculations that the particle momentum spectra can be presented in the Cooper-R-ye form despite freeze-out is not sharp and has the finite temporal width. The latter is equal to the inverse of the particle collision rate at points (t(sigma) (r, p), r) of the maximal emission at a fixed momentum p. The set of these points forms the hypersurfaces t(sigma)(r,p) which strongly depend on the values of p and typically do not enclose completely the initially dense matter. This is an important difference from the standard Cooper-Frye prescription (CFp), with a common freeze-out hypersurface for all p, that affects significantly the predicted spectra. Also, the well known problem of CFp as for negative contributions to the spectra from non-space-like parts of the freeze-out hypersurface is naturally eliminated in this improved prescription.

É do conhecimento geral que muitos professores ensinam os alunos a ouvir, cantar e compor, mas usam testes escritos para avaliar as aprendizagens. Este facto revela que, por um lado, há quem tenha a ideia que os testes escritos estão mesmo a avaliar as práticas musicais e, por outro lado, também há quem considere que é muito difícil e inconsistente avaliar as práticas musicais porque a Música tem um carácter transitório, efémero, imaterial. Além do problema ser interessante, ele é abrangente porque existe tanto entre os professores especialistas de Música como entre os professores generalistas de Educação de Infância e de 1º Ciclo. A experiência levada a cabo nos últimos três anos no Mestrado em Ensino de Educação Musical no Ensino Básico sobre a forma de avaliar as aprendizagens dos alunos em Educação Musical mantém os testes escritos para avaliar os conhecimentos teóricos, mas introduz um instrumento de avaliação das práticas musicais, as grelhas de descritores de desempenho. Estas grelhas, ainda que mantendo princípios orientadores comuns, são sempre feitas à medida para cada situação específica de obra, atividade musical, alunos e são aplicadas não só em observação direta, mas também sobre registos áudio/vídeo. Estes instrumentos são construídos pelos docentes e não em conjunto com os alunos, mas são-lhes apresentados e explicados desde o início de cada unidade didática, usados regulamente para autoavaliação formativa e nas apresentações finais para avaliação sumativa. Desta forma os alunos sabem desde o início onde se espera que cheguem e sabem em cada momento do processo em que ponto se encontram e que problemas/dificuldades musicais devem ser ultrapassados. Em cada atividade comparámos a autoavaliação dos alunos com a avaliação dos professores e verificámos em todas as situações uma elevada correlação positiva (r > 0,9). Teremos ainda que consolidar os dados já obtidos pelo que esperamos o envolvimento de mais professores. A possibilidade de divulgar a solução aqui apresentada, nomeadamente, através de ações de formação contínua, permitirá um evidente aumento da consistência e fiabilidade da avaliação das práticas musicais em Educação Musical.

El treball de recerca té com a principal objectiu l'estudi del cinema documental rus contemporani a través de l'obra cinematogràfica d'Alexander Sokurov, Sergei Dvortsevoi, Sergei Loznitsa i Victor Kossakovski. En un primer moment la investigació s'havia encaminat en un estudi comparatiu sobre les noves tendències del documental i els models de realisme proposats des de la Rússia post-comunista. El treball s'ha realitzat a partir de tres vies d'investigació. La primera ha consistit en una exhaustiva recerca bibliogràfica sobre cinema documental i cinema soviètic. La segona s'ha plantejat a partir d'un anàlisi acurat de les diverses pel•lícules. Finalment, la tercera via s'ha desenvolupat a partir d'un treball de camp realitzat durant una estada a Rússia, un període en el qual va ser possible entrevistar dos dels cineastes protagonistes de l'estudi, Sergei Dvortsevoi i Victor Kossakovski, així com el crític de cinema Andrei Xemijakin. També va ser fonamental l'assistència a la taula rodona i la master class impartida per Sergei Loznitsa en el marc del desè aniversari del Màster en Teoria i Pràctica de Documental Creatiu de la Universitat Autònoma de Barcelona. Tot i que es poden traçar vincles entre el treball dels quatre cineastes escollits i algunes de les pràctiques contemporànies en l'àmbit de la no-ficció, com pot ser l'experiència de Sergei Loznitsa en el terreny del found-footage, o els documentals experimentals de caràcter assagístic d'Alexander Sokurov, així com la tendència observacional i el pas al cinema de ficció de Segei Dvortsevoi, o l'ús de la tecnologia digital en les últimes pel•lícules de Victor Kossakovski. Tot i aquestes aproximacions, es pot afirmar que el model de realisme proposat per aquests cineastes troba el seu autèntic llegat en el cinema soviètic. Una herència que comença amb el cinema de Dziga Vertov –pioner del documental artístic i revolucionari- i acaba en el d'Artavadz Pelechian.

Mouse models are important tools to decipher the molecular mechanisms of mammary carcinogenesis and to mimic the respective human disease. Despite sharing common phenotypic and genetic features, the proper translation of murine models to human breast cancer remains a challenging task. In a previous study we showed that in the SV40 transgenic WAP-T mice an active Met-pathway and epithelial-mesenchymal characteristics distinguish low- and high-grade mammary carcinoma. To assign these murine tumors to corresponding human tumors we here incorporated the analysis of expression of transcription factor (TF) coding genes and show that thereby a more accurate interspecies translation can be achieved. We describe a novel cross-species translation procedure and demonstrate that expression of unsupervised selected TFs, such as ELF5, HOXA5 and TFCP2L1, can clearly distinguish between the human molecular breast cancer subtypes-or as, for example, expression of TFAP2B between yet unclassified subgroups. By integrating different levels of information like histology, gene set enrichment, expression of differentiation markers and TFs we conclude that tumors in WAP-T mice exhibit similarities to both, human basal-like and non-basal-like subtypes. We furthermore suggest that the low- and high-grade WAP-T tumor phenotypes might arise from distinct cells of tumor origin. Our results underscore the importance of TFs as common cross-species denominators in the regulatory networks underlying mammary carcinogenesis.

Although sources in general nonlinear mixturm arc not separable iising only statistical independence, a special and realistic case of nonlinear mixtnres, the post nonlinear (PNL) mixture is separable choosing a suited separating system. Then, a natural approach is based on the estimation of tho separating Bystem parameters by minimizing an indcpendence criterion, like estimated mwce mutual information. This class of methods requires higher (than 2) order statistics, and cannot separate Gaarsian sources. However, use of [weak) prior, like source temporal correlation or nonstationarity, leads to other source separation Jgw rithms, which are able to separate Gaussian sourra, and can even, for a few of them, works with second-order statistics. Recently, modeling time correlated s011rces by Markov models, we propose vcry efficient algorithms hmed on minimization of the conditional mutual information. Currently, using the prior of temporally correlated sources, we investigate the fesihility of inverting PNL mixtures with non-bijectiw non-liacarities, like quadratic functions. In this paper, we review the main ICA and BSS results for riunlinear mixtures, present PNL models and algorithms, and finish with advanced resutts using temporally correlated snu~sm

ABSTRACT : Fungal infections have become a major source of diseases in immuncompromised patients, but are quite benign in healthy individuals. As fungi are eukaryotes, and share many biological processes with humans, many antifungal drugs can cause toxicity in the patients. Therefore, the characterization of signaling pathways specific to the anti-fungal immune response is relevant for the better understanding of the disease and the development of new therapeutic approaches. Dectin-1 is the major mammalian pattern recognition receptor for the fungal component zymosan. Dectin-1 is an innate non-Toll-like receptor containing immunoreceptor tyrosine-based activation motifs (ITAMs). Card9, Bc110 and Maltl are proteins that have been shown to play a key role in the Dectin-l-induced signaliñg pathway by controlling Dectin-l-mediated cell activation, cytokine production and innate anti-fungal immunity in mice. Here we investigate the role of the Card9-Bc110-Maltl complex in humans using the monocytic cell line THP-1. We show that Card9 interacts with Bc110 through a CARD-CARD interaction and that interaction of Card9 with Bc110 is required for NF-xB activation. We further demonstrate that Card9 is phosphorylated in its C-terminal part on serine residues. The phosphorylation status of Card9 can influence its ability to active NF-xB, since mutation of the phosphorylation sites increases its ability to activate NF-xB. We find that Card9 is expressed in myeloid derived cells, such as the human monocytic cell lines THP1 and U937, and in human monocyte-enriched PBLs and monocyte-derived DCs. Our findings demonstrate that Card9 is implicated in anti-fungal responses, since silencing of Card9 as well as of Bc110 and Maltl diminishes the capacity of THP1 cells to produce TNF-a in response to zymosan. Interestingly, activation of the NF-xB and MAPK pathway remained normal and levels of TNF-a mRNA produced were also not affected in THP 1 cells silenced for the expression of Card9, Bc110 or Malt1. Using a Malt1 inhibitor, we provide evidence that the proteolytic activity of Malt1 is needed for zymosan-induced TNF-a production in THP 1 cells and bone marrow-derived macrophages of mice, but further experiments are required to confirm these findings and identify the substrate(s) of Malt1. In conclusion, our results reveal an important role for Card9 in the innate immune response of human macrophages to fungi. RÉSUMÉ : Les infections fongiques sont une source majeure de maladie chez les patients immunodéprimés, alors qu'elles sont plutôt bénignes chez les individus sains. Comme les champignons sont des eucaryotes et partagent beaucoup de processus biologiques avec les humains, les médicaments antifongiques peuvent être source de toxicité chez les patients. Il est donc important de mieux caractériser les voies de signalisation intracellulaire des réponses anti-fongiques pour pouvoir développer de nouvelles approches thérapeutiques. La protéine Dectin-1 est le récepteur principal du composé fongique zymosan. Les protéines Card9, Bc110 et Maltl ont été décrites comme jouant un rôle primordial dans les signaux d'activation induits par Dectin-l, en contrôlant l'activité cellulaire, la production de cytokines et la défense anti-fongique dans les souris. Dans cette étude, nous investiguons le rôle du complexe Card9-Bc110-Maltl dans la lignée monocytaire humaine THP1. Nous montrons que Card9 interagit avec Bc110 par une interaction CARD-CARD et que cette interaction est requise pour activer le facteur de transcription NF-xB. Nous observons que Card9 est phosphorylé dans sa partie C-terminale sur des résidus serine et que l'état de phosphorylation de Card9 influence sa capacité à activer NF-xB. En effet, sa capacité à activer NF-xB est augmentée, après mutation des sites de phosphorylation. La génération d'un anticorps spécifique dirigé contre Card9 nous a permis de démontrer que Card9 est exprimé dans des cellules myéloïdes comme les lignées cellulaires monocytiques THP-1 et U-937, ainsi que dans les cellules dendritiques humaines. Nos résultats démontrent que Card9 est impliqué dans la réponse immunitaire antifongique puisque la réduction de l'expression de Card9 ainsi que de Bc110 et de Malt1 diminue la capacité des THP-1 à produire du TNF-a en réponse au zymosan. Par contre, les voies de signalisation NF-xB et MAPK ainsi que les niveaux de mRNA de TNF-a produits en réponse au zymosan ne sont pas affectés dans ces cellules. En utilisant un inhibiteur de Malt1, nous montrons que l'activité protéolytique de Malt1 est nécessaire pour la production de TNF-a induite par le zymosan dans les cellules THP-1 ainsi que dans les macrophages de souris, mais d'autres expériences seront nécessaires pour confirmer cette observation et identifier le(s) substrat(s) de Malt1 responsables de cet effet. En conclusion, nos résultats révèlent un rôle important de la protéine Card9 dans la réponse immunitaire innée antifongique dans les macrophages humains.

Les gènes codant pour des protéines peuvent souvent être regroupés et intégrés en modules fonctionnels par rapport à un organelle. Ces modules peuvent avoir des composantes qui suivent une évolution corrélée pouvant être conditionnelle à un phénotype donné. Les gènes liés à la motilité possèdent cette caractéristique, car ils se suivent en cascade en réponse à des stimuli extérieurs. L’hyperthermophilie, d’autre part, est interreliée à la reverse gyrase, cependant aucun autre élément qui pourrait y être associé avec certitude n’est connu. Ceci peut être dû à un déplacement de gènes non orthologues encore non résolu. En utilisant une approche bio-informatique, une modélisation mathématique d’évolution conditionnelle corrélée pour trois gènes a été développée et appliquée sur des profils phylétiques d’archaea. Ceci a permis d’établir des théories quant à la fonction potentielle du gène du flagelle FlaD/E ainsi que l’histoire évolutive des gènes lui étant liés et ayant contribué à sa formation. De plus, une histoire évolutive théorique a été établie pour une ligase liée à l’hyperthermophilie.

This study investigates the production and on-line processing of English tense morphemes by sequential bilingual (L2) Turkish-speaking children with more than three years of exposure to English. Thirty nine 6-9-year-old L2 children and 28 typically developing age-matched monolingual (L1) children were administered the production component for third person –s and past tense of the Test for Early Grammatical Impairment (Rice & Wexler, 1996) and participated in an on-line word-monitoring task involving grammatical and ungrammatical sentences with presence/omission of tense (third person –s, past tense -ed) and non-tense (progressive –ing, possessive ‘s) morphemes. The L2 children’s performance on the on-line task was compared to that of children with Specific Language Impairment (SLI) in Montgomery & Leonard (1998, 2006) to ascertain similarities and differences between the two populations. Results showed that the L2 children were sensitive to the ungrammaticality induced by the omission of tense morphemes, despite variable production. This reinforces the claim about intact underlying syntactic representations in child L2 acquisition despite non target-like production (Haznedar & Schwartz, 1997).

Contemporary acquisition theorizing has placed a considerable amount of attention on interfaces, points at which different linguistic modules interact. The claim is that vulnerable interfaces cause particular difficulties in L1, bilingual and adult L2 acquisition (e.g. Platzack, 2001; Montrul, 2004; Müller and Hulk, 2001; Sorace, 2000, 2003, 2004, 2005). Accordingly, it is possible that deficits at the syntax–pragmatics interface cause what appears to be particular non-target-like syntactic behavior in L2 performance. This syntax-before-discourse hypothesis is examined in the present study by analyzing null vs. overt subject pronoun distribution in L2 Spanish of English L1 learners. As ultimately determined by L2 knowledge of the Overt Pronoun Constraint (OPC) (Montalbetti, 1984), the data indicate that L2 learners at the intermediate and advanced levels reset the Null Subject Parameter (NSP), but only advanced learners have acquired a more or less target null/overt subject distribution. Against the predictions of Sorace (2004) and in line with Montrul and Rodríguez-Louro (2006), the data indicate an overuse of both overt and null subject pronouns. As a result, this behavior cannot be from L1 interference alone, suggesting that interface-conditioned properties are simply more complex and therefore, harder to acquire. Furthermore, the data from the advanced learners demonstrate that the syntax–pragmatics interface is not a predetermined locus for fossilization (in contra e.g. Valenzuela, 2006).
